Abstract:AIM To observe whether the exogenous human vascular endothelial growth factor hVEGF can express and secrete endothelial growth factor EGF or not after it is guided in normal dermal fibroblasts which is the base of gene transfected artificial skin of tissue engineering as a new substitute for wound healing. METHODS This experiment was conducted in Jilin University from June 2001 to June 2005. ① The recombinant mammalian expression plasmid vector containing hVEGF cDNA pcDNA3.0-hVEGF165 was amplified in DH5a then purified and finally identified by agarose gel electrophoresis analysis after digestion with restriction endonuclease EcoRⅠand XbaⅠ.②Skin biopsies were obtained from two neonatal rabbits at the level of clean animal chipped into small pieces 0.3 cm in width. Dermal fibroblasts were isolated and cultured in vitro by digestion with collagenaseⅠ. Fibroblasts were transfected with the plasmids of pcDNA3.0-VEGF165 using lipo-mediated transfection and selected with G418 for 2 to 4 weeks. The positive clones of transfected cells were obtained and proliferated in vitro. ③The level of secreted VEGF165 protein in supernatant was determined with enzyme-linked immunosorbent assay ELISA The expression of VEGF165 mRNA in transfected cells was analyzed with in situ hybridization Flow cytometerFCM was used to detect cell cycle and apoptosis The ultrastructure of transfected dermal fibroblasts was observed with transmission electron microscopeTEM. RESULTS ①Two DNA segments about 5.4 kb and 0.57 kb in length were obtained after digestion with EcoRⅠand XbaⅠ. It was confirmed that the obtained vector was the recombinant plasmid pcDNA3.0-VEGF165. ②The G418-resistant colonies were pooled to generate the stable transfected cells in all 35 wells in 15 transfecting tests. It indicated that the pcDNA3.0-VEGF165 could be efficiently introduced into dermal fibroblasts using lipo-mediated transfection. ③When hVEGF165 detected with ELISA in cell culture there was a high level of hVEGF165 expressed by transfected fibroblasts in 24 hours (3 280±2 054 ng/L) which descended gradually in 48 and 72 hours ④ In situ hybridization analysis showed that there were several positive cells expressing hVEGF mRNA in transfected fibroblasts while a mass of positive cells expressing hVEGF mRNA were found in selected fibroblasts. The clones of pcDNA3.0-hVEGF165 transfected fibroblasts could be selected successfully after treated with G418 with 2-4 weeks. ⑤FCM analysis showed that the proportion of S phase-cells increased while that of G1 phase-cells and G2 phase-cells decreased in pcDNA3.0-hVEGF165 transfected fibroblasts. It indicated that there were enhancements of both DNA synthesis and cell proliferation in the transfected cells. The apoptosis ratios of fibroblasts increased gradually which was 1.26% 2.64% and 17.35% in the groups before transfection 2 days and 12 passages after transfection respectively. ⑥The ultrastructure changes of transfected cells analyzed by TEM included many microvilli on the membrane the nucleus in curved shapes numerous rough endoplasmic reticulum and mitochondria in the cytoplasm as well as some special enveloped granues near the membrane. CONCLUSION pcDNA3.0-hVEGF165 plasmids had been transfected into fibroblasts and high level of hVEGF165 had been expressed in the transfected cells in a short period. As a new cell source it shows a good foreground in the field of gene transfected artificial skin of tissue engineering and clinical therapy for ischemic diseases and wounds healing.
