Abstract:We have described a turn-on near-infrared fluorescent probe Cy-NO(2) based on nitro group reduction for intracellular H(2)S detection. The probe employs cyanine dye as a fluorophore, and is equipped with a nitro group as a fluorescent modulator. It is readily employed for assessing intracellular H(2)S level changes, and confocal imaging is achieved successfully.

Abstract:A novel selective fluorescent chemosensor 1 has been synthesized with a phenanthrene-fused dipyrromethene structure. Selective binding of Cu2+ by 1 results in a complex that displays high selectivity and sensitivity for H2S. The signal transduction occurs via reversible formation-separation of the Cu-1 complex and CuS. Its potential utility for biological applications was confirmed by fluorescence imaging of H2S in live cells.

Abstract:Since Hydrogen Sulfide (H2S) was recognized as a gas transmitter, its detection and quantification have become a hot research topic among chemists and biologists. In this area, fluorescent probes have shown great advantages: fast and strong response, low detection limit and easy manipulation. Here we developed a new fluorescent probe that detected H2S selectively among various bioactive and inorganic salts. This probe was based on the core structure of fluorescein and reacted with H2S through azide-reduction. Great linearity was achieved correlating fluorescence intensity and H2S concentrations in solution. The detection of H2S in cancer cells was also achieved.

Abstract:A coumarin-based fluorescence chemoprobe was developed and evaluated for the selective and sensitive detection of hydrogen sulfide in degassed PBS buffers and fetal bovine serum. Fluorescence detection of hydrogen sulfide in living cells was also successfully achieved using two-photon confocal fluorescence imaging. Further in situ visualization of endogenous H(2)S was realized in cardiac tissues of normal rats and atherosclerosis (AS) rats.

Abstract:A new Near-infrared fluorescent probe for hydrogen sulfide detection was synthesized by employing dicyanoisophorone based fluorescence dye as a fluorophore and methyl 3-(2-(carbonyl)phenyl)-2-cyanoacrylate group as the response unit. The Probe DCI-H 2 S showed a long emission wavelength (λem = 674 nm). Based on the H 2 S-induced addition–cyclization of deprotecting methyl 3-(2-(carbonyl)phenyl)-2-cyanoacrylate group, the probe DCI-H 2 S showed high selectivity, sensitivity and response speed toward hydrogen sulfide under room temperature. These numerous advantages of the probe DCI-H 2 S make it to potentially detect endogenous hydrogen sulfide in living organisms.

Abstract:We present a new two-photon fluorescent probe (T-HS) for the detection of H2S. With the addition of hydrogen sulfide, the absorption and fluorescence spectra of the probe show regular changes. The probe exhibited favorable properties, such as large turn-on fluorescence signal, good selectivity and low cytotoxicity. Moreover, the probe T-HS was successfully used for the fluorescence imaging of H2S in live cells and zebrafish.

Abstract:In this work, a series of novel fluorescent probes based on the fluorophores 3-benzothiazolyl-7-hydroxycoumarin/7-hydroxycoumarin were developed for detection of H2S. The probes exhibited excellent specific responses to H2S over other related competing species, including reactive sulfur, nitrogen and oxygen species. Moreover, the further successful imaging of the targetable fluorescent probe for sensing H2S in vivo was exciting and revealed that this targetable probe could serve as an efficient tool to study H2S-related chemical biology in physiological and pathological events.

Abstract:The rational design of effective tools capable of monitoring hydrogen sulfide (H2S) is of great importance to fully understand its physiological and pathological functions. Herein, a self-assembled fluorescent nanoprobe with both aggregation-induced emission (AIE) and excited-state intramolecular proton transfer (ESIPT) characteristics for H2S detection has been successfully developed via a modified nanoprecipitation method. The nanoprobe features high water dispersibility, a large Stokes shift (ca. 100 nm), good biocompatibility as well as high selectivity towards H2S over other putative interferents. Live cell imaging experiments demonstrate that the nanoprobe, with good cell-membrane permeability, can facilitate the visualization of exogenous and endogenous H2S levels. Moreover, the nanoprobe has also been found capable of detecting H2S in zebrafish. This nanoprobe with AIE and ESIPT characteristics can provide a novel method for the development of fluorescent probes for monitoring biological processes.

Abstract:Hydrogen sulphide (H2S), the third endogenous gaseous signalling molecule, has attracted attention in biochemical research. The selective detection of H2S in living systems is essential for studying its functions. Fluorescence detection methods have become useful tools to explore the physiological roles of H2S because of their real-time and non-destructive characteristics. Herein we report a near-infrared fluorescent probe, NIR-HS, capable of tracking H2S in living organisms. With high sensitivity, good selectivity and low cytotoxicity, NIR-HS was able to recognize both the exogenous and endogenous H2S in living cells. More importantly, it realized the visualization of endogenous H2S generated in cells overexpressing cystathionine β-synthase (CBS), one of the enzymes responsible for producing endogenous H2S. The probe was also successfully applied to detect both the exogenous and endogenous H2S in living mice. The superior sensing properties of the probe render it a valuable research tool in the H2S-related medical research.

Abstract:The discovery of hydrogen sulfide (H2S) as a novel gasotransmitter for cell signaling and other pathophysiological processes has spurred tremendous interest in developing analytical methods for its detection in biological systems. Herein, we report the development of a highly responsive and selective genetically encoded H2S probe, hsGFP, for the detection of H2S both in vitro and in living mammalian cells. hsGFP bestows a combination of favorable properties, including large fluorescence responses, high efficiency in folding and chromophore formation, and excellent sensitivity and selectivity toward H2S. As a genetically encoded probe, hsGFP can be readily and precisely localized to subcellular domains such as mitochondria, cell nuclei, and ion channels. hsGFP was further utilized to image H2S enzymatically produced from l-cysteine in human embryonic kidney (HEK) 293T cells.

Abstract:Hydrogen sulfide (H2S) is an important gaseous mediator in cellular physiology and pathology. For understanding the biological action of H2S to both healthy and disease states, it is important to develop H2S-selective sensor. Herein we report a colorimetric and fluorescent probe, which employs an NBD moiety as fluorophore, and is equipped with azide group as H2S-active unit. The probe exhibits selective and sensitive response to H2S in aqueous solution. By employing 1 mu M sensor, the detection limit was evaluated to be 680 nM with a signal-to-noise ratio of 3. Abstract:Hydrogen sulfide (H 2 S) is traditionally considered as an ecologically toxic gas, has been proven to be the third endogenous signaling gasotransmitter and plays important roles in physiological and pathophysiological conditions. Herein, we designed and synthesized a novel "turn off" fluorescent probe for the detection of hydrogen sulfide(H 2 S) in vitro. Probe A could detect H 2 S in CH 3 CN/PBS system with significant fluorescence quenching. Probe A also showed obviously aggregation- induced emission (AIE) effects. Fluorescence titration experiments revealed probe A combine with H 2 S in a 1: 1 ratio with a detection limit of 6.54 μM. Cell imaging results demonstrated the potential applicability of probe A for H 2 S detection in living cells. The supramolecular assembly of probe A and β-cyclodextrin further revealed the interaction with H 2 S.

Abstract:H2S is well-known as a colorless, acidic gas, with a notoriously rotten-egg smell. It was recently revealed that H2S is also an endogenous signaling molecule that has important biological functions, however, most of its physiology and pathology remains elusive. Therefore, the enthusiasm for H2S research remains. Fluorescence imaging technology is an important tool for H2S biology research. The development of fluorescence imaging technology has realized the study of H2S in subcellular organelles, facilitated by the development of fluorescent probes. The probes reviewed in this paper were categorized according to their chemical mechanism of sensing and were divided into three groups: H2S reducibility-based probes, H2S nucleophilicity-based probes, and metal sulfide precipitation-based probes. The structure of the probes, their sensing mechanism, and imaging results have been discussed in detail. Moreover, we also introduced some probes for hydrogen polysulfides.

Abstract:We report a sensitive and selective fluorescent probe SFP-BC for hydrogen sulfide detection based on BODIPY–coumarin platform. SFP-BC shows a significant emission increase in the fast response to sulfide at physiological pH. Utilizing this probe, we have successfully detected hydrogen sulfide in fresh mouse blood plasma and in live cells.
