Abstract:In this paper, we developed a novel fluorescent probe B601 for endogenous H2S detection. In the probe B601, the fast responding and highly selective reaction site towards H2S, incorporating the membrane permeable BODIPY fluorescent dye, enables the probe to be applied for the detection of endogenous H2S via the CSE enzymatic pathway.

Abstract:As a common gaseous signaling molecule, hydrogen sulfide (H2S) plays a vital role in physiology and pathology. The development of fluorescent probes for detecting H2S has attracted widespread attention. However, most of the reported fluorescent probes with nitrobenzoxadiazole (NBD) as the recognition group have been widely used to simultaneously detect biothiols and H2S, instead of specifically detecting H2S. Herein, a novel NBD-based near-infrared (NIR) fluorescent probe named CX-N for the detection of H2S is synthesized. The selectivity of CX-N for H2S is significantly higher than that for biothiols and other potential interferences. After reacting with H2S, CX-N shows a significant increase in NIR fluorescence (75-fold), large Stokes shift (155 nm) and fast response (4 min). And the possible response mechanism of CX-N to H2S is given and confirmed by HPLC and HRMS. Based on the low cytotoxicity of CX-N, it has been used for H2S imaging in live cells and zebrafish. More importantly, CX-N has also been successfully applied for the real-time imaging of H2S in inflammatory and tumor mice based on its NIR emission, which provides a reliable platform for the specific recognition of H2S in complex biological systems.

Abstract:In this work, a series of novel fluorescent probes based on the fluorophores 3-benzothiazolyl-7-hydroxycoumarin/7-hydroxycoumarin were developed for detection of H2S. The probes exhibited excellent specific responses to H2S over other related competing species, including reactive sulfur, nitrogen and oxygen species. Moreover, the further successful imaging of the targetable fluorescent probe for sensing H2S in vivo was exciting and revealed that this targetable probe could serve as an efficient tool to study H2S-related chemical biology in physiological and pathological events.

Abstract:Novel cyanine-based fluorescent probes for the detection of H2S were developed. The probes developed are stable under physiological conditions. The water soluble fluorescent probe 2 displayed ultrafast and specific response to H2S displaying NIR fluorescence of 115-fold turn-on with the detection limit of 11 nM without assistance of organic solvent or surfactant. Cell imaging experiments indicated that probe 2 was cell-permeable and was able to detect H2S sensitively in lysosomes. Moreover, our probe was able to detect H2S intrinsically produced H2S through enzymatic/non-enzymatic biosynthetic pathway from Cys/GSH. Moreover, we applied probe 2 to detect H2S in living mice and demonstrated the fast metabolism of H2S. Thus, probe 2 shows great promise as a reporter for H2S.

Abstract:A novel selective fluorescent chemosensor 1 has been synthesized with a phenanthrene-fused dipyrromethene structure. Selective binding of Cu2+ by 1 results in a complex that displays high selectivity and sensitivity for H2S. The signal transduction occurs via reversible formation-separation of the Cu-1 complex and CuS. Abstract:A selective probe with a significant regioselective turn-on response, CP-1, has been developed for detection of H2S from biothiols. CP-1 features superb selectivity to H2S, a wide pH range and low cytotoxicity. CP-1 has over 10000-fold selectivity for H2S from biothiols such as GSH. With the actual detected concentration low to 50 nM, CP-1 could provide a set of sectionalized linear standard curves to cover a 0 to 275 mu M detecting range accurately. NMR and HRMS spectra confirmed that the detection process was a regioselective reaction. Furthermore, mono-protected compound FL-1 has been synthesized and tested for Na2S and biothiols. All test results demonstrate that the detection can only occur in one side of CP-1. With high sensitivity and selectivity, CP-1 could be further used in rat urine samples system. Moreover, CP-1 has been applied in the imaging of exogenous and endogenous H2S in living cells and in living Caenorhabditis elegans.

Abstract:A coumarin-based fluorescence chemoprobe was developed and evaluated for the selective and sensitive detection of hydrogen sulfide in degassed PBS buffers and fetal bovine serum. Fluorescence detection of hydrogen sulfide in living cells was also successfully achieved using two-photon confocal fluorescence imaging. Abstract:Hydrogen sulfide (H 2 S) is traditionally considered as an ecologically toxic gas, has been proven to be the third endogenous signaling gasotransmitter and plays important roles in physiological and pathophysiological conditions. Herein, we designed and synthesized a novel "turn off" fluorescent probe for the detection of hydrogen sulfide(H 2 S) in vitro. Probe A could detect H 2 S in CH 3 CN/PBS system with significant fluorescence quenching. Probe A also showed obviously aggregation- induced emission (AIE) effects. Fluorescence titration experiments revealed probe A combine with H 2 S in a 1: 1 ratio with a detection limit of 6.54 μM. Cell imaging results demonstrated the potential applicability of probe A for H 2 S detection in living cells. The supramolecular assembly of probe A and β-cyclodextrin further revealed the interaction with H 2 S.

Abstract:Developing a ratiometric H2S fluorescent probe with fast response is of great importance for studying the H2S physiology. Herein, two hemicyanine-based H2S probes were constructed; the one with a propanoic acid group (CouPa) showed poor sensitivity while the other one with theN,N-diethylpropionamide moiety (CouDE) exhibited distinctly improved performance. CouDE showed the ability to detect mitochondrial H2S level fluctuation, which was triggered by alteration of CBS enzyme activity. Moreover, endogenous H2S change in solid tumours was monitored using CouDE.

Abstract:Aqueous sulphides, including hydrogen sulphide, have important roles in biological signalling and metabolic processes. Here we develop a selective sulphide-trapping strategy involving sulphide addition to an aldehyde; the resulting hemithioacetal undergoes a Michael addition with an adjacent unsaturated acrylate ester to form a thioacetal at neutral pH in aqueous solution. Employing this new strategy, two sulphide-selective fluorescent probes, SFP-1 and SFP-2, were synthesized on the basis of two different fluorophore templates. These probes exhibit an excellent fluorescence increase and an emission maximum shift (SFP-1) in response to Na 2 S and H 2 S in a high thiol background as found under physiological conditions. We show the utility of the probes for the selective detection of sulphides, and the capacity of our probes to monitor enzymatic H 2 S biogenesis and image free sulphide in living cells.

Abstract:Hydrogen sulfide (H 2 S) plays a key role in the physiology and pathology of organisms, and H 2 S in the environment is easily absorbed and harmful to health. It is of great significance to develop a probe with good selectivity, high sensitivity and good stability that can detect hydrogen sulfide inside and outside organisms. In this work, we designed a novel "turn-on" fluorescent probe CIM-SDB for the detection of H 2 S. The probe CIM-SDB used indene-carbazole as the fluorophore and 2,4-dinitrobenzenesulfonyl as the recognition site. The probe CIM-SDB exhibited high selectivity and sensitivity to H 2 S (detection limit as low as 123 nM). Moreover, the probe CIM-SDB was successfully applied to the detection of intracellular exogenous and endogenous H 2 S, and the test strips prepared by the probe CIM-SDB could realize the convenient and rapid detection of H 2 S.

Abstract:H2S has been reported to play essential roles in a variety of physiological and pathological procedures. In this work, a novel fluorescent probe, Rho-HS, for detecting H2S was developed by introducing the ortho-halogen to activate the least reactive recognition group 2,4-dinitrophenyl moiety. In combination of the structures from both Rhodamine B and fluorescein, Rho-HS could generate both the colorimetric and fluorescent responses. This feature was not frequently achieved and could lead to the quantitative and convenient for the end-user. In comparison with recent probes for H2S, the major advantages of Rho-HS included suiting wide pH range (6.0-10.0), relatively rapid response (within 15 min) and the high selectivity among the competing species including the biothiols. With low cytoxicity, Rho-HS was further applied in the biological imaging in living MCF7 cells and Caenorhabditis elegans. We hope that the designing strategy in this work might provide useful information for more preferable implements in this field.

Abstract:Since Hydrogen Sulfide (H2S) was recognized as a gas transmitter, its detection and quantification have become a hot research topic among chemists and biologists. In this area, fluorescent probes have shown great advantages: fast and strong response, low detection limit and easy manipulation. Here we developed a new fluorescent probe that detected H2S selectively among various bioactive and inorganic salts. This probe was based on the core structure of fluorescein and reacted with H2S through azide-reduction. Great linearity was achieved correlating fluorescence intensity and H2S concentrations in solution. The detection of H2S in cancer cells was also achieved.

Abstract:Hydrogen sulphide (H2S), the third endogenous gaseous signalling molecule, has attracted attention in biochemical research. The selective detection of H2S in living systems is essential for studying its functions. Fluorescence detection methods have become useful tools to explore the physiological roles of H2S because of their real-time and non-destructive characteristics. Herein we report a near-infrared fluorescent probe, NIR-HS, capable of tracking H2S in living organisms. With high sensitivity, good selectivity and low cytotoxicity, NIR-HS was able to recognize both the exogenous and endogenous H2S in living cells. More importantly, it realized the visualization of endogenous H2S generated in cells overexpressing cystathionine β-synthase (CBS), one of the enzymes responsible for producing endogenous H2S. The probe was also successfully applied to detect both the exogenous and endogenous H2S in living mice. The superior sensing properties of the probe render it a valuable research tool in the H2S-related medical research.

Abstract:The rational design of effective tools capable of monitoring hydrogen sulfide (H2S) is of great importance to fully understand its physiological and pathological functions. Herein, a self-assembled fluorescent nanoprobe with both aggregation-induced emission (AIE) and excited-state intramolecular proton transfer (ESIPT) characteristics for H2S detection has been successfully developed via a modified nanoprecipitation method. The nanoprobe features high water dispersibility, a large Stokes shift (ca. 100 nm), good biocompatibility as well as high selectivity towards H2S over other putative interferents. Live cell imaging experiments demonstrate that the nanoprobe, with good cell-membrane permeability, can facilitate the visualization of exogenous and endogenous H2S levels. Moreover, the nanoprobe has also been found capable of detecting H2S in zebrafish. This nanoprobe with AIE and ESIPT characteristics can provide a novel method for the development of fluorescent probes for monitoring biological processes.

Abstract:Hydrogen sulfide (H2S) is associated with numerous psychological and pathological processes, and related closely with human health and diseases. The detection of H2S is a key to better understand its roles. In this paper, we developed a 4-phenylselenium coumarin (PhSeCou) as a rapid-response fluorescent probe for the selective and sensitive detection of H2S. The probe PhSeCou is a stable and almost non-fluorescent molecule in a PBS buffer, and it can rapidly (< 60 s) sense H2S to give a strongly fluorescent product, Cou-H2S (Phi(f) = 0.11, centered at 563 nm), thereby achieving to a sensitive detection at an ultra-low limit of detection (8.8 nM). The fluorescent probe displays a high selectivity for H2S over biothiols and other relevant analytes. What is more, through fluorescence cell-imaging, PhSeCou was successfully applied to detect exo- and endogenous H2S in living cells.
