Abstract:Surface plasmon resonance (SPR) and localized SPR (LSPR) effects have been shown as the principles of some highlysensitive sensors in recent decades. Due to the advances in nano-fabrication technology, the plasmon nano-array sensors based on SPR and LSPR phenomena have been widely used in chemical and bioloical analysis. Sensing with surface-enhanced field and sensing for refractive index changes are able to identify the analytes quantitatively and qualitatively. With the newly developed ultrasensitive plasmonic biosensors, platforms with excellent performance have been built for various biomedical applications, including point-of-care diagnosis and personalized medicine. In addition, flexible integration of plasmonics nano-arrays and combining them with electrochemical sensing have significantly enlarged the application scenarios of the plasmonic nano-array sensors, as well as improved the sensing accuracy.

Abstract:The drive for a simultaneous analysis of multiple targets with excellent accuracy and efficiency, which is often required in both basic biomedical research and clinical applications, demands the development of multiplexed bioassays with desired throughput. With the development of nanotechnologies, innovative multiplex optical bioassays have been achieved. Nanomaterials exhibit unique physical and chemical properties such as easily tunable size, large surface-to-volume ratio, excellent catalysis and the desired signal transduction mechanism, which makes them excellent candidates for the fabrication of novel optical nanoprobes. This mini review summarizes nanomaterial-based optical multiplex sensors from the last 5 years. Specific optical techniques covered in this review are fluorescence, surface-enhanced Raman scattering (SERS), localized surface plasmon resonance (LSPR), chemiluminescence (CL), and the multimodality with fundamentals and examples.

Abstract:Multiplex bioassays have drawn more and more attention for the development of novel analytical techniques. Herein, we used photonic crystal (PC) and surface enhanced Raman scattering (SERS) as two encoding elements in different modes for the dual encoding of multiplex bioassays. In practice, PC beads and SERS nanotags act as carriers and labels, respectively, for the multiplex detection of antigens in a sandwich format. Except for the amplified capacity by two encoding modes, we also demonstrated that fine stability, low background and high sensitivity were realized for the quantitative analysis of multiple analytes, which holds great promise in biomedical applications like protein biomarker analysis.

Abstract:Since the colorimetric method has the characteristics of being simple and low cost, the fluorescence spectrum has the characteristics of a strong signal, and Surface-enhanced Raman scattering (SERS) detection has the characteristics of high sensitivity and strong specificity, people usually use these three methods for detection, but the detection of a single sample takes more time. If multiple samples can be tested at the same time, the detection efficiency and sensitivity can be improved, and the selectivity and reliability will be greatly improved. Multiplex sensing also provides a new direction for researchers. To fully understand the research of multiplex sensing based on the plasmonic optics of noble metal nanostructures, this review summarizes all the results previously reported in this field. It also discusses the principles of various detection methods and the biochemical application of multiple detections and finally summarizes the challenges and prospects.

Abstract:The advances of nanomaterials have provided exciting technologies and novel materials for protein detection, based on the unique properties associated with nanoscale phenomena such as plasmon resonance, catalysis and energy transfer. This article reviews a series of nanomaterials including nanoparticles, nanofibers, nanowires, and nanosheets, and evaluates their performances in the application for protein detection, focusing on approaches that realize ultrasensitive detection. Many of these nanomaterials were used to analyze clinically relevant protein biomarkers. Their detection in the picomolar, femtomolar or even zeptomolar regime has been realized, sometimes even with naked-eye readout. We summarize the detection methods and results according to materials and targets, review the current challenges, and discuss the solution in the context of technological integration such as combining nanomaterials with microfluidics, and classical analytical technologies.

Abstract:The drive to understand biology and medicine at the molecular level with accurate quantitation demands much of current high-throughput analysis systems. Nanomaterials and nanotechnology combined with modern instrumentation have the potential to address this emerging challenge. Using a variety of nanomaterials for multiplex diagnostics and imaging applications will offer sensitive, rapid and cost-effective solutions for the modern clinical laboratory. New nanomaterials have been developed with optical-encoding capabilities for selective tagging of a wide range of medically important targets, including bacteria, cancer cells and individual molecules, such as proteins and DNA, in a single assay. We envision further development in this field will provide numerous advanced tools with increased sensitivity and improved multiplexing capability, for unique applications in molecular biology, genomics and drug discovery.

Abstract:Localized surface plasmon resonance (LSPR) associated with metal nanostructures has developed into a highly useful sensor technique. Optical LSPR spectroscopy of nanostructures often shows sharp absorption and scattering peaks, which can be used to probe several bio-molecular interactions. Here, we report nanoplasmonic biosensors using LSPR on nanocup arrays (nanoCA) to recognize bio-molecular binding for biochemical detection. These sensors can be modified to quantify binding of small molecules to proteins for odorant and explosive detections. Electrochemical LSPR biosensors can also be designed by coupling electrochemistry and LSPR spectroscopy measurements. Multiple sensing information can be obtained and electrochemical LSPR property can be investigated for biosensors. In some applications, the electrochemical LSPR biosensor can be used to quantify immunoreactions and enzymatic activity. The biosensors exhibit better performance than those of conventional optical LSPR measurements. With multi-transducers, the nanoplasmonic biosensor can provide a promising approach for bio-detection in environmental monitoring, healthcare diagnostics, and food quality control.

Abstract:Photonic crystals (PhCs) and plasmonic nanostructures offer the unprecedented capability to control the interaction of light and biomolecules at the nanoscale. Based on PhC and plasmonic phenomena, a variety of analytical techniques have been demonstrated and successfully implemented in many fields, such as biological sciences, clinical diagnosis, drug discovery, and environmental monitoring. During the past decades, PhC and plasmonic technologies have progressed in parallel with their pros and cons. The merging of photonic crystals with plasmonics will significantly improve biosensor performances and enlarge the linear detection range of analytical targets. Here, we review the state-of-the-art biosensors that combine PhC and plasmonic nanomaterials for quantitative analysis. The optical mechanisms of PhCs, plasmonic crystals, and metal nanoparticles (NPs) are presented, along with their integration and potential applications. By explaining the optical coupling of photonic crystals and plasmonics, the review manifests how PhC-plasmonic hybrid biosensors can achieve the advantages, including high sensitivity, low cost, and short assay time as well. The review also discusses the challenges and future opportunities in this fascinating field.

Abstract:Multiplex assays have attracted considerable interest to meet the growing demand for clinical diagnosis, gene expression, drug discovery, and so on. Most of the assays are based on molecular binding or recognition events. In this point, different probe biomolecules could be immobilized to encoded carriers, which can be mixed and subjected to an assay simultaneously and then many binding events can be distinguished by their codes. Herein we summarize our work on photonic beads as novel encoded carriers of biomolecules in multiplex bioassays. We have successfully fabricated different kinds of encoded photonic beads with controlled size and monodispersity by microfluidic device. The beads with opal structure and inverse opal structure could be used in multiplex labeling detection and label-free detection of biomolecules, respectively. These photonic beads provide a new coding strategy of suspension array for low cost, sensitive and simultaneous multiplex bioassay.
Multispectral Spatial and Frequency Selective Sensing with Ultra-Compact Cross-Shaped Antenna Plasmonic Crystals

Abstract:We predict a novel multispectral spatial and frequency selective sensing based on a plasmonic sub-diffraction-limited (<λ/4) nanostructure. Via combining the strong plasmon near-field coupling effects, particle plasmon resonances (PPRs) and their hybridization by the metallic cross-shaped antennas (CSAs), a total of seven resonances with the minimum bandwidth<7nm are obtained in the visible and near-infrared region. Remarkably distinct biosensing behaviors at different spatial locations and different resonant wavelengths are simultaneously achieved, suggesting a new impressive sensing motif. High-quality biosensing with the maximal sensitivity (S=1134nm/RIU), figure of merit (FoM∼71.4), and high contrast ratio of spectral intensity difference (ΔR=44.2%) can be attained by detection a slight refractive index change of a thin biomolecular layer. These unique features of the proposed sensing motif could provide a powerful approach to develop desirable plasmon sensors with simultaneous multispectral spatial and frequency selective sensing, and hold potential applications in the high-integrated components for the high-performance plasmonic biosensing, detection and imaging.

Abstract:Simultaneous detection of different biomarkers from a single specimen in a single test, allowing more rapid, efficient, and low-cost analysis, is of great significance for accurate diagnosis of disease and efficient monitoring of therapy. Recently, developments in microfabrication and nanotechnology have advanced the integration of nanomaterials in microfluidic devices toward multiplex assays of biomarkers, combining both the advantages of microfluidics and the unique properties of nanomaterials. In this review, we focus on the state of the art in multiplexed detection of biomarkers based on nanomaterial-assisted microfluidics. Following an overview of the typical microfluidic analytical techniques and the most commonly used nanomaterials for biochemistry analysis, we highlight in detail the nanomaterial-assisted microfluidic strategies for different biomarkers. These highly integrated platforms with minimum sample consumption, high sensitivity and specificity, low detection limit, enhanced signals, and reduced detection time have been extensively applied in various domains and show great potential in future point-of-care testing and clinical diagnostics.

Abstract:A microfluidic chip integrated with pneumatically controlled valves was developed for multiplexed biomolecular detection via localized surface plasmonic resonance (LSPR) of single gold nanorod. The cost-effective microfluidic chip was assembled by polydimethylsiloxane layers and glass substrates with a precisely controlled thickness. The thin and flat microfluidic chip fitted the narrow space of dark-field microscopy and enabled the recording of single-nanoparticle LSPR responses. Aptamer-antigen-antibody sandwiched detection scheme was utilized to enhance the LSPR responses for label-free biomolecular detection. This microfluidic chip successfully demonstrated the multiplexed detection of three independent analytes (PSA, IgE, and thrombin).

Abstract:Protein chips are widely used for high-throughput proteomic analysis, but to date, the low sensitivity and narrow dynamic range have limited their capabilities in diagnostics and proteomics. Here we present protein microarrays on a novel nanostructured, plasmonic gold film with near-infrared fluorescence enhancement of up to 100-fold, extending the dynamic range of protein detection by three orders of magnitude towards the fM regime. We employ plasmonic protein microarrays for the early detection of a cancer biomarker, carcinoembryonic antigen, in the sera of mice bearing a xenograft tumour model. Further, we demonstrate a multiplexed autoantigen array for human autoantibodies implicated in a range of autoimmune diseases with superior signal-to-noise ratios and broader dynamic range compared with commercial nitrocellulose and glass substrates. The high sensitivity, broad dynamic range and easy adaptability of plasmonic protein chips presents new opportunities in proteomic research and diagnostics applications.

Abstract:Technological innovation, cost effectiveness, and miniaturization are key factors that determine the commercial adaptability and sustainability of sensing platforms. Nanoplasmonic biosensors based on nanocup or nanohole arrays are attractive for the development of various miniaturized devices for clinical diagnostics, health management, and environmental monitoring. In this review, we discuss the latest trends in the engineering and development of nanoplasmonic sensors as biodiagnostic tools for the highly sensitive detection of chemical and biological analytes. We focused on studies that have explored flexible nanosurface plasmon resonance systems using a sample and scalable detection approach in an effort to highlight multiplexed measurements and portable point-of-care applications.

Abstract:Plasmonic nanoparticles are commonly used as optical transducers in sensing applications. The optical signals resulting from the interaction of analytes and plamsonic nanoparticles are influenced by surrounding physical structures where the nanoparticles are located. This paper proposes inverse opal photonic crystal hydrogel as 3D structure to improve Raman signals from plasmonic staining. By hybridization of the plasmonic nanoparticles and photonic crystal, surface-enhanced Raman spectroscopy (SERS) analysis of multiplexed protein is realized. It benefits the Raman analysis by providing high-density hot spots in 3D and extra enhancement of local electromagnetic field at the band edge of PhC with periodic refractive index distribution. The strong interaction of light and the hybrid 3D nanostructure offers new insights into plasmonic nanoparticle applications and biosensor design.

Abstract:The robust identification and quantification of various biomarkers is of utmost significance in clinical diagnostics and precision medicine. Fluorescent immunoassays are widely used and considered as a gold standard for biomarker detection due to their high specificity and accuracy. However, current commercial immunoassay tests suffer from limited detection sensitivity and complicated, labor-intensive operation procedures, making them impractical for point-of-care diagnosis, particularly in resource-limited regions. Recently, microfluidic immunoassay devices integrated with plasmonic nanostructures have emerged as a powerful tool for sensitive detection of biomarkers, addressing specific issues, such as integration schemes, easy operation, multiplexed detection, and sensitivity enhancement. In this paper, we provide a discussion on the recent advances in the plasmonic nanostructures integrated with microfluidic devices for fluorescent immunoassays. We shed light on the nanofabrication strategies and various fluidic designs for rapid, sensitive, and highly efficient sensing of antigens. Finally, we share our perspectives on the potential directions of these integrated devices for practical applications.

Abstract:We report a nano-plasmonic biosensor with integrated optoelectronic architecture developed on a miniaturized chip. With simple means, the nano-plasmonic sensor demonstrated the capability to record the selective and specific binding of proteins to the bioreceptors immobilized on gold nanoparticles arrays placed directly on a photoconductive graphene-Perovskite quantum dots hybrid channel. The distinct combination of the nano-plasmonic structure, proteins and hybrid two-dimensional materials endowed the photoelectronic detection of biotin with advantages of label-free, ultrahigh sensitivity and rapid speed.
