Abstract:Background: Dopaminergic and serotonergic systems play crucial roles in the pathophysiology of schizophrenia and modulate response to antipsychotic treatment. However, previous studies of dopaminergic and serotonergic genes expression are sparse, and their results have been inconsistent. In this longitudinal study, we aim to investigate the expressions of Catechol-O-methyltransferase (COMT), serotonin 2A receptor (5-HTR2A), and serotonin transporter gene (SLC6A4) mRNA in first-episode antipsychotic-naïve schizophrenia and to test if these mRNA expressions are associated with cognitive deficits and treatment outcomes or not. Method: We measured COMT, 5-HTR2A, and SLC6A4 mRNA expressions in 45 drug-naive first-episode schizophrenia patients and 38 health controls at baseline, and repeated mRNA measurements in all patients at the 8-week follow up. Furthermore, we also assessed antipsychotic response and cognitive improvement after 8 weeks of risperidone monotherapy. Results: Patients were divided into responders (N = 20) and non-responders groups (N = 25) according to the Remission criteria of the Schizophrenia Working Group. Both patient groups have significantly higher COMT mRNA expression and lower SLC6A4 mRNA expression when compared with healthy controls. Interestingly, responder patients have significantly higher levels of COMT and 5-HTR2A mRNA expressions than non-responder patients at baseline. However, antipsychotic treatment has no significant effect on the expressions of COMT, 5-HTR2A, and SLC6A4 mRNA over 8-week follow up. Conclusion: Our findings suggest that dysregulated COMT and SLC6A4 mRNA expressions may implicate in the pathophysiology of schizophrenia, and that COMT and 5-HTR2A mRNA may be potential biomarkers to predict antipsychotic response.

Abstract:Schizophrenia is a common disease with complex mode of inheritance; great efforts have been made to identify the susceptible genes. Catechol-O-methyltransferase (COMT) gene has long been considered as a candidate gene mainly because of two reasons: First, it encodes a key dopamine catabolic enzyme. Second, it maps to the velocardiofacial syndrome (VCFS) region of chromosome 22q11, which is associated with schizophrenia predisposition. Numerous case-control and family-based studies have been conducted, majority of them focused on a functional Val/Met polymorphism (rs4680). Unfortunately, these studies have produced conflicting results. In a previous report, Shifman et al. found a three-marker haplotype (rs737865-rs4680-rs165599) that showed significant association with schizophrenia. Abstract:AIM:To investigate the correlation between the polymorphism of four candidate genes involved in dopaminergic neurotransmission- COMT, DBH, DAT, MAOB and the susceptibility of Shanghai Chinese Han population to Parkinson's disease(PD).METHODS: Genetic polymorthism of COMT, DBH, DAT and MAOB were investigated by PCR/RFLP in 144 unrelated idiopathic PD and 188 age- matched healthy controls. RESULTS: Compared with controls, the polymorphic distribution in DBH gene intron TaqI polymorphism in PD cases was significant differences . CONCLUSION: The polymorphism in DBH gene might play a role in the susceptibility of Shanghai Chinese Han population.

Abstract:Catechol-O-methyltransferase (COMT), an enzyme involved in the degradation and inactivation of the neurotransmitter dopamine, is associated with the sensory gating phenomenon, protecting the cerebral cortex from information overload. The COMT Val(108/158)Met polymorphism is essential for prefrontal cortex processing capacity and efficiency. The current study was designed to investigate the role of COMT Val(108/158)Met polymorphism in development, sensory gating deficit, and symptoms of schizophrenia in Han Chinese population. P50 gating was determined in 139 schizophrenic patients and 165 healthy controls. Positive and Negative Syndrome Scale (PANSS) was used to assess the clinical symptomatology in 370 schizophrenic subjects. COMT Val(108/158)Met polymorphism was genotyped by PCR-restriction fragment length polymorphism (PCR-RFLP). No significant differences in COMT allele and genotype distributions were observed between schizophrenic patients and control groups. Although P50 deficits were present in patients, there was no evidence for an association between COMT Val(108/158)Met polymorphism and the P50 biomarker. Moreover, PANSS negative subscore was significantly higher in Val allele carriers than in Met/Met individuals. The present findings suggest that COMT Val(108/158)Met polymorphism may not contribute to the risk of schizophrenia and to the P50 deficits, but may contribute to the negative symptoms of schizophrenia among Han Chinese.

Abstract:Several lines of evidence pointed to a locus for psychosis near the COMT gene on chromosome 22q11. We previously reported linkage disequilibrium between Chinese schizophrenic subjects and a Val158Met polymorphism in exon 4 of the COMT gene [Li et al., 1996]. We have now genotyped an additional four biallelic polymorphic markers (promoter/Hind III, exon 3/Pml I, exon 4/Bcl I, and exon 6/Bgl I) in 200 family trios, consisting of Han Chinese schizophrenic subjects and their parents. These five polymorphic markers (including of Val158Met) were in strong linkage disequilibrium with each other (P value from 0.0001 to 0) except between exon 4/Bcl I and exon6/Bgl I (P = 0.77). The data were analysed using the transmission disequilibrium test (TDT) for individual polymorphism markers and haplotypes between paired markers. We found evidence of linkage disequilibrium for one polymorphism alone in exon 3/Pml I (P = 0.02, two tailed), but others were not significant. When we used haplotype of pairs of markers for TDT analysis (ETDT2 program, David Curtis and Pak Sham), the results gave much stronger evidence of linkage disequilibrium between the COMT gene and schizophrenia. Seven haplotypes out of ten analysed and gave P value less than 0.05 (from 0.0006 to 0.036 for allele-wise). P values were 0.0006 for allele-wise and 0.004 for genotype-wise when using haplotype of Val158Met and exon 6/Bgl I for TDT analysis. Our results support the hypothesis that COMT is a susceptibility gene, or this region of chromosome 22 contains a susceptibility gene which is in linkage disequilibrium with the COMT gene.

Abstract:Aims: Genetics play a major role in the etiology of schizophrenia (SZ). Catechol-O-methyltransferase (COMT) is one of the promising candidate genes for SZ. A nonsynonymous single-nucleotide polymorphism (SNP), rs4680, causing a Valine (Val) to Methionine (Met) substitution, has been widely studied in relation to psychiatric phenotypes, including SZ, but with conflicting results. We conducted a two-stage study to examine the association of COMT polymorphisms with SZ in the Han Chinese population. Results: Association analysis of nine SNPs in 768 patients and 1348 controls failed to detect any positive markers or haplotypes. Then, we tested rs4680 in a validation sample of 963 patients and 992 controls, and no significant association was observed, but the cases significantly deviated from Hardy-Weinberg equilibrium (p = 5.7e-4). There was no association of rs4680 with SZ in the combined sample (n = 4071, p = 0.110, odds ratio = 1.08). Conclusions: Our results do not support the association of COMT with SZ in the Han Chinese population.

Abstract:Tourette's syndrome is a complex and heritable behavioral disorder with early onset (2-15 years), prominently characterized by motor and vocal ticks affecting up to 1/100 boys and 1/700 girls. Several neurotransmitter receptor pathways and related biosynthetic enzymes have been implicated in its etiology through previous genetic and/or biological analysis. Using genotypes from functional polymorphisms in the catechol-O-methyltransferase gene COMT (Val158Met), dopamine D4 receptor gene DRD4 (exon III VNTR and a promoter SNP), dopamine D2 receptor gene DRD2 (promoter SNP), monoamine oxidase A MAOA (promoter CA repeat sequence) and serotonin receptor gene HTR2A (promoter SNP), we performed association analysis using the Quantitative Transmission Disequilibrium approach with scores for the Conners, CBCL and Depression scale for the affected children and three QTD Tests proposed by Abecasis et al., Rabinowitz et al., and Allison et al. Significant associations (<0.05) were found between the 5-HTR2A gene and the traits 'delinquent' (p=0.0147, 66 probands), 'thought' (p=0.0417, 66 probands) and 'extrovert' (p=0.0347, 66 probands); COMT gene with the "compulsive" trait (p=0.0458, 69 probands); and DRD4 VNTR with the 'bad behavior' trait (p=0.048). Using the method proposed by Fulker et al. (1999) a test for population stratification was performed on all trios and no significant evidence for population stratification was found. Therefore, an overall test for association was performed (i.e. not a TDT). Significant associations (0.01) were found between the DRD2 promoter and the traits 'depression mood' (p=0.01, 94 probands), 'low confidence' (p=0.002); DRD4 VNTR and traits 'bad behavior' (p < 0.001, 95 probands), 'group activity' (p=0.008, 95 probands); MAO polymorphism and traits 'bad behavior' (p=0.007, 38 probands), 'somatic' (p=0.0046, 56 probands), 'aggressive' (p<0.001, 56 probands), 'extrovert' (p<0.001, 56 probands).

Abstract:Abstract Background There are three childhood disruptive behavior disorders (DBDs), attention deficit hyperactivity disorder (ADHD), oppositional defiant disorder (ODD), and conduct disorder (CD). The most common comorbid disorder in ADHD is ODD. DSM-IV describes three ADHD subtypes: predominantly inattentive type (ADHD-IA), predominantly hyperactive-impulsive type (ADHD-HI), and combined type (ADHD-C). Prior work suggests that specific candidate genes are associated with specific subtypes of ADHD in China. Our previous association studies between ADHD and functional polymorphisms of COMT and MAOA, consistently showed the low transcriptional activity alleles were preferentially transmitted to ADHD-IA boys. Thus, the goal of the present study is to test the hypothesis that COMT Val158Met and MAOA-uVNTR jointly contribute to the ODD phenotype among Chinese ADHD boys. Methods 171 Chinese boys between 6 and 17.5 years old (mean = 10.3, SD = 2.6) with complete COMT val158met and MAOA-uVNTR genotyping information were studied. We used logistic regression with genotypes as independent variables and the binary phenotype as the dependent variable. We used p < 0.05 as the level of nominal statistical significance. Bonferroni correction procedures were used to adjust for multiple comparisons. Results Our results highlight the potential etiologic role of COMT in the ADHD with comorbid ODD and its predominately inattentive type in male Chinese subjects. ADHD with comorbid ODD was associated with homozygosity of the high-activity Val allele, while the predominantly inattentive ADHD subtype was associated with the low-activity Met allele. We found no evidence of association between the MAOA-uVNTR variant and ADHD with comorbid ODD or the ADHD-IA subtype. Conclusion Our study of attention deficit hyperactivity disorder comorbid oppositional defiant disorder and its predominately inattentive type highlights the potential etiologic role of COMT for ADHD children in China. But we failed to observe an interaction between COMT and MAOA, which suggests that epistasis between COMT and MAOA genes does not influence the phenotype of ADHD-IA with comorbid ODD in a clinical sample of Chinese male subjects. To confirm our findings further studies with a larger number of subjects and healthy controls are needed.

Abstract:OBJECTIVE:To detect the relationship between heroin dependence and catechol-O-methyltransferase (COMT) gene.METHODS:Genotype and allele frequencies of 108 val/met and 900 Ins C/Del C polymorphisms of COMT gene were examined in 313 heroin-dependent subjects and 214 normal controls.RESULTS:No differences in genotype and allele frequencies of 108 val/met polymorphism of COMT gene were observed between heroin-dependent subjects and normal controls (genotype-wise: chi-square=1.67, P=0.43; allele-wise: chi-square=1.23, P=0.27). No differences in genotype and allele frequencies of 900 Ins C/Del C polymorphism of COMT gene were observed between heroin-dependent subjects and normal controls (genotype-wise: chi-square=3.73, P=0.16; allele-wise: chi-square=0.76, P=0.38).CONCLUSION:The results suggested that neither 108 val/met polymorphism nor 900 Ins C/Del C polymorphism of COMT gene was associated with heroin dependence.

Abstract:Objective To investigate the association of catechol-O-methyltransferase (COMT) gene Vl108/158Met SNP (rs4680)with cognitive function in children with attention deficit hyperactivity disorder(ADHD) in China. Methods A total of 203 DSM-1V ADHD children of Chinese Hart descent were included, both the data of neuropsychological tests ( Wechsler Memory Scale, Stroop test and cancellationtest) and COMT Val108/158Met genotyping results were obtained. The two-sample t test and Mann-Whitney test were used to compare the average scores between two groups defined by COMT Val158Met genotypes,high-enzymatic activity ( ValVal, n = 92) and mid-low enzymatic activity ( ValMet and MetMet, n = 111 ).Results The children with high-enzymatic activity (ValVal) performed significantly better on some aspects of Wechsler Memory Scale and Stroop test ( picture recall, and part C error number in Stroop test) than those with mid-low enzymatic activity (P ＜0.05-0.01 ). No statistically significant differences were found between two groups and index in other tests. Conclusion The results suggest that COMT gene Val108/158Met polymorphism may be related to the cognitive functions as memory, response in hibition and attention in ADHD children in China.
Associations between variants in levodopa metabolic pathway genes and levodopa-induced dyskinesia in Parkinson’s disease

Abstract:IntroductionLevodopa-induced dyskinesia (LID) is a common motor complication in Parkinson’s disease (PD). Several genes in the levodopa metabolic pathway, such as COMT, DRDx and MAO-B, were reported associated with LID. However, there has been no systematic analyses between common variants in levodopa metabolic pathway genes and LID in a large sample of the Chinese population.MethodsThrough the whole exome sequencing (WES) and target region sequencing, we aimed to explore the potential associations between common single nucleotide polymorphisms (SNPs) in the levodopa metabolic pathway and LID in Chinese PD individuals. Five hundred and two PD individuals were enrolled in our study, among them, 348 individuals underwent WES, and 154 individuals underwent target region sequencing. We acquired the genetic profile of 11 genes including COMT, DDC, DRD1–5, SLC6A3, TH and MAO-A/B. We established a stepwise strategy to filter SNPs, which finally included 34 SNPs in our analyses. And we used a two-stage study, with discovery (348 individuals with WES) and the replication (all 502 individuals) to confirm our findings.ResultsAmong the 502 PD individuals, 104 (20.7%) were diagnosed with LID. In the discovery stage, we found that COMT rs6269, DRD2 rs6275 and DRD2 rs1076560 were associated with LID. In the replication stage, associations between the three above-mentioned SNPs and LID were still present in all 502 individuals.ConclusionWe demonstrated that in the Chinese population, COMT rs6269, DRD2 rs6275 and rs1076560 were significantly associated with LID. And rs6275 was reported associated with LID for the first time.

Abstract:OBJECTIVE:To investigate the association between attention-deficit hyperactivity disorder (ADHD) in Han Chinese children and Val158Met polymorphism of catechol-O-methyltransferase (COMT) gene caused by the missense mutation of G158A in exon 4.METHODS:By using polymerase chain reaction-restriction fragment length polymorphisms the Val158Met polymorphism of COMT gene was tested in 117 children with the diagnosis of ADHD as defined by DSM-IV and in 105 healthy controls living in Shanghai.RESULTS:The frequencies of A allele were 25.21% and 23.81% in the ADHD group and the health controls respectively, which showed no significant difference between the two groups (Chi2=0.5197, P>0.05). There was also no significant difference in the distribution of all genotypes of COMT gene between the ADHD patients and the controls (P>0.05).CONCLUSION:It was suggested that for the Han Chinese children with ADHD in this study, there was no association between ADHD and Val158Met polymorphism of COMT gene.

Abstract:The catechol-O-methyltransferase (COMT) gene is a schizophrenia susceptibility gene. A common functional polymorphism of this gene, Val158/158Met, has been proposed to influence gray matter volume (GMV). However, the effects of this polymorphism on cortical thickness/surface area in schizophrenic patients are less clear. In this study, we explored the relationship between the Val158Met polymorphism of the COMT gene and the GMV/cortical thickness/cortical surface area in 150 first-episode treatment-naïve patients with schizophrenia and 100 healthy controls. Main effects of diagnosis were found for GMV in the cerebellum and the visual, medial temporal, parietal, and middle frontal cortex. Patients with schizophrenia showed reduced GMVs in these regions. And main effects of genotype were detected for GMV in the left superior frontal gyrus. Moreover, a diagnosis × genotype interaction was found for the GMV of the left precuneus, and the effect of the COMT gene on GMV was due mainly to cortical thickness rather than cortical surface area. In addition, a pattern of increased GMV in the precuneus with increasing Met dose found in healthy controls was lost in patients with schizophrenia. These findings suggest that the COMTMet variant is associated with the disruption of dopaminergic influence on gray matter in schizophrenia, and the effect of the COMT gene on GMV in schizophrenia is mainly due to changes in cortical thickness rather than in cortical surface area.

Abstract:OBJECTIVE To detect the relationship between heroin-dependence and -287 A/G polymorphism of catechol-O-methyltransferase(COMT) gene. METHODS Genotype and allele frequencies of -287 A/G polymorphism of COMT gene were examined in 268 heroin-dependent subjects and 177 normal controls. RESULTS Weak but significant difference in genotype of -287 A/G polymorphism of COMT gene was observed between heroin-dependent subjects and controls (chi(2)=7.41, P=0.025), and genotype AA was higher in the former. The frequency of allele A of -287 A/G polymorphism of COMT gene was also significantly higher in heroin-dependent subjects than in the controls (chi(2)=5.69, P=0.017). CONCLUSION The results suggested that liability to heroin-dependence was associated with -287 A/G polymorphism of COMT gene.

Abstract:Background: Mitochondrial DNA polymerase gamma (POLG1) mutations were associated with levodopa-responsive Parkinsonism. POLG1 gene contains a number of common nonsynonymous SNPs and intronic regulatory SNPs which may have functional consequences. It is of great interest to discover polymorphisms variants associated with Parkinson's disease (PD), both in isolation and in combination with specific SNPs.Materials and Methods: We conducted a case-control study and genotyped twenty SNPs and poly-Q polymorphisms of POLG1 gene including in 344 Chinese sporadic PD patients and 154 healthy controls. All the polymorphisms of POLG1 we found in this study were sequenced by PCR products with dye terminator methods using an ABI-3100 sequencer. Hardy-Weinberg equilibrium and linkage disequilibrium (LD) for association between twenty POLG1 SNPs and PD were calculated using the program Haploview.Principal Results: We provided evidence for strong association of four intronic SNPs of the POLG1 gene (new report: c. 2070-12T>A and rs2307439: c.2070-64G>A in intron 11, P = 0.00011, OR = 1.727; rs2302084: c. 3105-11T>C and rs2246900: c.3105-36A>G in intron 19, P = 0.00031, OR = 1.648) with PD. However, we did not identify any significant association between ten exonic SNPs of POLG1 and PD. Linkage disequilibrium analysis indicated that c.2070-12T>A and c.2070-64G>A could be parsed into one block as Haplotype 1 as well as c.3105-11T>C and c.3105-36A>G in Haplotype 2. In addition, case and control study on association of POLG1 CAG repeat (poly-Q) alleles with PD showed a significant association (P = 0.03, OR = 2.16) of the non-10/11Q variants with PD. Although intronic SNPs associated with PD didn't influence POLG1 mRNA alternative splicing, there was a strong association of c.2070-12T>A and c.2070-64G>A with decreased POLG1 mRNA level and protein levels.Conclusions: Our findings indicate that POLG1 may play a role in the pathogenesis of PD in Chinese populations.
Association Between the COMT Val158Met Polymorphism and Antipsychotic Efficacy in Schizophrenia: an Updated Meta-Analysis.

Abstract:BACKGROUND:Catechol-O-methyltransferase (COMT) contributes to the control of synaptic dopamine (DA) transmission by catalyzing DA degradation in the presynaptic space. The COMT Val158Met polymorphism (rs4680) substantially alters enzymatic activity and consequently synaptic DA concentration in the prefrontal cortex and hippocampus. The COMT genotype could, therefore, exert a major influence on antipsychotic treatment response as many of these agents also target dopaminergic transmission.OBJECTIVE:The present meta-analysis aimed to test a putative relationship between the COMT Val158Met polymorphism and antipsychotic response across different populations and antipsychotic types.METHODS:Searches of PubMed, Web of Science, EMBASE, OVID, Google Scholar, and Baidu Scholar databases yielded 30 peer-reviewed studies published before January 2020 with a pooled total of 6291 participants. The Lipták-Stouffer Z score method for meta-analysis was applied to combine data. The Z score was also calculated separately for Caucasian and Asian subgroups.RESULTS:Pooled results indicated a highly significant association between COMT Val158Met and antipsychotic response (Z = 6.709, P = 9.8 × 10-12). Further, this relationship remained significant in subgroup analyses of Caucasian patients (Z = 3.180, P = 7.4 × 10-4) and Asian patients (Z = 4.487, P = 3.6 × 10-6).CONCLUSION:Pooled evidence supports the hypothesis that the COMT Val158Met polymorphism influences the antipsychotic response in Caucasian and Asian schizophrenia patient populations. Prediction of antipsychotic response by patient genotyping may warrant closer consideration in randomized clinical trials of efficacy.
Association study of DRD4 exonⅢ48 bp variable number of tandem repeats polymorphism and tic disorder

Abstract:Objective　This study was to investigate whether the dopamine receptor-4 gene (DRD4)exonⅢ48 bp variable number of tandem repeats (VNTRs) polymorphism was associated with tic disorder. Methods　178 patients with tic disoder who met with DSM-VI criteria for tic disorder or Tourette Syndrome and 209 normal controls were enrolled the study. According to whether combining with attention deficit hyperactivy disorder(ADHD) or not, the 178 patients were divided into tic disorder (TC) or combined subgroup. Polymerase chain reaction and VNTR technique were conducted.Results　No associations between alleles of DRD4 exonⅢ48 bpVNTR and tic disorder were found in the overall sample and the two subgroups （χ2=5.81, P=0.21; χ2=3.76, P=0.44; χ2=6.78, P=0.15) respectively. When the D4 VNTR was divided into “longer allele”（5-7 repeats） and “shorter allele”（2-4 repeats）, a significant excess of the longer allele was observed in the combined subgroup with odds ratio of 2.75 （χ2=5.55,P=0.02). Conclusions　The longer allele of DRD4 exonⅢ48 bpVNTR polymorphism is exclusively associated with tic disorder combined with ADHD, therefor it is possibly a genetic risk factor of tic disorder combined with ADHD in Chinese.
Association Analysis of Catechol-O-methyltransferase Gene Polymorphism and Adolescent Outcome of Attention Deficit Hyperactivity Disorder

Abstract:Objective:To examine the potential role of COMT gene Val158Met polymorphisms in the adolescent course of attention deficit hyperactivity disorder(ADHD).Methods:A total of 77 DSM-Ⅳ ADHD boys of Chinese Han descent were followed up over 3 to 5 years from childhood to adolescent and were administered structured psychiatric interviews to make DSM-Ⅳ diagnoses evaluated at an average age of 14years.The Chi-square test was used to compare COMT Val158Met allele frequencies between different types of the outcome in ADHD,such as the remitted children and non-remitted children.Results:Of the 77 total children in the sample,41(53.2%) showed no remission,36(46.8%) met criteria for remission:13(16.9%) met criteria for syndromatic remission,7(9.1%) met criteria for symptomatic remission,and 16(20.8%) met criteria for functional remission.No significant differences in allele frequencies of the Val158Met polymorphism of COMT were observed among any of the remitted or non-remitted subjects.Conclusion:The Val158Met SNP of COMT had no potential role on the adolescent course of ADHD.To confirm our findings further studies with a larger number of subjects are needed.
