Abstract:An efficient and general protocol for the ortho-alkylation of pyridines via C-H addition to olefins has been developed, using cationic half-sandwich rare-earth catalysts, which provides an atom-economical method for the synthesis of alkylated pyridine derivatives. A wide range of pyridine and olefin substrates including α-olefins, styrenes, and conjugated dienes are compatible with the catalysts.

Abstract:Brønsted base catalyzed C-C bond formation reactions have been extensively utilized as reliable, efficient, and atom economical methods in organic synthesis. However, the electrophiles were mostly limited to polar ones such as imines, carbonyl compounds, α,β-unsaturated compounds, styrenes and conjugated dienes. The use of α-alkenes as electrophiles in the C-C bond formation reactions always needs transition metal catalysts. Herein, we reported an alkyl lithium-catalyzed benzylic C-H bond addition of alkyl pyridines to α-alkenes. The alkyl lithium catalyst displayed quite different selectivity from those of transition metal catalysts.

Abstract:The benzylic functionalization of alkylpyridines is an important pathway for pyridine derivatives synthesis. The reaction partners, however, were mostly limited to highly reactive polar electrophiles. Herein, we report a potassium amide-catalyzed selective benzylic C-H bond addition of alkylpyridines to styrenes. Potassium bis(trimethylsilyl)amide (KHMDS), a readily available Brønsted base, showed excellent catalytic activity and chemoselectivity. A series of alkylpyridine derivatives, including benzylic quaternary carbon substituted pyridines, were obtained in good to high yield. Preliminary mechanistic studies revealed that the deprotonation equilibrium is probably responsible for the excellent selectivity.

Abstract:We report herein an iridium-catalyzed activation of pyridine C-H bond and its nucleophilic addition to benzaldehydes. The reaction was applicable to various pyridine derivatives and aryl aldehydes. In contrast to most methods that selectively functionalize the C2 pyridine C-H bond, this reaction proceeded with unusual meta-selectivity. An independently synthesized silyl iridium complex was able to catalyze the reaction, indicating that a silyl iridium intermediate might be responsible for the novel reactivity and the unusual selectivity.

Abstract:The catalytic C–H bond addition of alkylpyridines to olefins represents one of the most efficient approaches to alkyl-substituted pyridine derivatives with 100% atom-economy efficiency. Various catalysts including late transition metal, early transition metal, and s-block metal complexes displayed rich and versatile activity. Potassium amide could selectively achieve the benzylic alkylation of alkylpyridines, displaying distinct activity and regioselectivity. Here, the mechanistic investigation via both kinetic experiments and DFT calculations revealed the dimer structure of potassium amide and confirmed alkylation via a kinetic deprotonative functionalization process.

Abstract:Co(III)-catalyzed highly regio- and stereoselective direct C6 olefination of 2-pyridones with alkynes has been developed with the assistance of chelation. Upon variation of the reaction conditions, 2-pyridones react well with diaryl alkynes via a C6 olefination/directing group migration pathway to give the tetrasubstituted 6-vinyl-2-pyridones, but the C6–H olefination with terminal alkynes works effectively to afford only the C6-olefinated 2-pyridones. A judicious choice of a solvent and an additive is crucial for catalysis. The protocols feature 100% atom economy, excellent site selectivity, high stereoselectivity, an ample substrate scope, and good compatibility of functional groups. Abstract:An oxidant promoted 1,3-dipolar cycloaddition of benzimidazolium ylides to alkenes was developed for the preparation of 4H-pyrrolo[1,2-a]benzimidazole derivatives in moderate yields under mild conditions. In the presence of a suitable oxidant, the most commercially available "normal" alkenes, instead of alkynes or "abnormal" alkenes, could be used as dipolarophiles successfully. Moreover, CrO3/Et3N has been proved to be a more effective dehydrogenating reagent than MnO2 or tetrakispyridine cobalt (II) dichromate (TPCD) in this procedure.

Abstract:The Authors report new pyrazole‐alkyl phosphine ligands enabling additive‐free, regio‐, and chemoselective C─H arylation of heterocycles, showing excellent α/β selectivity and unprecedented C─Cl over C─OTf chemoselectivity. Mechanistic studies, supported by experiments and density functional theory calculations, reveal that pyrazole phosphine ligands with optimal ring sizes lower the energy barrier of the reaction proceeding via a concerted metalation‐deprotonation pathway. A series of new pyrazole‐alkyl phosphine ligands with varying cycloalkyl ring sizes that enable additive‐free regio‐ and chemoselective C─H arylation of heterocycles are reported. Excellent α/β selectivity of various heterocycles such as benzo[b]thiophene, thiophene, furan, benzofuran, and thiazole can be achieved using these ligands, along with excellent chemoselectivity of C─Cl over C─OTf of chloroaryl triflates. Mechanistic studies supported by both experimental findings and density functional theory calculations indicate that the pyrazole phosphine ligands with optimal ring sizes allow the reaction to proceed with a lower energy barrier via a concerted metalation–deprotonation pathway.

Abstract:A highly efficient Ru(II)-catalyzed regio- and stereospecific hydro-oxycarbonylation of unsymmetrical internal alkynes bearing a difunctional 2-pyridyloxy directing group with carboxylic acids has been developed, which provides allylic (Z)-enol esters in good to excellent yields with a broad substrate scope under mild conditions. The difunctional directing group can be diversely derivatized, particularly undergoing allylic substitution with various nucleophiles to afford β-functionalized (Z)-enol esters without directing groups.

Abstract:We have developed a borane-catalyzed sequential addition of terminal alkynes to para-substituted phenols, which affords a wide range of ortho-propargylic alkylated phenols bearing congested quaternary carbons. Control experiments combined with DFT calculations suggest that the reaction undergoes a sequential phenol alkenylation/hydroalkynylation process. Further extension of this strategy to the construction of triaryl-substituted quaternary carbons demonstrates the broad utility of this method.

Abstract:[GRAPHICS] Simple heteroarenes such as pyrroles and indoles undergo addition reactions to C-C triple bonds in the presence of a catalytic amount of Pd(OAc)(2) under very mild conditions, affording cis-heteroarylalkenes in most cases. The cleavage of aromatic CH bonds is the possible rate-determining step in CH2Cl2, and the addition of heteroaromatic C-H bonds to CC triple bonds is in a trans-fashion.

Abstract:A highly enantioselective allylic C-H alkylation reaction of allylarenes with pyrazol-5-ones has been established by the cooperative catalysis of a chiral palladium complex and chiral Bronsted acid to afford a wide spectrum of functionalized chiral N-heterocycles with an all-carbon quaternary stereogenic center in high yields and with high levels of enantioselectivity (up to 96% ee), wherein the chiral ligand and phosphoric acid showed synergistic effect on the control of stereoselectivity. In addition, a palladium-catalyzed asymmetric allylic C-H alkylation of 1,4-pentadienes with pyrazol-5-ones has been realized to furnish highly functionalized pyrazol-5-ones in high enantioselectivities. In this case, the chiral ligand controls the stereoselectivity while the achiral Bronsted acid, 2-fluorobenzoic acid, turns out to be a better cocatalyst than the chiral phosphoric acid. The installation of electron-deficient substituents at 3,3'-positions of binaphthyl backbone of chiral phosphoramidites is actually beneficial to the allylic C-H oxidation due to their survival in the presence of quinone derivative oxidants. These allylic C-H alkylation reactions undergo smoothly under mild conditions and tolerate a wide range of substrates. The resultant highly functionalized chiral pyrazol-5-ones have been applied to the preparation of more structurally diverse heterocycles by classical transformations.
