Abstract:A novel hydrogel suspension array, which possesses the joint advantages of quantum-dot-encoded technology, bioresponsive hydrogels, and photonic crystal sensors with full multiplexing label-free DNA detection capability is developed. The microcarriers of the suspension array are quantum-dot-tagged DNA-responsive hydrogel photonic beads. In the case of label-free DNA detection, specific hybridization of target DNA and the crosslinked single-stranded DNA in the hydrogel grid will cause hydrogel shrinking, which can be detected as a corresponding blue shift in the Bragg diffraction peak position of the beads that can be used for quantitatively estimating the amount of target DNA. The results of the label-free DNA detection show that the suspension array has high selectivity and sensitivity with a detection limit of 10(-9) M. This method has the potential to provide low cost, miniaturization, and simple and real-time monitoring of hybridization reaction platforms for detecting genetic variations and sequencing genes.

Abstract:Multiplex bioassays have drawn more and more attention for the development of novel analytical techniques. Herein, we used photonic crystal (PC) and surface enhanced Raman scattering (SERS) as two encoding elements in different modes for the dual encoding of multiplex bioassays. In practice, PC beads and SERS nanotags act as carriers and labels, respectively, for the multiplex detection of antigens in a sandwich format. Except for the amplified capacity by two encoding modes, we also demonstrated that fine stability, low background and high sensitivity were realized for the quantitative analysis of multiple analytes, which holds great promise in biomedical applications like protein biomarker analysis.

Abstract:Barcode particles have a demonstrated value for multiplexed high-throughput bioassays. Here, a novel photonic crystal (PhC) barcode is presented that consists of hollow colloidal nanospheres assembled through microfluidic droplet templates. Due to their gas-filled core, the resultant barcode particles not only show increased refractive index contrast, but also remain in suspension by adjusting the overall density of the PhC to match that of a detection solution. In addition, magnetic nanoparticles can be integrated to give the barcodes a magnetically controllable motion ability. The encoding ability of the barcodes is demonstrated in microRNA detection with high specificity and sensitivity, and the excellent features of the barcodes make them potentially very useful for biomedical applications.

In this point, different probe biomolecules could be immobilized to encoded carriers, which can be mixed and subjected to an assay simultaneously and then many binding events can be distinguished by their codes. Herein we summarize our work on photonic beads as novel encoded carriers of biomolecules in multiplex bioassays. We have successfully fabricated different kinds of encoded photonic beads with controlled size and monodispersity by microfluidic device. The beads with opal structure and inverse opal structure could be used in multiplex labeling detection and label-free detection of biomolecules, respectively. Abstract:Suspension array attracted much attention around the world because of its benefits in simultaneous analysis of multiple targets. Photonic crystal encoded beads array was a type of optical encoded suspension array technology. Microbead with opal structure, named as silica colloidal crystal beads (SCCBs), was the most frequently used carrier in immunoassay previously, while microbead with inverse opal structure was used in molecular imprinting assay or label-free detection. However, inverse opal structure makes microcarrier to have larger specific surface area; and such structure makes microcarrier to have transformable dimension. With both characteristics, inverse opal structure microbead is more competent for fluorescent immunoassay compared with SCCB. Here a solvent responsive photonic encoded microbead with inverse opal structure, also named as responsive breathing microbead, was prepared, and simultaneously integrated with a special designed microfluidic chip, its excellent performance in multiplex fluorescent immunoassay was presented.

Abstract:In this study, a multiplex detection system was proposed by integrating a localized surface plasmon resonance (LSPR) sensing array and parallel microfluidic channels. The LSPR sensing array was fabricated by nanoimprinting and gold sputter on a polycarbonate (PC) substrate. The polydimethylsiloxane (PDMS) microfluidic channels and PC LSPR sensing array were bound together through (3-aminopropyl)triethoxysilane (APTES) surface treatment and oxygen plasma treatment. The resonant spectrum of the LSPR sensing device was obtained by broadband white-light illumination and polarized wavelength measurements with a spectrometer. The sensitivity of the LSPR sensing device was measured using various ratios of glycerol to water solutions with different refractive indices. Multiplex detection was demonstrated using human immunoglobulin G (IgG), IgA, and IgM. The anti-IgG, anti-IgA, and anti-IgM were separately modified in each sensing region. Various concentrations of human IgG, IgA, and IgM were prepared to prove the concept that the parallel sensing device can be used to detect different targets.

Abstract:A microfluidic device was used to generate droplets of monodispersed silica nanoparticles in poly (ethylene glycol) diacrylates (PEG-DA), which were then polymerized into hydrogel beads by UV irradiation. The nanoparticles in the beads were assembled into ordered lattices and locked in the network of the hydrogel. Therefore, the beads showed photonic crystal features, whose reflection peaks were used as the coding elements of biomolecular carriers. And huge coding capacity could be gained by changing the volume fraction of the silica nanoparticles in the droplets. The beads provided homogeneous water surroundings so that some shortcomings in bioassay introduced by the presence of solid surface of carriers could be avoided. Their application in oligonucleotide detection demonstrated potential advantages of hydrogel beads as encoded carriers in multiplex biomolecular detection.

Abstract:Barcode particles have a demonstrated value for multiplexed high-throughput bioassays. Attempts to develop this technology tend to focus on the generation of featured barcodes both with a large number of identifications to increase the throughput and with novel functions to improve the assays. Here, we report a new class of barcodes that are composed of multiple photonic crystal or magnetic-tagged ethoxylated trimethylolpropane triacrylate (ETPTA) cores and polyethylene glycol (PEG) hydrogel shells. These barcodes are prepared by polymerizing microfluidic multiple double emulsions. As the photonic crystal cores possess distinct reflection peaks, our barcodes allow for a substantial number of coding levels for multiplexing applications. The hydrogel shells surrounding the barcodes enable the creation of three-dimensional scaffolds for immobilizing probes. Moreover, the presence of magnetism in the barcodes confers their controllable movement under magnetic fields, which can be used to significantly increase the sensitivity of the bioassays and to simplify the processing. These features make photonic crystal barcodes ideal for biomedical applications. Biological systems are often studied through the use of assays, in which different experimental parameters are varied systematically. One method uses so-called barcode particles to track different samples. However this approach is complicated by the difficulty of producing barcode systems that allow for a sufficient number of configurations. Zhongze Gu and colleagues from Southeast University in Nanjing, China, have now developed a barcode system based on photonic crystals. These crystals, whose color reflection can be controlled during fabrication, are prepared with the help of a microfluidic system that also enables the simultaneous incorporation of the biomolecules to be probed. The barcodes particles also contain a magnetic tagged component that allows them to be moved in a magnetic field for easier read-out. By combining several types of photonic crystals in one unit, up to 1.28 million marker combinations are possible. Barcodes that composed of multiple colloidal crystal or magnetic-tagged ETPTA cores and PEG hydrogel shells were developed by using microfluidics. Attempts to develop this technology are focusing on the generation of novel particles with a large number of identifications to increase the throughput of the assays. Here we report a new type of barcode particles with quantum dot (QD)-photonic crystal (PC) joint spectrum encoding. These barcode particles are simply prepared by in-situ polymerization of CdTe QDs-contained acrylamide pre-gel in the colloidal photonic crystal beads (PCBs). By encapsulating different wavelength-and-intensity QDs into the PCBs with various-reflection spectra, thousands of available barcode particles could be obtained. Moreover, by employing magnetic nanoparticles into the PCBs, the achieved barcode particles could be conferred a function of controllable movement under magnetic field. This feature could significantly increase the sensitivity and simplify the processing of the bioassays.

Abstract:We present a handheld biosensor system for the label-free and specific multiplexed detection of several biomarkers employing a spectrometer-free imaging measurement system. A photonic crystal surface functionalized with multiple specific ligands forms the optical transducer. The photonic crystal slab is fabricated on a glass substrate by replicating a periodic grating master stamp with a period of 370 nm into a photoresist via nanoimprint lithography and deposition of a 70-nm titanium dioxide layer. Capture molecules are coupled covalently and drop-wise to the photonic crystal surface. With a simple camera and imaging optics the surface-normal transmission is detected. In the transmission spectrum guided-mode resonances are observed that shift due to protein binding. This shift is observed as an intensity change in the green color channel of the camera. Non-functionalized image sections are used for continuous elimination of background drift. In a first experiment we demonstrate the specific and time-resolved detection of 90.0 nm CD40 ligand antibody, 90.0 nM EGF antibody, and 500 nM streptavidin in parallel on one sensor chip. In a second experiment, aptamers with two different spacer lengths are used as receptor. The binding kinetics with association and dissociation of 250 nM thrombin and regeneration of the sensor surface with acidic tris-HCl-buffer (pH 5.0) is presented for two measurement cycles.

Abstract:Photonic crystals have wide applications not only in optoelectronic fields but also in bioassays. In this review, we summarized our work on colloidal photonic crystals as novel biomolecular supports in multiplex bioassays. Except for enhancing the fluorescence signal and encoding the biomolecular carriers in fluorphore labeled bioassays, photonic crystal can also encode suspended arrays. From the point view of practicality, the synthesis and self-assembly of monodispersed colloidal spheres for colloidal crystals is involved.

Abstract:A novel photonic suspension array was developed for multiplex immunoassay. The carries of this array were silica colloidal crystal beads (SCCBs). The codes of these carriers are the characteristic reflection peak originated from their structural periodicity, and therefore they do not suffer from fading, bleaching, quenching, and chemical instability. In addition, because no dyes or materials related with fluorescence are included, the fluorescence background of SCCBs is very low. With a sandwich format, the proposed suspension array was used for simultaneous multiplex detection of tumor markers in one test tube. The results showed that the four tumor markers, α-fetoprotein (AFP), carcinoembryonic antigen (CEA), carcinoma antigen 125 (CA 125) and carcinoma antigen 19-9 (CA 19-9) could be assayed in the ranges of 1.0–500ngmL−1, 1.0–500ngmL−1, 1.0–500UmL−1 and 3.0–500UmL−1 with limits of detection of 0.68ngmL−1, 0.95ngmL−1, 0.99UmL−1 and 2.30UmL−1 at 3σ, respectively. The proposed array showed acceptable accuracy, detection reproducibility, storage stability and the results obtained were in acceptable agreement with those from parallel single-analyte test of practical clinical sera. This technique provides a new strategy for low cost, automated, and simultaneous multiplex immunoassay.

Abstract:We demonstrate a label-free biosensor imaging approach that utilizes a photonic crystal (PC) surface to detect surface attachment of individual dielectric and metal nanoparticles through measurement of localized shifts in the resonant wavelength and resonant reflection magnitude from the PC. Using a microscopy-based approach to scan the PC resonant reflection properties with 0.6 μm spatial resolution, we show that metal nanoparticles attached to the biosensor surface with strong absorption at the resonant wavelength induce a highly localized reduction in reflection efficiency and are able to be detected by modulation of the resonant wavelength. Experimental demonstrations of single-nanoparticle imaging are supported by finite-difference time-domain computer simulations. The ability to image surface-adsorption of individual nanoparticles offers a route to single molecule biosensing, in which the particles can be functionalized with specific recognition molecules and utilized as tags.
