Abstract:An easily prepared "turn-on" -type fluorescent chemosensor for mercury based on Rhodamine-B, which exhibits high sensitivity and selectivity over other metal ions in aqueous systems, was obtained. The distinctive wavelengths in the UV/ Vis absorption range can sense Hg 2+ , Cu 2+ , and Fe 3+ separately. Furthermore, this "turn-on"-type fluorescent sensor, upon the addition of Hg 2+ over other competitive species, was successfully applied to bioimaging in yeast and HeLa cells. The potential of these types of chemosensors for use in environmental and biological systems is great.

Abstract:In this work, a fluorescent sensing strategy was developed for the detection of mercury(II) ions (Hg2+) in aqueous solution with excellent sensitivity and selectivity using a target-induced DNAzyme cascade with catalytic and molecular beacons (CAMB). In order to construct the biosensor, a Mg2+-dependent DNAzyme was elaborately designed and artificially split into two separate oligonucleotide fragments. In the presence of Hg2+, the specific thymine-Hg2+-thymine (T-Hg2+-T) interaction induced the two fragments to produce the activated Mg2+-dependent DNAzyme, which would hybridize with a hairpin-structured MB substrate to form the CAMB system. Eventually, each target-induced activated DNAzyme could catalyze the cleavage of many MB substrates through true enzymatic multiple turnovers. This would significantly enhance the sensitivity of the Hg2+ sensing system and push the detection limit down to 0.2 nM within a 20 min assay time, much lower than those of most previously reported fluorescence assays. Owning to the strong coordination of Hg2+ to the T-T mismatched pairs, this proposed sensing system exhibited excellent selectivity for Hg2+ detection, even in the presence of 100 times of other interferential metal ions. Furthermore, the applicability of the biosensor for Hg2+ detection in river water samples was demonstrated with satisfactory results. These advantages endow the sensing strategy with a great potential for the simple, rapid, sensitive, and specific detection of Hg2+ from a wide range of real samples.

Abstract:A water-soluble, biocompatible, and fluorescent chemosensor (1) for label-free, simple, and fast detection of mercury ions (Hg(2+)) in aqueous solutions and in HepG2 cells with high selectivity is reported herein. Chelation of 1 with Hg(2+) results in the disappearance of its fluorescence emission at 350 nm and the appearance of a new emission at 405 nm. Selectivity and interference studies indicated that 1 could be selectively chelated by Hg(2+) without interference from other metal ions. Insight into the mechanisms responsible for its fluorescence effect was gained from ultrafast transient absorption spectroscopy. With these properties, 1 was successfully applied for imaging Hg(2+) in living cells and for removing Hg(2+) from river water. Moreover, we also constructed a simple device for fast and effective removal of Hg(2+) from contaminated liquid samples.

Abstract:A highly sensitive and selective, "turn-on" and simple Hg2+ biosensor is reported by using water-soluble graphene oxide (WSGO) and dye-labeled mercury(II)-specific oligonucleotide (MSO) probe. The probe is rich of thymine (T) and can readily form the stem-loop structure which consists of the T-Hg2+-T configuration. In the absence of Hg2+, the probe exists as a random coil conformation which can be readily adsorbed on the surface of WSGO by strong noncovalent binding of bases, as a result, the fluorescence of the dye labeled on the terminus of the MSO is strongly quenched by the efficient electron/energy transfer from the dye to WSGO. Upon addition of Hg2+, the formation of the T-Hg2+-T structure releases the MSO from the surface of WSGO, resulting in a restoration of the fluorescence of dye-labeled MSO probe. Based on this observation, a highly sensitive and selective Hg2+ sensor is developed, which can work with "turn-on" mode in aqueous solutions at room temperature. By using the fluorometric method, the limit of detection for Hg2+ can reach picomolar range (187 pmol.L-1), and it is demonstrated that the biosensor is highly selective and only minimally perturbed by a wide range of non-specific metal ions.
A Novel Camphor-Based “Turn-on” Fluorescent Probe with High Specificity and Sensitivity for Sensing Mercury(II) in Aqueous Medium and Its Bioimaging Application

Abstract:Precise detection of Hg2+ in an ecological environment and biological system is of great importance. Hence, in this paper, we have constructed a simple fluorescent turn-on probe CP-Hg toward Hg2+ based on natural and renewable camphor. A significantly enhanced green fluorescence was clearly observed only with the participation of Hg2+ due to the Hg2+-mediated deprotection reaction of thioacetal. The probe CP-Hg could also sensitively detect Hg2+ with a detection limit of 19.3 nM and a large Stokes shift of 153 nm in a near-perfect aqueous solution. The reaction mechanism was fully analyzed through 1H NMR (proton nuclear magnetic resonance), ESI-MS (electrospray ionization mass spectrometry), and DFT (density functional theory) calculations. Moreover, the CP-Hg-coated test strips were successfully fabricated for naked-eye detection of Hg2+. Abstract:Hg2+ has a significant hazardous impact on the environment and ecosystem. There is a great demand for new methods with high selectivity and sensitivity to determine mercury in life systems and environments. In this paper, a novel turn-on Hg2+ fluorescent probe has been reported with a naphthalimide group. The Hg2+ fluorescent probe was designed by the inspiration of the well-known specific Hg2+-triggered thioacetal deprotection reaction. A 1,2-dithioalkyl group was chosen as the specific recognition site of Hg2+. The probe showed weak fluorescence without Hg2+, and the color of the solution was light yellow. In the presence of Hg2+, the probe reacted specifically with the mercury ion to produce an aldehyde and emitted strong fluorescence, and the color of the solution also turned light green, thus realizing the monitoring of the mercury ion. The Hg2+ fluorescent probe showed outstanding sensitivity and selectivity toward Hg2+. Furthermore, the Hg2+ fluorescent probe could work in a wide pH range. The linear relationship between the fluorescence intensity at 510 nm and the concentration of Hg2+ was obtained in a range of Hg2+ concentration from 2.5 × 10–7 to 1.0 × 10–5 M. The detection limit was found to be 4.0 × 10–8 M for Hg2+. Furthermore, with little cell toxicity, the probe was successfully applied to the confocal image of Hg2+ in PC-12 cells.This article has not yet been cited by other publications.

Abstract:Mercury(II) ions have emerged as a widespread environmental hazard in recent decades. Despite different kinds of detection methods reported to sense Hg(2+) , it still remains a challenging task to develop new sensing molecules to replenish the fluorescence-based apparatus for Hg(2+) detection. This communication demonstrates a novel fluorescent sensor using UiO-66-NH2 and a T-rich FAM-labeled ssDNA as a hybrid system to detect Hg(2+) sensitively and selectively. To the best of our knowledge, it has rarely been reported that a MOF is utilized as the biosensing platform for Hg(2+) assay.

Abstract:A fluorescence turn-on chemosensor based on rhodamine B derivative (FD10) has been developed as a highly sensitive chemosensor for Hg 2+ . A prominent fluorescence enhancement was measured in the presence of Hg 2+ , which was in agreement with the changes in the absorption spectrum. Furthermore, by means of laser scanning fluorescence microscopy experiments, it was demonstrated that FD10 was cell-permeable and could be used as a fluorescent probe for monitoring Hg 2+ in living cells.

Abstract:Mercury (Hg), as a highly harmful environmental pollutant, poses severe ecological and health risks even at low concentrations. Accurate and sensitive methods for detecting Hg2+ ions in aquatic environments are highly needed. In this work, we developed a highly sensitive fluorescence sensor for Hg2+ detection with an integrated use of biosynthetic CdSe/CdS quantum dots (QDs) and liposome carrier signal amplification. To construct such a sensor, three single-stranded DNA probes were rationally designed based on the thymine-Hg2+-thymine (T-Hg2+-T) coordination chemical principles and by taking advantage of the biocompatibility and facile-modification properties of the biosynthetic QDs. Hg2+ could be determined in a range from 0.25 to 100 nM with a detection limit of 0.01 nM, which met the requirements of environmental sample detection. The sensor also exhibited a high selectivity for Hg2+ detection in the presence of other high-level metal ions. A satisfactory capacity of the sensor for detecting environmental samples including tap water, river water, and landfill leachate was also demonstrated. This work opens up a new application scenario for biosynthetic QDs and holds a great potential for environmental monitoring applications.

Abstract:By combination of the AIEE-active fluorophore and coordination binding group, a new selective and sensitive probe (1) for Hg2+ detection has been synthesized. 1 exhibits weak fluorescence in dilute solution, but exhibits dramatically enhanced emission in aggregation. In the solid state, a remarkable red-shifted emission and longer PL lifetime of 1 are present, which is caused by the formation of excimers due to the pi-pi stacking interactions of the coumarin groups. This AIEE effect of 1 is utilized to design probe for the detection of Hg2+ in a luminescence "turn-on" mode. The functionalization of the AIEE-active molecule with chelating group provides the potential binding sites for Hg2+. Abstract:A “signal-on” electrochemical sensing strategy was designed for highly sensitive and selective detection of mercury (II) via its induction to three-way junction of DNA (DNA-TWJ). The TWJ consisted of the capture probe that was self-assembled on a gold electrode surface through SAu bond, the signal probe that was labeled with ferrocene (Fc) and contained single T–T mismatch to capture probe, and an assistant probe for the formation of DNA-TWJ upon the presence of mercury (II). This process caused the Fc tag approaching the electrode for fast electron transfer and thus increased the oxidation current. The “signal-on” sensing method could detect Hg2+ ranging from 0.005 to 100nM. The assay was simple and fast. It showed potential application in on-site and real-time Hg2+ detection.

Abstract:The simply constructed fluorescent sensor with inexpensive reagents and low toxicity has attracted increasing attention contributing to its practical application. However, the common construction methods usually required a few building blocks and complex procedures, which is inconvenient for their further application. Herein, a simply constructed fluorescent Hg2+ sensor has been developed based on the intrinsic fluorescence quenching power of G-quadruplex. Two components, AGRO 100 and AMT, were used to construct the sensor. AMT was selected as the fluorescent probe because of its distinct merits. The free AMT emits strongly. However, the fluorescence of AMT could be quenched by G-quadruplex DNA. Additionally, AMT is less toxic and inexpensive. AGRO 100 acts as both the quencher and the capture sequence because it consists of G-rich sequences and T-T mismatched base pairs. The fluorescence of AMT could be quenched by the formed G-quadruplex structure of AGRO 100 in the presence of K+. In the presence of Hg2+, G-quadruplex structure of AGRO 100 was switched to hairpin DNA structure because T-T mismatched base pairs in AGRO 100 could specifically recognize and capture Hg2+ with high affinity. Thus, AMT was released and the fluorescence of AMT was recovered. The developed sensing system was successfully applied to detect Hg2+ in human serum with good recovery and reproducibility.Graphical abstract

Abstract:A new fluorescent sensor 1, based on the naphthalimide chromophore and thioether-rich crown receptor, exhibited dual signaling behaviors for Hg2+ and Ag+ in aqueous solution. Upon addition of Hg2+, the fluorescence intensity enhanced in a linear fashion with a quantum yield increase of about 5-fold. Moreover, with the 1-Hg2+ complex, Ag+ was easily recognized by a marked fluorescence quenching. The living cells image experiments demonstrate the value of sensor 1 in fluorescent visualization of Hg2+ ions in biological systems.

Abstract:Herein, a novel fluorescent sensor QPE based on quinolimide was developed, which displayed the highly selective response to Hg 2+ through the turn-on signal in aqueous media. The corresponding recognition mechanism of QPE for Hg 2+ was investigated by the titration experiments, Job's plots, HRMS, and DFT calculations analysis. QPE could detect Hg 2+ in a broad pH range (pH = 4-9) with the LOD of 20 nM and the LOQ of 66 nM. Meanwhile, QPE -loaded test strips were demonstrated in the visual detection of Hg 2+ . Importantly, QPE exhibited the reliable capability to track Hg 2+ in real water and soil samples and yeast cells, indicating that QPE could be used as a prospective tool for monitoring Hg 2+ in wide fields.
