Abstract:From the fruits of Celastrus orbiculatus Thunb. a new beta-dihydroagarofuran sesquiterpene ester named 1beta,2beta,13-triacetoxy-9alpha-cinnamoyloxy-beta-dihydroagarofuran (1) has been isolated along with two known compounds, 6alpha-acetoxy-1beta, 9beta-dibenzoyloxy-8beta-hydroxy-beta-dihydroagarofuran (2), and 1beta, 6alpha-diacetoxy-9beta-benzoyloxy-8beta-hydroxy-beta-dihydroagarofuran ( 3). Abstract:Four novel cycloartanes, named sphaerophysone A (1), B (2), C (3) and D (4), were isolated from the ethanol extract of Sphaerophysa salsula (Pall.) DC. The structures were elucidated on the basis of spectral evidence, and the stereochemistry of compound 1 was defined by X-ray crystallographic analysis. Sphaerophysone B (2) may be an artifact formed in the isolation procedure.

Abstract:A new mixture of epimeric isomers furanosesquiterpenes (1 and 1a), together with two known furanosesquiterpenes were isolated from Xanthopappus subacaulis, which was used as Tibetan medicine in China. The structures of these compounds were elucidated on the comprehensive analysis of spectroscopic data including UV, IR, MS and NMR. The biosynthetic pathway of furanosesquiterpenes is proposed and the chemotaxonomic significance of these compounds is also discussed.

Abstract:A new eremophilane sesquiterpene, capnodiumone (1), along with five known eremophilane sesquiterpenes (2-6) and eight other compounds (7-14), have been isolated from a mangrove-associated fungus Capnodium sp. SZ-F22. The chemical structures were elucidated on the basis of extensive spectroscopic analysis. The broth extract of the fungus exhibited a good inhibitory effect on the mycelium growth against Fusarium graminearum at 100g/mL, however, all the 14 compounds showed no expected antifungal activity. The probable reasons were discussed. [GRAPHICS] .

Abstract:A new furanoeremophilane, 1α-chloro-6β-isobutyroxy-9-oxo-10β-hydroxy-furanoeremophilane (1) was isolated from the root of Ligularia atroviolacea and identified mainly on the basis of detailed spectral analyses including X-ray. Moreover, five known furanoeremophilanes,1α-hydroxy-6β-isobutyroxy-9-oxo-10β-H-furanoeremophilane (2), 1α-hydroxy-6β-isobutyroxy-9-oxo-10α-H-furanoeremophilane (3), 1α,10β-dihydroxy-6β-angeloyloxy-9-oxo-furanoeremophilane (4), furanoeremophil-3-en-15,6α-olide (5) and furanoeremophil-15β,6α-olide (6) were isolated from the same source.

Abstract:Fibraurea tinctoria Lour. (Menispermaceae) is a medicinal plant used by many local tribes in Indonesia and other countries. This species has pharmacological potential, such as antimalaria, antibacterial, antitumor, antioxidant, anticancer, antidiabetic, anti-inflammatory, and hepato-protector. However, the secretory structures and sites of secondary metabolite accumulation still need to be studied. This study aimed to explore the secretory structures and reveal the accumulation sites of the main classes of secondary metabolites in the root, stem, and leaves of F. tinctoria. Histochemical analysis using various specific reagents was performed on freehand sections of the fresh organs to detect alkaloids, phenolics, terpenoids, and lipophilic compounds. Observations of secretory structures were carried out using light microscopy. The result showed that idioblast was the predominant secretory structure in the plant organs and contained all the chemical groups investigated, while laticifer accumulated alkaloids were found only in the stem. Some secondary metabolites were localized in the xylem, intercellular space, and cuticle layer. Crystals were observed in the root, stem, and leaf of this species. These results might be helpful as guide information for extracting the particular plant parts to obtain the desired secondary metabolites. It suggested that leaves are a potential alternative source of medicinal raw material of this species, in addition to the stem. RESEARCH HIGHLIGHTS: The first study on secretory structures of Fibraurea tinctoria. Secondary metabolites in F. tinctoria organs accumulate in specialized and unspecialized structures. The idioblast is the primary secretory structure in the organs of F. tinctoria spreading over the root, stem, and leaves.
Chiral resolution of furofuran lignans and their derivatives from the stems of Dendrobium 'Sonia'

Abstract:Five new furofuran lignans and their derivatives, (-)-glaberide I 4-O-β-D-glucopyranoside (1a), (+)-glaberide I 4-O-β-D-glucopyranoside (1b), (+)-glaberide I 7'-ethoxy-4-O-β-D-glucopyranoside (2a), (-)-glaberide I 7'-ethoxy-4-O-β-D-glucopyranoside (2b), and (-)-isoeucommin A (3b), along with fifteen known analogs were isolated from the stems of Dendrobium 'Sonia'. These compounds were classified into ten pairs of enantiomers or diastereoisomers via chiral resolution, and their structures were determined based on extensive spectroscopic data. Their absolute configurations were determined by hydrolysis, comparison of experimental and calculated electronic circular dichroism (ECD) data, and single-crystal X-ray diffraction analysis. The isolates were evaluated for their ability to inhibit nitric oxide (NO) production in RAW264.7 cells. Among them, syringaresinol (5) exhibited prominent inhibition activity, with an IC50 value of 28.4 ± 3.0 μmol·L-1, and there was a slight difference between 5a, 5b and the racemic mixture 5.

Abstract:To investigate the chemical components from the stems of Casearia velutina Bl.,the constituents were isolated by repeated chromatography with silica gel,Sephadex LH-20,and ODS columns.The structures were elucidated by spectroscopic analysis.Eleven triterpenoids and its glycosides were isolated from the crude extract of C.velutina,and their structures were identified as friedelin-2,3-1actone (1),friedelane (2),epifriedelanol (3),friedelin (4),2α,3α,19α-trihydroxy-urs-12-cn-28-oic acid (5),2α,3β,19α-trihydroxy-urs-12-en-28-oic acid (6),2α,3α,23-trihydroxy-urs-12-en-28-oic acid (7),2α,3α,23-trihydroxyolean-12-en-28-oic acid (8),2α,3α,19α,23-tetrahydroxy-urs-12-en-28-oic acid (9),2α,3α,19α,23-tetrahydroxy-urs-12-en-28-oic acid-28-O-β-D-glucopyranosyl ester (10),and 3β,19α-dihydroxy-urs-12-en-28-oic acid 3-O-α-L-arabinopyranoside (11).All the compounds described above were isolated from this species for the first time.Compound 1 is a rarely occurred secofriedelolactone in Flacourtiaceae.

Abstract:Two diterpenoids and a few miscellaneous components (one phenethylpropanoid, one flavonoid, and one lignan) were isolated from the MeOH extract of the whole plant of the rare Chloranthaceae plant Chloranthus sessilifolius during a further phytochemical reinvestigation. Based on their spectroscopic data (i.e., IR, UV, 1D/2D NMR, ECD, and HR-ESIMS) and/or comparing with those reported physicochemical data in related literature, the structures were determined to be (3R,5S,6S,95,10S,135)-3,6,13-trihydroxy-ent-abieta-8(14)-en-7-one (1), 3 alpha,15-dihydroxy-13 alpha-methoxy-ent-abieta-8(14)-en-7-one (2), p-coumaroyl-beta-phenethyl amine (3), narigenin trimethyl ether (4), and (3R,4-R,3'R,4'R)-6,6'-dimethoxy-3,4,3',4'-tetrahydro-2H,2H'[3,3]-bichromenyl-4,4'-diol (5). Among them, compound 1 is new, while compounds 3 similar to 5 were isolated from the Chloranthus genus for the first time.

Abstract:New eremophilane sesquiterpenes, 6 beta-sarracinoyloxy-1 beta,10 beta-epoxy-furanoeremophilane (1), 6 alpha-angeloyloxy-10 beta H-furanoeremophil-1-one (2), 1 alpha-hydroxy-9-deoxycacalol (3), and 1 beta-hydroxy-11(R,S)-8-oxoeremophil-6,9-dien-12-al (4a+4b), together with five known sesquiterpenes (5-9) were isolated from the roots and rhizomes of Ligularia macrophylla. The structures were elucidated by spectroscopic methods including 2D NMR techniques, and the structure of 1 was confirmed by a single-crystal X-ray diffraction experiment. The compounds were also evaluated for cytotoxic activity against human lung carcinoma (A-549) and human breast adenocarcinoma (MCF-7) and were found to show only very weak cytotoxicity.

Abstract:Tetrapleura tetraptera fruit, used as spice in West Africa was studied chemically; five previously undescribed triterpenoid saponins 1 – 5 and four known compounds 6 – 9 were isolated from the n -Butanol fraction. The chemical structures of all nine isolates were determined by comprehensive analysis of HRMS, 1D & 2D NMR experiments and by comparison with data in the literature. All nine compounds were evaluated for antibacterial activity by the broth microdilution through the rapid p -iodonitrotetrazolium chloride (INT) colorimetric technique. The results showed that only compounds 5 (MIC = 64 μg/mL against P. aeruginosa and P. stuartii ) and 8 (MIC = 64 μg/mL against E. coli ) exhibited moderate antibacterial activity. The rest of the compounds displayed weak antibacterial activity against the tested organisms. Molecular docking studies was used to comprehend antibacterial activities.

Abstract:Four fawcettimine-type (palhicerines A-D, resp.) and two lycopodine-type (palhicerines E and F) Lycopodium alkaloids together with twenty known ones were isolated from the whole plant of Palhinhaea cernua. The structures and absolute configurations of the palhicerines A-F were determined by extensive spectroscopic methods, single-crystal X-ray diffraction analyses, chemical transformation, and electronic circular dichroism (ECD) calculations or induced electronic circular dichroism (IECD) spectra. Among the isolates, the new C/D-ring of the palhicerines A-C (trans-fused fawcettimine-type alkaloids) are rare, and each possesses a β-oriented C-16 methyl group and a distinctive tertiary methoxy group at C-13. Chemotaxonomy for differentiating species in the genus Palhinhaea is briefly discussed.

Abstract:Phytochemical study on the roots of a medicinal plant Ferula communis L. (Apiaceae) resulted in the isolation of 20 sesquiterpenes including 12 previously undescribed compounds, dauferulins A–L ( 1 − 12 ). The detailed spectroscopic analysis revealed 1 – 12 to be daucane-type sesquiterpenes with a p -methoxybenzoyloxy group at C-6. The absolute configurations of 1 – 12 were deduced by analysis of the ECD spectra. Dauferulins A–L ( 1 – 12 ), known sesquiterpenes ( 13 − 20 ), and analogues ( 14a – 14l ) derived from 6- O - p -methoxybenzoyl-10α-angeloyloxy-jeaschkeanadiol ( 14 ) were evaluated for their effects on AMPK phosphorylation in human hepatoma HepG2 cells as well as inhibitory activities against erastin-induced ferroptosis on human hepatoma Hep3B cells and IL-1β production from LPS-treated murine microglial cells.

Abstract:One new norsesquiterpenoid, namely tussfarfarin A (1), and four new artifacts resulting from extraction procedure, namely tussfarfarin B (2), 6-(1-ethoxyethyl)-2,2-dimethylchroman-4-ol (3), 5-ethoxymethyl-1H-pyrrole-2-carbaldehyde (4), and 3β-hydroxy-7α-ethoxy-24β-ethylcholest-5-ene (5), along with 18 known compounds, were isolated from the flower buds of Tussilago farfara. Their structures were elucidated by extensive spectroscopic analysis.

Abstract:In the previous work we reported five lanostane-type triterpenes from the CHCl(3) soluble fraction of Fomes officinalis. In further study on the isolation of constituents from the CHCl(3) soluble fraction, four new triterpenes, fomefficinic acids F (1), G (2) and fomefficinols A (3), B (4), together with seven known compounds officinalic acid (5), fomlactone A (6), fomlactone B (7), fomlactone C (8), laricinolic acid (9), ergosterol (10), ergota-7,22,dien-3beta-ol (11) were isolated. The structures of new compounds were determined by spectroscopic analyses and chemical methods including NMR spectroscopic techniques ((13)C, (1)H, heteronuclear multiple quantum coherence (HMQC), heteronuclear multiple bonding correlation (HMBC), correlation spectroscopy (COSY) and nuclear Overhauser effect spectroscopy (NOESY)).
