Abstract:Portions of corn, a commodity in which aflatoxin is frequently found, were held at 16, 26 and 32°C after the moisture contents were adjusted to achieve water activities (aw) ranging from too low to ample for support of mold growth. Suspensions of mold spores from toxigenic cultures of Aspergillus flavus, A. ochraceus, Penicillium citrinum, P. cyclopium and P. urticae were added to the test portions, either as A. flavus alone, as A. flavus with one of the other molds or as a mixture of all 5 species. Additional water was used to obtain the proper moisture levels. A temperature of 16°C was generally too low for aflatoxin production by either the added or native strains of A. flavus, although the mold was able to grow at 16°C at aw values as low as 0.80, 0.77 and 0.85 on corn, soybeans and pinto beans, respectively. Aflatoxin production was essentially the same at 26 and 32° C with limiting aw values in the range of 0.85–0.89. Limiting aw values for mold growth at 26 and 32°C were 0.73, 0.69 and 0.75 for corn, soybeans and pinto beans, respectively. This study provided no evidence that substrate suitability at limiting temperatures and aw levels is a factor in the observed difference in the risk of aflatoxin contamination for these commodities. The study did indicate that the associated mold flora, when the seed is exposed to mold invasion, is a risk determinant.

Abstract:The contamination of peanuts, with Aspergillus flavus and subsequent aflatoxins (AFs) is considered to be one of the most serious, safety problems in the world. Water activity (aw) and temperature are limiting, factors for fungal growth and aflatoxin production during storage. The objectives of this study were to integrate data on the effects of temperature (34, 37, and 42 °C) and water activity (aw; 0.85, 0.90, and 0.95) on growth rate aflatoxin B1 (AFB1) production and up- or-downregulation of the molecular expression of biosynthetic AFB1 genes divided into three types based on their A. flavus isolate composition and AFB1 capacity in vitro: A. flavus KSU114 (high producer), A. flavus KSU114 (low producer), and A. flavus KSU121 (non-producer). The A. flavus isolates were shown to be resilient in terms of growth on yeast extract sucrose agar media when exposed to temperature and water activity as pivotal environmental factors. The optimal conditions for the fungal growth of three isolates were a temperature of 34 °C and water activity of 0.95 aw; there was very slow fungal growth at the highest temperature of 42 °C, with different aw values causing inhibited fungal growth. The AFB1 production for the three isolates followed the same pattern with one exception: A. flavus KSU114 failed to produce any AFB1 at 42 °C with different aw values. All tested genes of A. flavus were significantly up- or downregulated under three levels of interaction between temperature and aw. The late structural genes of the pathway were significantly upregulated at 34 °C under aw 0.95, although aflR, aflS and most of the early structural genes were upregulated. Compared to 34 °C with an aw value of 0.95, most of the expressed genes were significantly downregulated at 37 and 42 °C with aw values of 0.85 and 0.90. Additionally, two regulatory genes were downregulated under the same conditions. The expression level of laeA was also completely associated with AFB1 production, while the expression level of brlA was linked to A. flavus colonization. This information is required to forecast the actual effects of climate change on A. flavus. The findings can be applied to improve specific food technology processes and create prevention strategies to limit the concentrations of potential carcinogenic substances in peanuts and their derivatives.

This is illustrated by a case presentation in which discrepancies of shape and diameter between sizer and valve mounted on the deployment system resulted in discarding the prosthesis and performing a biocomposite root replacement instead. Modification of the valve sizer will allow for safer sizing because the sizing process will mimic the implantation process more accurately.

Abstract:Aspergillus flavus is a saprophytic fungus that may colonize several important crops, including cotton, maize, peanuts and tree nuts. Concomitant with A. flavus colonization is its potential to secrete mycotoxins, of which the most prominent is aflatoxin. Temperature, water activity (aw) and carbon dioxide (CO₂) are three environmental factors shown to influence the fungus-plant interaction, which are predicted to undergo significant changes in the next century. In this study, we used RNA sequencing to better understand the transcriptomic response of the fungus to aw, temperature, and elevated CO₂ levels. We demonstrate that aflatoxin (AFB₁) production on maize grain was altered by water availability, temperature and CO₂. RNA-Sequencing data indicated that several genes, and in particular those involved in the biosynthesis of secondary metabolites, exhibit different responses to water availability or temperature stress depending on the atmospheric CO₂ content. Other gene categories affected by CO₂ levels alone (350 ppm vs. 1000 ppm at 30 °C/0.99 aw), included amino acid metabolism and folate biosynthesis. Finally, we identified two gene networks significantly influenced by changes in CO₂ levels that contain several genes related to cellular replication and transcription. These results demonstrate that changes in atmospheric CO₂ under climate change scenarios greatly influences the response of A. flavus to water and temperature when colonizing maize grain.

Abstract:Abstract Aflatoxin is a group of polyketide-derived carcinogenic and mutagenic secondary metabolites produced by Aspergillus flavus that negatively impact global food security and threaten the health of both humans and livestock. Aflatoxin biosynthesis is strongly affected by the fungal developmental stage, cultivation conditions, and environmental stress. In this study, a novel float culture method was used to examine the direct responses of the A. flavus transcriptome to temperature stress, oxidative stress, and their dual effects during the aflatoxin production stage. The transcriptomic response of A. flavus illustrated that the co-regulation of different secondary metabolic pathways likely contributes to maintaining cellular homeostasis and promoting cell survival under stress conditions. In particular, aflatoxin biosynthetic gene expression was downregulated, while genes encoding secondary metabolites with antioxidant properties, such as kojic acid and imizoquins, were upregulated under stress conditions. Multiple mitochondrial function-related genes, including those encoding NADH:ubiquinone oxidoreductase, ubiquinol-cytochrome C reductase, and alternative oxidase, were differentially expressed. These data can provide insights into the important mechanisms through which secondary metabolism in A. flavus is co-regulated and facilitate the deployment of various approaches for the effective control and prevention of aflatoxin contamination in food crops.

Abstract:Almonds are among the commodities at risk of aflatoxin contamination by Aspergillus flavus. Temperature and water activity are the two key determinants in pre and post-harvest environments influencing both the rate of fungal spoilage and aflatoxin production. Varying the combination of these parameters can completely inhibit or fully activate the biosynthesis of aflatoxin, so it is fundamental to know which combinations can control or be conducive to aflatoxin contamination. Little information is available about the influence of these parameters on aflatoxin production on almonds. The objective of this study was to determine the influence of different combinations of temperature (20 °C, 28 °C, and 37 °C) and water activity (0.90, 0.93, 0.96, 0.99 aw) on growth, aflatoxin B1 (AFB1) production and expression of the two regulatory genes, aflR and aflS, and two structural genes, aflD and aflO, of the aflatoxin biosynthetic cluster in A. flavus grown on an almond medium solidified with agar. Maximum accumulation of fungal biomass and AFB1 production was obtained at 28 °C and 0.96 aw; no fungal growth and AFB1 production were observed at 20 °C at the driest tested conditions (0.90 and 0.93 aw). At 20° and 37 °C AFB1 production was 70-90% lower or completely suppressed, depending on aw. Reverse transcriptase quantitative PCR showed that the two regulatory genes (aflR and aflS) were highly expressed at maximum (28 °C) and minimum (20 °C and 37 °C) AFB1 production. Conversely the two structural genes (aflD and aflO) were highly expressed only at maximum AFB1 production (28 °C and 0.96-0.99 aw). It seems that temperature acts as a key factor influencing aflatoxin production which is strictly correlated to the induction of expression of structural biosynthesis genes (aflD and aflO), but not to that of aflatoxin regulatory genes (aflR and aflS), whose functional products are most likely subordinated to other regulatory processes acting at post-translational level. The results of this study are useful to select conditions that could be used in the almond processing chain to suppress aflatoxin production in this important product.

Abstract:Corn genotypes evaluated for aflatoxin resistance in field studies must be artificially inoculated with Aspergillus flavus due to the sporadic nature of aflatoxin contamination from year to year. We compared a spray inoculation technique with the side-needle technique and natural infection for inducing aflatoxin contamination in commercial corn hybrids in two soil types from 2000 to 2006. Hybrids inoculated using the side-needle technique consistently had high levels of aflatoxin contamination. Hybrids inoculated with the spray technique and grown in a loam soil were more prone to drought stress and had higher levels of aflatoxin contamination than those grown in a silty clay loam soil with a high water-holding capacity. The association of maximum daily temperatures and cumulative rainfall during nine periods before or after A. flavus inoculation with aflatoxin contamination was determined. In the silty clay loam, maximum daily temperatures during the 14-day period following inoculation were positively correlated with aflatoxin contamination. Cumulative rainfall during periods of 14 to 28 days and 21 to 42 days following inoculation was negatively correlated with aflatoxin contamination. In the loam, maximum daily temperatures before and after inoculation were positively correlated with aflatoxin contamination. Also, cumulative rainfall during a period 21 to 42 days before inoculation was negatively correlated with aflatoxin contamination. Our results also demonstrated that temperature and moisture stress prior to inoculations may have a significant effect on aflatoxin contamination.

Mitigating aflatoxin B1 in high-moisture sorghum silage: Aspergillus flavus growth and aflatoxin B1 prediction

Abstract:Aspergillus flavus (A. flavus), a frequent contaminant in silage, is a significant producer of aflatoxins, notably the potent carcinogen aflatoxin B1. This contaminant poses a potential risk during the initial aerobic phase of ensiling. The present work studied the impact of temperature on A. flavus growth and aflatoxin B1 production in laboratory-scale sorghum silos during the initial aerobic phase. Growth curves of A. flavus were generated at various temperatures and modeled with the Gompertz model. Results indicated that the optimal temperature range for the maximum growth rate in sorghum mini-silos is between 25 and 30°C. Mold biomass and aflatoxin B1 levels were quantified using qPCR and HPLC, respectively. A predictive model for aflatoxin B1 synthesis in the initial ensiling phase was established, in function of grain moisture, external temperature, and time. Within the studied range, A. flavus's initial concentration did not significantly influence aflatoxin B1 production. According to the model maximum aflatoxin production is expected at 30% moisture and 25°C temperature, after 6 days in the aerobic phase. Aflatoxin B1 production in such conditions was corroborated experimentally. Growth curves and aflatoxin B1 production highlighted that at 48 h of incubation under optimal conditions, aflatoxin B1 concentrations in mini-silos exceeded national legislation limits, reaching values close to 100 ppb. These results underscore the risk associated with A. flavus presence in ensiling material, emphasizing the importance of controlling its development in sorghum silos.

The effect of temperature and relative humidity for Aspergillus flavus BIO 2237 growth and aflatoxin production on soybeans

Abstract:Aspergillus flavus (A. flavus) producing aflatoxin frequently contaminates crops such as soybeans. The growth of this mold on soybeans and other foodstuffs is affected by temperature and relative humidity (RH). The aim of this study was to measure the growth of A. flavus BIO 2237 and aflatoxin production at different temperatures and RH. Aspergillus flavus BIO 2237 was isolated from Indonesia origin foodstuffs. Aspergillus flavus BIO 2237 was inoculated in Czapek Dox Agar (CDA) and soybeans for 10 days at a temperature of 20, 30, and 40 o C with RH of 70, 80, and 90%. Aflatoxin analysis was conducted using RP-HPLC equipped with fluorescence detector and post column photochemical reactor. The limit of detection (LoD) for aflatoxin of B 1 , B 2 , G 1 , and G 2 was 0.45, 0.26, 0.05, and 0.13 ng/mL (ppb), while their limit of quantification (LoQ) was 1.50, 0.88, 0.18 and 0.43 ng/mL (ppb) respectively. The maximum growth for A. flavus BIO 2237 in CDA and soybeans was reached at a temperature of 30 o C with RH of 90%, and this was based on the highest diameter of colony and amount of cell mass formed in that condition. The maximum level of aflatoxin in contaminated soybeans was found at 999 ng/g (ppb), and this was produced at the same condition as its fungi’s growth. Aspergillus flavus BIO 2237 can not grow as well as produce aflatoxin in soybeans at high temperature (40oC) with low RH (70%). There was a significant difference (sig<0.05) in aflatoxin content (AFB1, AFB2, AFG1, and AFG2) between temperature and RH, meanwhile the difference on the growth of A. flavus BIO 2237 in CDA and soybeans caused by RH.

Abstract:Aspergillus section Flavi (Flavi) is a diverse group of fungal species whose common members include A. flavus and A. parasiticus. These are well-known for the production of aflatoxin (AF) B and G and other toxic metabolites, like cyclopiazonic acid (CPA). They are saprophytic soil dwellers and also become crop opportunistic epiphytes. The consequence is contamination of the crop with mycotoxins, such as carcinogenic AF. We investigated the Flavi community structure of maize and that of their surrounding soil, including their mycotoxigenicity. Furthermore, we investigated the link of the maize Flavi diversity with preharvest maize AF levels. The study was carried out in four selected districts of Zambia, in a low rainfall zone. The Flavi characterisation was triphasic, involving morphological (colony colour and sclerotia formation), metabolic (AF and CPA production) and genetic (calmodulin gene polymorphism) analyses. Flavi abundance was determined by dilution plate technique on modified rose Bengal agar. Results showed that Flavi communities on maize and in soil differed. Maize had a higher Flavi species diversity than soil. A. parasiticus dominated the soil community by frequency of field appearance (85%), while maize was dominated by A. minisclerotigenes (45%). CPA-producers with or without AF production dominated the maize (65%) while producers of only AF (B/G) dominated the soil (88%). The ratio between maize A. parasiticus and A. minisclerotigenes abundance seemed to have had a bearing on the levels of AF in maize, with a ratio close to 1:1 having higher levels than a pure community of either A. parasiticus or A. minisclerotigenes.
The Master Transcription Factor mtfA Governs Aflatoxin Production, Morphological Development and Pathogenicity in the Fungus Aspergillus flavus

Abstract:Aspergillus flavus produces a variety of toxic secondary metabolites; among them, the aflatoxins (AFs) are the most well known. These compounds are highly mutagenic and carcinogenic, particularly AFB₁. A. flavus is capable of colonizing a number of economically-important crops, such as corn, cotton, peanut and tree nuts, and contaminating them with AFs. Molecular genetic studies in A. flavus could identify novel gene targets for use in strategies to reduce AF contamination and its adverse impact on food and feed supplies worldwide. In the current study, we investigated the role of the master transcription factor gene mtfA in A. flavus. Our results revealed that forced overexpression of mtfA results in a drastic decrease or elimination of several secondary metabolites, among them AFB₁. The reduction in AFB₁ was accompanied by a decrease in aflR expression. Furthermore, mtfA also regulates development; conidiation was influenced differently by this gene depending on the type of colonized substrate. In addition to its effect on conidiation, mtfA is necessary for the normal maturation of sclerotia. Importantly, mtfA positively affects the pathogenicity of A. flavus when colonizing peanut seeds. AF production in colonized seeds was decreased in the deletion mtfA strain and particularly in the overexpression strain, where only trace amounts were detected. Interestingly, a more rapid colonization of the seed tissue occurred when mtfA was overexpressed, coinciding with an increase in lipase activity and faster maceration of the oily part of the seed.

Abstract:Climate change is primarily manifested by elevated temperature and carbon dioxide (CO2) levels and is projected to provide suitable cultivation grounds for pests and pathogens in the otherwise unsuitable regions. The impacts of climate change have been predicted in many parts of the world, which could threaten global food safety and food security. The aim of the present work was therefore to examine the interacting effects of water activity (aw) (0.92, 0.95, 0.98 aw), CO2 (400, 800, 1200 ppm) and temperature (30, 35 °C and 30, 33 °C for Fusarium verticillioides and F. graminearum, respectively) on fungal growth and mycotoxin production of acclimatised isolates of F. verticillioides and F. graminearum isolated from maize. To determine fungal growth, the colony diameters were measured on days 1, 3, 5, and 7. The mycotoxins produced were quantified using a quadrupole-time-of-flight mass spectrometer (QTOF-MS) combined with ultra-high-performance liquid chromatography (UHPLC) system. For F. verticillioides, the optimum conditions for growth of fumonisin B1 (FB1), and fumonisin B2 (FB2) were 30 °C + 0.98 aw + 400 ppm CO2. These conditions were also optimum for F. graminearum growth, and zearalenone (ZEA) and deoxynivalenol (DON) production. Since 30 °C and 400 ppm CO2 were the baseline treatments, it was hence concluded that the elevated temperature and CO2 levels tested did not seem to significantly impact fungal growth and mycotoxin production of acclimatised Fusarium isolates. To the best of our knowledge thus far, the present work described for the first time the effects of simulated climate change conditions on fungal growth and mycotoxin production of acclimatised isolates of F. verticillioides and F. graminearum.

Abstract:Increased frequencies of Aspergillus section Flavi and aflatoxins in cereal grains have been seen in recent years due to changes in climate circumstances, such as high temperatures and drought. To assess the microbiological risks of contamination, it is critical to have a reliable and accurate means of identifying the fungi. The main goal of this study was to characterize Aspergillus species from section Flavi obtained from twenty-three samples of barley and maize grains, gathered from different markets in Qena, Egypt, using morphological and molecular techniques. Twenty-three isolates were chosen, one isolate from each sample; they were identified as A. aflatoxiformans (4 isolates), A. flavus (18), and A. parasiticus (1) The existence of four aflatoxin biosynthesis genes was also investigated in relation to the strains' ability to produce total aflatoxins and aflatoxin B1, focusing on the regulatory gene aflR and the structural genes aflD and aflM . All strains producing aflatoxins were linked to the presence of aflR1 and/or aflR2 , except two isolates that exhibited aflatoxins but from which aflR1 or aflR2 were not detected, which may be due to one or more missing or unstudied additional genes involved in aflatoxin production. AflD and aflM genes were amplified by 10 and 9 isolates, respectively. Five samples of barley and maize were contaminated by aflatoxins. Fifteen isolates were positive for producing total aflatoxins in the range of 0.1–240 ppm. Antagonistic activity of Trichoderma viride against A. flavus (F5) was assessed at 31.3%. Trichoderma reduced total aflatoxins in all treated seeds, particularly those subjected to Trichoderma formulation.

Abstract:A currently utilized pre-harvest biocontrol method involves field inoculations with non-aflatoxigenic Aspergillus flavus strains, a tactic shown to strategically suppress native aflatoxin-producing strains and effectively decrease aflatoxin contamination in corn. The present in situ study focuses on tracking the invasion and colonization of an aflatoxigenic A. flavus strain (AF70), labeled with green fluorescent protein (GFP), in the presence of a non-aflatoxigenic A. flavus biocontrol strain (AF36), to better understand the competitive interaction between these two strains in seed tissue of corn (Zea mays). Corn kernels that had been co-inoculated with GFP-labeled AF70 and wild-type AF36 were cross-sectioned and observed under UV and blue light to determine the outcome of competition between these strains. After imaging, all kernels were analyzed for aflatoxin levels. There appeared to be a population difference between the co-inoculated AF70-GFP+AF36 and the individual AF70-GFP tests, both visually and with pixel count analysis. The GFP allowed us to observe that AF70-GFP inside the kernels was suppressed up to 82% when co-inoculated with AF36 indicating that AF36 inhibited progression of AF70-GFP. This was in agreement with images taken of whole kernels where AF36 exhibited a more robust external growth compared to AF70-GFP. The suppressed growth of AF70-GFP was reflected in a corresponding (upto 73%) suppression in aflatoxin levels. Our results indicate that the decrease in aflatoxin production correlated with population depression of the aflatoxigenic fungus by the biocontrol strain supporting the theory of competitive exclusion through robust propagation and fast colonization by the non-aflatoxigenic fungus.

Abstract:Abstract Aflatoxins count to the most toxic known mycotoxins and are a threat to food safety especially in regions with a warm and humid climate. Contaminated food reaches consumers globally due to international trade, leading to stringent regulatory limits of aflatoxins in food. While the formation of aflatoxin (AF) B 1 by the filamentous fungus Aspergillus flavus is well investigated, less is known about the formation kinetics of its precursors and further aflatoxins. In this study, autoclaved maize kernels were inoculated with A. flavus and incubated at 25 °C for up to 10 days. Aflatoxins and precursors were analyzed by a validated UHPLC-MS method. Additional to AFB 1 and AFB 2 , AFM 1 and AFM 2 were detected, confirming the ability of the formation of M-group aflatoxins on cereals by A. flavus . The measured relative levels of AFB 2 , AFM 1 , and AFM 2 on maize compared to the level of AFB 1 (mean of days 5, 7, and 10 of incubation) were 3.3%, 1.5%, and 0.2%, respectively. The occurrence and kinetics of the measured aflatoxins and their precursors sterigmatocystin, O-methylsterigmatocystin, 11-hydroxy-O-methylsterigmatocystin, aspertoxin, and 11-hydroxyaspertoxin (group 1) as well as of dihydrosterigmatocystin and dihydro-O-methylsterigmatocystin (group 2) supported the so far postulated biosynthetic pathway. Remarkable high levels of O-methylsterigmatocystin and aspertoxin (17.4% and 4.9% compared to AFB 1 ) were found, raising the question about the toxicological relevance of these intermediates. In conclusion, based on the study results, the monitoring of O-methylsterigmatocystin and aspertoxin as well as M-group aflatoxins in food is recommended.

Abstract:Aims: This study was to illustrate the relationship between the thermotolerance and the contents of hydrophobin-like or formic-acid-extractable (FAE) proteins in aerial conidia of Beauveria bassiana and Paecilomyces fumosoroseus produced on rice-based substrate. Methods and Results: Survival indices of 11 isolates were separately assessed as a ratio of the viability of conidia after 3-150 min thermal stress at 48degreesC over that of unstressed conidia and fitted well to a survival model (r(2) greater than or equal to 0.97). For a given isolate, the fitted model generated an LT50, the time for 50% viability loss under the stress. The LT(50)s of six B. bassiana isolates (10.1-61.9 min) and five P. fumosoroseus isolates (2.8-6.2 min) were correlated (r(2) = 0.81) with FAE protein contents (6.9-23.4 mug mg(-1)). The survival indices of a fixed B. bassiana isolate after 45-min thermal stress at 48degreesC were also correlated to the FAE protein contents from conidia produced on glucose-, sucrose-, or starch-based substrate (0.79 less than or equal to r(2) less than or equal to 0.86). Three bands of 12.0, 15.0 and 17.5 kDa were recognized from SDS-PAGE profiles of protein extracts from different isolates or from the fixed isolate grown on the three carbon sources, but the 12 kDa was less likely related to the thermotolerance. Conclusions: Approximately 80% of variability in conidial thermotolerance was attributed to either 15.0- or 17.5-kDa FAE protein or both. Significance and Impact of the Study: The relationship between conidial thermotolerance and FAE protein contents implicates a new way to search for fungal biocontrol agents with more thermotolerance and environmental persistence.

Abstract:Aspergillus flavus is an opportunistic pathogen responsible for millions of dollars in crop losses annually and negative health impacts on crop consumers globally. A. flavus strains have the potential to produce aflatoxin and other toxic secondary metabolites, which often increase during plant colonization. To mitigate the impacts of this international issue, we employ a range of strategies to directly impact fungal physiology, growth and development, thus requiring knowledge on the underlying molecular mechanisms driving these processes. Here we utilize RNA-sequencing data that are obtained from in situ assays, whereby Zea mays kernels are inoculated with A. flavus strains, to select transcription factors putatively driving virulence-related gene networks. We demonstrate, through growth, sporulation, oxidative stress-response and aflatoxin/CPA analysis, that three A. flavus strains with knockout mutations for the putative transcription factors AFLA_089270, AFLA_112760, and AFLA_031450 demonstrate characteristics such as reduced growth capacity and decreased aflatoxin/CPA accumulation in kernels consistent with decreased fungal pathogenicity. Furthermore, AFLA_089270, also known as HacA, eliminates CPA production and impacts the fungus’s capacity to respond to highly oxidative conditions, indicating an impact on plant colonization. Taken together, these data provide a sound foundation for elucidating the downstream molecular pathways potentially contributing to fungal virulence.
