Abstract:We report a novel micro-fiber Bragg grating (mu FBG) sensor that takes advantage of the degeneracy of stop-band and rapid emergence of spectral modes when an effective phase shift occurs. The phase shift can be enabled by a range of perturbations in a central segment of the grating, including monolayer immobilization of bio-molecules or change in refractive index in the surrounding, thereby constituting the possibility of a highly sensitive sensor with the merit of scalable performance. The use of mu FBG ensures strong evanescent field coupling to the surrounding in order to maximize signal transduction. Simulation results indicate very favorable sensor signal characteristics such as large wavelength shift and sharp reflection dips. A general relation between the peak position within the stop-band and the amount of effective phase shift is also provided, and may generally serve as helpful guideline for FBG sensor design. A typical mu FBG sensor device may detect surface protein/DNA adsorption with limit-of-detection (LOD) as low as 3.3 pg.mm(-2) for surface mass density and 51.8 fg for total mass. Abstract:A challenge for optofluidic absorbance detection is the high concentration limit of detection due to the short optical path length. Herein, we introduce a concept of utilizing the coiled optical nanofiber for highly sensitive and robust optofluidic absorbance detection. Investigated by measuring the absorbance of FeCl3 solutions, the sensor shows a detection limit down to 10 μM with excellent reversibility in a concentration range of 0-5 mM. The sensitivity is 10-fold higher than that of standard absorbance measurement by using a 1 cm cuvette. Also, highly sensitive chloramphenicol sensing was demonstrated by using the enzyme-linked immunosorbent assay (ELISA) method, achieving a detection limit below 0.5 ng/L. The higher sensitivity and lower detection limit are caused by the large fractional power of evanescent field outside the nanofiber and the long detection length, which can effectively improve the absorption of the evanescent field, while the excellent reversibility is caused by the support of a polydimethylsiloxane (PDMS) pillar rather than by suspending the nanofiber in the microchannel. We envision that the present work may open up new opportunities for ultrasensitive chemical and biological sensing.
Hollow-Core-Photonic-Crystal-Fiber-Based Miniaturized Sensor for the Detection of Aggregation-Induced-Emission Molecules

Abstract:A miniature sensor for detection of aggregation-induced-emission (AIE) molecules is proposed in this work. The sensing head is fabricated by use of hollow-core photonic crystal fiber with a core diameter of about 4.8 μm. The cladding holes are sealed with a fusion splicing technique, and the central hole remains open to allow the filtration of solution with AIE molecules. When the solution is excited by an ultraviolet lamp, the fluorescence is received by a fiber-optic spectrometer. The fluorescence intensity is associated with the concentration of AIE molecules and the infiltrated-core length. In the whole process of the experiments, the output-peak wavelength is stable, which indicates that the existing forms of AIE particles are stable, and the fluorescence reabsorption can be neglected. The experimental results obtained are in accordance with traditional microplate-spectrophotometer methods. The most exciting result is that the amount of sample measured can be as low as 0.36 nL, which allows the detection of AIE molecules at only 0.02 pmol. In addition, the miniature sensor was successfully applied to the detection of an AIE-based bioprobe for evaluating the activity of the dipeptidyl-peptidase 4 (DPP-4) inhibitor sitagliptin with an IC50 of 59.80 ± 3.06 nM. The advantages of small device size and nanoliter-scale sample volumes suggest that the proposed sensor is promising for many biosensing applications.

Abstract:A novel fiber Bragg grating (FBG) based chemical sensor using hydrogel, a swellable polymer, as sensitive element is demonstrated. The sensing mechanism relies on the shift of Bragg wavelength due to the stress resulted from volume change of sensitive swellable hydrogel responding to the change of external environment. A polyacrylamide hydrogel fiber grating chemical sensor is made, and the experiments on its sensitivity to the salinity are performed. The sensitivity is low due to the less stress from the shrinking or swelling of hydrogels. Reducing the cross diameter of the grating through etching with hydrofluoric acid can greatly improve the sensitivity of the sensor.
A Novel Biosensor for the Detection of Glucose Concentration Using the Dual-Peak Long Period Grating in the Near- to Mid-Infrared

Abstract:In this article, we demonstrate an improved efficient fibre sensor with a high sensitivity to measure glucose concentrations in the physiological range of human beings, operating in a broad spectral bandwidth from the near- to mid-infrared. The sensor consists of a dual-peak long period grating (DPLPG) with a period of 150 μm inscribed in an optical fibre with a diameter of 80 μm. The investigation of sensing for refractive index results in a sensitivity of ~−885.7 nm/refractive index unit (RIU) and ~2008.6 nm/RIU in the range of 1.30–1.44. The glucose measurement is achieved by the immobilisation of a layer of enzyme of glucose oxidase (GOD) onto the fibre surface for the selective enhancement of sensitivity for glucose. The sensor can measure glucose concentrations with a maximum sensitivity of −36.25 nm/(mg/mL) in the range of 0.1–3.0 mg/mL. To the best of our knowledge, this is the highest sensitivity ever achieved for a measurement of glucose with a long period grating-based sensor, indicating its potential for many applications including pharmaceutical, biomedical and food industries.

Abstract:A novel optical fiber glucose biosensor based on fluorescent carbon quantum dots (CQDs)-glucose oxidase (GOD)/cellulose acetate (CA) complex sensitive film was fabricated, in which the dip-coating method was adopted to immobilize the CQDs-GOD/CA complex sensitive film onto the end face of the optical fiber. The surface morphology, microstructure and optical performances of the sensitive film were characterized by field emission scanning electron microscope (FESEM), atomic force microscope (AFM), Zeiss Axiovert 25 inverted microscope, Fourier transform infrared spectroscopy (FTIR), Ultraviolet-visible spectrophotometer and fluorescence spectrophotometer, respectively. The developed fiber-optic biosensor exhibits high sensitivity and repeatability for continuous online detection of low concentration glucose, allowing visualization of real-time glucose fluctuations over a period of time. The change ratios in fluorescence intensity of the biosensor are linear with glucose concentration in various ranges including micromole and nanomole levels, and the relationship between relative fluorescence intensity ratio and glucose concentration complies well with the modified Stern-Volmer equation in the range of 10-200 μmol/L with the detection limit of 6.43 μM, and in the range of 10-100 nmol/L with the detection limit of 25.79 nM, respectively.

Abstract:We present the development of a label-free, highly sensitive fiber-optical biosensor for online detection and quantification of biomolecules. Here, the advantages of etched fiber Bragg gratings (eFBG) were used, since they induce a narrowband Bragg wavelength peak in the reflection operation mode. The gratings were fabricated point-by-point via a nonlinear absorption process of a highly focused femtosecond-pulsed laser, without the need of prior coating removal or specific fiber doping. The sensitivity of the Bragg wavelength peak to the surrounding refractive index (SRI), as needed for biochemical sensing, was realized by fiber cladding removal using hydrofluoric acid etching. For evaluation of biosensing capabilities, eFBG fibers were biofunctionalized with a single-stranded DNA aptamer specific for binding the C-reactive protein (CRP). Thus, the CRP-sensitive eFBG fiber-optical biosensor showed a very low limit of detection of 0.82 pg/L, with a dynamic range of CRP detection from approximately 0.8 pg/L to 1.2 μg/L. The biosensor showed a high specificity to CRP even in the presence of interfering substances. These results suggest that the proposed biosensor is capable for quantification of CRP from trace amounts of clinical samples. In addition, the adaption of this eFBG fiber-optical biosensor for detection of other relevant analytes can be easily realized.

Abstract:A Refractive index sensor based on tilted fiber Bragg grating and stimulated Brillouin scattering is proposed and demonstrated. The cladding modes of the tilted fiber Bragg grating is sensitive to the surrounding refractive index and the approach relies on refractive index dependent resonance wavelength shift of the modes. Stimulated Brillouin scattering is introduced to measure the wavelength shift due to its narrow bandwidth, which enhances the wavelength resolution to 0.25 pm and provides much higher refractive index sensitivity than traditional wavelength readout methods. This kind sensor is suitable to sense a very small variation of refractive index and the sensitivities can reach to a resolution of 1.27 × 10(-4) RIU with the refractive index ranging from 1.3405 to 1.4025 and a resolution of 2.49 × 10(-5) RIU ranging from 1.4025 to 1.4219, respectively.

Abstract:We report the first observation of Fano resonance in the Y-shaped cavity (YC), demonstrate that the sensitivity of the sensor is as high as 1160 nm/RIU, much higher than that of the aforementioned sensors, and observe that the quality factor and sensitivity of Fano resonance can be adjusted by changing the geometry of the sensor or adding silver nanoparticles. Traditional sucrose detection methods either waste resources or pollute the environment. This work shows that the sensor can be used to detect the concentration of sucrose. In addition, we found that the concentration of sucrose has a linear relationship with its corresponding refractive index. The sensor we designed can easily and rapidly calculate the concentration of a sucrose solution based on the Fano resonance wavelength shift, which is an important first step towards detecting the refractive index of the solution and identify the composition.

Abstract:A fiber-optic biosensing platform based on ultra-narrowband cladding mode resonances was developed on a high-reflectivity fiber Bragg grating (FBG) for targeting biomolecular detection. The multiple cladding modes with a high sensitivity to the refractive index (RI) were excited in the FBG by coupling between the forward-propagating guided core mode of the multimode fiber and the backward-propagating guided cladding mode of the FBG without any damage to the fiber structure or any change to the standard FBG manufacturing process. The full width at half maximum and the Q-factor of the typical cladding mode resonance operation of the proposed sensor are 80 pm and 19,270, respectively, which are better than those of most fiber-optic biosensors reported to date. In addition, the FBG sensor demonstrated a high sensitivity in protein detection and a high selectivity in serum sample assays. The sensitivity of this sensor was further increased simply by coating it with graphene oxide (GO) sheets on the sensing surface without using a signal amplification strategy. Furthermore, an ultra-low limit of detection (LOD) of 32 pM was obtained by the GO-coated FBG sensor for IgG detection. The proposed FBG sensor provides a competitive fiber-optic platform for biomolecular detection. It has a great potential for applications in label-free biosensing.

Abstract:We propose refractive index sensors based on Ag-metalized nanolayer in microstructured optical fibers. The surface plasmon resonance modes and the sensing properties are theoretically analyzed using finite element method (FEM). In the calculation, Drude–Lorentz model is used to describe the Metal Dielectric constant. The calculation results show that the sensitivity of Ag-metalized SPR sensor can reach 1500nm/RIU corresponding to a resolution of 6.67×10−5RIU. Comparing with conventional detecting material-Au under the same structure, the sensitivity and 3dB bandwidth of our device are better.

Abstract:We combine fiber Bragg grating (FBG) technology with a wet chemical etch-erosion procedure and demonstrate two types of refractive index sensors using single-mode optical fibers. The first index sensor device is an etch-eroded single FBG with a radius of 3μm, which is used to measure the indices of four different liquids. The second index sensor device is an etch-eroded fiber Fabry-Pérot interferometer (FFPI) with a radius of ∼1.5μm and is used to measure the refractive indices of isopropyl alcohol solutions of different concentrations. Due to its narrower resonance spectral feature, the FFPI sensor has a higher sensitivity than the FBG sensor and can detect an index variation of 1.4×10−5. Since we can measure the reflection signal, these two types of sensors can be fabricated at the end of a fiber and used as point sensors.

Abstract:A D-type fiber biosensor based on surface-plasmon resonance (SPR) technology and heterodyne interferometry is presented. The sensing device is a single-mode optical fiber in which half the core is polished away and a thin-film layer of gold is deposited. We measure the phase-difference variations instead of the light intensity as in traditional SPR techniques. Therefore the accuracy and resolution of our method are very high. Its sensitivity can reach 2 x 10(-6) refractive-index units. The sensor has some merits, e.g., tunable high sensitivity, small size, lower cost, smaller sample volume, and suitability for in vivo testing. This novel method of a D-type fiber biosensor based on SPR technology and heterodyne interferometry is valuable for chemical, biological, and biochemical sensing, and the novel method of D-type fiber biosensing is a feasible means of study.

Abstract:A refractive index sensor based on the thinned and microstructure fiber Bragg grating (ThMs-FBG) was proposed and realized as a chemical sensing. The numerical simulation for the reflectance spectrum of the ThMs-FBG was calculated and the phase shift down-peak could be observed from the reflectance spectrum. Many factors influencing the reflectance spectrum were considered in detail for simulation, including the etched depth, length, and position. The sandwich-solution etching method was utilized to realize the microstructure of the ThMs-FBG, and the photographs of the microstructure were obtained. Experimental results demonstrated that the reflectance spectrum, phase shift down-peak wavelength, and reflected optical intensity of the ThMs-FBG all depended on the surrounding refractive index. However, only the down-peak wavelength of the ThMs-FBG changed with the surrounding temperature. Under the condition that the length and cladding diameter of the ThMs-FBG microstructure were 800 and 14 mum, respectively, and the position of the microstructure of the ThMs-FBG is in the middle of grating region, the refractive index sensitivity of the ThMs-FBG was 0.79 nm/refractive index unit with the wide range of 1.33-1.457 and a high resolution of 1.2 x 10(-3). The temperature sensitivity was 0.0103 nm/ degrees C, which was approximately equal to that of common FBG.

Abstract:A D-shaped photonic crystal fiber surface plasmon resonance (PCF-SPR) sensor is proposed for highly sensitive liquid refractive index sensing. Taking sensing sensitivity into consideration, silver-titanium dioxide (Ag-TiO2) composite micro-grating is introduced and SPR is excited within Ag-TiO2 composite micro-grating at the flat D-shaped PCF surface. The finite element method (FEM) based on COMSOL is applied to analyzed numerically response characteristics of the D-shaped sensor, which was research dependent on wavelength and amplitude interrogation method. The results displayed that designed sensor reached excellent amplitude sensitivity of 2245 RIU−1 and wavelength sensitivity of 10,300 nm/RIU for liquid refractive index varying between 1.33 and 1.40. And it also shows that the maximum figure of merit was 480 RIU−1 for proposed sensor after optimization of structural parameters, which has a great prospect in the biological detection and chemical analysis.

Quantitative broadband chemical sensing in air-suspended solid-core fibers

Abstract:We demonstrate a quantitative broadband fiber sensor, based on evanescent field sensing in the cladding holes of an air-suspended solid-core photonic crystal fiber. We discuss the fabrication process, together with the structural- and optical characterization of a range of different fibers. Measured mode profiles are in good agreement with finite element method calculations made without free parameters. The fraction of the light in the hollow cladding can be tuned via the core diameter of the fiber. Dispersion measurements are in excellent agreement with theory and demonstrate tuning of the zero dispersion wavelength via the core diameter. Optimum design parameters for absorption sensors are discussed using a general parameter diagram. From our analysis, we estimate that a sensitivity increase of three orders of magnitude is feasible compared to standard cuvette measurements. Our study applies to both liquid and gas fiber sensors. We demonstrate the applicability of our results to liquid chemical sensing by measuring the broad absorption peak of an aqueous nickel chloride solution. We find striking agreement with the reference spectrum measured in a standard cuvette, even though the sample volume has decreased by three orders of magnitude. Our results demonstrate that air-suspended solid-core PCFs can be used in quantitative broadband chemical sensing measurements.

Abstract:A novel sensor is demonstrated to allow a real-time measurement of the physical wear applied to the surface of an object. Two different measurement methods are presented, both utilizing the reflected power from a sacrificial chirped fibre Bragg grating to give the wear measurement. The measurement systems are simple to implement with the possibility of low cost designs depending on the application. The sensor can measure wear with a resolution of 120 µm.

Abstract:A fiber-optic acetylene gas sensor based on a microstructured optical fiber Bragg grating (MOFBG) is presented. The microstructured optical fiber (MOF) is specially designed to have a photosensitive core and holey cladding for grating fabrication and gas detection. The micro-holes of the MOF serve as gas cells, in which the acetylene molecules interact with light through the evanescent-waves of guided modes. The MOFBG with a specific period is utilized to not only make the sensor work in reflection mode, but also selectively determine the wavelength and the order of the acting fiber mode for gas sensing. The MOF is prepared by using the well-known stack-and-draw method, and the Bragg gratings are fabricated by using 193-nm laser pulses. Experiments for the use of the MOFBG for acetylene gas detection have been successfully demonstrated.
Nano-Photonic Crystal D-Shaped Fiber Devices for Label-Free Biosensing at the Attomolar Limit of Detection

Abstract:Maintaining both high sensitivity and large figure of merit (FoM) is crucial in regard to the performance of optical devices, particularly when they are intended for use as biosensors with extremely low limit of detection (LoD). Here, a stack of nano-assembled layers in the form of 1D photonic crystal, deposited on D-shaped single-mode fibers, is created to meet these criteria, resulting in the generation of Bloch surface wave resonances. The increase in the contrast between high and low refractive index (RI) nano-layers, along with the reduction of losses, enables not only to achieve high sensitivity, but also a narrowed resonance bandwidth, leading to a significant enhancement in the FoM. Preliminary testing for bulk RI sensitivity is carried out, and the effect of an additional nano-layer that mimics a biological layer where binding interactions occur is also considered. Finally, the biosensing capability is assessed by detecting immunoglobulin G in serum at very low concentrations, and a record LoD of 70 aM is achieved. An optical fiber biosensor that is capable of attaining extraordinarily low LoD in the attomolar range is not only a remarkable technical outcome, but can also be envisaged as a powerful tool for early diagnosis of diseases.
In-situ Dual-Channel Surface Plasmon Resonance Fiber Sensor for Temperature-Compensated Detection of Glucose Concentration

Abstract:External temperature variations inevitably affect the accuracy of surface plasmon resonance (SPR) biosensors. To that end, we propose an ultra-compact label-free dual-channel SPR fiber sensor (DSPRFS) that can simultaneously measure the glucose concentration and ambient temperature in real-time. The proposed sensor is based on a unique dual-channel structure fabricated by etching a side-hole fiber (SHF), and has significantly higher spatial sensitivity than traditional SPR biosensors. After coating with silver and zinc oxide films, one channel was filled with polydimethylsiloxane (PDMS) to sense the ambient temperature, and the other channel was immobilized with glucose oxidase (GOx) enzyme for glucose sensing. The proposed sensor is analyzed theoretically, fabricated and characterized. Glucose concentration sensitivity and temperature sensitivity of the manufactured sensor sample were found to be as high as 6.156 nm/mMand -1.604 nm/°C with limits of detection (LOD) of 16.24 µM and 0.06 °C, respectively. The proposed sensor has an extremely compact structure, enables temperature compensation, and is suitable for in-situ monitoring and high-precision sensing of glucose and other biological analytes.
