Abstract:Here we report a mild Ag-catalyzed domino acetalization reaction using quinoline-2-carboxylates, which were substructures in a number of natural products. Methanol was directly utilized as the source of the acetal part. The main features of the domino reaction include the Minisci reaction, oxidation, and acetalization. Hydrogen fluoride generated in-situ participates in the Minisci reaction, and no external acids were needed. This domino reaction provides a simple and versatile route to heteroaromatic acetals under mild condition with good functional group tolerance using methanol. (C) 2021 Elsevier Ltd. Abstract:A novel protocol for the facile synthesis of chiral 3-substituted 1,3,4,5-tetrahydrobenzo[e] [1,4]diazepin-2-ones via a domino reaction has been developed, which involves a cascade of S(N)2 nucleophilic substitution and copper-catalyzed C-N coupling reactions of a-amino acid amides and orthohalobenzyl halides. This protocol has some merits, such as one-pot process, easy operation, a wide scope of substrates. Furthermore, no heavy racemization occurs during the reaction.

Abstract:An enantioselective one-pot Michael/Michael/Henry/hemiacetalization reaction between alpha,beta-unsaturated aldehydes, alpha-ketoamides, and nitroalkenes under mild conditions catalyzed by a diarylprolinol silyl ether has been developed. The sequential methodology provides a direct approach to a wide range of fully substituted chiral oxabicyclo[2.2.2]octanes with seven contiguous stereocenters in moderate to excellent yields (up to 99%), high to excellent diastereoselectivities (up to >25:1 dr), and high to excellent enantioselectivities (up to 99% ee).

Abstract:Mukaiyama-Aldol and Prins reactions have been identified as highly efficient methods in C-C bond formation since they were discovered several decades ago. Since both reactions gave the same common intermediate, oxocarbonium, we intend to combine the two reactions into a domino process, which means the formation of multiple C-C bonds and stereogenic centers in one pot without isolation of any intermediates. We envisage that the domino reactions involving the Mukaiyama-Aldol reaction of silyl enol ether and acetal treated with Lewis acid (TiBr(4)) will produce an oxonium intermediate which upon trapping by an alkene functionality in an intramolecular Prins cyclization will generate the cyclopentyl ring system.

Abstract:A highly regio- and stereoselctive palladium-catalyzed domino reaction of functionalized aryl allyl ethers has been developed. Various aryl allyl ethers derived from the phosphine-catalyzed addition of electron-deficient allenes with phenol are found to be efficient substrates for the synthesis of 2-substituted 2,3-dihydrobenzofurans and indolines. It is the first example of aryl allyl ether used as an ideal and practical precursor of hard to get functionalized 1,3-butadiene for the heterocyclic compound synthesis.

Abstract:Phosphine-catalyzed intermolecular sequential [4 + 3] domino annulation/allylic alkylation of MBH carbonates and N-tosyl azadienes is reported. A series of functionalized benzofuran-fused seven-membered nitrogen-heterocyclic compounds with one quaternary center were obtained in good to excellent yields. In this sequential reaction, three new bonds were formed in one pot, and the MBH carbonates serve as 1,2,3-C3 synthon and C1 synthon at the same time.

Abstract:Catalytic enantioselective domino alkenylation-heteroarylation of nonconjugated iododienes proceeded with excellent stereoselectivity and broad scope of substrates. The reaction enables stereoselective syntheses of substituted azacycles such as piperidine, pyrrolidine azepine and dihydropyrans carrying new quaternary stereocenters. Mechanistically, C-H bonds of heterocycles were activated by lithium alkoxides via reversible deprotonation, rather than conventional palladium(ii)-assisted metalation processes. Many types of heteroarenes can be used, including not only azoles (such as thiazoles, oxazoles, imidazoles and oxadiazoles), but also nonazoles (thiophene, furan and azine N-oxides).

Abstract:A tunable four-module coupling approach has been established for assembling the 20-membered macrolactone core related to the proposed structure of iriomoteolide-1a by using ring-closing metathesis as the key step. Two C1-C6 (2E)-diene acid fragments with (4R,5S)- and (4S,5R)-stereogenic centers, respectively, were synthesized via anti-selective aldol reaction and E-selective conjugate addition of Me2CuLi with an alkynoic ester in the presence of TMSCl. The ring-closing metathesis reaction was carried out in the presence of 10 mol% Grubbs second-generation initiator at room temperature to give exclusively the (6E)-cycloalkenes. Our four-module coupling strategy enables efficient synthesis of two diastereomers of the proposed iriomoteolide-1a with opposite chirality at C4 and C5.

Abstract:The Acylase "Amano" (AA)-catalyzed synthesis of valuable pyridin-2-ones via domino Knoevenagel condensation-Michael addition-intramolecular cyclization-oxidization reaction between aldehyde, cyanoacetamide and ethyl acetoacetate or cyclohexyl acetoacetate was developed in the sense of a one-pot strategy. Various aliphatic, aromatic and hetero-aromatic pyridin-2-ones could also be produced in the reaction. The mechanism was illustrated according to the controlled reaction, pyridin-2-one was formed via the oxidization by oxygen at the final step. This simple and efficient enzymatic domino reaction not only widens its application of AA to organic synthesis, but is also an attractive way for the synthesis of heterocyclic compounds.

Abstract:This article describes the design and optimization of an effective organocatalytic three-component domino alpha-methylenation/Diels-Alder reaction to produce vinyl-substituted cyclohexenecarboxaldehydes in a highly regioselective fashion. In these one-pot transformations, 2-formyl-1,3-butadienes (4) were prepared in situ from alpha,beta-unsaturated aldehydes and formalin and were subsequently trapped with a variety of buta-1,3-dienes. The outcomes of the reactions were dependent on the electronic properties of the dienes. 1-Vinylcyclohexenecarbaldehydes 6 were formed by use of acyclic electron-rich dienes, while the initially formed cycloadducts of 4 with cyclopentadiene underwent Cope rearrangements, leading to the formation of tetrahydro-3H-indene-5-carbaldehyde compounds 7. The mechanisms involved in these reactions were deduced from experimental findings. Furthermore, the method was also extended to one-pot domino methylenation/Diels-Alder reactions of dihydrofurans and dihydropyrans to yield spirocyclic lactols 22. In these reactions, the unstable intermediate hydroxyethyl and hydroxypropyl acroleins behaved as dienophiles, undergoing cycloaddition reactions with dienes with good yields and selectivities. The wide variety of functionalized 1-vinylcyclohex-3-enecarbaldehydes 6, 4-vinylcyclohex-1-enecarbaldehydes 7, and spiro lactols 22 generated through the use of these organocatalytic domino processes as a diversity-oriented synthesis provided useful intermediates for the construction of novel odorants.

Abstract:Studies in recent decade have shown that tertiary enamides, once thought of as unreactive and marginally useful enamine variants, are shelf-stable yet versatile nucleophilic intermediates in organic synthesis. Many compounds, especially N-heterocycles, have been synthesized by monofunctionalization reactions of tertiary enamides. By taking advantage of intramolecular and intermolecular interception of the putative acyliminium intermediates formed by the initial nucleophilic reactions of tertiary enamides, we have established several novel difunctionalization reaction methods for constructing diverse complex fused heterocyclic products. In this Account, we summarize our endeavors to develop difunctionalization reactions of tertiary enamides in a domino fashion.

Abstract:A direct method to construct 3,4-dihydropyridin-2-ones by enzymatic condensation of aldehyde with cyanoacetamide and 1,3-dicarbonyl compounds was developed. One ring and four new bonds (two C–C, one C–N, one CC) were formed in one pot. And reaction conditions involving hydrolases, solvents, substrate molar ratios, and hydrolase loading were optimized. A series of new compounds based on the 3,4-dihydropyridin-2-one core were synthesized by the unprecedented three-component domino reaction.
