Abstract:Cognitive neuroscience, which studies the biological basis of mental processes, widely uses neuroimaging technologies like functional magnetic resonance imaging and positron emission tomography (PET) to study the human brain. Small laboratory animals, like rodents, are commonly used in brain research and provide abundant models of human brain diseases. The development of high-resolution small-animal PET and various radiotracers together with sophisticated methods for analyzing functional brain imaging data have accelerated research on brain function and neurotransmitter release during behavioral tasks in rodents. In this review, we first summarize advances in the methodology of cognitive research brought about by the development of sophisticated methods for whole-brain imaging analysis and improvements in neuroimaging protocols. Then, we discuss basic mechanisms related to metabolic changes and the expression of neurotransmitters in various brain areas during task-induced neural activity. In particular, we discuss glucose metabolism imaging and brain receptor imaging for various receptor systems. Finally, we discuss the current status and future perspectives. Mechanisms of neurotransmitter expression will probably become an increasingly important field of study in the future, leading to more collaboration between investigators in fields such as computational and theoretical neuroscience.

Abstract:Molecular imaging develops faster in the field of neuroimaging of ischemic stroke.Many studies have shown that its values in the aspects of cerebral blood flow monitoring during ischemic stroke and brain tissue metabolism.Ischemic penumbra is the basis of reperfusion therapy for acute ischemic stroke.The imaging methods,including magnetic resonance perfusion-weighted imaging/diffusion-weighted imaging mismatch,have been widely used to reveal ischemic penumbra because of their fast and strong availability.However,they can only reveal the blood flow of penumbra tissue.By contrast,molecular imaging technology can describe and measure the in vivo biological processes on the cellular and molecular levels,thus it can more accurately reveal the penumbral tissue.

Abstract:With the speeding tendency of aging society, human neurological disorders have posed an ever increasing threat to public health care. Human neurological diseases include ischemic brain injury, Alzheimer's disease, Parkinson's disease, and spinal cord injury, which are induced by impairment or specific degeneration of different types of neurons in central nervous system. Currently, there are no more effective treatments against these diseases. Traditional Chinese medicine (TCM) is focused on, which can provide new strategies for the therapy in neurological disorders. TCM, including Chinese herb medicine, acupuncture, and other nonmedication therapies, has its unique therapies in treating neurological diseases. In order to improve the treatment of these disorders by optimizing strategies using TCM and evaluate the therapeutic effects, we have summarized molecular imaging, a new promising technology, to assess noninvasively disease specific in cellular and molecular levels of living models in vivo, that was applied in TCM therapy for neurological diseases. In this review, we mainly focus on applying diverse molecular imaging methodologies in different TCM therapies and monitoring neurological disease, and unveiling the mysteries of TCM.

Abstract:Our understanding on human neurodegenerative disease was previously limited to clinical data and inferences about the underlying pathology based on histopathological examination. Animal models and in vitro experiments have provided evidence for a cell-autonomous and a non-cell-autonomous mechanism for the accumulation of neuropathology. Combining modern neuroimaging tools to identify distinct neural networks (connectomics) with target-specific positron emission tomography (PET) tracers is an emerging and vibrant field of research with the potential to examine the contributions of cell-autonomous and non-cell-autonomous mechanisms to the spread of pathology. The evidence provided here suggests that both cell-autonomous and non-cell-autonomous processes relate to the observed in vivo characteristics of protein pathology and neurodegeneration across the disease spectrum. We propose a synergistic model of cell-autonomous and non-cell-autonomous accounts that integrates the most critical factors (i.e., protein strain, susceptible cell feature and connectome) contributing to the development of neuronal dysfunction and in turn produces the observed clinical phenotypes. We believe that a timely and longitudinal pursuit of such research programs will greatly advance our understanding of the complex mechanisms driving human neurodegenerative diseases.

Abstract:Brain is the central organ of the human nervous system with complex functionality and complicated structure. The chemicals in brain play important roles in metabolism and signal transduction. It is a challenge for scientists to characterize and analyze these chemicals due to their diversity in chemical property and concentration. Mass spectrometry imaging as a new technique is able to screen the spatial distribution of various molecules simultaneously without labeling and has been broadly applied in the study of neurodegenerative disease, brain tumor and pharmacokinetic in brain. This review introduces the principle and new technique of mass spectrometry imaging, and focuses on its typical applications in neuroscience published in recent years.

Abstract:Analyzing the complex structures and functions of brain is the key issue to understanding the physiological and pathological processes. Although neuronal morphology and local distribution of neurons/blood vessels in the brain have been known, the subcellular structures of cells remain challenging, especially in the live brain. In addition, the complicated brain functions involve numerous functional molecules, but the concentrations, distributions and interactions of these molecules in the brain are still poorly understood. In this review, frontier techniques available for multiscale structure imaging from organelles to the whole brain are first overviewed, including magnetic resonance imaging (MRI), computed tomography (CT), positron emission tomography (PET), serial-section electron microscopy (ssEM), light microscopy (LM) and synchrotron-based X-ray microscopy (XRM). Specially, XRM for three-dimensional (3D) imaging of large-scale brain tissue with high resolution and fast imaging speed is highlighted. Additionally, the development of elegant methods for acquisition of brain functions from electrical/chemical signals in the brain is outlined. In particular, the new electrophysiology technologies for neural recordings at the single-neuron level and in the brain are also summarized. We also focus on the construction of electrochemical probes based on dual-recognition strategy and surface/interface chemistry for determination of chemical species in the brain with high selectivity and long-term stability, as well as electrochemophysiological microarray for simultaneously recording of electrochemical and electrophysiological signals in the brain. Moreover, the recent development of brain MRI probes with high contrast-to-noise ratio (CNR) and sensitivity based on hyperpolarized techniques and multi-nuclear chemistry is introduced. Furthermore, multiple optical probes and instruments, especially the optophysiological Raman probes and fiber Raman photometry, for imaging and biosensing in live brain are emphasized. Finally, a brief perspective on existing challenges and further research development is provided.

Abstract:Neuronal synapses are functional nodes in neural circuits. Their organization and activity define an individual's level of intelligence, emotional state and mental health. Changes in the structure and efficacy of synapses are the biological basis of learning and memory. However, investigation of the molecular architecture of synapses has been impeded by the lack of efficient techniques with sufficient resolution. Recent developments in state-of-the-art nano-imaging techniques have opened up a new window for dissecting the molecular organization of neuronal synapses with unprecedented resolution. Here, we review recent technological advances in nano-imaging techniques as well as their applications to the study of synapses, emphasizing super-resolution light microscopy and 3-dimensional electron tomography.

Abstract:The olfactory system of organisms serves as a genetically and anatomically model for studying how sensory input can be translated into behavior output. Some neurologic diseases are considered to be related to olfactory disturbance, especially Alzheimer's disease, Parkinson's disease, multiple sclerosis, and so forth. However, it is still unclear how the olfactory system affects disease generation processes and olfaction delivery processes. Molecular imaging, a modern multidisciplinary technology, can provide valid tools for the early detection and characterization of diseases, evaluation of treatment, and study of biological processes in living subjects, since molecular imaging applies specific molecular probes as a novel approach to produce special data to study biological processes in cellular and subcellular levels. Recently, molecular imaging plays a key role in studying the activation of olfactory system, thus it could help to prevent or delay some diseases. Herein, we present a comprehensive review on the research progress of the imaging probes for visualizing olfactory system, which is classified on different imaging modalities, including PET, MRI, and optical imaging. Additionally, the probes' design, sensing mechanism, and biological application are discussed. Finally, we provide an outlook for future studies in this field.

Abstract:Modern brain imaging technologies play essential roles in our understanding of brain information processing and the mechanisms of brain disorders. Magnetic Resonance Imaging (MRI) and Diffusion Tensor Imaging (DTI) can image the anatomy and structure of the brain. In addition, functional MRI (fMRI) can identify active regions, patterns of functional connectivities and functional networks during either tasks that are specifically related to various aspects of brain function or during the resting state. The merging of such structural and functional information obtained from brain imaging may be able to enhance our understanding of how the brain works and how its diseases can occur. In this paper, we will review advances in both methodologies and clinical applications of multimodal MRI technologies, including MRI, DTI, and fMRI. We will also give our perspectives for the future in these fields. The ultimate goal of our study is to find early biomarkers based on multimodal neuroimages and genome datasets for brain disorders. More importantly, future studies should focus on detecting exactly where and how these brain disorders affect the human brain. It would also be also very interesting to identify the genetic basis of the anatomical and functional abnormalities in the brains of people who have neurological and psychiatric disorders. We believe that we can use brain images to obtain effective biomarkers for various brain disorders with the aid of developing computational methods and models.

Abstract:Unlocking the secrets of the brain is a task fraught with complexity and challenge - not least due to the intricacy of the circuits involved. With advancements in the scale and precision of scientific technologies, we are increasingly equipped to explore how these components interact to produce a vast range of outputs that constitute function and disease. Here, an insight is offered into key areas in which the marriage of neuroscience and nanotechnology has revolutionized the industry. The evolution of ever more sophisticated nanomaterials culminates in network-operant functionalized agents. In turn, these materials contribute to novel diagnostic and therapeutic strategies, including drug delivery, neuroprotection, neural regeneration, neuroimaging and neurosurgery. Further, the entrance of nanotechnology into future research arenas including optogenetics, molecular/ion sensing and monitoring, and piezoelectric effects is discussed. Finally, considerations in nanoneurotoxicity, the main barrier to clinical translation, are reviewed, and direction for future perspectives is provided.

This article reviews the clinical applications and new advances for nuclear medicine,nuclear magnetic resonance,optical and multimodal molecular imaging in brain tumor diagnosis and therapy.

Abstract:When neuroscience’s focus moves from molecular and cellular level to systems level, information technology mixes in and cultivates a new branch neuroinformatics. Especially under the investments of brain initiatives all around the world, brain atlases and connectomics are identified as the substructure to understand the brain. We think it is time to call for a potential interdisciplinary subject, brainsmatics, referring to brain-wide spatial informatics science and emphasizing on precise positioning information affiliated to brain-wide connectome, genome, proteome, transcriptome, metabolome, etc. Brainsmatics methodology includes tracing, surveying, visualizing, and analyzing brain-wide spatial information. Among all imaging techniques, optical imaging is the most appropriate solution to achieve whole-brain connectome in consistent single-neuron resolution. This review aims to introduce contributions of optical imaging to brainsmatics studies, especially the major strategies applied in tracing and surveying processes. After discussions on the state-of-the-art technology, the development objectives of optical imaging in brainsmatics field are suggested. We call for a global contribution to the brainsmatics field from all related communities such as neuroscientists, biologists, engineers, programmers, chemists, mathematicians, physicists, clinicians, pharmacists, etc. As the leading approach, optical imaging will, in turn, benefit from the prosperous development of brainsmatics.

Abstract:Imaging technologies for the analysis of the central nervous system are rapidly developing. Matrix-assisted laser desorption/ionization time-of-flight mass spectrometry imaging, tracer-based magnetic resonance imaging, CLARITY technology and optogenetics can be used to visualize small molecules in brain tissues, the interstitial system of the brain and neuronal circuits in whole-brain samples. These tools serve as powerful technical means to explore the mechanisms underlying disease models and to evaluate the effects of drugs. Here, we review the constituting principles of these imaging techniques and describe their applications in the field of neuroscience.

Abstract:The intricate mesh of neural connections in the central nervous system (CNS) necessitates high-resolution observations to gain insights into cellular composition and molecular interactions. Nevertheless, nanoscale imaging within a specified region may fail to capture the overarching functionality and systemic state. Traditional microscopes are somewhat inadequate in satiating the need for multi-scale imaging. In light of this, X-ray-based imaging and analysis techniques have surfaced as a formidable contender in bridging this gap in neuroscience. However, a comprehensive review evaluating the progression of these applications is notably absent. This article aims to provide an extensive overview of recent advancements in X-ray-based techniques, emphasizing innovative modalities and their applications in neuroscience. These promising techniques harbor significant potential to demystify the complexities inherent in the CNS, augmenting our understanding of brain functionality and neurological disorders. Finally, we also contemplate the impending challenges and future trajectory of X-ray-based neuroimaging technologies.
