Abstract:A highly regio-and stereoselective Pd-catalyzed cascade reaction of allenoate adducts has been developed. Various allenoate adducts of phosphine-catalyzed addition of allenoates are found to be efficient substrates for the preparation of 2,3-dihydrobenzofuran. It is the first example of allenoate adducts used as ideal and practical precursors of hard-to-get functionalized 1,3-butadienes for heterocycle synthesis.

Abstract:A highly enantioselective [4+2] annulation with respect to allenoates and 3-olefinic oxindoles catalyzed by Lewis base was reported, which proved to be an efficient way to synthesize chiral dihydropyran-fused indoles. The cycloaddition products were generally obtained in high yields (up to 98%) with very good enantioselectivities (up to 94% enantiomeric excess).

Abstract:We have developed a methodology for the greatly efficient construction of significant 2,3-dihydrobenzofuran scaffolds bearing a quaternary carbon center at the C2 position by means of [4 + 1] annulation reactions between p-quinone methides and α-aryl diazoacetates as C1 synthons through organocatalysis by readily accessible TfOH catalyst under mild and transition metal-free conditions. This metal-free protocol furnishes an operationally simple and swift process for the free assembly of diverse highly functionalized 2,3-dihydrobenzofurans and also features broad substrate scope, excellent functional group compatibility, and environmental friendliness. Mechanistic investigation suggested that the reaction undergoes a rapid cascade protonation/intermolecular Michael addition/intramolecular substitution process.

Abstract:A highly diastereoselective [3 + 2] cycloaddition of aziridines with difluorinated silyl enol ethers has been developed. This approach provides a facile methodology for highly functionalized gem-difluorinated pyrrolidines in good to excellent yields with good functional group tolerance. A one-pot, two-step approach for synthesis of structurally interesting fluorinated pyrroles has also been developed through a cycloaddition/aromatization/desulfonation sequence. Moreover, readily available substrates, mild reaction conditions, and easy scale-up synthesis show practical advantages.

Abstract:A highly regio- and diastereoselective visible light-promoted [2 + 2] cycloaddition between readily available 1,4-dihydropyridines and olefins has been developed. This strategy is operationally simple and atom-economical and enables the construction of strained polysubstituted 2-azabicyclo[4.2.0]octanes with three all-carbon quaternary centers with good functional group tolerance. These products can be easily converted to various structurally unique derivatives. The primary mechanistic studies demonstrated that the reaction proceeds through an energy transfer pathway.

Abstract:The first enantioselective formal [5+4] cycloaddition is realized under palladium catalysis to deliver benzofuran-fused nine-membered rings. These medium-sized heterocycles and derivatives undergo unique rearrangements induced by transannular bond formation, resulting in the production of two classes of densely substituted polycyclic heterocycles in excellent efficiency and stereoselectivity.

Abstract:A highly enantioselective β-isocupreidine (β-ICD) catalyzed synthesis of dihydropyran-fused benzofurans through [4+2] cycloaddition of allenoates and benzofuranone alkenes was developed. Switchable chirality inversion of cycloaddition products was achieved by replacing the phenolic proton of the catalyst with a methyl, demonstrating an amazing effect of minimal structural variation on inverting enantioselectivity. DFT calculations were utilized to elucidate the origin of the observed phenomena. Computation also provided a clue for a rational design in which the multi-hydrogen bond with the alcohol additive was found to improve the enantioselectivity of the cycloaddition. Finally, the substrate scope was examined, in which a number of functionalized dihydropyran-fused benzofurans could be obtained in high yields (up to 97 %) with very good regio- (>20:1) and enantioselectivities (up to 98:2 e.r.).

Abstract:Highly enantio- and diastereoselective phosphine-catalyzed [3+2] cycloadditions of terminal allenoates with β-perfluoroalkyl α,β-enones leading to a range of trifluoromethylated cyclopentenes with two contiguous chiral centers (up to 99% yield with 99% ee) have been developed. Moreover, these reactions could be performed at the gram scale by using only 1 mol% catalyst. The method developed here was also applied to the concise synthesis of trifluoromethylated DGAT-1 inhibitor.

Various aryl allyl ethers derived from the phosphine-catalyzed addition of electron-deficient allenes with phenol are found to be efficient substrates for the synthesis of 2-substituted 2,3-dihydrobenzofurans and indolines. It is the first example of aryl allyl ether used as an ideal and practical precursor of hard to get functionalized 1,3-butadiene for the heterocyclic compound synthesis.

Abstract:The first highly enantioselective [3 + 2] formal cycloaddition of 1-styrylnaphthols (or phenol) with quinones catalyzed by a chiral phosphoric acid has been reported. A class of trans-2,3-diarylbenzofurans were prepared efficiently (up to 99% yield, >20:1 dr, 99% ee). This organocatalytic procedure allows lowering of the catalyst loading to 0.5 mol % without considerable loss in reactivity and enantioselectivity.

Abstract:An efficient ferrocene-derived bifunctional phosphine-catalyzed enantioselective oxa-[4+2] cycloaddition of -substituted allenones with a broad range of enones is investigated for the preparation of stereodefined dihydropyrans in good to excellent yields (up to 99%) and excellent enantioselectivity (up to 99% ee). Furthermore, a series of valuable chiral polyheterocyclic frameworks can be efficiently achieved in good yields with excellent enantioselectivities.

Abstract:The construction of benzopyrone‐fused hydrobenzofuranones via DABCO‐catalyzed [3+2] cyclization/deformylation cascade of p‐quinols with 3‐formylchromones is described. The reaction works under mild reaction conditions to provide the desired products in 53–90% yields with complete diastereoselectivities. In addition, an enantioselective version with 81% ee is also realized in the presence of Takemoto's chiral thiourea catalyst.

Abstract:A highly diastereo- and enantioselective phosphine-catalyzed sequential [3 + 2]/[3 + 2] annulation of allenoates with arylidenemalononitriles has been developed. This reaction allows for the facile construction of multifunctionalized <i>cis</i>-fused bicyclic[3,3,0]octene scaffolds, encompassing three consecutive stereogenic centers with one quaternary carbon center, in a one-step operation from readily available materials. The reported protocol is scalable, operates under mild reaction conditions, and creates the core structural motif of a number of natural products.

Abstract:The highly exo- and enantioselective gold-catalyzed tandem heterocyclization/[4 + 3] cycloaddition of 2-(1-alkynyl)-2-alken-1-ones and 1,3-diphenylisobenzofuran was implemented by utilizing Ming-Phos, which provides a facile access to chiral seven-membered oxa-bridged rings in 80–98% yield with high exo selectivity (exo/endo up to 50:1) and up to 97% ee.
