Abstract:In a systematic effort to establish the relative as well as absolute configurations of two natural products isolated from Asomycete Daldinia concentrica, four independent (non-antipodal) diastereomers of hept-6-en-2,4,5-triol, the structure previously proposed for the natural products, were synthesized in enantiopure forms through a chiral-pool route and their optical rotation as well as NMR data were recorded. Although these four synthetic isomers cover all possible relative configurations the originally assigned triol may have, none of them gave spectroscopic data compatible with those reported for the natural products. Similar negative results were also obtained with a group of four non-antipodal diastereomers of hept-6-en-2,3,5-triol. The genuine structures of the natural products are therefore to be re-assigned.

Abstract:The proposed diastereoisomers (1 a-d) together with their C8'-epimers (1 e-h) of amipurimycin, a unique antifungal peptidyl nucleoside antibiotic, have been synthesized for the first time. The synthetic approach is efficient and stereodivergent, and features a stereoselective aldol condensation to build the branched C9 sugar amino acid skeleton and a regio- and stereocontrolled gold(I)-catalyzed N-glycosylation to furnish the purine nucleoside. Analysis of the NMR data suggests that the previously assigned configuration of the tertiary C3' in amipurimycin should be of opposite configuration.

Abstract:A diastereoselective approach for the total synthesis of an unusual atropisomer of the Schisandra triterpenoid (+/-)-schiglautoneA is described. The efficient synthetic strategy features three key transformations: 1) two sequential titanium(III)-catalyzed radical cyclization/homologation reactions to construct the trans-fused [6,7] bicycle as well as install the quaternary carbons at C10 and C14 with the desired stereo-chemistry; 2) a Claisen rearrangement followed by a ring-closing metathesis to forge the strained nine-membered ring; and 3) a substrate-controlled Michael addition to enable the introduction of the C17 side-chain with good diastereoselectivity.

The results of our synthesis suggest that euvesperin A may be a mixture of the (2 R ,3 R ,4 S ,7 S ) and (2 S ,3 S ,4 R ,7 S ) isomers and euvesperin B may be a mixture of the (2 R ,3 S ,4 S ,7 S ) and (2 S ,3 R ,4 R ,7 S ) isomers in consideration of their putative biosynthetic pathways.

Abstract:An enantioselective total synthesis of the alleged structure of (+)-fimbricalyxoid A is reported. The synthetic strategy features a pyridine-N-oxidate-mediated SN2' reaction to introduce an oxygen functionality at position C3 of the A-ring and a sequential three-step process via the cleavage of the C-O bonds and hemiketalization to form the 3,20-oxybridge. With this strategy, the target molecule was synthesized in 19% overall yield and 12 steps from our previously synthesized cis-fused octahydrophenanthrene (+)-6.

Abstract:The asymmetric total synthesis of (-)-retigeranic acid A was described, which relies on a crucial reductive skeletal rearrangement cascade for the controllable assembly of diverse angular triquinane subunits. Taken together with an intramolecular Michael/aldol cyclization, an ODI-[5 + 2] cycloaddition/pinacol rearrangement cascade, a Wolff ring contraction and a stereoselective HAT reduction, our synthetic approach has enabled the access to (-)-retigeranic acid A in a concise and practical manner.

Abstract:Biogenetically patterned total syntheses of the title alkaloids have been realised while a related synthesis of the racemic modification of the structure assigned to asperversiamide A suggests this is incorrect. The efficient oxidative rearrangement of (±)-asperversiamide D into the C8a epimer of asperversiamide C suggests this spiro-oxindole is likely to be a natural product-in-waiting as is ent-asperversiamide F, the co-product from the Stryker-type reduction of asperversiamide G.

Abstract:A biomimetic approach has been investigated and developed for the total synthesis of azonazine, an unusual marine natural cyclopeptide containing a rigid transannular 10-membered ring. A hypervalent iodine-mediated direct oxidative cyclization was successfully developed and applied to construct the highly strained core, which was the key step in the first total synthesis of ent-(-)-azonazine. Based on the physical evidences of synthesized diastereomer and enantiomer of azonazine, both the relative and absolute configurations of the natural product were revised. Two fluorinated azonazine derivatives were also synthesized in short convenient steps utilizing the same intermediate in this work. The established total synthesis opens a potential opportunity to study the structure activity relationship of natural azonazine. (c) 2014 Elsevier Ltd. All rights reserved.

Abstract:The efficient total synthesis of cudratricusxanthone B (1), a biologically interesting natural isoprenylated xanthone, has been achieved for the first time starting from commercially available 2,4-dihydroxybenzoic acid via a linear reaction sequence of 8 steps with the overall yield of 3.1%, wherein Claisen rearrangement and demethylation with AlCl3/pyridine are used as key reactions. This work definitely laid the foundation for the further pharmacological study of this natural compound. Meanwhile, another xanthone (2) has been synthesized, which proved the reported structure of the natural xanthone staud-tiixanthone D to be wrong.

Abstract:Concise total syntheses of smenothiazoles A (1) and B (2), two distinguished vinyl chloride containing natural products isolated from the marine sponge S. aurea, have been developed. Silastannation, Stille reaction and a carefully controlled desilylchlorination were employed as key steps to construct unique polyketide acid fragments, and the optimized reaction conditions avoided migration of 2,5-diene to a 2,4-conjugated system. Abstract:Based on the structural features of obyanamide, the target molecule was disconnected into four fragments by applying retrosynthetic analysis method, i.e., N-Me-Phe-N-Me-Val (fragment A), thiazole carboxy acid derivative (fragment B), lactic acid derivative (fragment Q and unnatural 3-aminopentanoic acid (fragment D). The total synthesis of a natural marine cytotoxic cyclic depsipeptide obyanamide was accomplished successfully from synthesized the fragments A, B, C and D. The structures of the synthesized intermediates and product was identified by H-1 NMR, C-13 NMR, HMBC and HRMS (FAB) spectra.

Abstract:Recently, several studies on the chemical synthesis of Brevianamide A (BA) were reported. Especially, a highly efficient and remarkably selective synthetic strategy was reported by Lawrence's group. However, a unified mechanistic understanding of these results is still lacking. We have carried out a DFT study, and proposed a unified mechanism to understand these experimental results. Starting from intermediate 2, the most favorable reaction sequence is a fast tautomerization, followed by a σ-migration of the base moeity, and a final inverse-electron demanding Diels–Alder reaction to result in the formation of BA product stereoselectively. This reaction mechanism can also be applied to understand the biosynthesis of BA that involves enzymatic catalysis.
