Expression of target gene in mesenchymal stem cells after transfection of basic fibroblast growth factor gene

Abstract:Aim: To observe the expression of adenovirus transfected basic fibroblast growth factor (bFGF) in rat mesenchymal stem cells (MSCs), and investigate the biological characteristics of bFGF function influencing MSCs. Methods: The experiment was carried out in the Medical Experimental Center of Second Affiliated Hospital of Sun Yat-sen University from March 2005 to February 2006. (1) Ten 4-week male SD rats of inbreed line were selected, and the MSCs were isolated and cultured with adherence method to construct the adenoviral vector containing bFGF (Ad.bFGF) gene. The MSCs were transfected with adenovirus. (2) After 2 passages, the cells were seeded onto 12-well plate, and the supernatant was removed when the cells grew to 90% fusion. The transfected adenovirus titer was arranged to 50-400. The transfected condition was observed under fluorescence microscope, and the transfection efficiency was tested by flow cytometry. (3) After 2 passages, the cells were put into a 6-well plate after digested with pankrin, with 5 × 105 cells each well. Two days later, the cells adhered on the coverslip and transfected Ad.bFGF and Ad.green fluorescent protein (GFP) with 200 titer; meanwhile, the control group was set up. The expression of bFGF was measured by immunohistochemical staining and western-blot; and ELISA was used to detect the concentration of bFGF in supernatant. The shape and growth conditon of the transfected MSCs were also examined. Results: (1) pcDNA3/bFGF identification: Adenovirus vector containing bFGF gene was constructed successfully. (2) Transfection efficiency of MSCs with Ad.GFP: After 48 hours transfection, the expression of GFP was significantly increased. When the titer ≤200, the efficiency was positively correlated with the dosage (P < 0.05), if the titer > 200, the efficiency was stable and more than 96%. (3) Expression in MSCs transfected with Ad. bFGF: Immunohistochemistry showed that it was expressed in MSCs plasma with brown staining, and the positive rate was 86%. But it could not be seen in the MSCs transfected by Ad.GFP and the control group. Western-blot showed bFGF expression in the transfected cells was obviously higher than that transfected by Ad.GFP and blank control. ELISA detection showed that the concentration of bFGF in the supernatant mounted to the peak level on the 15th day (1 467 ng/L) and decreased gradually thereafter, but still higher than those in the control group on the 28th day (441 ng/L, 128 ng/L, t=4.31, P < 0.01). (4) Effect of transfection with Ad.bFGF on the proliferation of MSCs: No obvious change was found in MSCs. Conclusion: Gene transfer technology mediated by adenoviral vector can transfect bFGF gene into MSCs with expression of exogenous protein, indicating MSCs are the ideal vectors for bFGF gene therapy.
