Abstract:We have achieved the first total synthesis of pallamolides A-E, of which pallamolides B-E possess intriguing tetracyclic skeletons with novel intramolecular transesterifications. Key transformations include highly diastereoselective sequential Michael additions to construct the bicyclo[2.2.2]octane core with simultaneous generation of two quaternary carbon centers, a one-pot SmI2-mediated intramolecular ketyl-enoate cyclization/ketone reduction to generate the key oxabicyclo[3.3.1]nonane moiety, and an acid-mediated deprotection/oxa-Michael addition/β-hydroxy elimination cascade sequence to assemble pallamolides tetracyclic skeletons. Kinetic resolution of ketone 14 via Corey-Bakshi-Shibata reduction enabled the asymmetric synthesis of pallamolides A-E.

Abstract:Main observation and conclusionA full account of the total synthesis of (–)‐pallambins A—D (1—6) is described. The strategy was devised by simulating their biosynthetic pathway. The left‐part bicyclo[3.2.1]octane system of pallambins C and D was efficiently constructed via a palladium‐ catalyzed oxidative cyclization. For construction of the right‐part tetrahydrofuran/γ‐lactone moiety (C/D rings), initial attempts to synthesize the allylic alcohol 15 for an one‐step Pd‐mediated alkoxycarbonylation have failed. However, during the course of this work, an unprecedented CH3Li‐mediated method for conversion of bromoisoxazoline to the corresponding β‐hydroxy nitrile has been discovered. Furthermore, a stepwise protocol was designed, namely an Eschenmoser‐Claisen rearrangement/Lactonization to generate the C ring, and a non‐classical Wittig reaction to form the D ring. During the course of this work, a palladium‐catalyzed method for dehydrobromination of bromide ketone was developed. Finally, an individual transformation of pallambins C (3) and D (4) generated pallambins A (5) and B (6) under mild UV irradiation. Thus, the first enantioselective total syntheses of (–)‐pallambins A—D have been achieved in 15 or 16 steps from the known chiral cyclohexenone 8. The described synthesis avoids protecting‐group manipulations by designing highly chemo‐ and stereoselective transformations.

Abstract:In this study, a concise and stereoselective approach for the divergent total synthesis of (-)-indoxamycins A-F is described. The key steps of the strategy include an Ireland-Claisen rearrangement, an enantioselective 1,6-enyne reductive cyclization, and a tandem 1,2-addition/oxa-Michael/methylenation reaction. The relative and absolute configuration of these natural products has been unambiguously elucidated, and their cytotoxic activities against HT-29 and A-549 tumor cell lines are also reported.

Abstract:Daphnezomines A and B are structurally unusual Daphniphyllum alkaloids that contain a unique aza-adamantane core skeleton. Herein, a modular approach to these alkaloids is presented that exploits a diverse array of reaction strategies. Commencing from a chiral pool terpene–(S)-carvone, the azabicyclo[3.3.1]nonane backbone, which occurs widely in Daphniphyllum alkaloids, was easily accessed through a Sharpless allylic amination and a palladium-catalyzed oxidative cyclization. A protecting group enabled a stereoselective B-alkyl Suzuki–Miyaura coupling sequence and an Fe-mediated hydrogen atom transfer (HAT)-based radical cyclization were then applied to construct C6 and C8 stereocenters. Abstract:(-)-Daphnilongeranin B and (-)-daphenylline are two hexacyclic Daphniphyllum alkaloids, each containing a complex cagelike backbone. Described herein are the first asymmetric total synthesis of (-)-daphnilongeranin B and a bioinspired synthesis of (-)-daphenylline with an unusual E ring embedded in a cagelike framework. The key features include an intermolecular [3+2] cycloaddition, a late-stage aldol cyclization to install the F ring of daphnilongeranin B, and a bioinspired cationic rearrangement leading to the tetrasubstituted benzene ring of daphenylline.

Abstract:Concise total syntheses of smenothiazoles A (1) and B (2), two distinguished vinyl chloride containing natural products isolated from the marine sponge S. aurea, have been developed. Silastannation, Stille reaction and a carefully controlled desilylchlorination were employed as key steps to construct unique polyketide acid fragments, and the optimized reaction conditions avoided migration of 2,5-diene to a 2,4-conjugated system. This report unambiguously confirmed the structures of both natural products.

Abstract:The first total syntheses of cyclic depsipeptides colletopeptide A and colletotrichamide A, have been accomplished. The key advanced intermediate, a cyclic tridepsipeptide derivative, was constructed using a sequence of transformations that features asymmetric Brown crotylation, cross metathesis, Yamaguchi esterification, ozonolysis, and macrolactamization. A late-stage incorporation of the mannose fragment completed the synthesis of colletotrichamide A, and the desilylation of the common intermediate gave rise to colletopeptide A, which led to unambiguous confirmation of the absolute stereochemistry of the aforementioned natural products.

Abstract:The structural puzzle of amipurimycin, a peptidyl nucleoside antibiotic, is solved by total synthesis and X-ray diffraction analysis, with the originally proposed configurations at C3' and C8' inverted and those at C6', C2'', and C3'' corrected. A similar structural revision of the relevant miharamycins is proposed via chemical transformations and then validated by X-ray diffraction analysis. The miharamycins bear an unusual trans-fused dioxabicyclo[4.3.0]nonane sugar scaffold, which was previously assigned as being in the cis configuration.

Abstract:A novel preparation of N-substituted pyrrole-2-carboxylates has been developed based upon 1,3-dipolar cycloaddition and a conventional hydrogenolysis. By using this method as the key step, total syntheses of natural alkaloids ( )-hanishin, ( )-longmide B, and ( )-longmide B methyl ester were accomplished in the highest overall yields, respectively. Pyrrole-2-carboxylic acid derivatives are a large family of natural alkaloids. Most brominated pyrrole-2-carboxamides were isolated from marine sponges, such as ( )-hanishin (1), ( )-longmide B (2), and ( )-longmide Bmethyl ester (3) (Figure 1). These alkaloids not only have a novel chiral dihydropyrrolo[1,2-a]pyrazin-1-one skeleton but also showed biologically important properties. For example, ( )-hanishin (1) was cytotoxic toward NSCLCN6 human nonsmall-cell-lung carcinoma (IC50 9.7 μg/mL); ( )-longmide B (2) displayed activity against African trypanosome (IC50 1.53 μ/mL); and (()-longmide B methyl ester (3) exhibited cytotoxic activity against P-388 lymphocytic leukemia cells (ED50 30 μg/mL), respectively. Therefore, they are highly attractive targets for the total synthesis. The first racemic total syntheses of 1 3 were accomplished a year after ( )-hanishin (1) was first isolated. Thereafter a number of enantioselective total syntheses were reported via chiral pool and asymmetric catalysis strategies. Abstract:The first total synthesis of incarnatapeptins A and B, two novel marine natural products, was accomplished from readily available (S)-1-benzyloxycarbonylhexahydropyridazine-3-carboxylic acid. This route, whose longest linear sequence was 12 steps, provided the incarnatapeptins A and B in yields of 26.5% and 19.7%, respectively, and enabled the structure and stereochemistry of both natural products to be unambiguously confirmed. Highlights of our synthesis include the photoredox-mediated decarboxylative 1,4-addition reaction and a novel and practical N-acylation paradigm promoted by silver carbonate. The unusual facile atropisomerism of some linear peptidic intermediates was also observed by TLC analysis in the course of this work.

Abstract:Huperserratines A (1) and B (2), two novel macrocyclic Lycopodium alkaloids, possess an aza-12-membered ring. Here, we describe the first total synthesis of these two natural products in 12 steps. Key elements of the synthesis include Suzuki-Miyaura coupling, Hosomi-Sakurai reaction, ring-closing metathesis, dihydroxylation, and Swern oxidation. A gram-scale ring-closing metathesis was pivotal in forging the challenging aza-12-membered ring. This total synthesis would enable future studies of the structure-activity relationship for 1 and 2.

Abstract:Enantioselective total syntheses of (+)-fawcettidine and (+)-lycoposerramine Q as well as the first total synthesis of (-)-lycopladine D from a common intermediate have been accomplished by a divergent path. The common intermediate was derived from a Hajos-Parrish-like diketone by a stereoselective Birch reduction and a Suzuki coupling. The synthesis of (-)-lycopladine D featured an allylic oxidation and a biomimetic aminoketalization while the route to (+)-fawcettidine and (+)-lycoposerramine Q highlighted an oxidative rearrangement.
