Abstract:Epsilon-poly-L-lysine (ε-PL) is an unusual biopolymer composed of L-lysine produced by several microorganisms, especially by the genus Streptomyces . Due to its excellent antimicrobial activity, good water solubility, high safety, and biodegradable nature, ε-PL with a GRAS status has been widely used in food and pharmaceutical industries. In the past years, studies have focused on the biotechnological production of ɛ-PL, the biosynthetic mechanism of microbial ɛ-PL, and its application. To provide new perspectives from recent advances, the review introduced the methods for the isolation of ɛ-PL producing strains and the biosynthetic mechanism of microbial ɛ-PL. We summarized the strategies for the improvement of ɛ-PL producing strains, including physical and chemical mutagenesis, ribosome engineering and gene engineering, and compared the different metabolic regulation strategies for improving ɛ-PL production, including medium optimization, nutrient supply, pH control, and dissolved oxygen control. Then, the downstream purification methods of ɛ-PL and its recent applications in food and medicine industries were introduced. Finally, we also proposed the potential challenges and the perspectives for the production of ε-PL.

Abstract:Poly(epsilon-L-lysine) (epsilon-PL) is an unusual biopolymer composed of L-lysine connected between alpha-carboxyl and epsilon-amino groups. It has been used as a preservative in food and cosmetics industries, drug carrier in medicines, and gene carrier in gene therapy. Modern biotechnology has significantly improved the synthetic efficiency of this novel homopoly( amino acid) on an industrial scale and has expanded its industrial applications. In the latest years, studies have focused on the biotechnological production and understanding the biosynthetic mechanism of microbial epsilon-PL. Herein, this review focuses on the current trends and future perspectives of microbial epsilon-PL. Information on the screening of epsilon-PL-producing strains, fermentative production of epsilon-PL, breeding of high-epsilon-PL-producing strains, genomic data of epsilon-PL-producing strains, biosynthetic mechanism of microbial epsilon-PL, and the control of molecular weight of microbial epsilon-PL is included. This review will contribute to the development of this novel homopoly(amino acid) and serve as a basis of studies on other biopolymers.

Abstract:ε-Poly-l-lysine (ε-PL) is a natural homo-poly(amino acid) which can be produced by microorganisms. With the advantages in broad-spectrum antimicrobial activity, biodegradability, and biocompatibility, ε-PL has been widely used as a preservative in the food industry. Different molecular architectures endow ε-PL and ε-PL-based materials with versatile applications. However, the microbial synthesis of ε-PL is currently limited by low efficiencies in genetic engineering and molecular architecture modification. This review presents recent advances in ε-PL production and molecular architecture modification of microbial ε-PL, with a focus on the current challenges and solutions for the improvement of the productivity and diversity of ε-PL. In addition, we highlight recent examples where ε-PL has been applied to expand the versability of edible films and nanoparticles in various applications. Commercial production and the challenges and future research directions in ε-PL biosynthesis are also discussed. Currently, although the main use of ε-PL is as a food preservative, ε-PL and ε-PL-based polymers have shown excellent application potential in biomedical fields. With the development of synthetic biology, the design and synthesis of ε-PL with a customized molecular architecture are possible in the near future. ε-PL-based polymers with specific functions will be a new trend in biopolymer manufacturing.

Abstract:The naturally occurring homo-polyamide biopolymer, ε-poly-L-lysine (ε-PL) consists of 25–35 L-lysine residues with amide linkages between α-carboxyl groups and ε-amino groups. ɛ-PL exhibits several useful properties because of its unusual structure, such as biodegradability, water solubility, no human toxicity, and broad-spectrum antibacterial activities; it is widely applied in the fields of food, medicine, clinical chemistry and electronics. However, current industrial production of ε-PL is only performed in a few countries. Based on an analysis of the physiological characteristics of ε-PL fermentation, current advances that enhance ε-PL fermentation, from strain improvement to product isolation are systematically reviewed, focusing on: (1) elucidating the metabolic pathway and regulatory mechanism of ε-PL synthesis; (2) enhancing biosynthetic performance through mutagenesis, fermentation optimization and metabolic engineering; and (3) understanding and improving the biological activity and functional properties of ε-PL. Finally, perspectives on engineering and exploiting ε-PL as a source material for the production of various advanced materials are also discussed, providing scientific guidelines for researchers to further improve the ε-PL fermentation process.

Abstract:ε-poly-L-lysine (ε-PL) is a naturally occurring poly(amino acid) of varying polymerization degree, which possesses excellent antimicrobial activity and has been widely used in food and pharmaceutical industries. To provide new perspectives from recent advances, this review compares several conventional and advanced strategies for the discovery of wild strains and development of high-producing strains, including isolation and culture-based traditional methods as well as genome mining and directed evolution. We also summarize process engineering approaches for improving production, including optimization of environmental conditions and utilization of industrial waste. Then, efficient downstream purification methods are described, including their drawbacks, followed by the brief introductions of proposed antimicrobial mechanisms of ε-PL and its recent applications. Finally, we discuss persistent challenges and future perspectives for the commercialization of ε-PL.

Abstract:Three kinds of poly amino acids, poly-γ-glutamic acid, poly(ɛ-L-lysine) and multi-L-arginyl-poly (L-aspartic acid) can be synthesized by enzymatic process independently from ribosomal protein biosynthesis pathways in microorganism. These biosynthesized polymers have attracted more and more attentions because of their unique properties and various applications. In this review, the current knowledge on the biosynthesis, biodegradations and applications of these three poly amino acids are summarized.

Abstract:ε-Poly-L-lysine (ε-PL) is a novel food biopreservative with broad antimicrobial activity. In the present study, single-factor experiments were applied to screen optimal carbon and nitrogen sources that affect the yield of a ε-PL-producing strain, Streptomyces violaceusniger, which was obtained in our laboratory recently. Box-Behnken design and response surface methodology (RSM) were adopted to derive a statistical model for optimizing the composition of the fermentation medium. The results showed that the main factors that affect ɛ-PL production were glucose, carbon source; peptone, organic nitrogen source; and (NH(4))(2)SO(4), inorganic nitrogen source. The optimum fermentation medium for ɛ-PL production of S. violaceusniger consisted of (g/L) glucose, 62.49; peptone, 5.71; (NH(4))(2)SO(4), 11.19; MgSO(4), 0.5; FeSO(4), 0.03; ZnSO(4), 0.04; KH(2)PO(4), 1.36; and Na(2)HPO(4).12H(2)O, 3.58. When cultured in the optimum medium, the ε-PL production was an average of 0.349 ± 0.025 mg/mL, which was 1.75 times higher than the initial medium.
Enhanced ε-Poly-L-Lysine Production in Streptomyces albulus through Multi-Omics-Guided Metabolic Engineering

Abstract:Safe and eco-friendly preservatives are crucial to preventing food spoilage and illnesses, as foodborne diseases caused by pathogens result in approximately 600 million cases of illness and 420,000 deaths annually. ε-Poly-L-lysine (ε-PL) is a novel food preservative widely used in many countries. However, its commercial application has been hindered by high costs and low production. In this study, ε-PL's biosynthetic capacity was enhanced in Streptomyces albulus WG608 through metabolic engineering guided by multi-omics techniques. Based on transcriptome and metabolome data, differentially expressed genes (fold change >2 or <0.5; p 1.2 or <0.8) were separately subjected to gene ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analysis. The integrative analysis of transcriptome, metabolome, and overexpression revealed the essential roles of isocitrate lyase, succinate dehydrogenase, flavoprotein subunit, diaminopimelate dehydrogenase, polyphosphate kinase, and polyP:AMP phosphotransferase in ε-PL biosynthesis. Subsequently, a strain with enhanced ATP supply, L-lysine supply, and ε-PL synthetase expression was constructed to improve its production. Finally, the resulting strain, S. albulus WME10, achieved an ε-PL production rate of 77.16 g/L in a 5 L bioreactor, which is the highest reported ɛ-PL production to date. These results suggest that the integrative analysis of the transcriptome and metabolome can facilitate the identification of key pathways and genetic elements affecting ε-PL synthesis, guiding further metabolic engineering and thus significantly enhancing ε-PL production. The method presented in this study could be applicable to other valuable natural antibacterial agents.

Abstract:ε-Poly-l-lysine (ε-PL) is a natural antimicrobial cationic peptide, which is generally recognized as safe for use as a food preservative. To date, the production capacity of strains that produce low-molecular weight ε-PL remains very low and thus unsuitable for industrial production. Here, we report a new low-molecular weight ε-PL-producing Kitasatospora aureofaciens strain. The ε-PL synthase gene of this strain was cloned into a high ε-PL-producing Streptomyces albulus strain. The resulting recombinant strain efficiently produced ε-PL with a molecular weight of 1.3-2.3 kDa and yielded of 23.6 g/L following fed-batch fermentation in a 5 L bioreactor. In addition, circular dichroism spectra showed that this ε-PL takes on a conformation similar to an antiparallel pleated-sheet. Moreover, it demonstrated better antimicrobial activity against yeast compared to the 3.2-4.5 kDa ε-PL. This study provides a highly efficient strategy for production of the low-molecular weight ε-PL, which helps to expand its potential applications.
Whole-cell biocatalysis for ε-poly-l-lysine production by a food-grade recombinant Bacillus subtilis

Abstract:ε-Poly-l-lysine (ε-PL), a natural food preservative with various advantages, is primarily produced by Streptomyces. It has attracted considerable attentions for the outstanding antibacterial activity, safety, heat stability, water solubility and other remarkable properties. In this study, a food-grade recombinant Bacillus subtilis was constructed for the biocatalysis of ε-PL. Firstly, the d-alanine racemase gene (alrA) was deleted from the genome of Bacillus subtilis 168 to construct an auxotrophic B. subtilis 168 (alrA-). Based on the shuttle plasmid pMA5, a food-grade plasmid pMA5a was constructed by replacing the genes of kanamycin resistance (Kanr) and ampicillin resistance (Ampr) with alrA and the gene encoding α-peptide of β-galactosidase (lacZα), respectively. Subsequently, codon-optimized ε-PL synthase gene (pls) and P-pls were ligated into pMA5a and transformed in E. coli DH5α and expressed in B. subtilis 168 (alrA-). Finally, the whole-cell biocatalysis conditions for ε-PL production by B. subtilis 168 (alrA-)/pMA5a-pls were optimized, and the optimal conditions were 30°C, pH 4, l-lysine concentration of 0.6 g/L, bacterial concentration of 15 % (w/v) and a catalytic time of 7 h. The ε-PL production reached a maximum of 0.33 ± 0.03 g/L. The product was verified to be ε-PL by HPLC and tricine-SDS-PAGE. The information obtained in this study shows critical reference for the food-grade heterologous expression of ε-PL.

Abstract:Poly-L-lysine (PLL) is a water-soluble biopolymer consist of repeating unit of L-lysine. It displays intrinsic non-antigenicity, antibacterial property, biocompatibility, and biodegradability, and facile synthesis procedure, therefore has attracted particular attention in various biomedical and pharmaceutical applications. Following a brief introduction to PLL synthesis, this review focuses on the significant advances of PLL and PLL-derived complexes utilized in cargo delivery, tissue adhesives, and antibacterial materials. Meanwhile, this review summarized the modification and optimization of PLL complexes for further clinical translational studies. The final section discusses the challenges and opportunities of PLL-based complexes in the biomedical areas.

Abstract:Over the past few decades, with the development of science and technology, the field of biomedicine has rapidly developed, especially with respect to biomedical materials. Low toxicity and good biocompatibility have always been key targets in the development and application of biomedical materials. As a degradable and environmentally friendly polymer, polylactic acid, also known as polylactide, is favored by researchers and has been used as a commercial material in various studies. Lactic acid, as a synthetic raw material of polylactic acid, can only be obtained by sugar fermentation. Good biocompatibility and biodegradability have led it to be approved by the U.S. Food and Drug Administration (FDA) as a biomedical material. Polylactic acid has good physical properties, and its modification can optimize its properties to a certain extent. Polylactic acid blocks and blends play significant roles in drug delivery, implants, and tissue engineering to great effect. This article describes the synthesis of polylactic acid (PLA) and its raw materials, physical properties, degradation, modification, and applications in the field of biomedicine. It aims to contribute to the important knowledge and development of PLA in biomedical applications.

Abstract:Polylysine is an important class of polyamino acids with a broad spectrum of applications in biomedical research and development. It can be divided into two classes, α-polylysine and ε-polylysine, the former is synthesized by artificial chemical synthesis and has limited applications due to its high toxicity, and the latter is produced by microbial synthesis as a class of natural polymers and is widely used in various food, medicinal, and electronics products. Another major class of synthetic polymers is dendrimers (after linear, cross-linked, and branched polymers). Dendrigraft poly-L-lysine (DGL) has the favorable properties of polylysine and dendrimers, with a broad spectrum of applications in drug discovery and development, including drug delivery, gene carriers, diagnostic imaging, diagnostics, biosensors, and special cancer therapies (such as boron neutron capture therapy and photodynamic therapy). As there are still some problems with the development of DGL, further research is warranted for its broad applications.

Combinatorial strain improvement and bioprocess development for efficient production of ε-poly-L-lysine in Streptomyces albulus

Abstract:ε-Poly-L-lysine (ε-PL) is an amino acid homopolymer with diverse potential applications in the food, pharmaceutical and cosmetic industries. To improve its biomanufacturing efficiency, strain engineering and bioprocess optimization were combined in this study. Firstly, a cocktail strain breeding strategy was employed to generate a ε-PL high-production mutant, Streptomyces albulus GS114, with enhanced L-lysine uptake capability. Subsequently, the L-lysine feeding conditions during fed-batch fermentation were systematically optimized to improve the L-lysine supply, resulting in ε-PL production reaching 73.1 ± 1.4 g/L in 5 L bioreactor. Finally, an engineered strain, S. albulus L2, with enhanced uptake capability and polymerization ability of L-lysine was constructed, achieving ε-PL production of 81.4 ± 5.2 g/L by fed-batch fermentation. This represents the highest reported production of ε-PL to date. This study provided an efficient production strategy for ε-PL and valuable insights into the high-value utilization of L-lysine.

Abstract:In recent years, biodegradable polymers have gained the attention of many researchers for their promising applications, especially in drug delivery, due to their good biocompatibility and designable degradation time. Poly (lactic-co-glycolic acid) (PLGA) is a biodegradable functional polymer made from the polymerization of lactic acid (LA) and glycolic acid (GA) and is widely used in pharmaceuticals and medical engineering materials because of its biocompatibility, non-toxicity, and good plasticity. The aim of this review is to illustrate the progress of research on PLGA in biomedical applications, as well as its shortcomings, to provide some assistance for its future research development.

Abstract:Natural antibacterial agents with antimicrobial properties have a broad potential to prevent bacterial from forming biofilms adhesion in marine environments. ε-Polylysine (E-PL) consist of homomeric polymer with 25–30 lysine residues with stability, nontoxicity, and biodegradability. ε-Polylysine is a natural cationic antibacterial peptide that can resist microbial forming biofilm adhesion. The current study investigated the action of E-PL against Pseudomonas aeruginosa biofilm isolated from a marine environment. Crystal violet staining was used to examine the effects of E-PL on the formation and destruction of mature biofilms. Scanning Electron and fluorescence microscopy revealed that E-PL treatment damaged the biofilm structure and affected the secretion of extracellular polymers. The CCK8 colorimetric assay showed that E-PL also decreased the metabolic activity and motility of biofilm bacteria. QPCR and transcriptome analysis revealed that E-PL affected biofilm formation and transcriptional regulation by downregulating genes involved in flagellar synthesis (flgE, PA4651, pilW), chemotaxis transduction (PA1251, PA4951, PA4788), biofilm biosynthesis (pelC, pelD, pslK, plsM), transcriptional regulation (PA3973, PA3508, PA0268), phenazine biosynthesis (phzM, phzH, phzS), and electron transfer (PA5401, PA5400, PA3492). This study used multiple methods to identify the mechanism of E-PL action against biofilm, informing the design of novel biofilm treatment methods.

Abstract:Synthesis and application of polylactic acid(PLA),a kind of eco-friendly polymers possessing excellent biocompatibility and biodegradability,has recently attracted greatly attention.PLA is manufactured by two methods,i.e.polycondensation and ring-opening.Commercially the catalysts used for the synthesis of PLA are stannous compounds as stannous chloride dehydrate,and stannous octoate.Research revealed that the stannous compounds are cytotoxic that has aroused concern about the bio-safety of the materials based on PLA and its use in humans.Key advances in synthesis of PLA were reviewed.

Abstract:The synthesis of poly(DL-lactic acid)-co-poly(ethylene glycol)-co-poly(L-lysine) copolymer was investigated by the reaction of amino acid-N-carboxy anhydride with lactic acid anhydrosulphite and methoxypolyoxyethylene amine (MPEG-NH2). The successful synthesis was obtained after adding dibutylmegnesium into the system. The product was proved to be copolymer. The solubility of the polymer revealed that it was ionic polymer.
