Abstract:In the past two decades, substantial advances have been made on the asymmetric alkyne functionalization by the activation of inert alkynes. However, these asymmetric transformations have so far been mostly limited to transition metal catalysis, and chiral Bronsted acid-catalyzed examples are rarely explored. Here, we report a chiral Bronsted acid-catalyzed dearomatization reaction of phenol- and indole-tethered homopropargyl amines, allowing the practical and atom-economical synthesis of a diverse array of valuable fused polycyclic enones and indolines bearing a chiral quaternary carbon stereocenter and two contiguous stereogenic centers in moderate to good yields with excellent diastereoselectivities and generally excellent enantioselectivities (up to >99% enantiomeric excess). This protocol demonstrates Bronsted acid-catalyzed asymmetric dearomatizations via vinylidene-quinone methides.

Abstract:The asymmetric B & uuml;chner reaction and related arene cyclopropanations represent one type of the powerful methods for enantioselective dearomatization. However, examples of asymmetric B & uuml;chner reactions via a non-diazo approach are quite scarce, and the related arene cyclopropanation based on alkynes has not been reported. Herein, we disclose an asymmetric B & uuml;chner reaction and the related arene cyclopropanation by copper-catalyzed controllable cyclization of N-propargyl ynamides via vinyl cation intermediates, leading to chiral tricycle-fused cycloheptatrienes and benzonorcaradienes in high yields and enantioselectivities. Importantly, this protocol represents an asymmetric arene cyclopropanation reaction of alkynes and an asymmetric B & uuml;chner reaction based on vinyl cations. Asymmetric B & uuml;chner reactions are an effective methodology to bring about synthetic complexity from the common benzene motif, but effective, general catalytic methods are scarce. Here, the authors present divergent asymmetric arene dearomatization from ynamides using copper catalysis, wherein the catalytic cycle proceeds via a putative vinyl cation intermediate.

Abstract:To study the intramolecular Diels-Alder cycloadditon of 2H-chromen-2-one as a diene, a series of chiral N-allyl-N-benzylamides that contain a 2H-chromen-2-one moiety were designed for the synthesis of benzo[f]isoindol-1-ones via an intramolecular Diels-Alder and a subsequent retro-Diels-Alder cycloaddition with the expulsion of CO2. Both the yield (80%-89%) and absolute stereocontrol of the tandem reaction were high when an electron-withdrawing group was attached to the dienophile. The double bond in the styrene substructure remained in the products could be further derivatized by dihydroxylation. Abstract:The asymmetric cross-coupling reaction is developed as a straightforward strategy toward 1,1-diaryl alkanes, which are a key skeleton in a series of natural products and bioactive molecules in recent years. Here we report an enantioselective benzylic C(sp(3))-H bond arylation via photoredox/nickel dual catalysis. Sterically hindered chiral biimidazoline ligands are designed for this asymmetric cross-coupling reaction. Readily available alkyl benzenes and aryl bromides with various functional groups tolerance can be easily and directly transferred to useful chiral 1,1-diaryl alkanes including pharmaceutical intermediates and bioactive molecules. This reaction proceeds smoothly under mild conditions without the use of external redox reagents.

Abstract:An efficient enantioselective hetero-Diels-Alder reaction was developed under catalysis of a chiral copper complex. A variety of spiropyrrolidones, which bear a tetra-substituted carbon stereocenter, can be obtained in good yields with excellent enantioselectivities by virtue of this method. Abstract:The 3-substituted 3-hydroxybenzofuran-2-one unit is encountered in a large variety of natural products with a wide spectrum of biological activities. Therefore, development of new methodologies to the construction of chiral quaternary center at the 3-position of benzofuran-2-one compounds were highly desired. A highly enantioselective Aldol reaction of benzofuran-2,3-ones with ketone catalyzed by N-(2-thiophenesulfonyl)prolinamide through enamine activation strategy was developed. As a result, a number of chiral 3-hydroxyl substituted benzofuran-2-ones derivatives were synthesized with very good yields(up to 96%) and moderate to good enantioselectivities(up to 91% e.e.). This methodology substantially broadens the benzofuranone chemistry and offer functionalized chiral products.

Abstract:A multisubstituted tetrahydrofuran building block bearing three vicinal chiral carbon centers widely exists in a broad spectrum of bioactive natural products, and the development of efficient and convenient methods to establish this skeleton remains a challenging task. Herein, we have developed an efficient method for the construction of significant tetrahydrofuran scaffolds bearing three vicinal and α-quaternary chiral carbon stereocenters through Pd-catalyzed asymmetric [3 + 2] annulation of vinylethylene carbonates with alkenes installed on cyclic N-sulfonyl imines. A series of multisubstituted tetrahydrofuran derivatives are obtained in high efficiencies with excellent enantioselectivities and diastereoselectivities. Density functional theory (DFT) studies are accomplished to rationalize the stereocontrol of the annulation process and disclose that methanol could be applied to stabilize the reactive zwitterionic π-allylpalladium via the H-bond interaction.

Abstract:A novel asymmetric Pd-catalyzed decarboxylative [3 + 2] cycloaddition of vinylethylene carbonates and alpha,beta-unsaturated pyrazolones has been developed to afford highly functionalized tetrahydrofuran-fused spiro-heterocyclic skeletons with excellent stereoselectivity. A diverse range of enantioenriched spiro-tetrahydrofuran-pyrazolones, possessing three contiguous stereogenic centers including one all-carbon quaternary, were obtained in high yields and excellent enantioselectivities (up to 90% yield and 98% ee). In addition, the practical application of this protocol was also demonstrated by gram-scale synthesis and further transformations into various compounds. Preliminary mechanistic experiments indicated that the allylic 1, 3-strain was crucial to the stereoselective control. We developed an asymmetric Pd-catalyzed [3 + 2] cycloaddition of alpha,beta-unsaturated pyrazolones with vinylethylene carbonates. An array of spiro-tetrahydrofuran-pyrazolones were obtained in high yields with excellent enantioselectivities.

Abstract:A convenient method was developed via an NHC mediated three-component cascade reaction for the construction of the medicinally important spirobenzofuranone derivatives. The reaction features mild reaction conditions and generates three consecutive stereocenters, one of which is an all-carbon substitution. The protocol can accommodate various substituents and substitution patterns, afford the products in moderate to good yields with excellent diastereoselectivities and moderate enantioselectivity.

Abstract:Employing a chiral spirodiphosphine monoxide ligand with 1,1'-spirobiindane backbone (SDP(O)), a desymmetrization strategy of Pd-catalyzed intramolecular asymmetric aryl C-O coupling of 2-(2-halophenoxy)propane-1,3-diols, was developed. The SDP(O) ligand shows much better results than its SDP counterpart. The protocol provides an efficient and highly enantioselective method for the synthesis of 2-hydroxymethyl-1,4-benzodioxanes. Density functional theory studies provide a model that accounts for the origin of the enantioselectivity.

Abstract:The merger of photoredox and transition-metal catalysis has evolved as a robust platform in organic synthesis over the past decade. The stereoselective 1,4-functionalization of 1,3-enynes, a prevalent synthon in synthetic chemistry, could afford valuable chiral allene derivatives. However, tremendous efforts have been focused on the ionic reaction pathway. The radical-involved asymmetric 1,4-functionalization of 1,3-enynes remains a prominent challenge. Herein, we describe the asymmetric three-component 1,4-dialkylation of 1,3-enynes via dual photoredox and chromium catalysis to provide chiral allenols. This method features readily available starting materials, broad substrate scope, good functional group compatibility, high regioselectivity, and simultaneous control of axial and central chiralities. Mechanistic studies suggest that this reaction proceeds through a radical-involved redox-neutral pathway.

Abstract:A series of novel chiral C2-symmetric bis (oxazoline) ligands have been synthesized.The copper and magnesium complexes,prepared in situ from copper(Ⅱ)-triflate of magnesium triflate with the new enantiopure oxazoline ligands,were evaluated as chiral catalysts in the enantioselective Diels-Alder reaction of cyclopentadiene with N-crotenoyl-oxazolidin-2-one.Primary results showed that diastereoselectivity up to 94% and enantioselectivity up to 68% ee for endo products were observed respectively with these ligands.
