Abstract:Common indoor and outdoor environmental fungi such as Aspergillus fumigatus produce asexual spores containing a collection of proteins that can bind IgE antibodies and trigger allergic reactions. We characterized the impact of sporulation temperature on the IgE-binding capacity (allergenicity) of A. fumigatus and explored the links between variable allergenicity and temperature-dependant expression of genes encoding these allergenic proteins. A 12-fold increase in A. fumigatus allergenicity per spore was observed when sporulation temperatures were decreased from 32°C to 17°C. Per spore protein mass and Asp f 1 allergen mass also followed this trend. Functional gene expression analysis of A. fumigatus sporulating cultures by real-time reverse-transcription PCR and gene expression microarrays revealed that a greater number of genes encoding known, major allergens are more highly expressed at lower sporulation temperatures. The results of this study indicate that environmental conditions at growth significantly influence the allergenicity of this common mould through the differential production of allergenic proteins, and highlight the importance of in vivo or in vitro allergenicity measurements for understanding environmental exposure to airborne allergenic fungi.

Abstract:Objective To determine the growth characteristics of Aspergillus fumigatus in different nutrient media and temperature.Methods Growth curves were used to determine the optimum medium,optimum and maximum growth temperatures for two strains of Aspergillus fumigatus:colony counts were used to determine the fatal temperatures for them.Results Three different nutrient media,yeast extract medium(YG),minimal medium(MM) and yeast nitrogen base(YNB),were tested.YG medium was the optimum medium for measuring the growth curves of Aspergillus fumigatus;In YG medium,the optimum growth temperature for Aspergillus fumigatus was 37℃,and the maximum growth temperature was 52℃.The slower growth rates of strains were observed with gradually elevated temperature.Unexpectedly,the responses of these two strains to high temperature were different:one strain's growth became slower markedly at 48℃ than at 37℃ during log phase,while the other one grew at 48℃ as well as at 37℃ during log phase.The strains of Aspergillus fumigatus were dead when exposed to 65℃ lasting more than 180 min or 70℃ lasting more than 120 min.Conclusions The optimum and the maximum growth temperature for Aspergillus fumigatus were 37℃ and 52℃,respectively.The fatal condition for Aspergillus fumigatus was 65℃ lasting more than 180 min or 70℃ lasting more than 120 min.

Abstract:CD4(+) T cells orchestrate immune responses against fungi, such as Aspergillus fumigatus, a major fungal pathogen in humans. The complexity of the fungal genome and lifestyle questions the existence of one or a few immune-dominant Ags and complicates systematic screening for immunogenic Ags useful for immunotherapy or diagnostics. In this study, we used a recently developed flow cytometric assay for the direct ex vivo characterization of A. fumigatus-specific CD4(+) T cells for rapid identification of physiological T cell targets in healthy donors. We show that the T cell response is primarily directed against metabolically active A. fumigatus morphotypes and is stronger against membrane protein fractions compared with cell wall or cytosolic proteins. Further analysis of 15 selected single A. fumigatus proteins revealed a highly diverse reactivity pattern that was donor and protein dependent. Importantly, the parallel assessment of T cell frequency, phenotype, and function allowed us to differentiate between proteins that elicit strong memory T cell responses in vivo versus Ags that induce T cell exhaustion or no reactivity in vivo. The regulatory T cell (Treg) response mirrors the conventional T cell response in terms of numbers and target specificity. Thus, our data reveal that the fungal T cell immunome is complex, but the ex vivo characterization of reactive T cells allows us to classify Ags and to predict potential immunogenic targets. A. fumigatus-specific conventional T cell responses are counterbalanced by a strong Treg response, suggesting that Treg-depletion strategies may be helpful in improving antifungal immunity.

Abstract:The effect of air temperature (4–30°C) and relative humidity (RH 11–96%) on the growth of two common fungi Aspergillus fumigatus and Penicillium sp. was studied in the laboratory. A short period of favorable conditions was sufficient to start fungal growth. Temperature was not a limiting factor for fungal growth on building materials, because fungi grew at even below 10°C. The relative humidity of air had no direct influence on the growth of fungi. Fungi may grow at very low levels of air humidity if water is available on the surface. Thus, repeated or persistent moisture condensation or water leakage is sufficient for fungal germination and growth on building materials.

Abstract:The contamination of peanuts, with Aspergillus flavus and subsequent aflatoxins (AFs) is considered to be one of the most serious, safety problems in the world. Water activity (aw) and temperature are limiting, factors for fungal growth and aflatoxin production during storage. The objectives of this study were to integrate data on the effects of temperature (34, 37, and 42 °C) and water activity (aw; 0.85, 0.90, and 0.95) on growth rate aflatoxin B1 (AFB1) production and up- or-downregulation of the molecular expression of biosynthetic AFB1 genes divided into three types based on their A. flavus isolate composition and AFB1 capacity in vitro: A. flavus KSU114 (high producer), A. flavus KSU114 (low producer), and A. flavus KSU121 (non-producer). The A. flavus isolates were shown to be resilient in terms of growth on yeast extract sucrose agar media when exposed to temperature and water activity as pivotal environmental factors. The optimal conditions for the fungal growth of three isolates were a temperature of 34 °C and water activity of 0.95 aw; there was very slow fungal growth at the highest temperature of 42 °C, with different aw values causing inhibited fungal growth. The AFB1 production for the three isolates followed the same pattern with one exception: A. flavus KSU114 failed to produce any AFB1 at 42 °C with different aw values. All tested genes of A. flavus were significantly up- or downregulated under three levels of interaction between temperature and aw. The late structural genes of the pathway were significantly upregulated at 34 °C under aw 0.95, although aflR, aflS and most of the early structural genes were upregulated. Compared to 34 °C with an aw value of 0.95, most of the expressed genes were significantly downregulated at 37 and 42 °C with aw values of 0.85 and 0.90. Additionally, two regulatory genes were downregulated under the same conditions. The expression level of laeA was also completely associated with AFB1 production, while the expression level of brlA was linked to A. flavus colonization. This information is required to forecast the actual effects of climate change on A. flavus. The findings can be applied to improve specific food technology processes and create prevention strategies to limit the concentrations of potential carcinogenic substances in peanuts and their derivatives.

Abstract:Fungal species within Aspergillus section Flavi contaminate food and feed with aflatoxins. These toxic fungal metabolites compromise human and animal health and disrupt trade. Genotypically and phenotypically diverse species co-infect crops, but temporal and spatial variation in frequencies of different lineages suggests that environmental factors such as temperature may influence structure of aflatoxin-producing fungal communities. Furthermore, though most species within Aspergillus section Flavi produce sclerotia, divergent sclerotial morphologies (small or S-type sclerotia vs. large or L-type sclerotia) and differences in types and quantities of aflatoxins produced suggest lineages are adapted to different life strategies. Temperature is a key parameter influencing pre- and post-harvest aflatoxin contamination of crops. We tested the hypothesis that species of aflatoxin-producing fungi that differ in sclerotial morphology will vary in competitive ability and that outcomes of competition and aflatoxin production will be modulated by temperature. Paired competition experiments between highly aflatoxigenic S-type species (A. aflatoxiformans and Lethal Aflatoxicosis Fungus) and L-type species (A. flavus L morphotype and A. parasiticus) were conducted on maize kernels at 25 and 30°C. Proportions of each isolate growing within and sporulating on kernels were measured using quantitative pyrosequencing. At 30°C, S-type fungi were more effective at host colonization compared to L-type isolates. Total aflatoxins and the proportion of B vs. G aflatoxins were greater at 30°C compared to 25°C. Sporulation by L-type isolates was reduced during competition with S-type fungi at 30°C, while relative quantities of conidia produced by S-type species either increased or did not change during competition. Results indicate that both species interactions and temperature can shape population structure of Aspergillus section Flavi, with warmer temperatures favoring growth and dispersal of highly toxigenic species with S-type sclerotia.

Abstract:Aspergillus flavus and A. parasiticus are the main causes of aflatoxin contamination in various foods, particularly grains, as they can thrive in environments with lower water activity and higher temperatures. The growth of Aspergillus and the formation of the mycotoxins aflatoxin and cyclopiazonic acid are strongly influenced by environmental stimuli and can be reduced by modulating parameters such as water activity, pH, temperature and light during the storage. This study has two objectives-on the one hand, to assess how global warming and an increase in exposure to sunlight affect growth and mycotoxin formation, and on the other hand, how the findings from these experiments can be used to reduce fungal growth and mycotoxin formation in stored foods. Using growth substrates with two different water activities (aw 0.95, aw 0.98), together with a light incubation device consisting of different chambers equipped with diodes emitting visible light of five different wavelengths (455 nm, 470 nm, 530 nm, 590 nm, 627 nm) plus white light, we analyzed the growth and mycotoxin formation of selected Aspergillus flavus and A. parasiticus isolates. It was shown that light with a wavelength of 455/470 nm alone, but especially in combination with a lower water activity of aw 0.95, leads to a significant reduction in growth and mycotoxin formation, which was accompanied by reduced transcriptional activity of the responsible mycotoxin biosynthetic genes. Therefore, these results can be used to significantly reduce the growth and the mycotoxin formation of the analyzed fungi during storage and to estimate the trend of fungal infestation by Aspergillus flavus and A. parasiticus in water activity- and light exposure-equivalent climate change scenarios. Mycotoxin-producing aspergilli can be effective and sustainably inhibited using a combination of short-wave light and lowered water activity in the substrate. A higher annual mean temperature accompanying climate change may lead to an increased spread of aflatoxin-producing fungi in areas that were previously too cold for them. On the other hand, there will be regions in the world where contamination with aflatoxin-producing fungi will be reduced due to increased drought and sun exposure.

Abstract:Portions of corn, a commodity in which aflatoxin is frequently found, were held at 16, 26 and 32°C after the moisture contents were adjusted to achieve water activities (aw) ranging from too low to ample for support of mold growth. Suspensions of mold spores from toxigenic cultures of Aspergillus flavus, A. ochraceus, Penicillium citrinum, P. cyclopium and P. urticae were added to the test portions, either as A. flavus alone, as A. flavus with one of the other molds or as a mixture of all 5 species. Additional water was used to obtain the proper moisture levels. A temperature of 16°C was generally too low for aflatoxin production by either the added or native strains of A. flavus, although the mold was able to grow at 16°C at aw values as low as 0.80, 0.77 and 0.85 on corn, soybeans and pinto beans, respectively. Aflatoxin production was essentially the same at 26 and 32° C with limiting aw values in the range of 0.85–0.89. Limiting aw values for mold growth at 26 and 32°C were 0.73, 0.69 and 0.75 for corn, soybeans and pinto beans, respectively. This study provided no evidence that substrate suitability at limiting temperatures and aw levels is a factor in the observed difference in the risk of aflatoxin contamination for these commodities. The study did indicate that the associated mold flora, when the seed is exposed to mold invasion, is a risk determinant.

Relationship between thermotolerance and hydrophobin-like proteins in aerial conidia of Beauveria bassiana and Paecilomyces fumosoroseus as fungal biocontrol agents.

Abstract:Aims: This study was to illustrate the relationship between the thermotolerance and the contents of hydrophobin-like or formic-acid-extractable (FAE) proteins in aerial conidia of Beauveria bassiana and Paecilomyces fumosoroseus produced on rice-based substrate. Methods and Results: Survival indices of 11 isolates were separately assessed as a ratio of the viability of conidia after 3-150 min thermal stress at 48degreesC over that of unstressed conidia and fitted well to a survival model (r(2) greater than or equal to 0.97). For a given isolate, the fitted model generated an LT50, the time for 50% viability loss under the stress. The LT(50)s of six B. bassiana isolates (10.1-61.9 min) and five P. fumosoroseus isolates (2.8-6.2 min) were correlated (r(2) = 0.81) with FAE protein contents (6.9-23.4 mug mg(-1)). The survival indices of a fixed B. bassiana isolate after 45-min thermal stress at 48degreesC were also correlated to the FAE protein contents from conidia produced on glucose-, sucrose-, or starch-based substrate (0.79 less than or equal to r(2) less than or equal to 0.86). Three bands of 12.0, 15.0 and 17.5 kDa were recognized from SDS-PAGE profiles of protein extracts from different isolates or from the fixed isolate grown on the three carbon sources, but the 12 kDa was less likely related to the thermotolerance. Conclusions: Approximately 80% of variability in conidial thermotolerance was attributed to either 15.0- or 17.5-kDa FAE protein or both. Significance and Impact of the Study: The relationship between conidial thermotolerance and FAE protein contents implicates a new way to search for fungal biocontrol agents with more thermotolerance and environmental persistence.

Abstract:Aspergillus flavus is the predominant species that produce aflatoxins in stored peanuts under favourable conditions. This study aimed to describe the growth and aflatoxin production by two A. flavus strains isolated from imported raw peanuts and to model the effects of temperature and aw on their colony growth rate as a function of temperature and aw in Peanut Meal Extract Agar (PMEA). A full factorial design with seven aw levels (0.85-0.98 aw) and five temperature levels (20-40 °C) was used to investigate the growth and aflatoxin production. Colony diameter was measured daily for 28 days while AFB1 and total aflatoxin were determined on day 3, 7, 14, and 21. The maximum colony growth rate, μmax (mm/day) was estimated by using the primary model of Baranyi, and the μmax was then fitted to the secondary model; second-order polynomial and linear Arrhenius-Davey to describe the colony growth rate as a function of temperature and aw. The results indicated that both strains failed to grow at temperature of 20 °C with aw <0.94 and aw of 0.85 for all temperatures except 30 °C. The highest growth rate was observed at 30 °C, with 0.98 aw for both strains. The analysis of variance showed a significant effect of strain, temperature, and aw on the fungal growth and aflatoxin production (p < 0.05). Furthermore, both secondary models were in good agreement with the observed μmax. However, the polynomial model was found to be a better predictor of the experimental data. A similar pattern was observed in aflatoxin production but in a narrower range of temperature (25-35 °C) and aw (0.92-0.98 aw). The highest production of aflatoxins was observed on day 21 at 30 °C with the aw level of 0.98 for both strains. Overall, the current findings may help in improving the mycotoxin management and intervention strategies in peanuts, especially during storage.

Abstract:Almonds are among the commodities at risk of aflatoxin contamination by Aspergillus flavus. Temperature and water activity are the two key determinants in pre and post-harvest environments influencing both the rate of fungal spoilage and aflatoxin production. Varying the combination of these parameters can completely inhibit or fully activate the biosynthesis of aflatoxin, so it is fundamental to know which combinations can control or be conducive to aflatoxin contamination. Little information is available about the influence of these parameters on aflatoxin production on almonds. The objective of this study was to determine the influence of different combinations of temperature (20 °C, 28 °C, and 37 °C) and water activity (0.90, 0.93, 0.96, 0.99 aw) on growth, aflatoxin B1 (AFB1) production and expression of the two regulatory genes, aflR and aflS, and two structural genes, aflD and aflO, of the aflatoxin biosynthetic cluster in A. flavus grown on an almond medium solidified with agar. Maximum accumulation of fungal biomass and AFB1 production was obtained at 28 °C and 0.96 aw; no fungal growth and AFB1 production were observed at 20 °C at the driest tested conditions (0.90 and 0.93 aw). At 20° and 37 °C AFB1 production was 70-90% lower or completely suppressed, depending on aw. Reverse transcriptase quantitative PCR showed that the two regulatory genes (aflR and aflS) were highly expressed at maximum (28 °C) and minimum (20 °C and 37 °C) AFB1 production. Conversely the two structural genes (aflD and aflO) were highly expressed only at maximum AFB1 production (28 °C and 0.96-0.99 aw). It seems that temperature acts as a key factor influencing aflatoxin production which is strictly correlated to the induction of expression of structural biosynthesis genes (aflD and aflO), but not to that of aflatoxin regulatory genes (aflR and aflS), whose functional products are most likely subordinated to other regulatory processes acting at post-translational level. The results of this study are useful to select conditions that could be used in the almond processing chain to suppress aflatoxin production in this important product.
The effect of temperature and relative humidity for Aspergillus flavus BIO 2237 growth and aflatoxin production on soybeans

Abstract:Aspergillus flavus (A. flavus) producing aflatoxin frequently contaminates crops such as soybeans. The growth of this mold on soybeans and other foodstuffs is affected by temperature and relative humidity (RH). The aim of this study was to measure the growth of A. flavus BIO 2237 and aflatoxin production at different temperatures and RH. Aspergillus flavus BIO 2237 was isolated from Indonesia origin foodstuffs. Aspergillus flavus BIO 2237 was inoculated in Czapek Dox Agar (CDA) and soybeans for 10 days at a temperature of 20, 30, and 40 o C with RH of 70, 80, and 90%. Aflatoxin analysis was conducted using RP-HPLC equipped with fluorescence detector and post column photochemical reactor. The limit of detection (LoD) for aflatoxin of B 1 , B 2 , G 1 , and G 2 was 0.45, 0.26, 0.05, and 0.13 ng/mL (ppb), while their limit of quantification (LoQ) was 1.50, 0.88, 0.18 and 0.43 ng/mL (ppb) respectively. The maximum growth for A. flavus BIO 2237 in CDA and soybeans was reached at a temperature of 30 o C with RH of 90%, and this was based on the highest diameter of colony and amount of cell mass formed in that condition. The maximum level of aflatoxin in contaminated soybeans was found at 999 ng/g (ppb), and this was produced at the same condition as its fungi’s growth. Aspergillus flavus BIO 2237 can not grow as well as produce aflatoxin in soybeans at high temperature (40oC) with low RH (70%). There was a significant difference (sig<0.05) in aflatoxin content (AFB1, AFB2, AFG1, and AFG2) between temperature and RH, meanwhile the difference on the growth of A. flavus BIO 2237 in CDA and soybeans caused by RH.

[Intraspecific Variations of Paecilomyces Fumosoroseus in Temperature Fitness and Carbendazim Resistance].

Abstract:A total of six wild strains of fungal biocontrol agent Paecilomyces fumosoroseus were grown at 15 degrees C-35 degrees C to reveal their variations in colony growth, conidiation, and conidial viability. The results showed that the test three indices were the best at 25 degrees C, but differed significantly at different temperatures for a given strain or at a given temperature among test strains. The strains Pfr116 and Pfr6206 behaved better or more stably than other strains at 15 degrees C-35 degrees C. The sensitivities of the fungal strains to gradient concentrations of fungicide carbendazim were assayed at the optimal temperature, based on their colony growth diameters and the counts of colony forming units (CFU). The inhibitory effect of carbendazim concentration on CFU counts fitted well to a Logistic model, with high coefficients of determination being achieved for all the strains (r2 > or = 0.90). The fitted minimal inhibitory concentration (MIC) values showed a low carbendazim resistance of strains Pfr4205 and Pfr116 (MIC < or = 20.0 microg x ml(-1)), whereas the other four strains fell within medium resistance to the fungicide (20 < or = MIC < or = 100). The MIC of strain Pfr6206 (93.5 microg x ml(-1) was very close to the high resistance level of MIC > 100 microg x ml(-1). Therefore, strain Pfr6206 was satisfactory with both the temperature fitness and the carbendazim resistance, and could be considered as a candidate strain for a fungal formulation adapting to more variable crop systems against insect pests.

In this study, we surveyed all Aspergillus species isolated from aspergillosis patients in Zhongshan Hospital Fudan University, Shanghai, China, from 2019 to 2021. Methods: We characterized the susceptibility profiles of these Aspergillus species to medical azoles (voriconazole, itraconazole and posaconazole) using YeastOne TM broth microdilution system. To determine the underlying antifungal resistance mechanisms in azole-resistant A. fumigatus (AR Af ) isolates, we characterized mutations in the cyp51A gene. Genotypic diversity of sampled A. fumigatus was investigated using CSP-typing. Results: A total of 112 Aspergillus isolates (81 A. fumigatus , 17 A. flavus , 5 A. niger , 2 A. terreus , 2 A. lentulus , 2 A. oryzae , 1 A. nidulans , 1 A. versicolor and 1 A. sydowii ) from 105 patients diagnosed with aspergillosis (including proven or probable invasive aspergillosis, chronic pulmonary aspergillosis, allergic bronchopulmonary aspergillosis and cutaneous aspergillosis) were obtained. Eight isolates (7 A. fumigatus and 1 A. niger ) from seven patients were either azole non-susceptible or non-wild type. Azole non-susceptible or non-wild type rate was 7.1%/isolate and 6.7%/patient analysed. Four AR Af harbored TR34/L98H mutation, whereas one carried TR46/Y121F/T289A allele. The 81 A. fumigatus isolates were spread across 8 CSP types with t01 to be the predominant type (53.1%). AR Af isolates were distributed over CSP types t01, t02, t04A and t11. Conclusion: Results from this study provided us with an understanding of the antifungal resistance and related characteristics of Aspergillus species in Eastern China. Further comparisons of our results with those in other countries reflect potential clonal expansion of A. fumigatus in our region. Further surveillance study is warranted to guide antifungal therapy and for epidemiological purposes. Factors Contributing to Sex Differences in Mice Inhaling Aspergillus fumigatus

Abstract:Aspergillus fumigatus is a respiratory fungal pathogen and an allergen, commonly detected in flooded indoor environments and agricultural settings. Previous studies in Balb/c mice showed that repeated inhalation of live and dry A. fumigatus spores, without any adjuvant, elevated allergic immune response and airway remodeling. Sex-specific differences can influence host-pathogen interactions and allergic-asthma related outcomes. However, the effect of host sex on immune response, in the context of A. fumigatus exposure, remains unknown. In this study, we quantified the multivariate and univariate immune response of C57BL/6J mice to live, dry airborne A. fumigatus spores. Our results corroborate previous results in Balb/c mice that repeated inhalation of live A. fumigatus spores is sufficient to induce mucus production and inflammation by day 3 post last challenge, and antibody titers and collagen production by day 28 post-challenge. Principal Component Analysis (PCA) showed that females exhibited significantly higher levels of immune components than males did. Taken together, our data indicate that host-sex is an important factor in shaping the immune response against A. fumigatus, and must be considered when modeling disease in animals, in designing diagnostics and therapeutics for A. fumigatus-associated diseases or while drafting evidence-based guidelines for safe mold levels.
Compatible solutes determine the heat resistance of conidia

Abstract:Abstract Background Asexually developed fungal spores (conidia) are key for the massive proliferation and dispersal of filamentous fungi. Germination of conidia and subsequent formation of a mycelium network give rise to many societal problems related to human and animal fungal diseases, post-harvest food spoilage, loss of harvest caused by plant-pathogenic fungi and moulding of buildings. Conidia are highly stress resistant compared to the vegetative mycelium and therefore even more difficult to tackle. Results In this study, complementary approaches are used to show that accumulation of mannitol and trehalose as the main compatible solutes during spore maturation is a key factor for heat resistance of conidia. Compatible solute concentrations increase during conidia maturation, correlating with increased heat resistance of mature conidia. This maturation only occurs when conidia are attached to the conidiophore. Moreover, conidia of a mutant Aspergillus niger strain, constructed by deleting genes involved in mannitol and trehalose synthesis and consequently containing low concentrations of these compatible solutes, exhibit a sixteen orders of magnitude more sensitive heat shock phenotype compared to wild-type conidia. Cultivation at elevated temperature results in adaptation of conidia with increased heat resistance. Transcriptomic and proteomic analyses revealed two putative heat shock proteins to be upregulated under these conditions. However, conidia of knock-out strains lacking these putative heat shock proteins did not show a reduced heat resistance. Conclusions Heat stress resistance of fungal conidia is mainly determined by the compatible solute composition established during conidia maturation. To prevent heat resistant fungal spore contaminants, food processing protocols should consider environmental conditions stimulating compatible solute accumulation and potentially use compatible solute biosynthesis as a novel food preservation target.

Abstract:In this study, we evaluated the effect of different temperatures (10, 20, 30, and 40 °C) and relative humidities (RHs; 12, 44, 76, and 98%) on populations of predominant grain fungi (Aspergillus candidus, Aspergillus flavus, Aspergillus fumigatus, Penicillium fellutanum, and Penicillium islandicum) and the biocontrol activity of Pseudomonas protegens AS15 against aflatoxigenic A. flavus KCCM 60330 in stored rice. Populations of all the tested fungi in inoculated rice grains were significantly enhanced by both increased temperature and RH. Multiple linear regression analysis revealed that one unit increase of temperature resulted in greater effects than that of RH on fungal populations. When rice grains were treated with P. protegens AS15 prior to inoculation with A. flavus KCCM 60330, fungal populations and aflatoxin production in the inoculated grains were significantly reduced compared with the grains untreated with strain AS15 regardless of temperature and RH (except 12% RH for fungal population). In addition, bacterial populations in grains were significantly enhanced with increasing temperature and RH, regardless of bacterial treatment. Higher bacterial populations were detected in biocontrol strain-treated grains than in untreated control grains. To our knowledge, this is the first report showing consistent biocontrol activity of P. protegens against A. flavus population and aflatoxin production in stored rice grains under various environmental conditions of temperature and RH.

Abstract:BACKGROUNDThe fungus Aspergillus fumigatus causes a variety of clinical phenotypes in patients with cystic fibrosis (pwCF). Th cells orchestrate immune responses against fungi, but the types of A. fumigatus-specific Th cells in pwCF and their contribution to protective immunity or inflammation remain poorly characterized.METHODSWe used antigen-reactive T cell enrichment (ARTE) to investigate fungus-reactive Th cells in peripheral blood of pwCF and healthy controls.RESULTSWe show that clonally expanded, high-avidity A. fumigatus-specific effector Th cells, which were absent in healthy donors, developed in pwCF. Individual patients were characterized by distinct Th1-, Th2-, or Th17-dominated responses that remained stable over several years. These different Th subsets target different A. fumigatus proteins, indicating that differential antigen uptake and presentation directs Th cell subset development. Abstract:Objective To analyze the growth characteristics of the clinical isolates of Aspergillus compared to standard strains in vitro.Methods The sabourauds agar medium (SDA),yeast extract medium(YG) and potato dextrose agar medium (PDA) were used to test two clinical isolates and one standard strain.The strains were incubated at 29 ℃C for 96 hours.In liquid medium,the optical density (OD) values at 0 hour,4,8,12,24,36,48,60,72 and 96 hours after culture were recorded to generate growth curves for each strain and culture medium.Meanwhile,on solid medium,the features of colony morphology were recorded.Results In liquid medium,the growth curves were not significantly different between clinical isolates and standard strain in PDA medium (all at P＜0.05),while the differences were obvious in SDA medium and YG medium (P＜0.05).The clinical isolates showed shorter lag phase and higher growth rate than standard strain.On solid medium,all strains appeared the best growth state on PDA medium and the poorest state on SDA medium.Color change time was later on YG medium than that on the other medium.The clinical isolates performed better on the aspects of colony sizes and color change than standard strain when growed on the same medium,especially on SDA medium.Conclusions Compared with standard strain,the clinical isolates of Aspergillus have higher adaptive capacity to poor nutrition condition.
