Abstract:BACKGROUND:E-cadherin is a subclass of the cadherin family that plays an important role in the maintenance of intercellular junctions in normal epithelium.Decreased expression of E-cadherin might be closely related to invasiveness and dedifferentiation in human cancers.There is increasing evidence that modulation of the E-cadherin-catenin cell-cell adhesion complex is an important step in the initiation and progression of human cancers.The aim of this study is to investigate the relationship between the level of E-cadherin mRNA expression and pathological grades and clinical stages of non-small cell lung cancer(NSCLC). METHODS:RT-PCR was used to measure the level of E-cadherin mRNA expression in 53 specimens of NSCLC,46 of para-cancer lung tissues,5 of benign nodal lung diseases,and the stages of disease was determined according to the results of surgery,pathology and imaging diagnoses.Then analyses were carried out between the level of E-cadherin mRNA expression and the clinical variables. RESULTS:45.3%(24/53) and 45.7%(21/46) specimens of NSCLC and para-cancer lung tissue were positive for E-cadherin mRNA expression respectively(P > 0.05);NSCLC with low differentiation,advanced stages and nodal metastases showed a magnificantly lower expression of E-cadherin mRNA(P < 0.05).The median survival time for E-cadherin mRNA positive and negative patients were 15.5 months and 46 months,respectively,but the expression of E-cadherin mRNA did not correlate with patient's survival(P > 0.05). CONCLUSIONS:E-cadherin expression is related to the differentiation,lymph node metastasis and pathological staging of NSCLC,but probably does not effectively affect its prognosis.

Abstract:Abstract Conclusion: Down-regulation of ECD may associate with infiltrative tumor growth and result in poor prognosis. However, ECD expression is not significantly associated with tumor metastasis. Aim: To investigate the prognostic role of E-cadherin (ECD) expression in laryngeal cancer. Methods: In a clinical study, a total of 79 laryngeal cancer patients who received surgery were included in this study. The expression of ECD in laryngeal cancer tissue was detected by immumohistochemical staining. In an in vitro study, ECD up-regulated cells (Ad-ECD) and ECD down-regulated cells (sh-ECD) were cultured, along with interference by siRNA. Cell apoptosis was detected by flow cytometry. In an in vivo study, nude mice were injected with Ad-ECD, sh-ECD, and Hep-2 cells. The ECD expression and tumor metastasis in the brain, liver, and lung tissues were detected. Results: Clinical data showed that expression of ECD was statistically correlated with survival rate (p < 0.05), but not with other clinical characteristics, such as age, gender, lymphatic metastasis, and TNM staging. ECD was down-regulated in sh-ECD cells and the down-regulation significantly reduced apoptosis of Hep-2 cells. Tumor growth was significantly inhibited in the Ad-ECD group. Except the tumor tissue, no positive ECD expression or significant tumor cells were found in the brain, liver, and lung tissues.
Clinicopathological Significance of E-Cadherin and PCNA Expression in Human Non-Small Cell Lung Cancer

Abstract:This study was designed to assess E-cadherin (E-cad)and proliferating cell nuclear antigen (PCNA)expression as well as their clinicopathological significance in human non small cell lung cancers (NSCLCs). Possible molecular mechanisms of differentiation and metastasis of NSCLCs are discussed. Immuno-histochemical and immunofluorescence double staining were performed to examine the expression of E-cad and PCNA in 68 primary NSCLCs cases. The E-cad expression in squamous cell carcinomas and adenocarcinomas showed no significant difference. E-cad expression had a positive correlation with the histological-diferentiated grade. A significant difference of E-cad expression was found between metastatic and non-metastatic groups. PCNA expression in squamous cell carcinomas and adenocarcinomas showed no significant difference. The PCNA expression had a reverse correlation with the histological-differentiated grade. A significant difference of PCNA expression was found between m etastatic and non-metastatic groups. The E-cad and PCNA expression presented a reverse correlation. E-cad expression is not associated with the histological type of NSCLC, but is associated with differentiation and metastasis of the cancer. Down-regulation of E-cad expression affects the proliferation of cancer cells. Conjoint analysis of E-cad and PCNA expression is a good way to evaluate tumor biological behavior.

Abstract:Objective:To investigate the expression of E-cadherin(ECD) and its clinical value in predicting the prognosis of laryngeal carcinoma.Methods:The expression of ECD was determined by immunohistochemistry with EnVision two-step method on tissue microarray slides in 79 cases of laryngeal carcinoma tissues;the relationship between the pathological features and the prognosis was analyzed.Results:Negative expression of ECD was detected in 11 cases of the tissue(13.3%);The ECD expression had no statistical differences between the clinical and the pathological features of the patients with the laryngeal carcinoma(P 0.05).The positive rates of ECD were 76.74% and 94.44% in the dead patients and the survival ones respectively;the difference was statistically significant(P 0.05).Conclusions:The ECD expression may be of great value in predicting the prognosis of patients with the laryngeal carcinoma.

Abstract:OBJECTIVE To elucidate the relationship between the expression of E-cadherin(E-cad)、 vascular endothelial growth factor(VEGF) and cathepsin D (Cath-D) in laryngeal squamous cell carcinoma and their significance in lymph node metastasis. METHODS Sixty specimens of laryngeal squamous cell carcinoma were studied with im munohistochemical staining method to explore the expression of E-cadherin、 VEGF and Cath-D. RESULTS Positive expression of E-cad 、 VEGF and Cath-D were found in 75 %, 83 % and 66 % of laryngeal squamous cell carcinoma specimens respectively . There was a positive relationship between VEGF and Cath-D, E-cad. The expression of E-cad, VEGF and Cath-D were significantly higher in patients with cervical lymph node metastasis than those without metastasis. No correlation was identified between E-cad and Cath-D staining, the expression of E-cad in VEGF-posi tive and Cath-D positive tumors was significantly high. These phenomena were more commonly found in patients with cervical lymph node metastasis than in patients without metastasis. CONCLUSION The expression of E-cad, VEGF and Cath-D could be an important monitor marker in laryngeal squamous cell carcinoma with cervical lymph node metastasis. There might be a regulated interaction among VEGF,Cath-D and E-cad.
Prognostic Significance Of E-Cadherin Expression In Hepatocellular Carcinoma: A Meta-Analysis

Abstract:Backgrounds: Hepatocellular Carcinoma (HCC) is one of the most common malignancy of liver and HCC-related morbidity and mortality remains at high level. Researchers had investigated whether and how reduced E-cadherin expression impacted the prognosis of patients with HCC but the results reported by different teams remain inconclusive.Methods: A systematic literature search was performed in all available databases to retrieve eligible studies and identify all relevant data, which could be used to evaluate the correlation between reduced E-cadherin expression and clinicopathological features and prognosis for HCC patients. A fixed or random effects model was used in this meta-analysis to calculate the pooled odds ratios (OR) and weighted mean differences (WMD) with 95% confidence intervals (CI).Results: Total 2439 patients in thirty studies matched the selection criteria. Aggregation of the data suggested that reduced E-cadherin expression in HCC patients correlated with poor 1-, 3- and 5-year overall survival. The combined ORs were 0.50 (n = 13 studies, 95% CI: 0.37-0.67, Z = 4.49, P<0.00001), 0.39 (n = 13 studies, 95% CI: 0.28-0.56, Z = 5.12, P<0.00001), 0.40 (n = 11 studies, 95% CI: 0.25-0.64, Z = 3.82, P = 0.0001), respectively. Additionally, the pooled analysis denoted that reduced E-cadherin expression negatively impacts recurrence-free survival (RSF) with no significant heterogeneity. The pooled ORs for 1-, 3- and 5-year RSF affected by down-regulated E-cadherin were 0.73 (n = 6 studies, 95% CI: 0.54-1.00, Z = 1.95, P = 0.05), 0.70 (n = 6 studies, 95% CI: 0.52-0.95, Z = 2.32, P = 0.02), 0.66 (n = 5 studies, 95% CI: 0.48-0.90, Z = 2.64, P = 0.008). And what's more, reduced E-cadherin expression tended to be significantly associated with metastasis (OR = 0.31, 95% CI: 0.16-0.60, Z = 3.50, P = 0.0005), vascular invasion (OR = 0.76, 95% CI: 0.59-0.98, Z = 2.14, P = 0.03), advanced differentiation grade (OR = 0.31, 95% CI: 0.21-0.45, Z = 6.04, P<0.00001) and advanced TMN stage (T3/T4 versus T1/T2) (OR = 0.61,95% CI: 0.38-0.98, Z = 2.05, P = 0.04).Conclusions: Reduced E-cadherin expression indicates a poor prognosis for patients with HCC, and it may have predictive potential for prognosis of HCC patients.

Abstract:OBJECTIVE:To investigate the expression of E-cadherin(E-cad), N-cadherin(N-cad), β-catenin(β-cat), which are the markers of Epithelial-mesenchymal transition and analyze their relationships with the clinicopathological features and the prognosis of the laryngeal squamous cell carcinoma.METHODS:The expression levels of E-cad, N-cad, β-cat in 76 tumor tissues and their corresponding adjacent normal laryngeal tissues were determined by immunohistochemistry method. Relationships between the proteins' expression and clinicopathological features were analyzed. Survival curves were calculated using the Kaplan-Meier method. Differences in the survival rates were analyzed by the log-rank test among different expression groups. Cox's regression model was used to examine the independent predictor of the prognosis of the laryngeal cancer.RESULTS:The expression levels of E-cad, N-cad, β-cat in tumor tissues and adjacent normal tissues were statistically significant(P<0.001). The expression level of E-cad and β-cat in the laryngeal carcinoma was related to lymph node metastasis, clinical pathological stage and differentiation degree (P<0.05), while N-cad expression level was associated with clinical stage and differentiation degree (P<0.05). The expression of E-cad was correlated with the expression of β-cat (P=0.001), and the expression of N-cad was correlated with β-cat (P=0.02), but the expression of E-cad was not correlated with N-cad. There were four subgroups of patterns of E-cad and N-cad expression: E-cad (+ )/N-cad (-), E-cad (+ ) /N-cad (+ ), E-cad (-) /N-cad (-), E-cad (-) /N-cad (+ ). The expression level of each group was related to the clinical pathological stage and differentiation degree (P<0.05). The expression level of E-cad/β-cat was associated with lymph node metastasis, clinical pathological stage and differentiation degree (P<0.01). Log-rank analysis showed that the prognosis of negative and positive groups was statistically different (P<0.05), and the combined analysis showed that the prognosis of E-cad/N-cad or E-cad/β-cat group was significantly different (P<0.01). Cox's regression model analysis showed that the clinical stage and β-cat were independent predictors of the prognosis of laryngeal carcinoma.CONCLUSIONS:The low expression of E-cad, high abnormal expression of N-cad and β-cat played an important role in the occurrence and development of laryngeal carcinoma. It can provide a reference for evaluating clinical prognosis. The clinical pathological stage and β-cat can be used as independent predictors for the prognosis of laryngeal carcinoma.

Abstract:Reduced expression of E-cadherin, a cell-cell adhesion molecule, was frequently observed in several types of human carcinomas, and the protein plays a role as an invasion suppressor in vitro. In an attempt to evaluate the significance of E-cadherin gene in non-small cell lung cancer (NSCLC), we undertook the immunohistochemical and molecule structural analyses of E-cadherin gene in 40 resection specimens of NSCLC and the corresponding paracarcinoma controls. E-cadherin expression was explored by immunohistochemistry with a monoclonal antibody, and the E-cadherin gene was studied by polymerase chain reaction single-strand conformation polymorphism analysis (PCR-SSCP). The analysis represented in this study demonstrated clear reduction in the expression of E-cadherin proteins in the cancer tissues. However, only in one amplicon were aberrant bands detected, which was a single polymorphic site (codon 692; exon 13), and no somatic mutation was found. These results indicated that defected E-cadherin expression might play a role in the development of malignant phenotype in NSCLC, even though the genetic mutation of E-cadherin gene is not involved in the pathogenesis of NSCLC and does not appear to be direct cause for the reduced expression of E-cadherin gene.

Abstract:Objective:To investigate the expression of E-cadherin and β-catenin as prognostic factors in stage IIIa non-small-cell lung carcinoma(NSCLC).Methods: Tumor samples were constructed by tissue microarray(TMA) and immunohistochemistry(IHC) to analyze the relationship between the patients'clinical significance and the E-cadherin,β-catenin expression.Results: A total of 22 cases survived after 3 years' follow up,the over-all survival was 30.67% and the disease-free survival was 29.33%.The number of trace,low,intermediate,and high-level expression of E-cadherin and β-catenin were19,28,28,12 and 28,32,33,4,respectively.There was a positive correlation between E-cadherin and β-catenin(P<0.001).Dedifferentiation was associated with reduced expression of both E-cadherin and β-catenin(P=0.001,P<0.001).E-cadherin expression was an independent prognostic factor for disease-free survival the hazard ratio(HR) was 0.48(P=0.015).Conclusions: Reduced expression of E-cadherin and β-catenin correlates with poor differentiation.E-cadherin is an independent prognostic factor for stage IIIa NSCLCs 3 years'disease-free survival.
Prognostic Value of High FoxC2 Expression in Resectable Non-Small Cell Lung Cancer, Alone or in Combination with E-cadherin Expression.

Abstract:BACKGROUND:FoxC2 is an epithelial-mesenchymal transition (EMT) regulator which induces metastasis. The purpose of this study is to assess the prognostic value of FoxC2 expression in non-small cell lung cancer (NSCLC), alone or in combination with E-cadherin expression.METHODS:A retrospective study was conducted using immunohistochemistry to investigate FoxC2 and E-cadherin expression in a cohort of 309 patients with surgically resected NSCLCs. The prognostic value of FoxC2 and E-cadherin on overall survival (OS) and recurrence-free survival (RFS) was determined by Kaplan-Meier analysis and Cox proportional hazard models.RESULTS:High FoxC2 expression was detected in 26.5% of tumors, and significantly correlated with tobacco use (p = 0.047), adenocarcinoma (p = 0.008) and nodal involvement (p < 0.001). Univariate analysis revealed its association with OS (p = 0.036) and RFS (p = 0.011). By multivariate analysis, high FoxC2 expression lost its significance as an independent predictor of recurrence (p = 0.077), while TNM stage, nodal status and the presence of high FoxC2 and impaired E-cadherin expression retained independent prognostic significance in relation to both OS and RFS. Subset analyses indicated that high FoxC2 expression was significantly associated with disease outcome in node-positive, but not in node-negative patients.CONCLUSION:Evaluation of FoxC2 expression, alone or in combination with E-cadherin expression, may help to stratify NSCLC patients for risk of disease progression, pointing to this EMT regulator as a potential prognostic marker.

Abstract:Ezrin is known to regulate cellular survival, adhesion, migration, and invasion and has been identified as 1 of the key components of tumor progression and metastasis. The purpose of this study was to evaluate the clinicopathologic and prognostic significance of ezrin expression in non-small-cell lung cancer (NSCLC).We investigated the expression pattern of ezrin immunohistochemically in 89 paraffin samples of NSCLC between January 1998 and December 2006 and conducted survival analyses. In addition, 73 frozen specimens (including tumorous and precancerous tissues) of NSCLC and 28 frozen specimens of benign pneumonic diseases collected between January 2009 and December 2009 were analyzed by Western blot and reverse transcriptase-polymerase chain reaction (RT-PCR).In 89 paraffin samples, ezrin was expressed in 40 cases, either in the cytoplasm or on the membrane. Ezrin-positive expression was significantly associated with increased tumor stage and lymph node (LN) metastasis. The positive rate of cytoplasm expression was significantly associated with LN metastasis. Importantly, ezrin-positive expression independently predicted inferior overall survival (OS) and disease-free survival (DFS). In 73 frozen specimens of NSCLC and 28 frozen specimens of benign pneumonic diseases, the ezrin mRNA, protein, and phospho-ezrin protein expressions in tumorous tissues were higher than they were in precancerous tissues and benign pneumonic tissues.These results suggested that high-level ezrin expression contributed to NSCLC progression and that phosphorylation and subcellular translocation of ezrin might be the important mechanisms. Ezrin might be a potential prognostic marker of progression in NSCLC.

Expression and clinical study of vascular endothelial growth factor and its receptors FLT-1, FLK-1/KDR in non-small cell lung cancer

Abstract:Objective To investigate the relationship between the expression of vascular endothelial growth factor (VEGF), its receptors FLT-1, FLK-1/KDR and tumor angiogenesis, clinical staging, lymph node metastasis in non-small cell lung cancer (NSCLC). Methods Immunohistochemical staining and enzyme-linked immunosorbent assay (ELISA) were used to detect the levels of VEGF and its receptors FLT-1,FLK-1/KDR in the tissue and serum of 112 patients with NSCLC, and analysed their clinical significances. Results The expression of VEGF and its receptors FLT-1, FLK/KDR in lung tissue of NSCLC were significantly higher than that in normal alveolar tissue and paracancerous tissue (P<0.05) and they increased significantly in proportion with clinical stages Ⅰ, Ⅱ, Ⅲ-Ⅳ (P<0.05). The positive rates of VEGF and its receptors FLT-1, FLK-1/KDR in lymph node metastasis group were significantly higher than those without lymph node metastasis group (P<0.05). Se- rum VEGF in NSCLC was significantly higher than that in the control group and also increased according to clinical staging (Ⅰ, Ⅱ, Ⅲ-Ⅳ; P<0.05). Serum VEGF was significantly higher in the lymph node metastasis group than that of the absence group (P<0.05) . Conclusions The expression of VEGF and its receptors FLT-1, FLK/KDR in NSCLC increased significantly, and positively correlated with tumor development, progression and lymph node metastasis. Serum VEGF is an important marker of hyperplasia and metastasis in NSCLC.

Abstract:OBJECTIVE:To investigate the expression of E-cadherin (ECD) and uPA in laryngeal cancer and evaluate their clinical value in prognosis.METHODS:ECD and uPA were determined by immunohistochemistry of Envision methods in carcinoma tissues of 51 patients of laryngeal carcinoma. All patients were followed and the prognostic factors were analyzed.RESULTS:Among 51 patients' tumor tissues, 24 (47.1%) were negative expression of ECD, and 26 (51.0%) were positive uPA immunoreaction was observed. There were four subgroups of patterns of ECD and uPA expression: 14 (27.1%) ECD-positive/uPA-negative, 13 (25.5%) ECD-negative/uPA-negative, 11 (21.6%) ECD-positive/uPA-positive, and 13 (25.5%) ECD-negative/uPA-positive. The tumor tissues with ECD-negative and uPA-positive expression were significant associated with lymph nodes metastasis (chi(2) = 5.545, 5.79, P = 0.019, 0.016 respectively). Patients with ECD-negative expression had a shorter survival than the patients with ECD positive expression but no statistic difference (chi(2) = 2.534, P > 0.05). Patients with uPA-positive expression had a significantly shorter survival time than those with uPA-negative expression (chi(2) = 6.259, P < 0.05). There was a difference for the median survival time between the patients with uPA-negative/ECD-positive and the patients with uPA-positive/ECD-negative in laryngeal cancer tissue (chi(2) = 6.559, P = 0.01), and the survival curves between these two groups was also statistically significant difference. Multivariate analysis of Cox revealed that clinical stage and ECD/uPA (P = 0.009, 0.007 respectively) were two independent prognostic factors.CONCLUSIONS:The combination analysis of uPA and ECD immunohistochemical expression in the laryngeal cancer tissue may be useful for predicting tumor metastatic risks and patient's prognosis.
Expression and clinical significance of GS,E-cadherin and β-catenin in hepatocellular carcinoma

Abstract:Objective To investigate the expression of glutamine synthetase(GS),E-cadherin and β-catenin in hepatocellular carcinoma and its relation to clinicopathological characterisitics and prognosis.Methods With the method of immunohistochemical Envision,the expressions of GS,E-cadherin and β-catenin were examined in 182 cases of hepatocellular carcinoma and 92 cases of adjacent non-neoplastic liver tissue.We then analyzed their relations to the clinicopathological data of hepatocellular carcinoma.Results In the hepatocellular carcinoma group,77.5% samples expressed GS,significantly higher than in surrounding liver tissue(4.3% P<0.05).The expression of GS was positively correlated with TNM stage,tumor metastasis and recurrence(P<0.05).In hepatocellular carcinoma,aberrant expression of E-cadherin and β-catenin was 59.3% and 58.8% respectively,higher than in adjacent non-neoplastic liver tissue(30.4% and 26.1%,P<0.05).Aberrant expression of E-cadherin and β-catenin was related to vascular cancer embolus,TNM stage,tumor metastasis and recurrence(P<0.05).The expression of GS correlated with aberrant expression of E-cadherin and β-catenin in hepatocellular carcinoma.Conclusion The overexpression of GS,E-cadherin and β-catenin may be important mechanisms which promote the invasion and metastasis of hepatocellular carcinoma.Combined detection of GS,E-cadherin and β-catenin may be helpful to judge the degree of malignancy and potential metastasis,and to evaluate the prognosis.
Wnt3a Expression Is Associated with Epithelial-Mesenchymal Transition and Impacts Prognosis of Lung Adenocarcinoma Patients.

Abstract:Background: Epithelial-mesenchymal transition (EMT) plays an important role in the invasion and migration during cancer metastasis. Wnt3a is one of the ligands in canonical Wnt/beta-catenin signaling pathway, which contributes to the carcinogenesis and progression of lung cancer cell lines. The aim of this study was to evaluate the association between Wnt3a and EMT-related proteins (E-cadherin and N-cadherin), and to further investigate its impact on prognosis of lung adenocarcinoma patients. Methods: A total of 147 lung adenocarcinoma patients were included and their clinicopathological characteristics were collected in this retrospective study. The expression levels of Wnt3a, E-cadherin and N-cadherin in post-surgery cancerous and adjacent normal tissues were assessed by immunohistochemistry. The association between Wnt3a and EMT-related proteins and their prognostic values were systematically evaluated. HCC827 and PC9 cell lines were treated with Wnt3a to detect the expression of EMT-related and Wnt/beta-catenin signaling-associated proteins, as well as the in vitro migration and invasion abilities. Results: High Wnt3a expression level was significantly associated with low E-cadherin (P<0.001) and high N-cadherin (P<0.001) expression levels in lung adenocarcinoma tissues. Besides, high Wnt3a level predicted poorer lung adenocarcinoma survival by univariate Cox analysis (P=0.001), while the multivariate result was not significant (P=0.355). Subgroup analysis suggested that the prognostic value of Wnt3a expression level was significant in stage T1-T2 (log rank P=0.003) and stage N0 (log rank P=0.031) patients. The multivariate Cox analysis suggested N-cadherin was an independent prognostic factor for lung adenocarcinoma patients (P=0.012). After including these markers into a nomogram, the Harrell's C-index of the nomogram was 0.755. The decision-curve analysis of our nomogram performed net benefit at the threshold probability from 21.6% to 82.0%, and the current model had a better prognostic value than TNM-classification with a lower Akaike information criterion (AIC) value of 166.54. In vitro experiments suggested that Wnt3a could regulate EMT-related proteins and promotes in vitro invasion and migration abilities. Conclusions: Wnt3a could regulate EMT-related proteins and promote the migration and invasion process of lung adenocarcinoma. Although its value as an independent prognostic factor was limited, the combined model suggested good prognostic performance for lung adenocarcinoma patients.

Abstract:Objective: To investigate the expression of serum response factor(SRF),E-cadherin and βcatenin in hepatocellular carcinoma and its relation to clinicopathological characterisitics of the tumor and prognosis.Methods: Tissue microarray and immunohistochemical study(EnVision method) for SRF,E-cadherin and βcatenin was performed on tissue microarray which consisted of 182 cases of hepatocellular carcinoma,92 cases of adjacent non-neoplastic liver tissues.Results: In the hepatocellular carcinoma group 75.8% tumor samples expressed SRF,and the levels is significantly higher than that in surrounding liver tissue(21.7%,P<0.05).The expression of SRF was positively correlated with the TNM stage,tumor metastasis and recurrence(P<0.05).In the hepatocellular carcinoma group,aberrant expression of E-cadherin and β-catenin was 59.3% and 58.8% respectively,and these levels were higher than those in the adjacent non-neoplastic liver tissue group(30.4% and 26.1% P<0.05).Aberrant expression of E-cadherin andβ-catenin was related to vascular cancer embolus,the TNM stage,tumor metastasis and recurrence(P<0.05).The expression of SRF correlated with aberrant expression of E-cadherin and β-catenin in hepatocellular carcinoma.Conclusion: In hepatocellular carcinoma,the overexpression of SRF and the downregulated expression of E-cadherin and β-catenin may be important mechanisms for promoting the invasion and metastasis of hepatocellular carcinoma and may also play a key role in the occurrence and development of hepatocellular carcinoma.
Expression of E-cadherin in the tumor tissue and serum of patients with esophageal squamous cell carcinoma

Abstract:Objective: To survey E-cadherin (E-cad) expression in tumor tissue and serum of esophageal squamous cell carcinoma patients, and to observe the clinical significance of their expression. Methods: Forty-eight samples of esophageal squamous cell carcinoma tissue, 23 samples of erosive esophagitis tissue, 24 samples of normal esophagus tissue and the corresponding sera were obtained. We used immunohistochemistry (IHC) to detect expression of E-cad in the tissues and enzyme-linked immunosorbent assay (ELISA) to examine expression of E-cad in the serum. Furthermore, we collected complete clinicopathological data from the participating patients. Results: The expression level of E-cad in the esophageal squamous cell carcinoma tissue was lower than that in normal esophagus tissues and erosive esophagitis tissues (P<0.05). Moreover, the expression level of E-cad was related to the depth of invasion, the status of lymph node metastasis and the level of differentiation of esophageal squamous cell carcinoma (P<0.05). The expression level of serum E-cad of esophageal squamous cell carcinoma patients was obviously higher than that in the serum of normal esophagus controls and erosive esophagitis patients (P<0.05). But the expression level of E-Cad in the serum of esophageal squamous cell carcinoma patients was unrelated to clinicopathological features. The expression level of E-cad in the tissue was not correlated with that in the serum(P=0.134). Conclusion: The expression of E-cad in tissues may assistin the diagnosis and prognosis of esophageal squamous cell carcinoma. The expression of E-cad in the serum may assistin the diagnostic screening of esophageal squamous cell carcinoma.
The Impact of E-cadherin Expression on Non-Small Cell Lung Cancer Survival: a Meta-Analysis

Abstract:E-cadherin has been implicated in invasiveness and metastasis. However, the clinical prognostic value of decreased E-cadherin expression in patients with non-small cell lung cancer (NSCLC) remains unsettled. A meta-analysis of eligible studies was performed to quantitatively review the correlation of decreased E-cadherin expression with survival in patients with NSCLC. Thirteen studies, including 2,274 patients, were subjected to final analysis. The rate of decreased E-cadherin expression was 47.6 % overall and 41.4 % for stage I disease. The combined hazard ratio (HR) was 1.41 (95 % CI 0.18-1.65; P = 0.001), indicating that decreased E-cadherin expression had an unfavorable impact on the survival of patients with NSCLC. Further, in the stratified analysis by ethnicity, the combined HR in Asians was 1.49 (95 % CI 1.27-1.71) and in non-Asians was 1.01 (95 % CI 1.00-1.02). However, when only the stage I studies were considered, the combined HR was 1.19 (95 % CI 0.90-1.47; P = 0.576), suggesting that decreased E-cadherin expression has no impact on survival. Decreased E-cadherin expression was associated with poor survival in patients with NSCLC, especially among Asians, but was not significantly correlated with survival for stage I NSCLC patients.
