Abstract:Inhibition of lycopene cyclization decreased the salt tolerance of the euhalophyte Salicornia europaea L. We isolated a β-lycopene cyclase gene SeLCY from S. europaea and transformed it into Arabidopsis with stable expression. Transgenic Arabidopsis on post-germination exhibited enhanced tolerance to oxidative and salt stress. After 8 and 21 d recovery from 200 mM NaCl treatment, transgenic lines had a higher survival ratio than wild-type (WT) plants. Three-week-old transgenic plants treated with 200 mM NaCl showed better growth than the WT with higher photosystem activity and less H(2)O(2) accumulation. Determination of endogenous pigments of Arabidopsis treated with 200 mM NaCl for 0, 2 or 4 d demonstrated that the transgenic plants retained higher contents of carotenoids than the WT. Furthermore, to compare the difference between SeLCY and AtLCY from Arabidopsis, we used viral vector mediating ectopic expression of SeLCY and AtLCY in Nicotiana benthamiana. Although LCY genes transformation increased the salt tolerance in tobacco, there is no significant difference between SeLCY- and AtLCY-transformed plants. These findings indicate that SeLCY transgenic Arabidopsis improved salt tolerance by increasing synthesis of carotenoids, which impairs reactive oxygen species and protects the photosynthesis system under salt stress, and as a single gene, SeLCY functionally showed no advantage for salt tolerance improvement compared with AtLCY.
Gm NAC15 Overexpression in Hairy Roots Enhances Salt Tolerance in Soybean

Abstract:The NAC（NAM, ATAF1/2 and CUC2） transcription factor family plays a key role in plant development and responses to abiotic stress. GmN AC15（Glyma15 g40510.1）, a member of the NAC transcription factor family in soybean, was functionally characterized, especially with regard to its role in salt tolerance. In the present study, q RT-PCR（quantitative reverse transcription PCR） analysis indicated that Gm NAC15 was induced by salt, drought, low temperature stress, and ABA treatment in roots and leaves. Gm NAC15 overexpression in soybean（Glycine max） hairy roots enhanced salt tolerance. Transgenic hairy roots improved the survival of wild leaves; however, overexpression of Gm NAC15 in hairy root couldn’t influnce the expression level of Gm NAC15 in leaf. Gm NAC15 regulates the expression levels of genes responsive to salt stress. Altogether, these results provide experimental evidence of the positive effect of Gm NAC15 on salt tolerance in soybean and the potential application of genetic manipulation to enhance the salt tolerance of important crops.

Abstract:The CALCINEURIN B-LIKE PROTEIN family genes ( CBLs ) encode a group of plant-specific calcium sensor proteins that play critical roles in plant development and stress response. Although genome-wide analysis of CBL family genes has been conducted in several diploid plant species, less is known about their functions in allopolyploid plant species. This study conducted a detailed analysis of tobacco CBL family genes. Of the 24 NtCBL genes identified, 14 were derived from the maternal genome donor N . sylvestris , and 10 were from the paternal genome donor N . tomentosiformis . Phylogenetic analysis revealed that NtCBLs could be clustered into three different groups with distinct N-terminal characteristics. All NtCBLs contain EF-hand motifs, but amino acid substitutions and configuration changes may be responsible for variations in their Ca 2+ -binding abilities. Alternative splicing is common for NtCBL gene transcripts, which may contribute to variations in translation efficiency or protein interaction preferences. Several NtCBL genes exhibited shoot- or root-predominant expression patterns, and some were responsive to salt and/or drought stress. NtCBL4A-1 , an ortholog of Arabidopsis thaliana AtCBL4 (SOS3), was predominantly expressed in roots and exhibited a lower gene expression level under salt stress. Further functional analysis showed that overexpression of NtCBL4A-1 increased the sensitivity of transgenic tobacco to Na + -induced salt stress, which is inconsistent with the role of AtCBL4 in contributing to salt tolerance. This study provides data for identifying CBL gene functions and selecting candidate stress-tolerant genes in N . tabacum . Additionally, the results may aid gene function studies in other allopolyploid plant species.

Abstract:Calcineurin B-like (CBL) proteins are a class of important Ca2+ receptors that play key roles in plant stress response. CBLs have been shown to participate in responses to abiotic stresses such as drought, salt, and cold in many plant species, including Arabidopsis and rice. However, little is known about their potential functions in the desert halophyte Nitraria tangutorum. Here, we have identified 11 CBL genes distributed across six chromosomes of N. tangutorum and categorized them into four groups through phylogenetic analysis. Synteny analysis showed that they have strong collinear relationships and have undergone purifying selection during their evolution. NtCBL promoter regions contain a variety of cis-acting elements related to hormone and environmental stress responses. Real-time quantitative PCR showed that the expression of NtCBLs differed significantly among various tissues and was upregulated by salt and drought stress. We chose NtCBL1-1 for an in-depth functional characterization and observed that transgenic Arabidopsis plants expressing NtCBL1-1 exhibited increased tolerance to both drought and salt stress. Compared to wild-type Arabidopsis, transgenic lines showed higher germination rates, slower chlorophyll degradation, more soluble proteins, and reduced accumulation of the oxidative stress marker malondialdehyde. These findings indicate that NtCBL1-1 plays a significant role in responding to drought and salt stress, laying the foundation for further investigations into the functional mechanisms of NtCBL genes in N. tangutorum.
Halophyte Nitraria billardieri CIPK25 mitigates salinity-induced cell damage by alleviating H2O2 accumulation

Abstract:The plant-specific module of calcineurin B-like proteins (CBLs) and CBL-interacting protein kinases (CIPKs) play a crucial role in plant adaptation to different biotic and abiotic stresses in various plant species. Despite the importance of the CBL-CIPK module in regulating plant salt tolerance, few halophyte CIPK orthologs have been studied. We identified NbCIPK25 in the halophyte Nitraria billardieri as a salt-responsive gene that may improve salt tolerance in glycophytes. Sequence analyses indicated that NbCIPK25 is a typical CIPK family member with a conserved NAF motif, which contains the amino acids: asparagine, alanine, and phenylalanine. NbCIPK25 overexpression in salt-stressed transgenic Arabidopsis seedlings resulted in enhanced tolerance to salinity, a higher survival rate, longer newly grown roots, more root meristem cells, and less damaged root cells in comparison to wild-type (WT) plants. H2O2 accumulation and malondialdehyde (MDA) content were both deceased in NbCIPK25-transgenic plants under salt treatment. Furthermore, their proline content, an important factor for scavenging reactive oxygen species, accumulated at a significantly higher level. In concordance, the transcription of genes related to proline accumulation was positively regulated in transgenic plants under salt condition. Finally, we observed a stronger auxin response in salt-treated transgenic roots. These results provide evidence for NbCIPK25 improving salt tolerance by mediating scavenging of reactive oxygen species, thereby protecting cells from oxidation and maintaining plant development under salt stress. These findings suggest the potential application of salt-responsive NbCIPK25 for cultivating glycophytes with a higher salt tolerance through genetic engineering.

Abstract:Although extensive studies and remarkable progress have been made with Arabidopsis calcineurin B-like proteins (CBLs), knowledge of their functions in other plant species is still limited. Here we isolated gene GmCBL1 from soybean, a homolog of AtCBL1 in Arabidopsis. GmCBL1 was differentially induced by multiple abiotic stress and plant hormones, and its transcripts were abundant in seedlings and mature roots. We over-expressed GmCBL1 in Arabidopsis and found that it enhanced tolerances to both high salt and drought stresses in the transgenic plants. Overexpression of GmCBL1 also promoted hypocotyl elongation under light conditions. GmCBL1 may regulate stress tolerance through activation of stress-related genes, and may control hypocotyl development by altering the expression of gibberellin biosynthesis-related genes. This study identifies a putative soybean CBL gene that functions in both stress tolerance and light-dependent hypocotyl development.

Abstract:The calcineurin B-like (CBL)-interacting protein kinases (CIPKs), a type of plant-specific genes in the calcium signaling pathway, function in response to adverse environments. However, few halophyte derived CIPKs have been studied for their role in plant physiological and developmental adaptation during abiotic stresses, which inhibits the potential application of these genes to improve environmental adaptability of glycophytes. In this study, we constructed Nitraria billardieri CIPK25 overexpressing Arabidopsis and analyzed the seedling development under salt treatment. Our results show that Arabidopsis with NbCIPK25 expression exhibits more vigorous growth than wild type plants under salt condition. To gain insight into the molecular mechanisms underlying salt tolerance, we profiled the transcriptome of WT and transgenic plants via RNA-seq. GO and KEGG analyses revealed that upregulated genes in NbCIPK25 overexpressing seedlings under salt stress are enriched in photosynthesis related terms; Calvin-cycle genes including glyceraldehyde-3-phosphate dehydrogenases (GAPDHs) are significantly upregulated in transgenic plants, which is consistent with a decreased level of NADPH (GAPDH substrate) and increased level of NADP + . Accordingly, NbCIPK25 overexpressing plants exhibited more efficient photosynthesis; soluble sugar and proteins, as photosynthesis products, showed a higher accumulation in transgenic plants. These results provide molecular insight into how NbCIPK25 promotes the expression of genes involved in photosynthesis, thereby maintaining plant growth under salt stress. Our finding supports the potential application of halophyte-derived NbCIPK25 in genetic modification for better salt adaptation.

Abstract:Many tobacco ( Nicotiana tabacum ) cultivars are salt-tolerant and thus are potential model plants to study the mechanisms of salt stress tolerance. The CALCINEURIN B-LIKE PROTEIN (CBL) is a vital family of plant calcium sensor proteins that can transmit Ca 2+ signals triggered by environmental stimuli including salt stress. Therefore, assessing the potential of NtCBL for genetic improvement of salt stress is valuable. In our studies on NtCBL members, constitutive overexpression of NtCBL5A was found to cause salt supersensitivity with necrotic lesions on leaves. NtCBL5A -overexpressing (OE) leaves tended to curl and accumulated high levels of reactive oxygen species (ROS) under salt stress. The supersensitivity of NtCBL5A -OE leaves was specifically induced by Na + , but not by Cl − , osmotic stress, or drought stress. Ion content measurements indicated that NtCBL5A -OE leaves showed sensitivity to the Na + accumulation levels that wild-type leaves could tolerate. Furthermore, transcriptome profiling showed that many immune response-related genes are significantly upregulated and photosynthetic machinery-related genes are significantly downregulated in salt-stressed NtCBL5A -OE leaves. In addition, the expression of several cation homeostasis-related genes was also affected in salt-stressed NtCBL5A -OE leaves. In conclusion, the constitutive overexpression of NtCBL5A interferes with the normal salt stress response of tobacco plants and leads to Na + -dependent leaf necrosis by enhancing the sensitivity of transgenic leaves to Na + . This Na + sensitivity of NtCBL5A -OE leaves might result from the abnormal Na + compartmentalization, plant photosynthesis, and plant immune response triggered by the constitutive overexpression of NtCBL5A . Identifying genes and pathways involved in this unusual salt stress response can provide new insights into the salt stress response of tobacco plants.

Abstract:Calcineurin B-like protein-interacting protein kinases (CIPKs) play essential roles in plant abiotic stress response. In order to better understand salt tolerance, we cloned and analyzed the NtCIPK9 gene from the halophyte Nitraria tangutorum. Phylogenetic analysis shows that NtCIPK9 belongs to a sister clade with the Arabidopsis AtCIPK9 gene and is thought to localize to the plasma membrane. NtCIPK9 shows the highest expression level in the Nitraria tangutorum root under normal growth conditions, whereas after NaCl treatment, the highest expression was found in the blade. NtCIPK9-overexpressing Arabidopsis plants have a higher seed germination rate, longer root length, and displayed higher salt tolerance than wild type seedlings under salt stress conditions. Furthermore, NtCIPK9 overexpression might enhance the expression of genes related to K+ transportation after NaCl treatment. Thus, we conclude that NtCIPK9 increases transgenic plant salt tolerance and reduces damage associated with salt stress by promoting the expression of genes controlling ion homeostasis. Our results suggest that NtCIPK9 could serve as an ideal candidate gene to genetically engineer salt-tolerant plants.

Abstract:[Objective]It is reported that the Calcineurin B-like proteins (CBLs)play important roles in the regu-lating plant growth and stress tolerant.In this study,a CBL gene (ThCBL4)was cloned from Tamarix hispida.And the salt tolerance function of ThCBL4 was studied.[Method]The word"Calcineurin B-like protein"was used as a query to search against the functional annotation of non-redundant unigenes based on the previous constructed T. hispida transcriptomes for identification of the ThCBL4.The expression patters of ThCBL4 genes in the roots and leaves of T.hispida under 0.4 mol·L-1 NaCl,20%(w/v)PEG6000 and 100 μmol·L-1 ABA treatment by using the real time RT-PCR.Furthermore,the plant overexpression vector and knockdown vector were constructed and transient transformed into T.hispida.At the same time,the vector pROKII was also transient infected into T. hispida as a control.The MDA contents,POD,SOD activities and DAB,NBT and Evans blue staining under NaCl stress were measured and compared among the pROKⅡ-ThCBL4 transient transformed T.hispida,ThCBL4 knock-down transient transformed T.hispida and the control.[Result]The cDNA length of ThCBL4 was 1432 bp,con-taining a length of 675 bp open reading frame encoding 224 amino acids.The relative molecular weight of ThCBL4 protein was 25.57 KDa,with isoelectric point 5.0.The expressions of ThCBL4 in T.hispida were upregulated in roots and downregualted in the leaves under NaCl,PEG or ABA stress conditions.Furthermore,the relative expres-sion levels of ThCBL4 among three transient expression lines was the highest in the overexpression lines while the lowest in the knockdown transient expression lines.The results of physiological indicators and physiological staining showed that the protective enzymes SOD and POD activity in ThCBL4 overexpression plants were significantly higher than the control and suppression of expression lines under salt stress.NBT and DAB staining showed that the O2-and H2 O2 contents in overexpression plants were significantly lower than the control.On the contrary,the O2-and H2 O2 contents in the inhibited expression lines were significantly higher than the control.At the same times,Evans blue staining and MDA assay results showed that the ThCBL4 overexpressing plants colored more shallow compared with the control,MDA contents is lower than the control,and colored deeper and higher content of MDA in the in-hibited expression of ThCBL4 plants.These results showed that the protective enzyme activities of ThCBL4 transient overexpression lines were higher than the control,the active oxygen contents were lower,and the degree of cell inju-ry were lighter.[Conclusion]The transient overexpression of ThCBL4 lines increased the salt tolerance,and Th-CBL4 gene may be involved in the response to salt stress in T.hispida.In the future,its function and the regulation mechanism need to be further studied.

Abstract:NACs are one of the major transcription factor families in plants which play an important role in plant growth and development, as well as in adverse stress responses. In this study, we cloned a salt-inducible NAC transcription factor gene ( NAC13 ) from a poplar variety 84K, followed by transforming it into both Nicotiana tabacum and Arabidopsis thaliana . Stable expression analysis of 35S::NAC13-GFP fusion protein in Arabidopsis indicated that NAC13 protein was localized to the nucleus. We also obtained five transgenic tobacco lines. Evidence from morphological and physiological characterization and salt treatment analyses indicated that in the transgenic tobacco the salt tolerance was enhanced, suggesting that NAC13 gene may function as a positive regulator in tobacco responses to salt stress.
The SlNAC8 gene of the halophyte Suaeda liaotungensis enhances drought and salt stress tolerance in transgenic Arabidopsis thaliana

Abstract:NAC (NAM, ATAF1/2 and CUC) transcription factors play an important role in resisting abiotic stress in plants. In this study, a novel NAC gene, designated SlNAC8 from Suaeda liaotungensis K. was characterized. SlNAC8 protein is localized in the nucleus, and the yeast one-hybrid screening showed that it contains an activation domain in its C-terminus and functions as a transcriptional activator. Gene expression analysis revealed that it is induced by drought and salt stress. Arabidopsis plants overexpressing SlNAC8 demonstrated enhanced tolerance to drought and salt stress, showing significant advantages in seed germination, root growth, shoot growth, and survival rate compared with controls. Moreover, transgenic plants had a significantly higher proline concentration, antioxidant enzyme activity (superoxide dismutase, peroxidase, and catalase), and level of chlorophyll fluorescence than wild-type, and a significantly lower malondialdehyde concentration and electrolyte leakage under drought and salt stress. The overexpression of SlNAC8 in transgenic plants also enhanced the expression of stress-responsive genes such as RD20, GSTF6, COR47, RD29A, RD29B, and NYC1. In summary, SlNAC8, as a transcription factor, may change the physiological-biochemical characteristic of plants by regulating the expression of stress-responsive genes and enhance the drought and salt stress tolerance of plants. SlNAC8 can be utilized for developing drought and salinity tolerance in crop plants through genetic engineering.
Calcineurin B-Like Proteins CBL4 and CBL10 Mediate Two Independent Salt Tolerance Pathways in Arabidopsis

Abstract:In Arabidopsis, the salt overly sensitive (SOS) pathway, consisting of calcineurin B-like protein 4 (CBL4/SOS3), CBL-interacting protein kinase 24 (CIPK24/SOS2) and SOS1, has been well defined as a crucial mechanism to control cellular ion homoeostasis by extruding Na+ to the extracellular space, thus conferring salt tolerance in plants. CBL10 also plays a critical role in salt tolerance possibly by the activation of Na+ compartmentation into the vacuole. However, the functional relationship of the SOS and CBL10-regulated processes remains unclear. Here, we analyzed the genetic interaction between CBL4 and CBL10 and found that the cbl4 cbl10 double mutant was dramatically more sensitive to salt as compared to the cbl4 and cbl10 single mutants, suggesting that CBL4 and CBL10 each directs a different salt-tolerance pathway. Furthermore, the cbl4 cbl10 and cipk24 cbl10 double mutants were more sensitive than the cipk24 single mutant, suggesting that CBL10 directs a process involving CIPK24 and other partners different from the SOS pathway. Although the cbl4 cbl10, cipk24 cbl10, and sos1 cbl10 double mutants showed comparable salt-sensitive phenotype to sos1 at the whole plant level, they all accumulated much lower Na+ as compared to sos1 under high salt conditions, suggesting that CBL10 regulates additional unknown transport processes that play distinct roles from the SOS1 in Na+ homeostasis.
CIPK11: a Calcineurin B-like Protein-Interacting Protein Kinase from Nitraria Tangutorum, Confers Tolerance to Salt and Drought in Arabidopsis

Abstract:BACKGROUND:The CIPKs are a group of plant-specific Ser/Thr protein kinases acting in response to calcium signaling, which plays an important role in the physiological and developmental adaptation of plants to adverse environments. However, the functions of halophyte-derived CIPKs are still poorly understood, that limits a potential application of CIPKs from halophytes for improving the tolerance of glycophytes to abiotic stresses.RESULTS:In this study, we characterized the NtCIPK11 gene from the halophyte Nitraria tangutorum and subsequently analyzed its role in salt and drought stress tolerance, using Arabidopsis as a transgenic model system. NtCIPK11 expression was upregulated in N. tangutorum root, stem and blade tissues after salt or drought treatment. Overexpressing NtCIPK11 in Arabidopsis improved seed germination on medium containing different levels of NaCl. Moreover, the transgenic plants grew more vigorously under salt stress and developed longer roots under salt or drought conditions than the WT plants. Furthermore, NtCIPK11 overexpression altered the transcription of genes encoding key enzymes involved in proline metabolism in Arabidopsis exposed to salinity, however, which genes showed a relatively weak expression in the transgenic Arabidopsis undergoing mannitol treatment, a situation that mimics drought stress. Besides, the proline significantly accumulated in NtCIPK11-overexpressing plants compared with WT under NaCl treatment, but that was not observed in the transgenic plants under drought stress caused by mannitol application.CONCLUSIONS:We conclude that NtCIPK11 promotes plant growth and mitigates damage associated with salt stress by regulating the expression of genes controlling proline accumulation. These results extend our understanding on the function of halophyte-derived CIPK genes and suggest that NtCIPK11 can serve as a candidate gene for improving the salt and drought tolerance of glycophytes through genetic engineering.
Calcineurin B-like Protein 5（sicbl5） in Setaria Italica Enhances Salt Tolerance by Regulating Na + Homeostasis

Abstract:Salinity, a major abiotic stress, reduces plant growth and severely limits agricultural productivity. Plants regulate salt uptake via calcineurin B-like proteins（CBLs）. Although extensive studies of the functions of CBLs in response to salt stress have been conducted in Arabidopsis, their functions in Setaria italica are still poorly understood. The foxtail millet genome encodes seven CBLs, of which only SiCBL4 was shown to be involved in salt response. Overexpression of SiCBL5 in Arabidopsis thaliana sos3-1 mutant rescued its salt hypersensitivity phenotype, but that of other SiCBLs（SiCBL1, SiCBL2, SiCBL3, SiCBL6, and SiCBL7） did not rescue the salt hypersensitivity of the Atsos3-1 mutant. SiCBL5 harbors an N-myristoylation motif and is located in the plasma membrane. Overexpression of SiCBL5 in foxtail millet increased its salt tolerance,but its knockdown increased salt hypersensitivity. Yeast two-hybrid and firefly luciferase complementation imaging assays showed that SiCBL5 physically interacted with SiCIPK24 in vitro and in vivo. Cooverexpression of SiCBL5, SiCIPK24, and SiSOS1 in yeast conferred a high-salt-tolerance phenotype.Compared to wild-type plants under salt stress conditions, SiCBL5 overexpressors showed lower accumulations of Na + and stronger Na + efflux, whereas RNAi-SiCBL5 plants showed higher accumulations of Na + and weaker Na + efflux. These results indicate that SiCBL5 confers salt tolerance in foxtail millet by modulating Na + homeostasis.

Abstract:NAC (NAM, ATAF1/2 and CUC2) proteins are key regulators of various plant stress tolerances. However, knowledge of NAC genes remains largely unknown in Miscanthus. Here, we characterized a novel NAC gene MlNAC10 from M. lutarioriparius than plays a role in abiotic stress tolerance. MlNAC10 encodes a nuclear-localized protein with a C-terminal transactivation domain, and has a specific binding affinity to the NAC recognition sequence (NACRS). Ectopic expression of MlNAC10 in Arabidopsis led to increased sensitivity to abscisic acid (ABA) at early seedling growth stages. In addition, the proline content was significantly increased and the reactive oxygen species (ROS) scavenging capability was significantly enhanced in MlNAC10 overexpression lines under ABA treatment. Moreover, the drought and salt stress tolerance was significantly improved in MlNAC10 overexpression lines. Consistently, the activities of three antioxidant enzymes, namely catalase (CAT), peroxidase (POD), and superoxide dismutase (SOD), were dramatically stimulated in the overexpression lines compared to the wild type (WT). Correspondingly, the accumulation of ROS was dramatically decreased and malondialdehyde (MDA) was accumulated at a much lower level in the transgenic lines. Meanwhile, the expression of six abiotic stress-related genes was dramatically stimulated in the overexpression lines in comparison to the WT. Together, our results demonstrated that MlNAC10 acts as an important regulator of drought and salinity stress tolerance by stimulating antioxidant enzymes and alleviating ROS damage via the ABA signaling pathway.
The NAC-type transcription factor CaNAC46 regulates the salt and drought tolerance of transgenic Arabidopsis thaliana

Abstract:Background: The NAC (NAM, ATAF1/ATAF2, and CUC2) transcription factors belong to a large family of plant-specific transcription factors in monocot and dicot species. These transcription factors regulate the expression of stress tolerance-related genes that protect plants from various abiotic stresses, including drought, salinity, and low temperatures. Results: In this study, we identified the CaNAC46 transcription factor gene in Capsicum annuum. Its open reading frame was revealed to comprise 921 bp, encoding a protein consisting of 306 amino acids, with an isoelectric point of 6.96. A phylogenetic analysis indicated that CaNAC46 belongs to the ATAF subfamily. The expression of CaNAC46 was induced by heat, cold, high salt, drought, abscisic acid, salicylic acid, and methyl jasmonate treatments. Thus, CaNAC46 may be important for the resistance of dry pepper to abiotic stresses. A subcellular localization analysis confirmed that CaNAC46 is localized in the nucleus. The overexpression of CaNAC46 improved the tolerance of transgenic Arabidopsis thaliana plants to drought and salt stresses. The CaNAC46-overexpressing lines had longer roots and more lateral roots than wild-type lines under prolonged drought and high salt stress conditions. Additionally, CaNAC46 affected the accumulation of reactive oxygen species (ROS). Moreover, CaNAC46 promoted the expression of SOD, POD, RD29B, RD20, LDB18, ABI, IAA4, and P5CS. The malondialdehyde contents were higher in TRV2-CaNAC46 lines than in wild-type plants in response to drought and salt stresses. Furthermore, the expression levels of stress-responsive genes, such as ABA2, P5CS, DREB, RD22, CAT, and POD, were down-regulated in TRV2-CaNAC46 plants. Conclusions: Under saline and drought conditions, CaNAC46 is a positive regulator that activates ROS-scavenging enzymes and enhances root formation. The results of our study indicate CaNAC46 is a transcriptional regulator responsible for salinity and drought tolerance and suggest the abiotic stress-related gene regulatory mechanisms controlling this NAC transcription factor are conserved between A. thaliana and pepper.

Abstract:Lilium pumilum is a perennial herb with ornamental edible and medicinal value. It is an excellent wild germplasm resource with wide distribution and strong resistance. The NAC family of transcription factors is unique to higher plants. The NAC family plays a regulatory role in plant growth and development and participates in plant responses to biotic and abiotic stresses. The LpNAC17 gene of L. pumilum was cloned and transformed into tobacco to investigate the response of transgenic tobacco to salt stress. The results showed that the net photosynthetic rate and contents of chlorophyll in LpNAC17 over-expressed tobacco were higher than those in the control plants, while the stomatal conductance, transpiration rate and intercellular CO 2 concentration were lower than those in the controls. The activity of superoxide dismutase, peroxidase, catalase, and the content of proline in LpNAC17 over-expressed tobacco were higher than those in the controls, while the content of malondialdehyde, superoxide anion, and hydrogen peroxide were lower than that in the control. Nitro-blue tetrazolium staining and 3,3′-diaminobenzidine tissue localization showed that the contents of O 2 - and H 2 O 2 in transgenic tobacco was lower than in the controls. The expression levels of NtSOD, NtPOD, NtCAT, NtHAK1, NtPMA4 , and NtSOS1 in the transgenic tobacco were higher than those in the controls. Therefore, this study provides a gene source for molecular breeding of salt-tolerant plants through genetic engineering, and lays a foundation for further research on salt-tolerant Lily.
