Abstract:Epigenetics refer to the collective heritable changes in phenotype due to processes that arise independently of primary DNA sequence. It is comprised of DNA methylation, histone proteins modifications, non-coding and so on. Aberrant epigenetic modifications are involved in the development of neurodegenerative disorders. Recently, many epigenetics-based medications have been developed and provided us new strategies for treatment of those diseases, including Alzheimer's disease, Parkinson's disease and Huntington's disease. In this review, we would like to summarize the recent studies of epigenetic modification in neurodegeneration, mainly focusing on DNA methylation and histone acetylation and exploring the possibility of using epigenetics-based therapeutics to treat neurodegenerative disorders.

Small laboratory animals, like rodents, are commonly used in brain research and provide abundant models of human brain diseases. The development of high-resolution small-animal PET and various radiotracers together with sophisticated methods for analyzing functional brain imaging data have accelerated research on brain function and neurotransmitter release during behavioral tasks in rodents. In this review, we first summarize advances in the methodology of cognitive research brought about by the development of sophisticated methods for whole-brain imaging analysis and improvements in neuroimaging protocols. Then, we discuss basic mechanisms related to metabolic changes and the expression of neurotransmitters in various brain areas during task-induced neural activity. In particular, we discuss glucose metabolism imaging and brain receptor imaging for various receptor systems. Finally, we discuss the current status and future perspectives. Mechanisms of neurotransmitter expression will probably become an increasingly important field of study in the future, leading to more collaboration between investigators in fields such as computational and theoretical neuroscience.

The source and metabolism, the general situation and possible mechanism of improvement of learning and memory of PUFAs were discussed. The results indicated that PUFAs were significant component of brain tissue, and played an extremely important role in growth,me-tabolism and health,especially in strengthening learning and memory of brain. Researching on mechanism of PUFAs in improving the learning and memory could not only provide scientific basis for the develop-ment of health products,medicines etc,but also provide a breakthrough in treatment of diseases related to brain.

Abstract:The possibility of expanding memory or cognitive capabilities above the levels in high functioning individuals is a topic of intense discussion among scientists and in society at large. The majority of animal studies use behavioral endpoint measures; this has produced valuable information but limited predictability for human outcomes. Accordingly, several groups are pursuing a complementary strategy with treatments targeting synaptic events associated with memory encoding or forebrain network operations. Transcription and translation figure prominently in substrate work directed at enhancement. Notably, the question of why new proteins would be needed for a now-forming memory given that learning-driven synthesis presumably occurred throughout the immediate past has been largely ignored. Despite this conceptual problem, and some controversy, recent studies have reinvigorated the idea that selective gene manipulation is a plausible route to enhancement. Efforts to improve memory by facilitating synaptic encoding of information have also progressed, in part due of breakthroughs on mechanisms that stabilize learning-related, long-term potentiation (LTP). These advances point to a reductionistic hypothesis for a diversity of experimental results on enhancement, and identify under-explored possibilities. Cognitive enhancement remains an elusive goal, in part due to the difficulty of defining the target. The popular view of cognition as a collection of definable computations seems to miss the fluid, integrative process experienced by high functioning individuals. The neurobiological approach obviates these psychological issues to directly test the consequences of improving throughput in networks underlying higher order behaviors. The few relevant studies testing drugs that selectively promote excitatory transmission indicate that it is possible to expand cortical networks engaged by complex tasks and that this is accompanied by capabilities not found in normal animals.

Monitoring these compounds is pivotal, yet the intricate nature of the central nervous system poses challenges. Researchers have devised methods, notably electrochemical sensing with micro-nanoscale electrodes, offering high-resolution monitoring despite low concentrations and rapid changes. Implantable sensors enable precise detection in brain tissues with minimal damage, while microdialysis-coupled platforms allow in vivo sampling and subsequent in vitro analysis, addressing the selectivity issues seen in other methods. While lacking temporal resolution, techniques like HPLC and CE complement electrochemical sensing's selectivity, particularly for structurally similar neurochemicals. This review covers essential neurochemicals and explores miniaturized electrochemical sensors for brain analysis, emphasizing microdialysis integration. It discusses the pros and cons of these techniques, forecasting electrochemical sensing's future in neuroscience research. Overall, this comprehensive review outlines the evolution, strengths, and potential applications of electrochemical sensing in the study of neurochemicals, offering insights into future advancements in the field.

Abstract:Cannabinoids and the endocannabinoid system (ECS) have been intensively studied for their neuroregulatory roles in the central nervous system (CNS), especially in regulating learning and memory. However, many experimental and clinical studies obtained conflicting results indicating a complex network of interaction underlying the regulation of learning and memory by different cannabinoids and the ECS. The ECS influences neuronal synaptic communications, and therefore may exert different regulation via their different impact on other neurotransmitters. The monoaminergic system includes a variety of neurotransmitters, such as dopamine, norepinephrine, and serotonin, which play important roles in regulating mood, cognition, and reward. The interaction among cannabinoids, ECS and the monoaminergic system has drawn particular attention, especially their contributions to learning and memory. In this review, we summarized the current understanding of how cannabinoids, ECS and the monoaminergic system contribute to the process of learning and memory, and discussed the influences of monoaminergic neurotransmission by cannabinoids and ECS during this process.

Abstract:Memory is crucial for the acquisition of the tremendous amount of knowledge and skills and thus it is the basis of human survival and development. Then, how do learning and memory performance and neural mechanisms change with age? Around the core issue, this article first summarized the research results in recent years, especially those about some important memory subsystems such as working memory and long-term memory (including explicit and implicit memory). Then we revealed the research status in the field of developmental neuroscience of memory and predicted the future direction. On the basis of the work mentioned above, we put forward following suggestions to promote research work in China: (i) making further studies on the neural mechanisms of learning and memory and solving critical problems in this field; (ii) strengthening research on brain mechanisms of learning and memory of children and the elderly; (iii) paying attention to longitudinal studies; (iv) establishing more comprehensive brain-behavior models; (v) strengthening applied studies.

Abstract:Memory is a way for people to acquire information through personal experiences and from media such as books, TV, and the internet. Relying on memory and the ability to gain new ones, the human brain is capable of adapting to survive in the ever-changing environment and can create marvels and build objects out of nature. Therefore, it is memory that creates modern civilization. Memory consists of synapses and neural ensembles in the brain which integrate multisensory information both spatially and temporally. Thus, memory bridges the link between cognitive domain information and physical domain neural connections, making them easier to track and study. Memory is the foundation of the brain's cognitive functions. Consequently, research into how the brain stores and retrieves memories reveals not only how the brain works to generate intelligence, but also suggests possible avenues for creating efficient machine learning algorithms that have evolved over millions of years. Recent breakthroughs in biotechnology have led to discoveries about the mechanisms underlying memory formation and retrieval, providing insight into neural memories at the cellular and synaptic levels as well as understanding neural coding for circuitry integration and operation. Particularly, the discovery of cellular memory engrams in the neocortex has revealed a highly parallel arrangement of memory circuits, which are located in layer ll of widely distributed cortical areas, including sensory cortices, primary visual cortices, and retrosplenial cortices. In this review, we summarize the most recent findings on memory engrams in the hippocampus and neocortex. We highlight the correlation between neural oscillations and cortical engram activities during memory formation. We also explore the evidence that long-range synchrony of theta-coupled gamma oscillations plays a key role in modulating cortical engram activity to create a brain-wide memory network. We explain how memory engrams in the neocortex are integrated and organized by indexing signals from the hippocampus, allowing memories to be registered allocated and retrieved in the brain. Lastly, we shed light on the molecular mechanisms especially epigenetic regulators involved in the physical regulation of memory persistency and flexibility. Deficiencies in the epigenetic regulation of these engrams may lead to memory deficits in various brain diseases. Additionally, the laminar-specific distribution of memory engrams in local cortical columns and the global organization of memory traces in distinct functional cortical regions may inspire the development of novel computing algorithms in machine learning to achieve brain-level intelligence. Taken together, the advances in methodology are paving the way for us to directly investigate the neural mechanisms of memory storage and retrieval in mammalian brain, giving insight into human cognition and inspiring new approaches in machine learning.

Abstract:Alzheimer's disease (AD) is the most frequent cause of cognitive impairment in middle-aged and older populations. There is a lack of drugs that demonstrate significant efficacy in AD, so the study of the pathogenesis of AD is of great importance. More efficacious interventions are needed, as reflected by our population's fast aging. Synaptic plasticity is the capacity of neurons to adjust their connections, and it is strongly tied to learning and memory, cognitive function, and brain injury recovery. Changes in synaptic strength, such as long-term potentiation (LTP) or inhibition (LTD), are thought to represent the biological foundation of the early stages of learning and memory. The results of numerous studies confirm that neurotransmitters and their receptors play an important role in the regulation of synaptic plasticity. However, so far, there is no definite correlation between the function of neurotransmitters in aberrant neural oscillation and AD-related cognitive impairment. We summarized the AD process to understand the impact of neurotransmitters in the progression and pathogenesis of AD, including the current status of neurotransmitter target drugs, and the latest evidence of neurotransmitters' function and changes in the AD process.

Abstract:Activity-dependent synaptic plasticity has since long been proposed to represent the subcellular substrate of learning and memory, one of the most important behavioral processes through which we adapt to our environment. Despite the undisputed importance of synaptic plasticity for brain function, its exact contribution to learning processes in the context of cellular and connectivity modifications remains obscure. Causally bridging synaptic and behavioral modifications indeed remains limited by the available tools to measure and control synaptic strength and plasticity in vivo under behaviorally relevant conditions. After a brief summary of the current state of knowledge of the links between synaptic plasticity and learning, we will review and discuss the available and desired tools to progress in this endeavor.

Abstract:Animals acquire motor skills to better survive and adapt to a changing environment. The ability to learn novel motor actions without disturbing learned ones is essential to maintaining a broad motor repertoire. During motor learning, the brain makes a series of adjustments to build novel sensory-motor relationships that are stored within specific circuits for long-term retention. The neural mechanism of learning novel motor actions and transforming them into long-term memory still remains unclear. Here we review the latest findings with regard to the contributions of various brain subregions, cell types, and neurotransmitters to motor learning. Aiming to seek therapeutic strategies to restore the motor memory in relative neurodegenerative disorders, we also briefly describe the common experimental tests and manipulations for motor memory in rodents.

Abstract:Long-term potentiation (LTP) and long-term depression (LTD) are two major forms of synaptic plasticity that are widely considered as important cellular models of learning and memory. Metaplasticity is defined as the plasticity of synaptic plasticity and thus is an advanced form of plasticity. The history of synaptic activity can affect the subsequent synaptic plasticity induction. Therefore, it is important to study metaplasticity to explore new mechanisms underlying various brain functions including learning and memory. Since the concept of metaplasticity was proposed, it has aroused widespread concerns and attracted numerous researchers to dig more details on this topic. These new-found experimental phenomena and cellular mechanisms have established the basis of theoretical studies on metaplasticity. In recent years, researchers have found that metaplasticity can not only affect the synaptic plasticity, but also regulate the neural network to encode specific content and enhance the learning and memory. These findings have greatly enriched our knowledge on plasticity and opened a new route to study the mechanism of learning and memory. In this review, we discuss the recent progress on metaplasticity on following three aspects: (1) the molecular mechanisms of metaplasticity; (2) the role of metaplasticity in learning and memory; and (3) the outlook of future study on metaplasticity.

Abstract:Recent studies have reported the benefits of food-derived peptides for memory dysfunction. Beyond the physiological effects of peptides, their bioavailability to the brain still remains unclear since the blood-brain barrier (BBB) strictly controls the transportation of compounds to the brain. Here, updated transportation studies on BBB transportable peptides are introduced and evaluated using in vitro BBB models, in situ perfusion, and in vivo mouse experiments. Additionally, the mechanisms of action of brain health peptides in relation to the pathogenesis of neurodegenerative diseases, particularly Alzheimer's disease, are discussed. This discussion follows a summary of bioactive peptides with neuroprotective effects that can improve cognitive decline through various mechanisms, including anti-inflammatory, antioxidative, anti-amyloid β aggregation, and neurotransmitter regulation.

Abstract:Biological research has unraveled many of the molecular and cellular mechanisms involved in the formation of long-lasting memory, providing new opportunities for the development of cognitive-enhancing drugs. Studies of drug enhancement of cognition have benefited from the use of pharmacological treatments given after learning, allowing the investigation of mechanisms regulating the consolidation phase of memory. Modulatory systems influencing consolidation processes include stress hormones and several neurotransmitter and neuropeptide systems. Here, we review some of the findings on memory enhancement by drug administration in animal models, and discuss their implications for the development of cognitive enhancers.

Abstract:Extensive research has shed light on the cellular and functional underpinnings of higher cognition as influenced by the prefrontal cortex. Neurotransmitters act as key regulatory molecules within the PFC to assist with synchronizing cognitive state and arousal levels. The monoamine family of neurotransmitters, including dopamine, serotonin, and norepinephrine, play multifaceted roles in the cognitive processes behind learning and memory. The present review explores the organization and signaling patterns of monoamines within the PFC, as well as elucidates the numerous roles played by monoamines in learning and higher cognitive function.
