Abstract:A novel electrochemical immunoassay based on nanobody heptamer for ultrasensitive detection of alpha fetoprotein (AFP) is proposed. First, AFP heptamer fusion protein (A1-C4bp α) is prepared via fusing the AFP-specific nanobody (A1) with the C-terminal fragment of C4-binding protein (C4bp α). Then A1-C4bp α heptamer with higher affinity is used as the molecule recognizing probe to capture numerous the AFP, increasing the sensitivity of the immunosensor. Moreover, AuNPs decorated zeolitic imidazolate framework () with large surface area, and abundant binding sites serves an ideal nanocarrier to increase heptamer loading. Multiwalled carbon nanotube (MWCNTs) with excellent electrical conductivity was immobilized on glassy carbon electrode (GCE) to enhance electrochemical signals. The coupling between the @MWCNTs and heptamer technologies constructed an ultrasensitive and specific immunosensor for AFP detection in the range of 0.1 to 105 pg mL−1 with a lower detection limit of 0.033 pg mL−1. The as-prepared electrochemical sensing exhibited excellent performance and could be employed to determine AFP with satisfactory results in actual serum samples, suggesting that the proposed strategy for nanobody multimerization provides an alternative scheme for enhancing sensitivity in important biomarkers detection.

Abstract:Multifunctional nanolabels with both facile signal amplification and high selectivity are essential for the sensitive determination of the vital tumor marker alpha fetoprotein (AFP). Here, we prepared an immunosensing nanolabel (Ab2-PDA-AF) for convenient AFP detection by grafting the detection antibody (Ab2) and the electroactive redox probe aminoferrocene (AF) onto polydopamine (PDA) nanospheres through Michael addition and Schiff base reactions. The electrochemical signal is dually amplified by the Ab2-PDA-AF nanolabels when the nanolabels are selectively bound to the electrode. The electrochemical signal can be first amplified by oxidizing AF of the Ab2-PDA-AF on the electrode. The signal can be further amplified by the redox reaction of PDA nanospheres with the linked AF/AF+ on the nanospheres. More importantly, the integration of AF-PDA-Ab2 favors a quick electron transfer between AF and PDA nanospheres. Redox probes of AF/AF+ are bonded to the nanolabels instead of dispersing in an electrolyte solution, thus decreasing the electron-transfer resistance and increasing the current signal. The interfacial kinetic analysis demonstrates that the charge-transfer resistance is decreased by 80% by using Ab2-PDA-AF nanolabels. The sensor displays a low detection limit of ca. 0.01 pg/mL, as well as excellent selectivity, reproducibility, and stability. The linear calibration plot of the AFP immunosensor has been obtained in a range of 0.5–1000.0 pg/mL. The capability of the immunosensor has been verified by implementing the detection of AFP in human serum samples.

Abstract:The conventional electrochemical detection strategy for alpha-fetoprotein (AFP) is limited by the antigen-antibody (Ag-Ab) reactions and suffers from low sensitivity and poor reproducibility due to the inconsistency of Ab-modified electrodes. Herein, we designed and explored a sandwich-type electrochemical sensor for highly sensitive detection of AFP based on aptamer (Apt)-AFP-Ab interaction mode with silver@gold (Ag@Au) core-shell nanoparticles (NPs) as a signal amplifier. AuNPs were electrodeposited onto MXene (Ti3C2TX)-modified glassy carbon electrode (GCE) to get AuNPs/MXene/GCE and further used as the signal amplification substrate. The tetrahedral DNA-linked AFP aptamers were immobilized onto AuNPs/MXene/GCE surface via Au-S bonds and used as the sensing and recognition platform for AFP capturing. Ag@AuNPs with core-shell structures were synthesized, characterized, and bound with Ab as detection elements by catalyzing H2O2 reduction. In the presence of AFP, a stable Apt-AFP-Ab sandwich structure was formed owing to the high affinities of aptamer and Ab toward the target AFP. The catalytic current produced by H2O2 reduction increased linearly with the logarithm of AFP concentration from 5 × 10-4 ng/mL to 1 × 105 ng/mL, accompanied by a low detection limit (1.6 × 10-4 ng/mL). Moreover, the novel sandwich-type electrochemical sensor shows high sensitivity, outstanding selectivity, and promising performance in the analysis of actual samples, displaying a broad application prospect in bioanalysis.

Abstract:BACKGROUND: Liver cancer is one of the most common cancers in the world, and it seriously threatens human life and health. Alpha-fetoprotein (AFP), as a carcinogenic glycoprotein, is an important serum marker for detecting liver cancer. Therefore, the accurate and sensitive determination of AFP is crucial for the early diagnosis and treatment of liver cancer. To this end, a label-free fluorescence aptasensor for detecting AFP based on the use of a novel organic Compound D with an aggregation-induced emission activity and aptamer-modified magnetic microparticles was constructed.RESULTS: Compound D could combine with the complementary short chain of the aptamer (CSC-Apt) of AFP to form the D/CSC-Apt complex and realize the fluorescence enhancement of Compound D. Then, magnetic particles modified by the Apt of AFP (Apt-Fe3O4) were prepared. When AFP (or nontarget substance) and D/CSC-Apt were successively added to the Apt-Fe3O4 solution, Apt-Fe3O4 selectively bound to AFP or the D/CSC-Apt complex. Magnetic separation technology showed the changes in the fluorescence intensity of the supernatant. The research results revealed a good linear relationship between the changes in the fluorescence intensity of the supernatant and concentration of AFP within the concentration range of 10-10000 pg mL-1. The proposed aptasensor could achieve high-sensitivity and high-specificity detection of AFP, and its limit of detection was 3 pg mL-1 (S/N = 3).SIGNIFICANCE AND NOVELTY: The sensor combines the advantages of high selectivity of Apt, high sensitivity of fluorescence analysis, AIE effect and good water solubility of Compound D, and rapid separation using magnetic separation technology. And it can be directly used for the detection of AFP in actual serum samples with high accuracy, whereas most of the methods reported in the literature can only detect AFP in spiked serum samples.

Abstract:Alpha-fetoprotein (AFP) is one of the most important biomarkers associated with primary liver cancer and the development of accurate and sensitive methods for detecting the serum level of AFP is of great importance in cancer diagnosis. In this study, three self-assembling peptides (SAPs) tagged AFP specific affibody dimer (ZAFP D2)2 recombinant protein, (ZAFP D2)2-ELK16, (ZAFP D2)2-L6KD and (ZAFP D2)2-R18A were successfully produced and characterized. The (ZAFP D2)2-ELK16 affibody aggregates showed higher and more specific binding affinity to AFP compared to (ZAFP D2)2 and other two SAP-tagged (ZAFP D2)2 and in addition displayed enhanced AFP-detection sensitivity and improved stability. A sensitive two-site enzyme-linked immunosorbent assay for AFP using (ZAFP D2)2-ELK16 as the detection reagent was also established. The AFP detection limit and linear range using (ZAFP D2)2-ELK16 based two-site ELISA were 9.7 ng mL−1 and 20–600 ng mL−1, which were significantly improved compared to those of the (ZAFP D2)2 based two-site ELISA for AFP. To detect AFP in serum samples, the average recovery ranged from 95.02% to 110.41% and the relative standard deviation was in the range of 2.45%-3.97%, which meet the standard clinical testing requirements. This proposed strategy provides a high yield, easy purification and green biosynthesis method to prepare SAP-based affibody aggregates for enhanced detection sensitivity by improving the immunoassay signal amplification activity.

Abstract:The enzyme-linked immunosorbent assay (ELISA) offers several advantages, including simple operation, high throughput, and low cost, making it an ideal immunoassay method for efficient screening of disease-related biomarkers in clinical samples. However, the traditional colorimetric ELISA has relatively low sensitivity, which promotes the continuous emergence of various novel signal amplification technologies. In this work, we fused the AFP-specific nanobody (A1) with the streptavidin-binding peptide (SBP) to develop a fusion protein (A1-SBP) as biorecognition element in a colorimetric ELISA for detecting AFP. Besides, to further improve the sensitivity of the traditional colorimetric ELISA, the streptavidin-conjugated polymerized horseradish peroxidase (SA-PolyHRP) were selected as a detection probe for signal amplification. The proposed signal enhancement strategy demonstrated a limit of detection (LOD) of 0.597 ng/mL for the SA-polyHRP-based ELISA, which is 7.67-fold lower than that of the traditional SA-HRP-based ELISA without additional steps. Furthermore, the proposed SA-polyHRP-based ELISA showed a good correlation with the detection of clinical samples using the Roche E601 chemiluminescence immunoassay analyzer. Therefore, the proposed signal enhancement strategy is an attractive approach for improving the sensitivity of immunoassay without requiring additional steps.

Abstract:An ultrasensitive multiplexed electrochemiluminescence immunoassay method was developed for the detection of tumor markers by combining a functionalized graphene nanosheets and gold-coated magnetic Fe3O4 nanoparticles (GMPs) labeled alpha-fetoprotein (AFP) antibody (GMP similar to Ab1). The functionalized graphene nanosheets (PDDA-G) with poly (diallyldimethylammonium chloride) (PDDA) were synthesized and used to combine with luminol-capped gold nanoparticles (Lu-Au NPs). The resulting PDDA-G@Lu-Au composite displayed an enhanced capability for the immobilization of horseradish peroxidase (HRP) and signal antibody (Ab2) to realize its electrochemiluminescence immunoassay. Great signal amplification was achieved since PDDA-G could adsorb large amount of reporter molecules (Lu-Au). Besides gold nanoparticles were not only used as carriers of Ab2 and HRP but also catalyzed the ECL reaction of luminol, which further amplified the ECL signal of luminol in the presence of H2O2. GMPs as supporting material, not only been performed the rapid separation and purification of signal antibody on magnetic field, but also enhanced the fixed capacity of Ab1 to improve detection range. In addition, the magnetic probes were readily immobilized on the working electrode of screen-printed carbon electrode (SPCEs) by magnets. Under the optimized conditions, the ECL method shows a linear range of alpha-fetoprotein (AFP) from 0.002 to 20 ng mL(-1) and an extremely low detection limit of 0.2 pg mL(-1) (3 sigma). Moreover, the proposed method is also simple, stable, specific, and time-saving, avoiding the complicated process modification on the electrodes' surface, which may open a new door to ultrasensitive detection of tumor markers in clinical analysis.

Abstract:Early finding and diagnosis are critical for prevention and treatment of hepatocellular carcinoma (HCC). Alpha-fetoprotein (AFP) is a typical biomarker of HCC. Since AFP level can reflect the severity of HCC, it is essential to ensure the accurate detection of AFP. In this study, through a combination of the advantages exhibited by ordered mesoporous carbon (OMC)@gold nanoparticles (AuNPs) composites and AuPt-methylene blue (AuPt-MB), a disposable ultrasensitive sandwich-configuration electrochemical immunosensor for determination of AFP was designed. Characterized by excellent conductivity, highly ordered pore distribution and great surface area, OMC can be effective in promoting electron transfer and loading a large number of AuNPs. In the meantime, AuNPs can also immobilize AFP-Ab1 through Au-N bonds. As a new redox-active species, rod-like AuPt-MB demonstrates high conductivity, uniform morphology and excellent biocompatibility, which makes it capable not only to fix AFP-Ab2, but also to release electrochemical signals. A wide linearity of 10 fg mL-1-100 ng mL-1 and a low detection limit of 3.33 fg mL-1 (S/N=3) were obtained. Moreover, the proposed immunosensor exhibited acceptable selectivity, high stability and reproducibility. The excellent performance in detecting serum samples endows the proposed immunosensor with broad prospects of extensive application in the detection of disease biomarkers.

Abstract:We propose an ultrasensitive leaky surface acoustic wave (LSAW) immunosensor based on molybdenum disulfide @ cuprous oxide—gold (MoS2@Cu2O-Au) nanoparticles and subsequent gold staining for the detection of alpha-fetoprotein (AFP). MoS2@Cu2O-Au nanoparticles, with their large specific surface area and good biocompatibility, not only capture the secondary antibodies (Ab2) but also amplify the mass loading effect of the acoustic wave sensor in the detection of AFP. The immunosensor signals are further amplified upon injection of gold staining solution. The developed immunosensor achieved a low detection limit of 5.5 and 25.0 pg/mL with and without gold staining, respectively. The immunosensor demonstrated its efficiency for the quantitative detection of AFP in complex biological fluids, including human serum and saliva samples, with excellent selectivity and long-term stability, showing great potential for the quantification of AFP in clinical diagnosis.

Ultrasensitive lab-on-paper electrochemical device via heterostructure copper/cuprous sulfide@N-doped C@Au hollow nanoboxes as signal amplifier for alpha-fetoprotein detection

Abstract:Rapid and accurate detection of tumor markers at extremely low levels is crucial for the early diagnosis of cancers. In this work, we developed a portable label-free sliding electrochemical paper-based analytical device (ePAD) using copper/cuprous sulfide@N-doped C@Au nanoparticles (Cu/Cu2S@NC@Au) hollow nanoboxes as the signal amplifier for the ultrasensitive detection of alpha-fetoprotein (AFP). Cu/Cu2S@NC nanoboxes were synthesized by sacrificial template and interface reaction methods, on which Au nanoparticles were electrodeposited to construct unique heterostructure for effectively capturing anti-AFP and serving as signal amplifier. The designed ePAD incorporates sliding microfluidic paper chips to form a flexible three-electrode system, enabling highly sensitive detection of AFP with a wide linear range of 0.005-50 ng mL-1 and a low detection limit of 0.62 pg mL-1. The practicality of the prepared ePAD was validated through AFP detection in clinical human serum, which was consistent with chemiluminescence immunoassay. In addition, the developed immunosensor demonstrates excellent specificity, repeatability and stability. This novel platform exhibits significant potential for rapid on-site analysis and point-of-care diagnosis.

Abstract:The authors describe a sensitive electrochemical immunoassay for the cancer biomarker α-fetoprotein (AFP). It is based on a combination of an immunoassay with DNA-based signal amplification. Two-dimensional MnO2 nanosheets modified with gold nanoclusters (AuNC-MnO2) were synthesized through one-pot process and utilized as an electrochemical probe. Bovine serum albumin served as templating agent to guide the formation and assembly of the modified sheets. The detection antibody against AFP and a polycytosine DNA sequence (dC20) were immobilized onto the modified nanosheets. The electrochemical assay follows the usual sandwich protocol. The antibodies on the nanosheets then bind to AFP, while dC20 causes signal amplification. The reaction of the phosphate backbone of dC20 with molybdate leads to the formation of redox-active molybdophosphate which generates an electrochemical current, typically measured at 0.20 V (vs. Ag/AgCl). The method allows AFP to be determined in the 0.01 to 10 ng·mL−1 concentration range, and the detection limit is as low as 5 pg·mL−1. This strategy overcomes the drawbacks of conventional immunoassays whose sensitivity is often limited because many immunoassays are rather difficult to amplify. The method has a wide scope in that various other DNA signal amplification methods such as rolling circle amplification and hybridization chain reactions may also be applied.
An Amperometric Immunosensor for Α-Fetoprotein Based on Antibody Coated Nano ZrO2-Au-Poly Lysine Composite Membrane Modified Electrode

Abstract:One novel amperometric immunosensor for α-fetop- rotein was fabricated which was modified by antibody immobilized on nano ZrO2-Au-polyLysine composite membrane. Firstly, the deoxyribonucleic acid – phenoxyacetic acid isoniazid- Schiff base Co(II) complex (DNA-CoL) mixture film was casted on surface of multi carbon nanotubes(MWCNTs) modified glassy carbon electrode(GCE/CNTs/DNA-CoL). Then the composite nanoparticles (ZrO2-Au-pLL) was employed to immobilize the antibody of α-fetoprotein(AFP) to produce the probes(ZrO2-Au-pLL-anti AFP) for AFP. Finally the probes were modified on GCE/CNTs/DNA-CoL through the specific connection between DNA and ZrO2 to form a novel nanocomposite membrane immunosensor for AFP(GCE/CNTs/ DNA-CoL/ZrO2-Au–pLL-anti AFP). The function of immunosensor was investigated by scanning electron microscopy (SEM), cyclic voltammetry(CV), differential pulse voltammety (DPV) and electrochemical impedance spectroscopy experiment (EIS). The results indicated that combining the advantages of the MWNTs-DNA material and ZrO2-Au-pLL-anti AFP nano probes, the immunosensor with excellent sensitivity and selectivity to AFP was prepared successfully. The modified electrode was sensitive to AFP with a linear relationship between 0.05 and 10 ng · mL−1 and a correl- ation coefficient of 0.9905. The detection limit at a signal to noise ratio of 3 was 0.01 ng·mL−1 under the optimal conditions. The described immunosensor preparation and immunoassay methods offer promise for simple and cost-effective analysis of AFP in serum samples.
Ion-selective electrode-based potentiometric immunoassays for the quantitative monitoring of alpha-fetoprotein by coupling rolling cycle amplification with silver nanoclusters

Abstract:Potentiometric immunoassays have been utilized for the quantitative detection of alpha-fetoprotein (AFP) in hepatocellular carcinoma, but most of them involve low sensitivity and enzyme labels, and thus are unfavorable for routine use. In this work, we report the proof-of-concept of a sensitive and powerful ion-selective potentiometric sensing method for AFP detection with an in situ amplified signal readout. This potentiometric immunoassay mainly contains a silver nanocluster-functionalized single-stranded DNA (AgNC-DNA), a short DNA primer and two antibodies. A sandwich-type immunoreaction is employed for AFP determination on an anti-AFP capture antibody-coated microplate using a biotinylated human AFP secondary antibody. Coupling with a typical biotin-avidin system, the biotinylated DNA initiator strands are conjugated on the microplate in the presence of AFP to induce the rolling cycle amplification (RCA) reaction, followed by AgNC-DNA hybridization. Upon addition of HNO3, the hybridized AgNCs are dissolved into numerous Ag(I) ions, which can be readily determined on a portable handheld silver-ion selective electrode (Ag-ISE). Under optimal conditions, the electrode potential increases with an increase in AFP concentration and exhibits a good linear range of 0.01-100 ng mL-1 at a detection limit of 7.9 pg mL-1. Moreover, the Ag-ISE-based potentiometric immune assay also shows good reproducibility, high specificity and long-term storage stability. Importantly, 18 human serum specimens containing the AFP analyte are screened using the potentiometric immunoassay, giving well-matched experimental results relative to the referenced enzyme-linked immunosorbent assay (ELISA) method.

Abstract:The Anesthesia Simulator-Recorder is a computer program that trains and evaluates anesthesiologists' management of critical incidents. The program executes on IBM compatible personal computers, combining a graphic display of the operating room with mouse-driven input and using an integrated set of physiological and pharmacological models to predict patient responses. The program records the simulated patient's vital signs and all management decisions, and produces a printed case summary. The Anesthesia Simulator-Recorder was evaluated by 44 resident and attending anesthesiologists at seven different anesthesia training centers. These anesthesiologists found the simulator easy to use with clear presentation of the case and management options. The physiological and pharmacological models produced clinically realistic predictions of patient behavior (mean score = 8.5, where 10 is highly realistic and 1 is unrealistic). The Anesthesia Simulator-Recorder was appraised as an excellent training device (mean score = 8.5, where 10 is outstanding and 1 is poor) because it provides the ability to repeatedly practice the management of critical incidents. The simulator was judged to be a good evaluation device (mean score = 6.6, where 10 is outstanding and 1 is poor). No significant differences were found in evaluations between the institution where the program was developed and other institutions, or between residents and attendings.

Abstract:A renewable and magnetic electrochemiluminescence (ECL) immunosensor for alpha-fetoprotein (AFP) was fabricated by using RuL-MWNTs (Tris(2,2'-bipyridine) ruthenium(II) complex immobilized on multiwall carbon nanotubes through pi-pi stacking function between the carbon aromatic systems) doped Au (RuL-MWNTs@Au) composite as labels. Polyelectrolyte poly(dimethyldiallylammonium chloride) (PDDA) was employed to covered on the surfaces of RuL-MWNTs which made it have positive electric charge and can absorb negative colloid Au nanoparticles to fabricate RuL-MWNTs@Au through electric attraction. The primary antibody, anti-AFP was first immobilized on the Fe3O4(core)/gold(shell) nanoparticles (Fe3O4@Au, GMP) as probes. Then the antigen and the RuL-MWNTs@Au composite nanoparticles labeled Ru(bpy)(3)(2+)-conjuged secondary antibody (RuL-MWNTs@Au similar to RuLAb2) were combined successively to form RuL-MWNTs@Au similar to RuLAb2/AFP/anti-AFP-GMP sandwich-type immunocomplexes through the specific interaction. The resulting magnetic sandwich immune complexes can be absorbed on the planar screen printed carbon electrode (SPCE) due to the external magnetic field added on the bottom of it. The magnetic sandwich complexes can be easily washed away from the surface of SPCE when the magnetic field was removed which made the immunosensor reusable. The prepared RuL-MWNTs@Au composite nanoparticles own large surface area, good biocompatibility and highly effective ECL properties. The morphologies of the RuL-MWNTs@Au and its antibody labeled composite nanoparticles were characterized by transmission electronic microscope (TEM). The sensor performed high sensitivity and wide liner for detection AFP in the range of 0.01-50 ng.mL(-1) and the limit detection was 3 pg.mL(-1) (defined as S/N=3). Furthermore the sandwich immunocomplexes can be performed the rapid separation from the unbound secondary antibody quickly by magnetic field. So the immunosensor can simultaneously realize separation, enrichment and determination, with high sensitivity, which would be valuable for clinical immunoassay for AFP in human serum.
Dual-signal Amplification Strategy for Ultrasensitive Photoelectrochemical Immunosensing of Α-Fetoprotein.

Abstract:An ultrasensitive photoelectrochemical immunoassay of cancer biomarker α-fetoprotein (AFP) is proposed that uses titanium dioxide (TiO(2)) coupled with AFP-CdTe-GOx bioconjugate, which featured AFP antigen and glucose oxidase (GOx) labels linked to CdTe quantum dots (QDs) for signal amplification. The synthesized CdTe QDs yielded a homogeneous and narrow size distribution, which allowed the binding of AFP and GOx on CdTe QDs. Greatly enhanced sensitivity for AFP came from a dual signal amplification strategy. First, an effective matching of energy levels between the conduction bands of CdTe QDs and TiO(2) allowed for fast electron injection from excited CdTe QDs to TiO(2) upon irradiation, which reduced the recombination process of electron-hole pairs and prompted photoelectrochemical performance. Second, GOx enzyme could catalyze glucose to produce H(2)O(2), which acted as a sacrificial electron donor to scavenge the photogenerated holes in the valence band of CdTe QDs, further causing an enhanced photocurrent. Thus, on the basis of the dual signal amplification strategy, the competitive immunosensor based on the specific binding of anti-AFP antibodies to AFP and AFP-CdTe-GOx bioconjugates was achieved. This proposed biosensor for AFP possessed largely increased linear detection range from 0.5 pg/mL to 10 μg/mL with a detection limit of 0.13 pg/mL. The proposed amplification strategy shows high sensitivity, stability, and reproducibility and can become a promising platform for other protein detection.

Abstract:highly sensitive electrochemiluminescence immunosensor was originally proposed based on lead sulfide nanocrystals (PbS NCs) to detect alpha fetoprotein (AFP). Herein, PbS NCs, as a kind of nanoenzyme, was synthesized and applied to the luminol-hydrogen peroxide (L-H2O2) system for the first time. Additionally, PbS NCs with superior electrocatalytic activity toward H2O2 can enhance the electrochemiluminescence response of luminol. In this protocol, chitosan was utilized to immobilize PbS NCs through its film-forming characteristics, and further developed an ECL immunosensor by amination reaction combined with AFP antibodies. When the modified electrode was incubated with different concentrations of AFP antigen, the electrochemiluminescence intensity decreased obviously. In addition, under optimal conditions, the sensor can sensitively and selectively detect the alpha fetoprotein ranging from 1.0x10(-5) to 800 ng/mL with the detection limit of 3.0 fg/mL. On this basis, the proposed ECL immunosensor will open up a new way to sensitively detect AFP and exhibit its application prospect in biological analysis.

Abstract:Alpha-fetoprotein (AFP) is one of the most important biomarkers associated with primary liver cancer, and the main approaches for diagnosis are based on immunoassay. Affibody is a 58 amino acids peptide derived from the Z domain of staphylococcal protein A and generally applied in imaging diagnosis, clinical therapeutics and biotechnology research. The aim of this study was therefore to develop a novel affibody-based ELISA for detection of AFP. After three rounds of biopanning, six AFP-binding affibody peptides were selected using phage display technology, among them affibody ZAFPD2 showed high and specific binding affinity to AFP. An affibody dimer of ZAFPD2 was created, named (ZAFP D2)2, expressed in E.coli and the purified (ZAFP D2)2 recombinant protein showed higher binding affinity to AFP, as well as high thermal stability. A novel affibody-based two-site ELISA method using ZAFPD2 or (ZAFP D2)2 and polyclonal antibody to detect AFP was developed, the detection limit of the immunoassay using (ZAFP D2)2 was 2 ng mL-1 that was 4 times lower than ZAFPD2, which meets the requirements for practical application. Therefore, this concept of affibody-based ELISA may provide a new method for the detection of various cancer biomarkers.

Abstract:Sporozoite-induced experimental infections of Haemoproteus meleagridis produced a moderate to severe myositis and significant effects on weight gain and growth in domestic turkey poults. Pathological effects occurred in both low- and high-dose infections (4,400 and 57,500 sporozoites, respectively). Low-dose birds weighed significantly less than controls at 3 wk postinfection (PI) when peripheral parasitemia reached a peak and had significantly shorter tarsometatarsal lengths at both 1 and 3 wk PI. High-dose birds were significantly lighter and smaller than control and low-dose birds throughout the course of the 8-wk study. Infected birds were not anemic in spite of high parasitemias that often exceeded 50% of circulating erythrocytes. The most serious pathological effects occurred prior to patency and were associated with development of megaloschizonts in skeletal muscle. Microscopic lesions in 4 high-dose birds that died between 19 and 22 days PI were characteristic of a severe, acute hemorrhagic myositis. Megaloschizonts were surrounded by a hemorrhagic inflammatory infiltrate composed of macrophages, heterophils, giant cells, and red blood cells. Muscle fibers adjacent to megaloschizonts were swollen, hyaline, and contained prominent calcium deposits. Other observations included enlargement of the spleen, deposition of pigment in macrophages of the lung and spleen, and secondary bacterial and fungal infections in the intestine and lungs. Necrotic and calcified muscle fibers and degenerating megaloschizonts were still present at 8 wk PI when the experiment ended. Our results demonstrated significant pathological changes in H. meleagridis-infected domestic turkeys that were associated primarily with preerythrocytic stages of development.

Abstract:Accurate detection of tumor biomarkers in blood is crucial for diagnosing and treating tumor disease. In this study, a metal enzyme-linked immunosorbent assay (MeLISA) was fabricated for the ultrasensitive and naked-eye detection of tumor biomarker alpha-fetoprotein (AFP) in clinical serum samples. Herein, novel copper metal-organic frameworks and gold platinum nanoparticles composites (Cu-MOFs@AuPtNPs) were synthesized for the first time by in-situ method, which showed an enormous specific surface area and excellent peroxidase (POx) mimicking property. Cu-MOFs@AuPtNPs linked with antibodies targeting AFP performed as signal nanoprobe to amplify the detection signal. Additionally, the specificity of MeLISA was significantly enhanced through conventional antigen-antibody reaction and efficient blocking of non-specific sites with BSA. In optimal conditions, the sandwich-type MeLISA exhibited a wide range from 0.001 to 1000 ng mL-1 (R2 = 0.997) and a low detection limit of 0.86 pg mL-1 (S/N = 3) with acceptable stability, selectivity, and reproducibility. It was noteworthy that the suggested MeLISA performed exceptionally well in detecting clinical serum samples, which were visible to the naked eye and did not require complex platforms. To sum up, the innovative MeLISA based on Cu-MOFs@AuPtNPs provides an alternative method for early cancer diagnosis, particularly in economically backward areas where simple diagnostic apparatus is extremely desirable.
