Abstract:Herein, we report a silver-catalyzed halogenation and electrophilic cyclization reaction based on the trifunctionalization of terminal alkynols with NBS or iodine monochloride in a step-efficient synthetic way to produce homo/heterodihalogenated naphthalene derivatives bearing two different halogen atoms in moderate to good yields. This methodology readily accommodates various functional groups, including electron-withdrawing nitro, cyano, acid-sensitive dioxazolyl, and alkoxy groups.

Abstract:Dual activation of both C-I and vicinal C-H bonds of diaryliodonium salts allowing for diarylation is a subject of rapid construction of π-extended frameworks. Here, we report palladium-catalyzed cascade of C8-arylation/intramolecular Friedel-Crafts acylation of α-naphthoic acids in the synthesis of benzanthrone derivatives. The step-economical protocol tolerates various substrates, which resulted in a potential molecular library for developing functional polycyclic scaffolds. The approach relies on the synergistic action of strong acid with palladium catalysts to form two bonds in a one-pot procedure.

Abstract:An efficient sequential intramolecular hydroamination/asymmetric hydrogenation reaction under catalysis of a single chiral ruthenium complex or a binary system consisting of achiral gold complex and chiral ruthenium complex has been reported. A diverse range of enantioen-riched benzo-fused N-heterocycles, including 1,2,3,4-tetrahydroquinoline, indoline, and 2,3,4,5-tetrahydro-1H-benzo[b]-azepine derivatives, were obtained from anilino-alkynes in high yields (up to 98%) with moderate to excellent enantioselectivities (up to 98% ee) under mild conditions. This protocol features good functional group tolerance and high atom economy. Furthermore, this catalytic protocol is applicable to gram-scale synthesis of a naturally occurring alkaloid, (-)-Angustureine.

Abstract:We have established an efficient ruthenium(ii)-catalyzed cis carbohalogenation and carbooxygenation cyclization of internal alkyne-tethered cyclohexadienones with halide and acetate ions, which produces a series of alkenyl halides and acetates with hydrobenzofuran skeletons in high yields and features high atom economy and regio- and stereoselectivity. Furthermore, the obtained hydrobenzofuran products can easily undergo aromatization and subsequent cross-coupling reactions, which provide a novel and stereoselective approach to synthesize tetrasubstituted alkenes, particularly diarylalkenes and triarylalkenes.

Abstract:AbstractThe cover picture shows the efficient synthesis of polysubstituted naphthalene derivatives and the mechanism, which proceeds through the gold‐catalyzed cyclization of 1‐arylalka‐2,3‐dienyl acetates. The six‐membered cyclohexenyl gold species is formed from the coordination of the allene moiety to the gold followed by nucleophilic attack of the electron‐rich benzene to the metal‐activated electrophilic C=C bond. Subsequent elimination of acetic acid affords the β‐naphthyl gold intermediate, which may be trapped by iodonolysis to release the gold catalyst into the catalytic cycle and afford the target iodonaphthalene. Details are discussed in the article by S. Ma et al. on p. 6545 ff. The background picture depicts the typical scenery of West Lake in Hangzhou during the summer season where the lake is covered with lotus leaves. Zhejiang University, a university with a history of over 110 years, is located very close to West Lake.

Abstract:The development of protocols for constructing chiral medium-sized heterocycles with high efficiency and excellent stereocontrol is of great interest owing to their ubiquitous occurrence in natural products and biologically active pharmaceuticals. Nonetheless, current synthetic approaches are limited due to unfavorable enthalpy and entropy factors, as well as transannular interactions. The present work addresses this issue by designing an asymmetric allylation/ring expansion reaction of 2-(1-hydroxyallyl)phenols and cyclobutanone carboxamides enabled by sequential iridium/zinc/bifunctional squaramide catalysis, affording a series of 8-membered benzo[b]oxocines in high yields with high diastereo- and enantioselectivities. Mechanistic investigation reveals that the enantioselectivity is controlled by the chiral iridium catalyst, while density functional theory calculations demonstrate that the diastereoselectivity is controlled by the chiral bifunctional squaramide catalyst. Abstract:A domino annulation/oxidation of heterocyclic ketene aminals (HKAs) and 2-aminochalcones has been developed for the selective synthesis of poly-substituted benzo[f]imidazo[2,1-a][2,7]naphthyridines and 3-azaheterocyclic substituted 2-arylquinolines. These reactions proceed well under mild conditions without any additives. Plausible mechanisms for such a polycyclic ring system assembly were also proposed. Moreover, benzo[f]imidazo[2,1-a][2,7]naphthyridine 3g displayed a fluorescence effect, demonstrating the potential applications in organic optical materials.

This transformation not only features transition-metal free and without using other bases and additives but also provides a new synthetic application for ortho-alkynyl benzyl alcohols, i.e., as C6 synthons for the construction of PAHs.

Abstract:Efficient strategies to assemble enantioenriched pyridine derivatives are highly important, given their significance in both synthetic and medicinal chemistry. Here, we report an enantioselective nickel-catalyzed intermolecular [2 + 2 + 2] cycloaddition of alkyne-tethered malononitriles with alkynes for the synthesis of densely substituted pyridines. The α-all-carbon quaternary center adjacent to pyridine is introduced by desymmetrizing the two cyano groups of disubstituted malononitriles. Notably, terminal alkynes are also tolerated with good regioselectivity to afford tetrasubstituted pyridines. Zinc halide is essential to enable the occurrence of the transformation by promoting the hetero-cyclometallation step. The reaction uses bio-renewable 2-MeTHF as the solvent and features mild reaction conditions, good functional group compatibilities, and enantioselectivities. This study offers a straightforward approach to the valuable enantioenriched heteroarenes from feedstock chemicals by forming three bonds and one quaternary stereocenter simultaneously in a single reaction step.

Abstract:4H‐Pyran units are frequently present in molecules with significant biological and pharmaceutical activities. Herein, we present the first enantioselective formal [3+3] cycloaddition between 2‐(1‐alkynyl)‐2‐alken‐1‐ones and β‐keto esters catalyzed by a cyclohexyldiamine‐based thiourea‐tertiary amine bifunctional catalyst. Under the mild and eco‐friendly conditions, a wide range of polysubstituted 4H‐pyrans were obtained in moderate yields with good enantioselectivities.

Abstract:Polycyclic alkaloids bearing all-carbon quaternary centers possess a diversity of biological activities and are challenging targets in natural product synthesis. The development of a general and asymmetric catalytic method applicable to the efficient syntheses of a series of complex polycyclic alkaloids remains highly desirable in synthetic chemistry. Herein we describe an efficient palladium-catalyzed enantioselective dearomative cyclization which is capable of synthesizing two important classes of tricyclic nitrogen-containing skeleton, chiral dihydrophenanthridinone and dihydrocarbazolone derivatives bearing all-carbon quaternary centers, in excellent yields and enantioselectivities. The P-chiral monophosphorus ligand AntPhos is crucial for the reactivity and enantioselectivity, and the choice of the N-phosphoramide protecting group is essential for the desired chemoselectivity. This method has enabled the enantioselective total syntheses of three distinctive and challenging biologically important polycyclic alkaloids, specifically a concise and gram-scale synthesis of (-)-crinine, an efficient synthesis of indole alkaloid (-)-aspidospermidine and a formal enantioselective synthesis of (-)-minfiensine.

Abstract:An efficient protocol for the direct transformation of chrornan-4-ones to tricyclic fused pyridines with the skeleton of cassiarins, a family of alkaloids with antimalarial activity, was developed. Also, a general strategy for modular construction of polyheterocydes with diverse natural product-like skeletons was developed by using ketone-alkyne bifunctional substrates. These reactions involved a one-pot cascade oximation of ketones, rhodium-catalyzed C-H activation, and intermolecular/intramolecular alkyne annulations under mild conditions with high atom, step, and redox economy.
