Abstract:Objective: To discuss expression of E-cadherin, N-cadherin in rectal cancer and their relationship with clinical and pathological factors. Methods: Expression of N-cadherin and E-cadherin in 86 rectal cancer samples and normal rectal tissues was studied by immunohistochemical stain. Results: Both E-cadherin and N-cadherin expression were positive in 39 (45%) patients and negative in 47 (55%) patients. The positive rate of N-cadherin or negative rate of E-cadherin was positively related to the malignant degree, stage, invasion, metastasis and prognosis of the cancer .There was prominent significance (P ＜0.01) between the two groups; N-cadherin was not expressed in normal 86 rectal tissues, while E-cadherin was highly expressed. Conclusion: The abnormal expression of N-cadherin and low-level or non-expression of E-cadherin is an important factor for the invasion and metastasis of rectal cancer, it influences the 5-year survival rate. N and E -cadherin may act as indicators to the malignant degree and the prognosis of the rectal cancer.
E-cadherin Expression in Prostate Cancer and Its Correlation with PSA

Abstract:Objective To investigate the expression of E-cadherin in prostate cancer and its relationship with PSA. Methods E-cadherin expression of 56 prostate cancer samples were studied by immunohistochemical stain and its expression level was analyzed with respect to T-PSA, F-PSA and F/T ratio. Results 24 patients (43%) were normal and 32 patients (57%) were aberrant in E-cadherin expression. E-cadherin was related significantly to the grade and stage of cancer and the changes of F/T ratio.There were no significant relationship between E-cadherin expression and T-PSA or F-PSA. Conclusions E-cadherin expression may act as a marker to the malignant degree and the prognosis of the prostate cancer.
[Expression and clinical significance of Mucin and E-cadherin in colorectal tumors]

Abstract:Background & objective: Mucin (MUC), a glycoprotein with high molecular weight, can lubricate and protect the epithelium. E-cadherin (E-cad) is helpful in keeping the polarity and integrity of the epithelium. The abnormal expression of Mucin and E-cad is involved in the genesis of many tumors. This study was to investigate the expression of MUC1, MUC2, MUC5AC and E-cad in different colorectal tumor tissues, and explore their correlations to clinicopathologic features of colorectal cancer and the correlations of MUC1, MUC2, and MUC5AC expression to E-cad expression. Methods: The expression of MUC1, MUC2, MUC5AC and E-cad in 150 specimens of normal colorectal mucosa, 150 specimens of colorectal adenoma and 150 specimens of colorectal adenocarcinoma was detected by immunohistochemistry. Patients' survival was analyzed by Kaplan-Meier method. The correlations of MUC1, MUC2, and MUC5AC expression to E-cad expression were analyzed by spearman's rank correlation. Results: The positive rates of MUC1 were 0.07% in normal colorectal mucosa, 12.7% in colorectal adenoma, and 45.3% in colorectal adenocarcinoma. Those of MUC2 were 100%, 90.0% and 52.6%, respectively. Those of MUC5AC were 8.7%, 30.7% and 44.0%, respectively. Those of E-cad were 98.7%, 82.0% and 54.0%, respectively. In colorectal adenocarcinoma, the expression of MUC1 and MUC2 was correlated to tumor differentiation, invasion, lymph node metastasis and Dukes' stage (P<0.05); the expression of MUC5AC was correlated to tumor differentiation and invasion (P<0.01); the expression of E-cad was correlated to tumor differentiation (P<0.01). The 5-year survival rate was significantly higher in MUC1-negative group, MUC2-positive group and E-cadherin-positive group than in their counterparts (P<0.05). In colorectal adenocarcinoma, MUC1 expression was negatively correlated to E-cad expression (r=-0.234, P=0.004), MUC2 and MUC5AC expression were positively correlated to E-cad expression (r=0.170, P=0.038; r=0.198, P=0.015). Conclusions: In colorectal adenocarcinoma, MUC expression is obviously correlated to E-cad expression. The up-regulation of MUC1 and MUC5AC expression and the down-regulation of MUC2 and E-cad expression may be involved in the genesis of colorectal tumors and reflect the prognosis to a certain extent.
Downregulated E-Cadherin Expression Indicates Worse Prognosis in Asian Patients with Colorectal Cancer: Evidence from Meta-Analysis

Abstract:Background: Epithelial-mesenchymal transition (EMT) plays a crucial role in the progression and aggressiveness of colorectal carcinoma. E-cadherin is the best-characterized molecular marker of EMT, but its prognostic significance for patients with CRC remains inconclusive.Methodology: Eligible studies were searched from the PubMed, Embase and Web of Science databases. Correlation between E-cadherin expression and clinicopathological features and prognosis was analyzed. Subgroup analysis was also performed according to study location, number of patients, quality score of studies and cut-off value.Principal Findings: A total of 27 studies comprising 4244 cases met the inclusion criteria. Meta-analysis suggested that downregulated E-cadherin expression had an unfavorable impact on overall survival (OS) of CRC (n = 2730 in 14 studies; HR = 2.27, 95%CI: 1.63-3.17; Z = 4.83; P = 0.000). Subgroup analysis indicated that low E-cadherin expression was significantly associated with worse OS in Asian patients (n = 1054 in 9 studies; HR = 2.86, 95%CI: 2.13-3.7, Z = 7.11; P = 0.000) but not in European patients (n = 1552 in 4 studies; HR = 1.14, 95%CI: 0.95-1.35, Z = 1.39; P = 0.165). In addition, reduced E-cadherin expression indicated an unfavorable OS only when the cut off value of low E-cadherin expression was >50% (n = 512 in 4 studies; HR = 2.08, 95%CI 1.45-2.94, Z = 4.05; P = 0.000). Downregulated E-cadherin expression was greatly related with differentiation grade, Dukes' stages, lymphnode status and metastasis. The pooled OR was 0.36(95%CI: 0.19-0.7, Z = 3.03, P = 0.002), 0.34(95%CI: 0.21-0.55, Z = 6.61, P = 0.000), 0.49(95%CI: 0.32-0.74, Z = 3.02, P = 0.002) and 0.45(95%CI: 0.22-0.91, Z = 3.43, P = 0.001), respectively.Conclusions: This study showed that low or absent E-cadherin expression detected by immunohistochemistry served as a valuable prognostic factor of CRC. However, downregulated E-cadherin expression seemed to be associated with worse prognosis in Asian CRC patients but not in European CRC patients. Additionally, this meta-analysis suggested that the negative threshold of E-cadherin should be >50% when we detected its expression in the immunohistochemistry stain.

Abstract:OBJECTIVE:To investigate the expressions of E-cadherin (E-cd) and alpha-catenin (alpha-cat) proteins in benign and malignant prostate tumors, and determine whether they could be used as molecular markers for the prognosis of prostate cancer (PCa). METHODS:We detected the expressions of E-cd and alpha-cat in the prostatic tissues from 45 cases of PCa and 10 cases of benign prostatic hyperplasia (BPH) by immunohistochemical Elivision staining, and analyzed the relationships of E-cd and alpha-cat expressions with the PCa stage, PCa grade, preoperative PSA, results of endocrine therapy and prognosis. RESULTS:The E-cd protein was abnormally expressed in 86.7% of the PCa and 10.0% of the PSA patients, and the E-cd expression was significantly lower in the former than in the latter (P < 0.05). The abnormal expressions of E-cd in the PCa patients with metastasis, non-metastasis, Gleason score < or = 7 and > 7 were 85.0, 87.5, 100.0 and 86.7%, respectively, with no significant between-group differences (P > 0.05), those in the PCa patients with PSA < or = 10 and > 10 microg/L were 40.0 and 97.1%, respectively, significantly higher in the former than in the latter (P < 0.05), and those in the PCa patients with and without response to endocrine therapy were 93.8 and 72.7%, respectively, with no significant differences between the two groups (P > 0.05). The alpha-cat protein was abnormally expressed in 93.3% of the PCa and 30.0% of the BPH patients, respectively, and the alpha-cat expression was significantly lower in the former than in the latter (P < 0.05). The abnormal alpha-cat expressions in the PCa patients with metastasis, non-metastasis, Gleason score > 7 and < or = 7 were 90.0, 100.0, 90.0 and 100.0%, respectively, with no significant between-group differences (P > 0.05), those in the PCa patients with PSA < or = 10 and > 10 microg/L were 40.0 and 94.3%, respectively, significantly higher in the former than in the latter (P < 0.05), and those in the PCa patients with and without response to endocrine therapy were 100.0 and 81.8%, respectively, with no significant differences between the two groups (P > 0.05). CONCLUSION:The expressions of E-cd and alpha-cat are significantly lower in PCa than in BPH, and they are not associated with cancerous metastasis, but negatively correlated with the PSA level in PCa patients.

Abstract:Objective To investigate the expression of E-cadherin in human pancreatic cancer tissues,and its cli-nical implication.Methods The expression of E-cadherin in 60 patients with pancreatic cancer admitted during the priod from January 2001 to June 2006,was determined by tissue microarray and EnVision immunohistochemical staining.Results The expression of E-cadherin in the memberane of normal pancreatic cells was higher than that in cancer cells.The rate of reduced E-cadherin expression in pancreatic cancer tissues was 61.7%(37/60).The reduced E-cadherin expression was closely related with cancerous tissue differentiation,lymph node status,TNM stage and poor prognosis(P<0.05).Conclusions E-cadherin might be a sensitive predictor to evaluate the metastasic potential and the outcome of pancreatic cancer patients.

Abstract:OBJECTIVE:To observe the expressions of E-cadherin and beta-catenin in LNCaP and ARCaP cell lines and explore their relationship with the metastasis of human prostate cancer.METHODS:The expressions and distribution of E-cadherin and beta-catenin in LNCaP and ARCaP cell lines (IF11 and IA8) were detected by Western blot and immunofluorescent staining.RESULTS:The expression of E-cadherin was high in LNCaP, but absent in IF11 and IA8, while beta-catenin was expressed highly in IF11 and LA8, but lowly in LNCaP. Immunofluorescent staining showed that E-cadherin was mainly in the membrane of LNCaP, while beta-catenin both in the membrane of LNCaP and in the nuclei of IF11 and IA8.CONCLUSION:E-cadherin and beta-catenin are differently expressed and distributed in prostate cancer cell lines with different characteristics of epithelial-mesenchymal transition (EMT), and the abnormal activation of the beta-catenin signal pathway may be involved in the EMT of prostate cancer cells.
Expression of N-cadherin Protein in Urothelial Bladder Cancer and Its Relation to Prognosis

Abstract:Objective To explore the expression and significance of N-cadherin in urothelial bladder cancer and analyse its relation to clinicpathologic and prognosis of bladder cancer.Methods The expression of N-cadherin in 145 urothelial bladder cancer and 25 normal bladder tissues was detected by immunuhistochemisty,and correlations between N-cadherin and clinicopathologic features were analysed.Results The positive rate of N-cadherin protein was significantly higher in bladder cancer than in normal bladder tissue (P＜0.01).The positive rate of N-cadherin protein was 38.6％ in G1 bladder cancer,58.4％ in G2-G3 bladder cancer,and its difference was significant (P =0.028).The expression was significantly lower in non-muscle-invasive bladder cancer than in muscle invasive bladder cancer (45.7％ vs 64.7％,P=0.029).The muscle-invasive bladder cancer patients were followed up 4-103 months.Among those,the overall survival with positive expression of N-cadherin protein was 24.2％ (8/33),and the overall survival with negative expression of N-cadherin protein was 66.7 ％ (12/18).Kaplan-Meier analysis showed the positive expression of N-cadherin was significantly associated with overall survival of patients with muscle-invasive-bladder cancer (P=0.002 2).Multivariate Cox analysis showed that N-cadherin expression was an important prognostic factor.Conclusions The expression of N-cadherin protein was high in bladder cancer.The detection of the expression of N-cadherin protein is associated with the diagnosis and prognosis of bladder cancer.
Expression of E-, P- and N-Cadherin and Its Clinical Significance in Cervical Squamous Cell Carcinoma and Precancerous Lesions

Abstract:Aberrant expression of classical cadherins has been observed in tumor invasion and metastasis, but its involvement in cervical carcinogenesis and cancer progression is not clear. We investigated E-, P- and N-cadherin expression and its significance in cervical squamous cell carcinoma (SCC) and cervical intraepithelial neoplasia (CIN). This retrospective study enrolled 508 patients admitted to Women's Hospital, School of Medicine, Zhejiang University with cervical lesions between January 2006 and December 2010. Immunochemical staining was performed in 98 samples of normal cervical epithelium (NC), 283 of CIN, and 127 of early-stage SCC. The association of cadherin staining with clinical characteristics and survival of the patients was evaluated by univariate and multivariate analysis. We found gradients of decreasing E-cadherin expression and increasing P-cadherin expression from NC through CIN to SCC. Aberrant E-cadherin and P-cadherin expression were significantly associated with clinical parameters indicating poor prognosis and shorter patient survival. Interestingly, we found very low levels of positive N-cadherin expression in CIN and SCC tissues that were not related to CIN or cancer. Pearson chi-square tests showed that E-cadherin expression in SCC was inversely correlated with P-cadherin expression (E-P switch), and was not correlated with N-cadherin expression. More important, patients with tissues exhibiting an E-P switch in expression had highly aggressive phenotypes and poorer prognosis than those without E-P switch expression. Our findings suggest that E-cadherin and P-cadherin, but not N-cadherin staining, might be useful in diagnosing CIN and for predicting prognosis in patients with early-stage SCC.
Clinicopathological Significance of E-Cadherin and PCNA Expression in Human Non-Small Cell Lung Cancer

Abstract:This study was designed to assess E-cadherin (E-cad)and proliferating cell nuclear antigen (PCNA)expression as well as their clinicopathological significance in human non small cell lung cancers (NSCLCs). Possible molecular mechanisms of differentiation and metastasis of NSCLCs are discussed. Immuno-histochemical and immunofluorescence double staining were performed to examine the expression of E-cad and PCNA in 68 primary NSCLCs cases. The E-cad expression in squamous cell carcinomas and adenocarcinomas showed no significant difference. E-cad expression had a positive correlation with the histological-diferentiated grade. A significant difference of E-cad expression was found between metastatic and non-metastatic groups. PCNA expression in squamous cell carcinomas and adenocarcinomas showed no significant difference. The PCNA expression had a reverse correlation with the histological-differentiated grade. A significant difference of PCNA expression was found between m etastatic and non-metastatic groups. The E-cad and PCNA expression presented a reverse correlation. E-cad expression is not associated with the histological type of NSCLC, but is associated with differentiation and metastasis of the cancer. Down-regulation of E-cad expression affects the proliferation of cancer cells. Conjoint analysis of E-cad and PCNA expression is a good way to evaluate tumor biological behavior.

Abstract:BACKGROUND:E-cadherin is a subclass of the cadherin family that plays an important role in the maintenance of intercellular junctions in normal epithelium.Decreased expression of E-cadherin might be closely related to invasiveness and dedifferentiation in human cancers.There is increasing evidence that modulation of the E-cadherin-catenin cell-cell adhesion complex is an important step in the initiation and progression of human cancers.The aim of this study is to investigate the relationship between the level of E-cadherin mRNA expression and pathological grades and clinical stages of non-small cell lung cancer(NSCLC). METHODS:RT-PCR was used to measure the level of E-cadherin mRNA expression in 53 specimens of NSCLC,46 of para-cancer lung tissues,5 of benign nodal lung diseases,and the stages of disease was determined according to the results of surgery,pathology and imaging diagnoses.Then analyses were carried out between the level of E-cadherin mRNA expression and the clinical variables. RESULTS:45.3%(24/53) and 45.7%(21/46) specimens of NSCLC and para-cancer lung tissue were positive for E-cadherin mRNA expression respectively(P > 0.05);NSCLC with low differentiation,advanced stages and nodal metastases showed a magnificantly lower expression of E-cadherin mRNA(P < 0.05).The median survival time for E-cadherin mRNA positive and negative patients were 15.5 months and 46 months,respectively,but the expression of E-cadherin mRNA did not correlate with patient's survival(P > 0.05). CONCLUSIONS:E-cadherin expression is related to the differentiation,lymph node metastasis and pathological staging of NSCLC,but probably does not effectively affect its prognosis.
Prognostic Significance Of E-Cadherin Expression In Hepatocellular Carcinoma: A Meta-Analysis

Abstract:Backgrounds: Hepatocellular Carcinoma (HCC) is one of the most common malignancy of liver and HCC-related morbidity and mortality remains at high level. Researchers had investigated whether and how reduced E-cadherin expression impacted the prognosis of patients with HCC but the results reported by different teams remain inconclusive.Methods: A systematic literature search was performed in all available databases to retrieve eligible studies and identify all relevant data, which could be used to evaluate the correlation between reduced E-cadherin expression and clinicopathological features and prognosis for HCC patients. A fixed or random effects model was used in this meta-analysis to calculate the pooled odds ratios (OR) and weighted mean differences (WMD) with 95% confidence intervals (CI).Results: Total 2439 patients in thirty studies matched the selection criteria. Aggregation of the data suggested that reduced E-cadherin expression in HCC patients correlated with poor 1-, 3- and 5-year overall survival. The combined ORs were 0.50 (n = 13 studies, 95% CI: 0.37-0.67, Z = 4.49, P<0.00001), 0.39 (n = 13 studies, 95% CI: 0.28-0.56, Z = 5.12, P<0.00001), 0.40 (n = 11 studies, 95% CI: 0.25-0.64, Z = 3.82, P = 0.0001), respectively. Additionally, the pooled analysis denoted that reduced E-cadherin expression negatively impacts recurrence-free survival (RSF) with no significant heterogeneity. The pooled ORs for 1-, 3- and 5-year RSF affected by down-regulated E-cadherin were 0.73 (n = 6 studies, 95% CI: 0.54-1.00, Z = 1.95, P = 0.05), 0.70 (n = 6 studies, 95% CI: 0.52-0.95, Z = 2.32, P = 0.02), 0.66 (n = 5 studies, 95% CI: 0.48-0.90, Z = 2.64, P = 0.008). And what's more, reduced E-cadherin expression tended to be significantly associated with metastasis (OR = 0.31, 95% CI: 0.16-0.60, Z = 3.50, P = 0.0005), vascular invasion (OR = 0.76, 95% CI: 0.59-0.98, Z = 2.14, P = 0.03), advanced differentiation grade (OR = 0.31, 95% CI: 0.21-0.45, Z = 6.04, P<0.00001) and advanced TMN stage (T3/T4 versus T1/T2) (OR = 0.61,95% CI: 0.38-0.98, Z = 2.05, P = 0.04).Conclusions: Reduced E-cadherin expression indicates a poor prognosis for patients with HCC, and it may have predictive potential for prognosis of HCC patients.
Expression of E-cadherin and PCNA in pancreatic adenocarcinoma and their clinic significance

Abstract:Objective: To study the expression of E-cadherin and PCNA in pancreatic adenocarcinoma and their clinical significance. Methods: E-cadherin and PCNA in 59 cases of pancreatic adenocarcinoma was studied by immunohistochemistry. Results: The decreased expression of E-cadherin in pancreatic adenocarcinoma was significantly lower than that in normal pancreas; the higher rate of expression only was 13.6% (8/59); and the expression of E-cadherin in various groups was statistically significant with the differentiation of carcinoma; the metastasis of lymph nodes and the clinical stages (P<0.05); the increased expression of PCNA in pancreatic adenocarcinoma was significantly higher than that in normal pancreas; the higher positive rate of PCNA was 67.8% (40/59); and the expression of PCNA in various groups was significant in statistics with the differentiation of carcinoma; the metastasis of lymph nodes and the clinical stages(P<0.05); the correlation between E-cadherin and PCNA was negative in pancreatic adenocarcinoma. (P<0.05). Conclusion: The abnormal expression of E-cadherin and PCNA are closely associated with the differentiation, proliferation, invasion and metastasis of nancreatic adenocarcinoma.
Expressions of N-cadherin, E-cadherin and P63 in Cervical Squamous Cell Carcinoma and Their Significances

Abstract:Purpose To investigate the expression of N-cadherin,E-cadherin and p63 protein in cervical spuamous cell carcinoma and the relationship between their expression and clinicopathologic parameters and the prognosis.Methods Immuohistochemistry was used to detect the expressions of N-cadherin,E-cadherin and p63 in 131 cervical spuamous cell carcinoma and 30 normal cervical tissues.Results N-cadherin,E-cadherin and p63 positive expression rates in 131 cases of cervical squamous cell carcinoma tissue samples were associated significantly with tumor differentiation,clinical stages,lymph node metastasis and postoperative recurrence(P0.05),but had no relationship with age(P0.05).30 cases of normal cervical epithelial cells didn't express N-cadherin and E-cadherin and p63 was strong positive expression.The survival of patients with strong positive expression of N-cadherin was significantly lower than those with negative expression of N-cadherin,however,the survival of patients with negative expression of E-cadherin and p63 was significantly lower than those with strong positive expression of E-cadherin and p63.Conclusion The low expression of E-cadherin,p63 and high expression of N-cadherin may play a role in cervical cancer carcinogenesis and metastasis.Detection of the expression level of N-cadherin,E-cadherin and p63 could be one of the important markers for predicting the metastasis and prognosis of cervical squamous cell carcinoma.
Clinical Significance of E-cad and #Beta#-Cat Expressions in CIN and Cervical Squamous Cell Carcinoma

Abstract:OBJECTIVE:To study E-cad and β-catenin (β-cat) expressions and their clinical significance in cervical intraepithelial neoplasia (CIN) and cervical squamous cell carcinoma.METHODS:E-cad and β-cat expressions were investigated by the SP method in normal cervical epithelia,low-grade CIN, high-grade CIN and cervical squamous cell carcinoma.RESULTS:In normal cervical epithelia,E-cad and β-cat expressed at the membranes. E-cad membranous expression rates in normal cervical epithelia, CIN Ⅰ, CIN Ⅱ-Ⅲ and squamous cell carcinomas were 86%(12/14), 82%(9/11), 54%(7/13) and 21%(8/38), respectively; β-catmembranous expression rates were 79%(11/14), 64%(7/11), 23%(3/13) and 11%(4/38), respectively. The difference of E-cad expression between normal cervical epithelia and invasive squamous cell carcinoma was as statistically significant as that between CINⅠ and carcinoma,P0.05. The difference of E-cad expression in histological classification of invasive carcinoma was as statistically significant as that in depth of muscular invasion,P0.05.The difference of β-cat expression between normal cervical epithelia and CINⅡ-Ⅲ was as statistically significant as that between normal cervical epithelia and invasive squamous cell carcinoma,CINⅠ and carcinoma,P0.05.The differences of β-cat expression in histological classifications of invasive carcinoma were statistically significant,P0.05. The expression patterns between E-cad and β-cat in invasive carcinoma were correlated with each other, P0.05.CONCLUSIONS:The abnormal expressions of E-cad and β-cat are early events in the progress of carcinoma.In cervical squamous cell carcinoma,their abnormal expressions may be related to the invasion of carcinoma.

Abstract:OBJECTIVE:To investigate the expression of E-cadherin(E-cad), N-cadherin(N-cad), β-catenin(β-cat), which are the markers of Epithelial-mesenchymal transition and analyze their relationships with the clinicopathological features and the prognosis of the laryngeal squamous cell carcinoma.METHODS:The expression levels of E-cad, N-cad, β-cat in 76 tumor tissues and their corresponding adjacent normal laryngeal tissues were determined by immunohistochemistry method. Relationships between the proteins' expression and clinicopathological features were analyzed. Survival curves were calculated using the Kaplan-Meier method. Differences in the survival rates were analyzed by the log-rank test among different expression groups. Cox's regression model was used to examine the independent predictor of the prognosis of the laryngeal cancer.RESULTS:The expression levels of E-cad, N-cad, β-cat in tumor tissues and adjacent normal tissues were statistically significant(P<0.001). The expression level of E-cad and β-cat in the laryngeal carcinoma was related to lymph node metastasis, clinical pathological stage and differentiation degree (P<0.05), while N-cad expression level was associated with clinical stage and differentiation degree (P<0.05). The expression of E-cad was correlated with the expression of β-cat (P=0.001), and the expression of N-cad was correlated with β-cat (P=0.02), but the expression of E-cad was not correlated with N-cad. There were four subgroups of patterns of E-cad and N-cad expression: E-cad (+ )/N-cad (-), E-cad (+ ) /N-cad (+ ), E-cad (-) /N-cad (-), E-cad (-) /N-cad (+ ). The expression level of each group was related to the clinical pathological stage and differentiation degree (P<0.05). The expression level of E-cad/β-cat was associated with lymph node metastasis, clinical pathological stage and differentiation degree (P<0.01). Log-rank analysis showed that the prognosis of negative and positive groups was statistically different (P<0.05), and the combined analysis showed that the prognosis of E-cad/N-cad or E-cad/β-cat group was significantly different (P<0.01). Cox's regression model analysis showed that the clinical stage and β-cat were independent predictors of the prognosis of laryngeal carcinoma.CONCLUSIONS:The low expression of E-cad, high abnormal expression of N-cad and β-cat played an important role in the occurrence and development of laryngeal carcinoma. It can provide a reference for evaluating clinical prognosis. The clinical pathological stage and β-cat can be used as independent predictors for the prognosis of laryngeal carcinoma.
Expression and clinical significance of GS,E-cadherin and β-catenin in hepatocellular carcinoma

Abstract:Objective To investigate the expression of glutamine synthetase(GS),E-cadherin and β-catenin in hepatocellular carcinoma and its relation to clinicopathological characterisitics and prognosis.Methods With the method of immunohistochemical Envision,the expressions of GS,E-cadherin and β-catenin were examined in 182 cases of hepatocellular carcinoma and 92 cases of adjacent non-neoplastic liver tissue.We then analyzed their relations to the clinicopathological data of hepatocellular carcinoma.Results In the hepatocellular carcinoma group,77.5% samples expressed GS,significantly higher than in surrounding liver tissue(4.3% P<0.05).The expression of GS was positively correlated with TNM stage,tumor metastasis and recurrence(P<0.05).In hepatocellular carcinoma,aberrant expression of E-cadherin and β-catenin was 59.3% and 58.8% respectively,higher than in adjacent non-neoplastic liver tissue(30.4% and 26.1%,P<0.05).Aberrant expression of E-cadherin and β-catenin was related to vascular cancer embolus,TNM stage,tumor metastasis and recurrence(P<0.05).The expression of GS correlated with aberrant expression of E-cadherin and β-catenin in hepatocellular carcinoma.Conclusion The overexpression of GS,E-cadherin and β-catenin may be important mechanisms which promote the invasion and metastasis of hepatocellular carcinoma.Combined detection of GS,E-cadherin and β-catenin may be helpful to judge the degree of malignancy and potential metastasis,and to evaluate the prognosis.

Abstract:Objective To determine the expression of E-cadherin and CAR in bladder cancer cell lines and explore their significance and relation.Methods Expression of E-cadherin and CAR in bladder cancer cell lines was detected by Western blot.Results E-cadherin and CAR expressions were high in RT4 and 5637 cell,but low in 253J.E-cadherin was not detected in J82 and T24 cell.Expression of CAR was low in J82 cell,and no expression in T24 cell.Conclusion The expressions of E-cadherin and CAR were coincident in bladder cancer cell lines,indicating they may cooperate in the invasion and metastasis of bladder cancer.

Abstract:OBJECTIVE:To explore the expression patterns of cadherins (N-cadherin and E-cadherin) in urothelial carcinomas and understand the values of N-E cadherin switch in the predictions of postoperative recurrence and prognosis. METHODS:The expressions of N-cadherin and E-cadherin were measured by immunohistochemistry in 64 transurethral bladder tumor resection (TURBT) or partial cystectomy samples (48 cases) in 2005 by the method of streptavidin-biotin peroxidase. RESULTS:A positive expression of N-cadherin and a negative expression of E-cadherin were noted in 27 (42.2%) and 16 (25.0%) specimens respectively. Patients with more poorly differentiated bladder cancer were accompanied with a higher expression of N-cadherin and a lower expression of E-cadherin (both P < 0.05). A positive expression of N-cadherin decreased the recurrence-free and cancer-related survival rates while a negative expression of E-cadherin was correlated with a lower cancer-related survival rate (all P < 0.05). No significant association was found between the expression of E-cadherin and the recurrence-free survival rate. Furthermore, the N-E cadherin switch (a negative expression of E-cadherin and a positive expression of N-cadherin) showed a higher predictive power in both recurrence-free and cancer-related survival according to multivariate analysis (HR = 0.428, 95%CI: 0.217 - 0.845, HR = 0.098, 95%CI: 0.013 - 0.767). No significant association was found between the expressions of two cadherins and postoperative progression. CONCLUSION:Although the expressions of E-cadherin and N-cadherin appear to be correlated with survival outcomes in bladder cancer, N-E cadherin switch may be a better predicator for postoperative recurrence and cancer-related survival.

Abstract:OBJECTIVE:To investigate a potential mechanism of invasion and metastasis of oral cancers, and the possibility of the adhesion molecules being markers for evaluating the prognosis of oral cancers. METHODS:Randomly selecting achieved paraffin blocks, including 60 cases of squamous cell carcinoma(SCC), 30 cases of verrucous carcinoma(VC) of oral mucosa and 30 cases of basal cell carcinoma (BCC) of epidermis. All cases were followed up. Using immunohistochemistry method (Envision) to detect the expression of E-cadherin antibody and observe under light microscope. RESULTS:The studies demonstrated that 38.2% of SCC had recurrence and/or metastases after operation. However, only 0.0% and 0.04%, in VC and BCC, respectively, had recurrence and/or metastases. There was significant difference on prognoses between SCC and VC or BCC. The immunostaining showed significant difference of E-cad expression between SCC and VC (P<0.01), SCC and BCC (P<0.05). The former was weaker than VC and BCC. However, no difference was showed in E-cad expression between VC and BCC (P>0.05). CONCLUSION:E-cad plays an important role in maintaining the phenotype of epithelial cell of SCC and normal oral mucosa. The degree of expression of E-cad was correlated with the ability of invasion and/or metastasis of SCC, VC and BCC. It indicates that E-cad may be as a helpful marker to evaluate the prognosis of oral cancers.
