Abstract:Although extensive studies and remarkable progress have been made with Arabidopsis calcineurin B-like proteins (CBLs), knowledge of their functions in other plant species is still limited. Here we isolated gene GmCBL1 from soybean, a homolog of AtCBL1 in Arabidopsis. GmCBL1 was differentially induced by multiple abiotic stress and plant hormones, and its transcripts were abundant in seedlings and mature roots. We over-expressed GmCBL1 in Arabidopsis and found that it enhanced tolerances to both high salt and drought stresses in the transgenic plants. Overexpression of GmCBL1 also promoted hypocotyl elongation under light conditions. GmCBL1 may regulate stress tolerance through activation of stress-related genes, and may control hypocotyl development by altering the expression of gibberellin biosynthesis-related genes. This study identifies a putative soybean CBL gene that functions in both stress tolerance and light-dependent hypocotyl development.
Stress-Induced Expression of an Activated Form of Atbzip17 Provides Protection from Salt Stress in Arabidopsis

Abstract:Membrane-associated basic-leucine zipper (bZIP) transcription factors that reside in the endoplasmic reticulum represent a newly described class of plant stress sensor/transducers. The bZIP factors are anchored in endoplasmic reticulum (ER) membranes with their C-terminal tails facing the ER lumen and their N terminii, which contain transcriptional components, facing the cytosol. In response to stress, cytosolic components of the transcription factors are released by proteolysis and move to the nucleus where they promote the up-regulation of stress response genes. One such stress sensor/transducer in Arabidopsis is AtbZIP17, which is activated in response to salt stress. With the aim of enhancing salt tolerance, a constitutively activated form of AtbZIP17, truncated before its membrane-anchor domain, was introduced into transgenic plants. When placed under the control of a constitutive promoter, the activated form of AtbZIP17 up-regulated stress response genes under unstressed conditions, but caused a substantial delay in plant development. When the activated form of AtbZIP17 was placed under the control of stress-inducible promoter, development was normal under unstressed conditions. Under salt stress conditions, the stress-inducible expression of the activated AtbZIP17 enhanced salt tolerance as demonstrated by chlorophyll bleaching and seedling survival assays.
TaCIPK29, a CBL-interacting Protein Kinase Gene from Wheat, Confers Salt Stress Tolerance in Transgenic Tobacco.

Abstract:Calcineurin B-like protein-interacting protein kinases (CIPKs) have been found to be responsive to abiotic stress. However, their precise functions and the related molecular mechanisms in abiotic stress tolerance are not completely understood, especially in wheat. In the present study, TaCIPK29 was identified as a new member of CIPK gene family in wheat. TaCIPK29 transcript increased after NaCl, cold, methyl viologen (MV), abscisic acid (ABA) and ethylene treatments. Over-expression of TaCIPK29 in tobacco resulted in increased salt tolerance, which was demonstrated by higher germination rates, longer root lengths and better growth status of transgenic tobacco plants compared to controls when both were treated with salt stress. Physiological measurements indicated that transgenic tobacco seedlings retained high K(+)/Na(+) ratios and Ca(2+) content by up-regulating some transporter genes expression and also possessed lower H2O2 levels and reduced membrane injury by increasing the expression and activities of catalase (CAT) and peroxidase (POD) under salt stress. Moreover, transgenic lines conferred tolerance to oxidative stress by increasing the activity and expression of CAT. Finally, TaCIPK29 was located throughout cells and it preferentially interacted with TaCBL2, TaCBL3, NtCBL2, NtCBL3 and NtCAT1. Taken together, our results showed that TaCIPK29 functions as a positive factor under salt stress and is involved in regulating cations and reactive oxygen species (ROS) homeostasis.

Abstract:The CCCH type zinc finger proteins are a super family involved in many aspects of plant growth and development. In this study, we investigated the response of one CCCH type zinc finger protein AtZFP1 (At2g25900) to salt stress in Arabidopsis. The expression of AtZFP1 was upregulated by salt stress. Compared to transgenic strains, the germination rate, emerging rate of cotyledons and root length of wild plants were significantly lower under NaCl treatments, while the inhibitory effect was significantly severe in T-DNA insertion mutant strains. At germination stage, it was mainly osmotic stress when treated with NaCl. Relative to wild plants, overexpression strains maintained a higher K+, K+/Na+, chlorophyll and proline content, and lower Na+ and MDA content. Quantitative real-time PCR analysis revealed that the expression of stress related marker genes KIN1, RD29B and RD22 increased more significantly in transgenic strains by salt stress. Overexpression of AtZFP1 also enhanced oxidative and osmotic stress tolerance which was determined by measuring the expression of a set of antioxidant genes, osmotic stress genes and ion transport protein genes such as SOS1, AtP5CS1 and AtGSTU5. Overall, our results suggest that overexpression of AtZFP1 enhanced salt tolerance by maintaining ionic balance and limiting oxidative and osmotic stress.

Abstract:In plants, CBL mediated calcium signaling is widely involved in the response to plant stresses of adversity. However, to date, no comprehensive studies have been conducted on CBL family members in Salvia miltiorrhiza . Herein, we identified 8 SmCBLs in S. miltiorrhiza , and phylogenetic analysis classified SmCBLs into four groups. Analysis of cis-acting elements revealed that SmCBLs mostly have light-responsive and hormone-responsive elements. Tissue expression analysis indicated that almost all of SmCBLs were expressed in roots than in leaves and flowers. SmCBL3 responded to Abscisic Acid (ABA), polyethylene glycol (PEG), and NaCl treatments. Transgenic Arabidopsis thaliana that overexpressed SmCBL3 had higher germination rates and longer roots than the wild type (WT) when exposed to salt stress. Additionally, the transgenic lines exhibited higher levels of chlorophyll, proline, superoxide dismutase (SOD), peroxidase (POD), and catalase (CAT) activity and SOS1 , NHX1 and P5CS1 expression than WT, and lower levels of malondialdehyde (MDA). Furthermore, SmCBL3 interacts with SmCIPK9. In conclusion, we analyzed the protein physicochemical properties, evolutionary relationships, gene structures, and expression profiles of the SmCBL gene families in S. miltiorrhiza . Overexpression of SmCBL3 improves the salt tolerance of transgenic Arabidopsis. This study demonstrated that SmCBL3 is a positive regulator of plant salt tolerance, so the use of overexpressed SmCBL3 may serve as a potential strategy to enhance plant salt tolerance.

Abstract:Plasma membrane (PM) H+-ATPase (AHA) provides energy to some important physiological and biochemical processes in plant cells, playing a key role in plant growth and development. Here, we found that the growth and PM H+-ATPase activity of the Arabidopsis cbl10 mutant was lower than the wild-type plant (WT) under salinity stress, suggesting that the calcineurin B-like protein 10 (CBL10) might be involved in the regulation of AHA. In contrast, cbl10 grew better than WT when exposure to external higher pH stress, and the growth of cbl10 plant expressing CBL10 gene was similar as WT under external high pH. Under alkaline conditions, the proton influx rate of cbl10 mutant was slower than that of WT, and the pH in the plate area where cbl10 mutant grew was lower compared to WT plates. Yeast two-hybrid and BiFC assays showed that CBL10 bound to the C-terminus of AtAHA4 or AtAHA11, such that the growth of yeast cells coexpressing CBL10 and AHA4/AHA11 was worse than transgenic cells with a single AHA4 or AHA11. This suggests that CBL10 negatively regulates PM H+-ATPase activity by directly interacting with AtAHA4 or AtAHA11, which decreases the ability of Arabidopsis to adapt to alkaline environment. Overall, this study indicates that CBL10 may function as an interconnecting regulator to coordinate plant responses to saline and alkaline stresses.

Abstract:CBL-interacting protein kinases are involved in plant responses to abiotic stresses, including salt stress. However, the negative regulating mechanism of this gene family in response to salinity is less reported. In this study, we evaluated the role of TaCIPK25 in regulating salt response in wheat. Under conditions of high salinity, TaCIPK25 expression was markedly down-regulated in roots. Overexpression of TaCIPK25 resulted in hypersensitivity to Na+ and superfluous accumulation of Na+ in transgenic wheat lines. TaCIPK25 expression did not decline in transgenic wheat and remained at an even higher level than that in wild-type wheat controls under high-salinity treatment. Furthermore, transmembrane Na+/H+ exchange was impaired in the root cells of transgenic wheat. These results suggested that TaCIPK25 negatively regulated salt response in wheat. Additionally, yeast-one-hybrid, beta-glucuronidase activity and DNA-protein-interaction-enzyme-linked-immunosorbent assays showed that the transcription factor TaWRKY9 bound W-box in the TaCIPK25 promoter region. Quantitative real-time polymerase chain reaction assays showed concomitantly inverted expression patterns of TaCIPK25 and TaWRKY9 in wheat roots under salt treatment, ABA application and inhibition of endogenous ABA condition. Overall, based on our results, in a salt stress condition, the negative salt response in wheat involved TaCIPK25 with the expression regulated by TaWRKY9.

Abstract:For plant growth under salt stress, sensing and transducing salt signals are central to cellular Na+ homoeostasis. The calcineurin B‐like protein (CBL)‐CBL‐interacting protein kinase (CIPK) complexes play critical roles in transducing salt signals in plants. Here, we show that CBL5, an ortholog of CBL4 and CBL10 in Arabidopsis, interacts with and recruits CIPK8/CIPK24 to the plasma membrane. Yeast cells coexpressing CBL5, CIPK8/CIPK24 and SOS1 demonstrated lesser Na+ accumulation and a better growth phenotype than the untransformed or SOS1 transgenic yeast cells under salinity. Overexpression of CBL5 improved the growth of the cipk8 or cipk24 single mutant but not the cipk8 cipk24 double mutant under salt stress, suggesting that CIPK8 and CIPK24 were the downstream targets of CBL5. Interestingly, seed germination in cbl5 was severely inhibited by NaCl, which was recovered by the overexpression of CBL5. Furthermore, CBL5 was mainly expressed in the cotyledons and hypocotyls, which are essential to seed germination. Na+ efflux activity in the hypocotyls of cbl5 was reduced relative to the wild‐type under salt stress, enhancing Na+ accumulation. These findings indicate that CBL5 functions in seed germination and protects seeds and germinating seedlings from salt stress through the CBL5‐CIPK8/CIPK24‐SOS1 pathways.

Abstract:Drought is one of the major environmental stresses to plants. The calcium sensor, calcineurin B-like (CBL) proteins, and their interacting protein kinases (CIPK) play important roles in responding to abiotic stresses. In this study, we functionally characterized a CIPK gene from Triticum aestivum designated TaCIPK27. The transcriptional levels of TaCIPK27 were increased both in roots and leaves after treatment with polyethylene glycol 8000, abscisic acid and H2O2. Besides, TaCIPK27 interacted with AtCBL1, AtCBL3, AtCBL4, AtCBL5 and AtCBL9 in yeast two-hybrid assays. Ectopic overexpression of TaCIPK27 positively regulates drought tolerance in transgenic Arabidopsis compared with controls, which was demonstrated by seed germination and survival rates experiments, as well as the detection of physiological indices including ion leakage, malonic dialdehyde and H2O2 contents and antioxidant enzyme activities under normal and drought conditions. Moreover, higher concentration of endogenous abscisic acid was detected under drought in TaCIPK27 transgenic plants. In addition, TaCIPK27 transgenic plants were more sensitive to exogenous abscisic acid treatment at seed germination and seedling stage. The expression levels of somedrought stress and abscisic acid related genes were up-regulated in TaCIPK27 transgenic plants. The results suggest that TaCIPK27 functions as a positive regulator under drought partly in an ABA-dependent pathway.
Genome-Wide Identification of Members of the Soybean CBL Gene Family and Characterization of the Functional Role of GmCBL1 in Responses to Saline and Alkaline Stress

Abstract:Calcium ions function as key messengers in the context of intracellular signal transduction. The ability of plants to respond to biotic and abiotic stressors is highly dependent on the calcineurin B-like protein (CBL) and CBL-interacting protein kinase (CIPK) signaling network. Here, a comprehensive effort was made to identify all members of the soybean CBL gene family, leading to the identification of 15 total genes distributed randomly across nine chromosomes, including 13 segmental duplicates. All the GmCBL gene subfamilies presented with similar gene structures and conserved motifs. Analyses of the expression of these genes in different tissues revealed that the majority of these GmCBLs were predominantly expressed in the roots. Significant GmCBL expression and activity increases were also observed in response to a range of stress-related treatments, including salt stress, alkaline stress, osmotic stress, or exposure to salicylic acid, brassinosteroids, or abscisic acid. Striking increases in GmCBL1 expression were observed in response to alkaline and salt stress. Subsequent analyses revealed that GmCBL1 was capable of enhancing soybean salt and alkali tolerance through the regulation of redox reactions. These results offer new insight into the complex mechanisms through which the soybean CBL gene family regulates the responses of these plants to environmental stressors, highlighting promising targets for efforts aimed at enhancing soybean stress tolerance.

Halophyte Nitraria billardieri CIPK25 promotes photosynthesis in Arabidopsis under salt stress

Abstract:The calcineurin B-like (CBL)-interacting protein kinases (CIPKs), a type of plant-specific genes in the calcium signaling pathway, function in response to adverse environments. However, few halophyte derived CIPKs have been studied for their role in plant physiological and developmental adaptation during abiotic stresses, which inhibits the potential application of these genes to improve environmental adaptability of glycophytes. In this study, we constructed Nitraria billardieri CIPK25 overexpressing Arabidopsis and analyzed the seedling development under salt treatment. Our results show that Arabidopsis with NbCIPK25 expression exhibits more vigorous growth than wild type plants under salt condition. To gain insight into the molecular mechanisms underlying salt tolerance, we profiled the transcriptome of WT and transgenic plants via RNA-seq. GO and KEGG analyses revealed that upregulated genes in NbCIPK25 overexpressing seedlings under salt stress are enriched in photosynthesis related terms; Calvin-cycle genes including glyceraldehyde-3-phosphate dehydrogenases (GAPDHs) are significantly upregulated in transgenic plants, which is consistent with a decreased level of NADPH (GAPDH substrate) and increased level of NADP + . Accordingly, NbCIPK25 overexpressing plants exhibited more efficient photosynthesis; soluble sugar and proteins, as photosynthesis products, showed a higher accumulation in transgenic plants. These results provide molecular insight into how NbCIPK25 promotes the expression of genes involved in photosynthesis, thereby maintaining plant growth under salt stress. Our finding supports the potential application of halophyte-derived NbCIPK25 in genetic modification for better salt adaptation.

Abstract:The CALCINEURIN B-LIKE PROTEIN family genes ( CBLs ) encode a group of plant-specific calcium sensor proteins that play critical roles in plant development and stress response. Although genome-wide analysis of CBL family genes has been conducted in several diploid plant species, less is known about their functions in allopolyploid plant species. This study conducted a detailed analysis of tobacco CBL family genes. Of the 24 NtCBL genes identified, 14 were derived from the maternal genome donor N . sylvestris , and 10 were from the paternal genome donor N . tomentosiformis . Phylogenetic analysis revealed that NtCBLs could be clustered into three different groups with distinct N-terminal characteristics. All NtCBLs contain EF-hand motifs, but amino acid substitutions and configuration changes may be responsible for variations in their Ca 2+ -binding abilities. Alternative splicing is common for NtCBL gene transcripts, which may contribute to variations in translation efficiency or protein interaction preferences. Several NtCBL genes exhibited shoot- or root-predominant expression patterns, and some were responsive to salt and/or drought stress. NtCBL4A-1 , an ortholog of Arabidopsis thaliana AtCBL4 (SOS3), was predominantly expressed in roots and exhibited a lower gene expression level under salt stress. Further functional analysis showed that overexpression of NtCBL4A-1 increased the sensitivity of transgenic tobacco to Na + -induced salt stress, which is inconsistent with the role of AtCBL4 in contributing to salt tolerance. This study provides data for identifying CBL gene functions and selecting candidate stress-tolerant genes in N . tabacum . Additionally, the results may aid gene function studies in other allopolyploid plant species.

Abstract:Abstract Plant salt-stress response involves complex physiological processes. Previous studies have shown that some factors promote salt tolerance only under high transpiring condition, thus mediating transpiration-dependent salt tolerance (TDST). However, the mechanism underlying crop TDST remains largely unknown. Here, we report that ZmSTL1 ( Salt-Tolerant Locus 1 ) confers natural variation of TDST in maize. ZmSTL1 encodes a dirigent protein (termed ZmESBL) localized to the Casparian strip (CS) domain. Mutants lacking ZmESBL display impaired lignin deposition at endodermal CS domain which leads to a defective CS barrier. Under salt condition, mutation of ZmESBL increases the apoplastic transport of Na + across the endodermis, and then increases the root-to-shoot delivery of Na + via transpiration flow, thereby leading to a transpiration-dependent salt hypersensitivity. Moreover, we show that the ortholog of ZmESBL also mediates CS development and TDST in Arabidopsis. Our study suggests that modification of CS barrier may provide an approach for developing salt-tolerant crops.

Abstract:C2H2 zinc finger proteins (ZFPs) play important roles in plant development and response to abiotic stresses, and have been studied extensively. However, there are few studies on ZFPs in mangroves and mangrove associates, which represent a unique plant community with robust stress tolerance. MpZFP1, which is highly induced by salt stress in the mangrove associate Millettia pinnata, was cloned and functionally characterized in this study. MpZFP1 protein contains two zinc finger domains with conserved QALGGH motifs and targets to the nucleus. The heterologous expression of MpZFP1 in Arabidopsis increased the seeds’ germination rate, seedling survival rate, and biomass accumulation under salt stress. The transgenic plants also increased the expression of stress-responsive genes, including RD22 and RD29A, and reduced the accumulation of reactive oxygen species (ROS). These results indicate that MpZFP1 is a positive regulator of plant responses to salt stress due to its activation of gene expression and efficient scavenging of ROS.

Abstract:Characterization of a new tomato (Solanum lycopersicum) T-DNA mutant allowed for the isolation of the CALCINEURIN B-LIKE PROTEIN 10 (SlCBL10) gene whose lack of function was responsible for the severe alterations observed in the shoot apex and reproductive organs under salinity conditions. Physiological studies proved that SlCBL10 gene is required to maintain a proper low Na+/Ca2+ ratio in growing tissues allowing tomato growth under salt stress. Expression analysis of the main responsible genes for Na+ compartmentalization (i.e. Na+/H+ EXCHANGERs, SALT OVERLY SENSITIVE, HIGH-AFFINITY K+ TRANSPORTER 1;2, H+-pyrophosphatase AVP1 [SlAVP1] and V-ATPase [SlVHA-A1]) supported a reduced capacity to accumulate Na+ in Slcbl10 mutant leaves, which resulted in a lower uploading of Na+ from xylem, allowing the toxic ion to reach apex and flowers. Likewise, the tomato CATION EXCHANGER 1 and TWO-PORE CHANNEL 1 (SlTPC1), key genes for Ca2+ fluxes to the vacuole, showed abnormal expression in Slcbl10 plants indicating an impaired Ca2+ release from vacuole. Additionally, complementation assay revealed that SlCBL10 is a true ortholog of the Arabidopsis (Arabidopsis thaliana) CBL10 gene, supporting that the essential function of CBL10 is conserved in Arabidopsis and tomato. Together, the findings obtained in this study provide new insights into the function of SlCBL10 in salt stress tolerance. Thus, it is proposed that SlCBL10 mediates salt tolerance by regulating Na+ and Ca2+ fluxes in the vacuole, cooperating with the vacuolar cation channel SlTPC1 and the two vacuolar H+-pumps, SlAVP1 and SlVHA-A1, which in turn are revealed as potential targets of SlCBL10.
ZmILI1 confers salt stress tolerance by regulating genes of phytohormone response in maize

Abstract:Salt stress is a major abiotic stress that has severe adverse effects on the growth, development, yield and quality of crop plants. Molecular regulatory mechanisms underlying salt stress response in maize are still elusive. Understanding salt stress tolerance mechanisms is essential for the development of high-yielding maize cultivars with improved salt tolerance. Here, we identified a gene, INCREASED LEAF INCLINATION1 ( ZmILI1 ), encoding a bHLH transcription factor, which positively regulates maize response to salt stress. Salt stress induces the expression of ZmILI1 , and the overexpression of ZmILI1 enhances catalase (CAT), peroxidase (POD) and superoxide dismutase (SOD) activities, thus enhancing salt stress tolerance. DNA affinity purification sequencing (DAP-Seq), RNA sequencing (RNA-Seq), dual-luciferase (LUC), yeast one hybrid, and Real-time polymerase chain reaction (RT-qPCR) assays revealed that ZmILI1 positively regulates salt stress response in maize by modulating the expression of the transcription factor family genes ( ZmAP2-197 ), hormone-related genes ( ZmLOX6 , ZmDHN1 , ZmCaDK2C ) and antioxidant enzyme system-related genes ( ZmPOD5 ), thus maintaining cellular homeostasis. Moreover, ZmILI1 is directly or indirectly implicated in maize response to salt stress by regulating the expression of genes involved in jasmonic acid (JA) biosynthesis, such as ZmLOX6 , leading to elevated levels of JA hormone in maize under salt-stressed conditions. Our findings not only provide genetic evidence for the role of the bHLH protein family in agricultural production but also reveal potential regulatory mechanisms for the development of salt-tolerant genetic resources, offering new insights into the breeding of salt-tolerant maize varieties.

Abstract:Calcineurin B-like protein-interacting protein kinases (CIPKs) play essential roles in plant abiotic stress response. In order to better understand salt tolerance, we cloned and analyzed the NtCIPK9 gene from the halophyte Nitraria tangutorum. Phylogenetic analysis shows that NtCIPK9 belongs to a sister clade with the Arabidopsis AtCIPK9 gene and is thought to localize to the plasma membrane. NtCIPK9 shows the highest expression level in the Nitraria tangutorum root under normal growth conditions, whereas after NaCl treatment, the highest expression was found in the blade. NtCIPK9-overexpressing Arabidopsis plants have a higher seed germination rate, longer root length, and displayed higher salt tolerance than wild type seedlings under salt stress conditions. Furthermore, NtCIPK9 overexpression might enhance the expression of genes related to K+ transportation after NaCl treatment. Thus, we conclude that NtCIPK9 increases transgenic plant salt tolerance and reduces damage associated with salt stress by promoting the expression of genes controlling ion homeostasis. Our results suggest that NtCIPK9 could serve as an ideal candidate gene to genetically engineer salt-tolerant plants.

Abstract:Calcineurin B-like proteins (CBLs) as specific calcium sensors that interact with CBL-interacting protein kinases (CIPKs) play a key role in the regulation of plant development and abiotic stress tolerance. In this study, we isolated and characterized the CIPK32 gene from Zea mays. ZmCIPK32 showed that it comprised 440 amino acids and a conserved NAF motif responsible for the interaction with CBLs localized in the cytoplasm and cell membrane. The interaction of ZmCIPK32 with ZmCBL1 and ZmCBL9 demonstrated using yeast two-hybrid system and bimolecular fluorescence complementation assay required the presence of the NAF domain. Overexpression of ZmCIPK32 promoted early germination in transgenic Arabidopsis seeds relative to that observed in wild-type (WT) plants under mannitol treatment. In addition, ZmCIPK32-overexpressing plants were insensitive to treatments with exogenous abscisic acid and paclobutrazol (PBZ) at seed germination and early seedling stages. Expression levels of the key genes GA20ox and GA3ox involved in the synthesis of gibberellin (GA) were increased, whereas expression levels of genes involved in the conversion of active GA to inactive forms and GA signaling were reduced in ZmCIPK32-overexpressing plants relative to those in WT plants under mannitol and PBZ treatments. Furthermore, overexpression of ZmCIPK32 increased GA level but decreased abscisic acid level in transgenic lines compared to the respective levels in WT plants under PBZ or mannitol treatments. Our results suggest that ZmCIPK32 positively regulates seed germination under stressed conditions by modulating GA signals.

Abstract:Salinity stands as a significant environmental stressor, severely impacting crop productivity. Plants exposed to salt stress undergo physiological alterations that influence their growth and development. Meanwhile, plants have also evolved mechanisms to endure the detrimental effects of salinity-induced salt stress. Within plants, Calcineurin B-like (CBL) proteins act as vital Ca2+ sensors, binding to Ca2+ and subsequently transmitting signals to downstream response pathways. CBLs engage with CBL-interacting protein kinases (CIPKs), forming complexes that regulate a multitude of plant growth and developmental processes, notably ion homeostasis in response to salinity conditions. This review introduces the repercussions of salt stress, including osmotic stress, diminished photosynthesis, and oxidative damage. It also explores how CBLs modulate the response to salt stress in plants, outlining the functions of the CBL-CIPK modules involved. Comprehending the mechanisms through which CBL proteins mediate salt tolerance can accelerate the development of cultivars resistant to salinity.

Abstract:The NAC gene family has transcription factors specific to plants, which are involved in development and stress response and adaptation. In this study, ZmNAC89, an NAC gene in maize that plays a role in saline–alkaline tolerance, was isolated and characterized. ZmNAC89 was localized in the nucleus and had transcriptional activation activity during in vitro experiments. The expression of ZmNAC89 was strongly upregulated under saline–alkaline, drought and ABA treatments. Overexpression of the ZmNAC89 gene in transgenic Arabidopsis and maize enhanced salt tolerance at the seedling stage. Differentially expressed genes (DEGs) were then confirmed via RNA-sequencing analysis with the transgenic maize line. GO analyses showed that oxidation–reduction process-regulated genes were involved in ZmNAC89-mediated salt–alkaline stress. ZmNAC89 may regulate maize saline–alkali tolerance through the REDOX pathway and ABA signal transduction pathway. From 140 inbred maize lines, 20 haplotypes and 16 SNPs were found in the coding region of the ZmNAC89 gene, including the excellent haplotype HAP20. These results contribute to a better understanding of the response mechanism of maize to salt–alkali stress and marker-assisted selection during maize breeding.
