Abstract:Hydroxyapatite (HA) is an attractive bioceramic for hard tissue repair and regeneration due to its physicochemical similarities to natural apatite. However, its low fracture toughness, poor tensile strength and weak wear resistance become major obstacles for potential clinical applications. One promising method to tackle with these problems is exploiting graphene and its derivatives (graphene oxide and reduced graphene oxide) as nanoscale reinforcement fillers to fabricate graphene-based hydroxyapatite composites in the form of powders, coatings and scaffolds. The last few years witnessed increasing numbers of studies on the preparation, mechanical and biological evaluations of these novel materials. Herein, various preparation techniques, mechanical behaviors and toughen mechanism, the in vitro/in vivo biocompatible analysis, antibacterial properties of the graphene-based HA composites are presented in this review.

Abstract:In this study, self-assembled peptide nanofibers (PNFs) were firstly created with a specially designed peptide molecule. Graphene oxide (GO) nanosheet, as a two-dimensional scaffold, was then modified with the prepared PNFs to fabricate a new type of GO–PNF nanohybrid, which was further utilized as a template for biomimetic mineralization. The produced GO–PNF nanohybrid and its minerals were characterized by atomic force microscopy, scanning electron microscopy, Fourier transform infrared spectroscopy, X-ray diffraction, Raman spectroscopy, and thermogravimetric analysis. The obtained results indicate that the created GO–PNF nanohybrid facilitates the nucleation and growth of hydroxyapatite (HA) crystals. The created PNFs promote the formation of HA nanocrystals along the axis of PNFs within short-term incubation and the GO nanosheet mediates the formation of HA microsphere after long-term mineralization. The effects of the produced GO–PNF nanohybrid and biomimetic minerals (GO–PNF–HA) on the adhesion and proliferation of L-929 cells and MC3T3-E1 cells were further investigated. The cell culture result indicates that the produced GO–PNF–HA minerals have good biocompatibility and can enhance the proliferation ability of the studied cells. We believe this novel biocompatible nanohybrid will show great potentials in tissue engineering.

Abstract:PurposeIn recent years, tissue engineering scaffolds have gained popularity as a replacement for metallic/synthetic implants and tissue grafts.MethodsA porous tricomponent scaffold of graphene oxide (GO)-xanthan-hydroxyapatite (HAP) was fabricated using the freeze-drying process for bone tissue engineering.ResultsThe physicochemical analysis (FTIR and XRD) revealed that the composite was formed by hydrogen bonding and electrostatic interactions, and also determined a decreased crystallinity, similar to that of real bone apatite. TG/DTA analysis proved the presence of all the raw materials in the scaffold. Morphological studies supported the porous nature of HAP as well as its uniform distribution. The scaffold negatively charged functional group produced positive in vitro results in terms of cell proliferation, biocompatibility, and cell adhesion.ConclusionThe positive in vitro results confirmed that the fabricated scaffold materials could be a useful biomaterial for bone tissue engineering.

Abstract:Motivated by the success of using graphene oxide (GO) as a nanofiller of composites, there is a drive to search for this new kind of carbon material as a reinforcing phase in biocomposites. In the present work, graphene oxide and chitosan (CS) functionalized graphene oxide were introduced as templates to fabricate hydroxyapatite (HA) using a facile solution-based in situ synthesis method, and GO-HA and CS-GO-HA nanocomposites were successfully prepared for the first time. It was found that the spindle like HA nanoparticles with a diameter of about 27 ± 7 nm and a length around 150 ± 25 nm were decorated randomly and strongly on the surface or aggregated at the edges of the pristine and chitosan functionalized GO matrix. Compared with HA, the prepared GO-based HA nanocomposites displayed an increased elastic modulus and hardness. The in vitro cytotoxicity of the prepared nanocomposites was investigated using CCK-8 assay on murine fibroblast L-929 cell line and human osteoblast-like MG-63 cell line, respectively. Both of the nanocomposites exhibited a high cell proliferation rate for L-929 and MG-63, and the CS-GO-HA could provide significantly higher cell viability and alkaline phosphatase activity compared to the GO-HA composite. These findings may provide new prospects for utilizing the GO-based hydroxyapatite biocomposites in bone repair, bone augmentation, as well as coating of biomedical implants and broaden the application of GO sheets in biological areas.

Nanocomposites based on hydroxyapatite/lithium oxide and graphene oxide nanosheets for medical applications

Abstract:Metal-based bioceramics are developed to imitate the natural bones to be used as bone substitutes. Different nanocomposites are synthesized using hydroxyapatite (HAP), Li 2 O and GO to fabricate HAP, Li 2 O, HAP/Li 2 O, HAP/GO and HAP/Li 2 O/GO. XRD shows the change in crystal structure upon adding the three nanocomposites to each other. Besides, FTIR and XPS explain the functional groups of the compositions and analyze their surface composition. The TEM explains the morphological changes for particles dimensions of HAP/Li 2 O/GO showing a length of 20 nm for HAP and a size of 6 nm for Li 2 O sphere. In addition, SEM graphs explained the enhancement of HAP/Li 2 O/GO porosity showing a particle size of 0.3–0.4 μm and length of 0.8 μm. Further, roughness parameters are improved upon adding HAP and Li 2 O to GO, showing a roughness average value of 57.80 nm. Moreover, the cell viability of HAP/Li 2 O/GO is examined and shows a maximum value of 97.4 ± 1.2%. In addition, the antibacterial activity exhibited by HAP/Li 2 O/GO against ( Escherichia coli = E. coli ) and ( Staphylococcus aureus = S. aureus ) showed an inhibition zone of 13.4 ± 1.2 mm 12.3 ± 1.3 mm. Adding HAP presents an improvement in surface roughness as well as cell viability and antibacterial activity. Besides, the microhardness has been improved from 2.6 ± 0.1 GPa to 3.1 ± 0.2 for pure HAP and HAP/Li 2 O/GO, respectively. Therefore, combining HAP, GO and Li 2 O results in a nanocomposite of enhanced biocompatibility and can be suggested for bone biomedical applications.

Abstract:It is urgently needed to develop composite bone scaffold with excellent mechanical properties and bioactivity in bone tissue engineering. Combining graphene oxide (GO) and hydroxyapatite (HAP) for the reinforcement of biopolymer bone scaffold has emerged as a promising strategy. However, the dispersion of GO and HAP remains to be a big challenge.In this present work, the mechanical properties of GO and the bioactivity of and HAP were combined respectively via in situ synthesis for reinforcing biopolymer bone scaffold.GO nanosheets were employed to in situ synthesize GO-HAP nanocomposite via hydrothermal reaction, in which their abundant oxygen-containing groups served as anchor sites for the chelation of Ca2+ and then Ca2+ absorbed HPO42- via electrovalent bonding to form homogeneously dispersed HAP nanorods. Thereby, the GO-HAP nanocomposite was blended with biopolymer poly-L-lactic acid (PLLA) for fabricating biopolymer scaffold by selective laser sintering (SLS).GO nanosheets were uniformly decorated with HAP nanorods, which were about 60 nm in length and 5 nm in diameter. The compressive strength and modulus of PLLA/12%GO-HAP were significantly increased by 53.71% and 98.80% compared to the pure PLLA scaffold, respectively, explained on the base of pull out, crack bridging, deflection and pinning mechanisms. Meanwhile, the mineralization experiments indicated the PLLA/GO-HAP scaffold displayed good bioactivity by inducing the formation of apatite layer. Abstract:: This paper provides a review of the applications of some graphene oxide-based hydrogels in bone tissue engineering. Hydrogels used in bone tissue engineering usually require suitable biocompatibility, porosity, and mechanical strength. Under the original hydrogel condition, by adding GO, the various indexes of the composite hydrogel can reach more suitable conditions for inducing bone defect healing. This article also introduces some high-performance graphene oxide-based hydrogels, which are likely to play an important role in bone tissue engineering in the future.
Graphene Oxide/hydroxyapatite Composite Coatings Fabricated by Electrophoretic Nanotechnology for Biological Applications

Abstract:Graphene oxide (GO) was firstly employed as nanoscale reinforcement fillers in hydroxyapatite (HA) coatings by a cathodic electrophoretic deposition process, and GO/HA coatings were fabricated on pure Ti substrate. The transmission electron microscopy observation and particle size analysis of the suspensions indicated that HA nanoparticles were uniformly decorated on GO sheets, forming a large GO/HA particle group. The addition of GO into HA coatings could reduce the surface cracks and increase the coating adhesion strength from 1.55±0.39MPa (pure HA) to 2.75±0.38MPa (2 wt.% GO/HA) and 3.3±0.25MPa (5 wt.% GO/HA), respectively. Potentiodynamic polarization and electrochemical impedance spectroscopy studies indicated that the GO/HA composite coatings exhibited higher corrosion resistance in comparison with pure HA coatings in simulated body fluid. In addition, superior (around 95% cell viability for 2 wt.% GO/HA) or comparable (80–90% cell viability for 5 wt.% GO/HA) in vitro biocompatibility were observed in comparison with HA coated and uncoated Ti substrate.
Collagen Functionalized With Graphene Oxide Enhanced Biomimetic Mineralization and in Situ Bone Defect Repair

Abstract:Biomimetic mineralization using simulated body fluid (SBF) can form a bonelike apatite (Ap) on the natural polymers and enhance osteoconductivity and biocompatibility, and reduce immunological rejection. Nevertheless, the coating efficiency of the bonelike apatite layer on natural polymers still needs to be improved. Graphene oxide (GO) is rich in functional groups, such as carbonyls (-COOH) and hydroxyls (-OH), which can provide more active sites for biomimetic mineralization and improve the proliferation of the rat bone marrow stromal cells (r-BMSCs). In this study, we introduced 0%, 0.05%, 0.1%, and 0.2% w/v concentrations of GO into collagen (Col) scaffolds and immersed the fabricated scaffolds into SBF for 1, 7, and 14 days. In vitro environment scanning electron microscopy (ESEM), energy-dispersive spectrometry (EDS), thermogravimetric analysis (TGA), micro-CT, calcium quantitative analysis, and cellular analysis were used to evaluate the formation of bonelike apatite on the scaffolds. In vivo implantation of the scaffolds into the rat cranial defect was used to analyze the bone regeneration ability. The resulting GO-Col-Ap scaffolds exhibited a porous and interconnected structure coated with a homogeneous distribution of bonelike apatite on their surfaces. The Ca/P ratio of 0.1% GO-Col-Ap group was equal to that of natural bone tissue on the basis of EDS analysis. More apatites were observed in the 0.1% GO-Col-Ap group through TGA analysis, micro-CT evaluation, and calcium quantitative analysis. Furthermore, the 0.1% GO-Col-Ap group showed significantly higher r-BMSCs adhesion and proliferation in vitro and more than 2-fold higher bone formation than the Col-Ap group in vivo. Our study provides a new approach of introducing graphene oxide into bone tissue engineering scaffolds to enhance biomimetic mineralization.

Abstract:Layer-by-layer (LBL) assembly is a simple and effective method for the fabrication of a three-dimensional (3D) scaffold for nanotechnological and biomedical applications. Herein, a novel 3D scaffold based on an alternate LBL assembly of graphene oxide (GO) nanosheets and fibrinogen nanofibers (Fg NFs) on a silicon substrate was fabricated and utilized to create a 3D hydroxyapatite (HA) scaffold by biomimetic mineralization in 1.5x simulated body fluid for different nucleation periods. The obtained 3D (GO-NF)(n)-HA scaffold was characterized using atomic force microscopy, scanning electron microscopy, transmission electron microscopy, X-ray diffraction, Fourier transform infrared spectroscopy, and X-ray photoelectron spectroscopy. The results demonstrate that the Fg NFs could promote the nucleation and growth of HA crystals along the axis. The 3D (GO-NF)(10)-HA scaffold composed of 10 layers of GO alternating with 10 layers of NFs was successfully created by LBL assembly and subsequent biomimetic mineralization. The effects of the created 3D (GO-NF)(10)-HA scaffolds on the adhesion, morphology, and proliferation of L-929 cells were investigated. The in vitro cell culture indicates that the 3D (GONF)(10)-HA scaffold has a higher proliferation ability and better cytocompatibility than the other control samples.
A graded graphene oxide-hydroxyapatite/silk fibroin biomimetic scaffold for bone tissue engineering

Abstract:To better mimic natural bone, a graphene oxide-hydroxyapatite/silk fibroin (cGO-HA/SF) scaffold was fabricated by biomineralizing carboxylated GO sheets, blending with SF, and freeze-drying. The material has increasing porosity and decreasing density from outside to inside. Analysis of GO mineralization in simulated body fluid indicated that carboxylation and Chitosan may synergistically regulate HA growth along the c-axis of weakly crystalline, rod-like GO-HA particles. Compared with HA/SF gradient composites, a cGO-HA gradient scaffold with cGO:HA mass ratio 1:4 has 5-fold and 2.5-fold higher compressive strength and compressive modulus, respectively. Additionally, the cGO-HA/SF composite stimulated mouse mesenchymal stem cell adhesion and proliferation, alkaline phosphatase secretion, and mineral deposition more strongly than HA/SF and pure HA scaffolds. Hence, the material may prove to be an excellent and versatile scaffold for bone tissue engineering.

Abstract:<h2>Abstract</h2>The objective of this work is to design and development of graphene based bioceramic composite. The composition of graphene bioceramic composite is Hydroxyapatite/β-Tricalcium phosphate reinforced Graphene oxide (HAp/ β-TCP/GO) The crystalline structure, chemical composition, morphology and elemental analysis of the developed composite were characterized using analytical techniques such as X-Ray Diffraction (XRD), Fourier Transform Infra red (FTIR) Spectroscopy, Raman Spectroscopy, Field Emission Scanning Electron Microscopy (FESEM) and Energy Dispersive Spectroscopy (EDS). In order to investigate the effect of graphene oxide on the in-vitro bio mineralization, the developed composite was immersed in Simulated Body Fluid (SBF) solution for various time periods (0, 7, 14 and 21 days) and the results indicate that the reinforcement of graphene oxide promotes the growth of hydroxyl apatite layer on the surface of composite discs. Further the developed composite of β-TCP/HAp reinforced graphene oxide can be used as a potential candidate material for biomedical applications.

Abstract:To obtain three dimensional mesh nano-hydroxyapatite/graphene oxide composites, a surface modified hydroxyapatite (SHA) obtained by grafting with citric acid. The SHA particles were covered by GO in SHA/GO composites, and the structure of composites is three-dimensional structures. The biomimetic mineralization in SBF solution showed the bone-like apatite layer on composites surface. Aspirin was loaded onto the SHA/GO composites. The change of GO content in composites has a great influence on the adsorption performance of Aspirin.

The graphene–zinc oxide–calcium phosphate composite microspheres were characterized by X-ray diffraction (XRD), field emission scanning electron microscopy (FE-SEM), X-ray photoelectron spectroscopy (XPS), energy dispersion spectroscopy (EDS), water contact angle measurement, degradation and pH determination, and differential thermal analysis (DiamondTG/DTA). The biocompatibility, osteogenic activity, and antibacterial activity of GO/ZnO/nHAp composite microspheres were further studied. The results of the cell experiment and antibacterial experiment showed that 0.5% and 1% GO-ZnO-nHAp composite microspheres not only had good biocompatibility and osteogenic ability but also inhibited Escherichia coli and Staphylococcus aureus by more than 45% and 70%. Therefore, GO/ZnO/nHAp composite microspheres have good physical and chemical properties and show good osteogenic induction and antibacterial activity, and this material has the possibility of being used as a bone regeneration material.

Abstract:Hybrid nanomaterials with tailored functions, consisting of self-assembled peptides, are intensively applied in nanotechnology, tissue engineering, and biomedical applications due to their unique structures and properties. Herein, a peptide-mediated biomimetic strategy is adopted to create the multifunctional 3D graphene foam (GF)-based hybrid minerals. First, 2D peptide nanosheets (PNSs), obtained by self-assembling a motif-specific peptide molecule (LLVFGAKMLPHHGA), are expected to exhibit biofunctionality, such as the biomimetic mineralization of hydroxyapatite (HA) minerals. Subsequently, the noncovalent conjugation of PNSs onto GF support is utilized to form 3D GF-PNSs hybrid scaffolds, which are suitable for the growth of HA minerals. The fabricated biomimetic 3D GF-PNSs-HA minerals exhibit adjustable shape, superlow weight (0.017 g cm(-3)), high porosity (5.17 m(2) g(-1)), and excellent biocompatibility, proving potential applications in both bone tissue engineering and biomedical engineering. To the best of the authors' knowledge, it is the first time to combine 2D PNSs and GF to fabricate 3D organic-inorganic hybrid scaffold. Further development of these hybrid GF-PNSs scaffolds can potentially lead to materials used as matrices for drug delivery or bone tissue engineering as proven via successful 3D scaffold formation exhibiting interconnected pore-size structures suitable for vascularization and medium transport.
Additive manufacturing of graphene oxide/hydroxyapatite bioceramic scaffolds with reinforced osteoinductivity based on digital light processing technology

Abstract:In the field of bone tissue engineering, porous hydroxyapatite (HA) bone repair scaffolds with controlled architecture and porosity are challenging to manufacture and lack sufficient osteoinductive activity, which limit its wider clinical application. This study fabricated porous graphene oxide (GO)/HA composite ceramic scaffolds using digital light processing (DLP) technology. The properties of the composite ceramic slurry and the process parameters of curing, degreasing, and sintering were initially examined to ensure molding accuracy. The research showed that a small amount of GO (0.1–0.4 wt%) could better maintain the pore structure and improve the mechanical properties of composite ceramics. Porous scaffolds with pore sizes of 300–400 μm were prepared for co-culture with rat bone marrow mesenchymal stem cells to detect biocompatibility. The results showed that all scaffolds have no cytotoxicity. Compared with HA, the GO/HA ceramic scaffold significantly promoted the cell adhesion, proliferation, and expression of osteogenesis-related genes; GO (0.1–0.2 wt%) /HA scaffolds exhibited superior alkaline phosphatase activity and more effective bone mineralization, showing enhanced osteoinductivity. The porous GO/HA composite scaffold fabricated by DLP presented a strong potential for repairing bone defects.
