Abstract:We have firstly demonstrated fiber surface enhanced Raman scattering (SERS) sensor based on gold–silver nanoalloys fabricated by a simple and effective photochemical co-reduction method. The nanoalloys are separated into two phases: one phase located in the central circular area of fiber end puts particular emphasis on showing the dispersibility of gold, while the other one in the concentric ring region reflects more on the high enhancement effect of silver. Combining superiorities of gold and silver, fiber SERS sensor based on the biphasic gold–silver nanoalloys shows good reproducibility and high sensitivity.

Abstract:Understanding of the influence of nanotextured surface topographies on surface-enhanced Raman scattering (SERS) is still rather limited, even after intensive research in recent years; this is mainly due to the absence of a synthesis system to control the surface morphologies on metal meso-/nanoparticles. Here, a facile particle-mediated aggregation protocol was reported to synthesize a series of uniform gold mesoparticles with a nanotextured rough surface. The surface topographies can be precisely controlled by varying the temperatures and concentrations of dopa and Au ions. For example, the tips can be well changed from half-sphere to spike, and then to blade, and the size of the tips also can be tuned, such that the thickness of the blade-like tips can be artificially controlled from 9 to 14 nm. Single-particle SERS analysis reveals that the uniform Au mesoparticles with highly nanotextured topography have enhancement factors greater than 1.0 x 10(8) with narrow distribution. The effect of the precisely controlled mesoparticle topographies on SERS properties was deeply studied using Raman analysis and finite-difference time-domain (FDTD) simulation. The result indicated that a strong electric field will be induced at the tips and slits on the flower-like mesoparticle surface by the confined light in the cavities between the petals. As such, the current protocol opens up avenues for the fabrication of structurally reproducible mesosuperstructure-based SERS sensors.

Abstract:Surface-enhanced Raman scattering (SERS) probes based on gold nanoparticles modifying the core of the optical fiber were made by a surfactantless photochemical deposition method. The growth kinetics and shape evolution of gold nanoparticles depending on different experimental conditions were studied. It was found that, under the condition of detectable gold nanoparticle deposition, increasing the concentration of chloroauric acid (HAuCl(4)) was not conducive to the deposition whereas increasing the concentration of sodium citrate (Na(3)Ct) would speed up the deposition. By controlling the concentration of the reaction solution and irradiation time, we obtained fused spherical-like, spherical, and flowerlike gold nanoparticles. To test the SERS activity of the probes, the SERS spectra of a rhodamine 6G aqueous solution were recorded in direct detection mode and remote mode. We have also developed a new approach to improving the SERS sensitivity when detecting in remote mode.

Abstract:The surface-enhanced Raman scattering (SERS) optical fiber probes were successfully prepared by self-assembling on polyelectrolyte multilayers. Gold nanorods (Au NRs) were used as SERS enhancement material to give excellent biological affinity and stability to the SERS optical fiber probes. Au NRs were synthesized by seed growth method. The synergistic effect between AgNO3 and surfactant was investigated, and the highest yield was found when AgNO3 was 500 uL. Meanwhile, different SERS optical fiber probes were obtained by selecting silane coupling agent, polyelectrolyte multilayer and graphene oxide (GO) to treat quartz fiber. It was found that the SERS optical fiber probes obtained by the self-assembled on polyelectrolyte multilayers method performed better than those by other methods. In addition, Mapping was combined with finite element simulation to analyze the electromagnetic field distribution at the fiber end face. The electromagnetic field distribution of Au NRs was investigated, the difference of electromagnetic field intensity around the Au NRs with different arrangements was compared, the strongest signal was obtained when the Au NRs were head-to-head. Finally, sensitivity of the optimized SERS optical fiber probes could reach 10−9 mol/L, with excellent stability and repeatability.

Abstract:Remote sensing remains a challenge due to its demand for high sensitivity, convenient sampling and rapid response time. Surface enhanced Raman scattering (SERS) spectroscopy is a powerful analytical method for the detection of various samples. Here, aiming at increasing the sensitivity, a novel strategy for the preparation of a SERS probe is demonstrated by using hollow optical fiber tips decorated by layer-by-layer assembly of two kinds of nanoparticles. Specifically, Au@Ag core-shell nanorods and Ag nanospheres with opposite surface charge were assembled layer-by-layer on the tip of hollow optical fibers through electrostatic interaction. Then, much more hotspots are generated due to the close gap between the nanorods and nanospheres in the resultant 3D structure, which can lead to a dramatically enhanced SERS activity of the probe compared with that fabricated by pure silver sphere nanoparticles or nanorods. On the other hand, taking the advantages of the vibration spectroscopic fingerprints property of SERS spectra and the long-distance communication capacity of optical fibers, the remote online detection of biological species including proteins, funguses and cells can be easily achieved within a few minutes. Therefore, such a novel kind of optical fiber-SERS sensor holds great potential for the rapid detection of a wide range of samples due to its superiority of simplicity and high sensitivity.

Abstract:The development of intrinsic SERS fiber optic sensors, i.e., fiber optical sensors that also serve as SERS active platforms, is challenging in that an easy, robust method that integrates the SERS active platform with fiber optics is still largely missing. There is a trade-off between implementing optimal morphology of SERS active nanostructures for best enhancement effect and preserving optical transparency that allows maximum transmission of the excitation radiation and the detected signals. In the present work, highly sensitive and reproducible silver nanorod arrays (AgNRs) have been integrated to a fiber optic probe for SERS detection. The films underlying the AgNR coating have been tailored to allow maximum light transmission while maintaining optimal SERS activity. The intense spectral background from the probe fiber core is largely eliminated by using a GRIN lens to produce a tight focus of the incident radiation at the AgNR coating. The performance of the AgNR fiber optic probes has been evaluated in a forward scattering optical configuration using BPE and adenine. The low detection limit of BPE and adenine is 10−7M. Reproducibility and sampling methods are also discussed.

Abstract:We report on surface-enhanced Raman scattering (SERS) probes based on silver nanoparticles which are deposited on the core of the distal end of standard single mode fibers, by means of the very simple, versatile, and low-cost laser-induced nanoparticle technique. The morphological features of the Ag nanoparticles vary according to the experimental conditions such as laser power, illumination time, and concentration of the reaction solution. The SERS activity of the probes is demonstrated with the detections of rhodamine 6G aqueous solutions. The detections are made from both ends of the probes, i.e., in direct detection mode from the end with nanoparticles and in remote detection mode from the distal end, respectively.

Abstract:The optical properties of silver and gold sols with different sizes of nanoparticles and the method of their chemical deposition on the surface of silicon, silica, glass, and aluminum foil were studied in order to obtain SERS substrates, promising platforms for the development of aptamer sensors and immunoassay of various pathogens. It was determined that, for operation of lasers with exciting radiation wavelengths of 532, 638, and 785 nm, it is possible to create universal SERS substrates based on colloidal solutions obtained by the liquid-phase chemical method with an average silver particle size of 40 nm and by the Leopold–Lendl method with an average size of 20 nm.

Abstract:Biocompatible optical fibers and waveguides are gaining attention as promising platforms for implantable biophotonic devices. Recently, the distinct properties of silk fibroin were extensively explored because of its unique advantages, including flexibility, process compatibility, long-term biosafety, and controllable biodegradability for in vitro and in vivo biomedical applications. In this study, we developed a novel silk fiber for a sensitive optical sensor based on surface-enhanced Raman spectroscopy (SERS). In contrast to conventional plasmonic nanostructures, which employ expensive and time-consuming fabrication processes, gold nanoparticles were uniformly patterned on the top surface of the fiber employing a simple and cost-effective convective self-assembly technique. The fabricated silk fiber-optic SERS probe presented a good performance in terms of detection limit, sensitivity, and linearity. In particular, the uniform pattern of gold nanoparticles contributed to a highly linear sensing feature compared to the commercial multi-mode fiber sample with an irregular and aggregated distribution of gold nanoparticles. Through further optimization, silk-based fiber-optic probes can function as useful tools for highly sensitive, cost-effective, and easily tailored biophotonic platforms, thereby offering new capabilities for future implantable SERS devices.

Abstract:As a highly functional platform, lab-on-fiber (LOF) has exhibited amazing potential to be used in ultrasensitive surface-enhanced Raman spectroscopy (SERS) detection. However, the low detection efficiency and high cost of most SERS-based LOF platforms have severely limited their further applications in environmental monitoring, biomedicine, etc. Here, we report a reusable SERS-based LOF platform through a Au-coated optical fiber integrated into a patterned microfluidic chip. With the assistance of laser-induced thermophoresis, we successfully obtained rich “hot spots” on the Au-coated optical fiber by reversible accumulation of gold nanoparticles within 5 min. Experimental results indicate that it has a low detection limitation of 10–10 mol/L (M) for environmental pollutant molecules. Fortunately, with the dynamic and reversible accumulation of gold nanoparticles controlled by laser on/off, the platform shows excellent reversibility and stability under multiple detection–cleaning cycles, which are better than those of the LOF platform based on chemical modification and photochemical deposition. The thermophoresis-based LOF platform with the advantages of easy operation, miniaturization, and high reusability provides a cost-effective approach for SERS detection.

Fabrication of Silver Nano-Dendrites on Optical Fibre Core by Laser-Induced Method for Surface-Enhanced Raman Scattering Applications

Abstract:In this work, we present a novel fabrication method for making the surface-enhanced Raman scattering (SERS) probe based on silver (Ag) nano-dendrites which are grown and deposited on the end of multi-mode fibre core by a simple and low-cost laser-induced technique. The morphology of the Ag-nanoparticles (AgNPs) could be controlled by the experimental conditions such as laser power, illumination time, and concentration of the reaction solution. The morphology and chemical composition of SERS fibre probes are characterized by high-resolution scanning electron microscope (HR-SEM) and Energy dispersive X-ray spectroscopy (EDX), respectively. These results confirmed how the Ag nanostructures morphology is modified as a function of illumination time of laser irradiation, and the growth and deposition of Ag nanostructures occur only in the main laser-irradiated part on the end of multi-mode fibre core. The achieved SERS-activity substrates on the fibre probes are testing with the detection of low concentration of Rhodamine 6G aqueous solutions in the range of 10-5-10-10 M. This study shows that SERS activity coupled with Ag nano-dendrites substrate on the fibre probe has the best enhancement factor of 1.93×107 for Rhodamine 6G due to the creation of many of hot-spots for amplifying Raman signals by Ag nano-dendrite structures, which is a promising candidate with low-cost SERS probe of chemical compact optical fibre sensors for direct, rapid, real-time and non-destructive detection of chemical compounds in liquid environment.

Abstract:A simple ethanol sol-based method for the synthesis of gold nanosheets (AuNSs) and gold nanoparticle-over-nanosheet (AuNP/NS) is developed. Gold nanoparticles (AuNPs) with average sizes of approximate to 8 nm are grown in situ on the surface of the AuNS, which forms a NP/NS structure that obtains strong, significantly improved, surface-enhanced Raman spectroscopy activity with the magnitude approximate to 2 and approximate to 6 orders higher than the simplex AuNP and AuNS, respectively. This performance is mainly attributed to uniform AuNPs that are closely packed over AuNS and coupled with NP-NS and NP-NP interactions. The NP-NS-GP (the gap between NP-NS) is narrower than NP-NP-GP in which much stronger and steadier plasmon resonance is obtained that can significantly enhance the Raman signal. The results show that single-crystalline AuNS is an ideal substrate, which can be further coated with other metallic NPs to form a new flexible, high-activity and AuNS-based nanocomposite for a wide variety of applications.

Abstract:Uniform silver-containing metal nanostructures with strong and stable surface enhanced Raman scattering (SERS) signals hold great promise for developing ultrasensitive probes for biodetection. Nevertheless, the direct synthesis of such ready-to-use nanoprobes remains extremely challenging. Herein we report a DNA-mediated gold silver nanomushroom with interior nanogaps directly synthesized and used for multiplex and simultaneous SERS detection of various DNA and RNA targets. The DNA involved in the nanostructures can act as hot only gap DNA (mediated DNA) but also probe DNA (hybridized DNA), and DNA's involvement enables the nanostructures to have the inherent ability to recognize DNA and RNA targets. Importantly, we were the first to establish a new method for the generation of multicolor SERS probes using two different strategies. First Raman-labeled alkanethiol probe DNA was assembled on gold nanoparticles, and second, thiol-containing Raman reporters were coassembled with the probe DNA. The ready-to-use probes also give great potential to develop ultrasensitive detection methods for various biological molecules.
Photoreduced Silver Nanoparticles Grown on Femtosecond Laser Ablated, D-shaped Fiber Probe for Surface-Enhanced Raman Scattering.

Abstract:Surface-enhanced Raman scattering (SERS) probes are made by facile photochemical deposition of silver nanoparticles on a femtosecond (fs) laser ablated, D-shaped fiber. The structure and surface morphology of the probe are investigated by scanning electron microscopy. High-quality SERS signals are detected using Rhodamine 6G molecules via an in situ sensing mode. Experimental results show that the SERS signals increase with the increase of the length of fs laser ablated, D-shaped zone. Our D-shaped fiber SERS probe shows a feasible method for a large active area, high performance, and real-time and remote measurement of SERS signals in biochemical analysis.

Abstract:Surface-enhanced Raman spectroscopy (SERS) can boost the pristine Raman signal significantly which could be exploited for producing innovative sensing devices with advanced properties. However, the inherent complexity of SERS systems restricts their further applications in rapid detection, especially in situ detection in narrow areas. Here, we construct an efficient and flexible SERS-based Lab-on-Fiber (LOF) sensor by integrating Ag/Au nanocap arrays obtained by Ag/Au coating polystyrene nanospheres on the optical fiber face. We obtain rich "hot spots" at the nanogaps between neighboring nanocaps, and further achieve SERS performance with the assistance of laser-induced thermophoresis on the metal film that can achieve efficiency aggregation of detected molecules. We achieve a high Raman enhancement with a low detection limitation of 10−7 mol/L for the most efficient samples based on the above sensor. This sensor also exhibits good repeatability and stability under multiple detections, revealing the potential application for in situ detection based on the reflexivity of the optical fiber.

Abstract:The detection performances of noble metal-based surface enhanced Raman spectroscopy (SERS) devices are determined by the compositions and geometries of the metal nanostructures, as well as the substrates. In the current study, long spiky Au-Ag alloy nanostars were synthesized, and both core diameters and spike lengths were controlled by Lauryl sulfobetaine concentrations (as the nanostructure growth skeleton). The long spiky star geometries were confirmed by transmission electron micrograph images. Elements energy dispersive spectrometer mapping confirmed that Au and Ag elements were inhomogeneously distributed in the nanostructures and demonstrated a higher Ag content at surface for potential better SERS performance. Selected synthesized spiky nanostars were uniformly assembled on multi-mode silica fiber for probe fabrication by silanization. The SERS performance were characterized using crystal violet (CV) and rhodamine 6G (R6G) as analyte molecules. The lowest detection limit could reach as low as 10−8 M, with a 6.23 × 106 enhancement factor, and the relationship between analyte concentrations and Raman intensities was linear for both CV and R6G, which indicated the potential qualitative and quantitative molecule detection applications. Moreover, the fiber probes also showed good reproducibility and stability in the ambient atmosphere.

Abstract:The quest to enhance Raman spectroscopic signals through the rational design of plasmonic substrates has enabled the detection and characterization of pharmaceutically important molecules with low scattering cross-sections, such as amino acids and proteins, and is helping in making forays into the diverse field of biomedical sciences. This work presents a simple strategy for synthesizing silver nanoparticles-incorporated alumina nanofibers (Ag-AlNFs) utilizing controlled microwave synthesis for enhancing the surface-enhanced Raman chemical enhancement factor through photo-induced charge accumulation at the plasmonic-dielectric interface. The plasmonic-dielectric fibers serve as excellent charge carrier trappers, as evident from the ultrafast transient absorption spectroscopy studies. Apart from chemical enhancement, the increase in electronic surface charge also enables the protein disulfide bonds to capture these electrons and form a transient disulfide electron adduct radical, which converts to free thiol radical on dissociation. This allows protein molecules to bind to the nanoparticle's surface with the favorable silver thiol bond leading to greater surface affinity and larger SERS enhancement. The proposed Ag-AlNFs represent a cost-effective material that can be potentially used to probe biological systems in a label-free manner by photoactivating the SERS substrate for obtaining higher enhancement factors.
Controlling silver morphology on a cramped optical fiber facet via a PVP-assisted silver mirror reaction for SERS fiber probe fabrication

Abstract:Controlling the morphology of silver nanoparticles on a cramped and curved optical fiber facet is urgently needed to obtain SERS optical fiber probes with high performance. In this work, a fast and simple method, PVP-assisted silver mirror reaction, is employed to synthesize SERS fiber probes. The morphology of silver on a cramped optical fiber facet can be controlled by optimizing the reaction conditions. The influences of reaction conditions, including the PVP/AgNO3 molar ratio, ammonium hydroxide concentration, silver nitrate concentration, type of the reducing saccharide and the pH value of Tollens’ solution, on the morphology and SERS performance of the fiber probes were investigated. The results show that silver nanoparticles can be densely distributed on the optical fiber facet with narrow interparticle gaps by varying these reaction parameters. The resultant SERS fiber probe shows high sensitivity and good reproducibility by using 4-aminothiophenol (4-ATP) as the probing molecule. Furthermore, the localized electromagnetic field of silver nanoparticles giving rise to enormous SERS signal amplification was simulated to clarify the SERS performance of the fiber probe. To the best of our knowledge, in situ detection of the analyte in a confined area and a curved channel using the SERS fiber probe is verified for the first time. The obtained SERS fiber probe shows promising applications in flexible detection for early diagnosis of major diseases and critical conditions including confined, remote and hazardous environments.

Aggregated gold nanoparticles rich in electromagnetic field "hotspots" for surface enhanced Raman scattering

Abstract:A simple method for one-step synthesis of aggregated gold nanoparticles (a-AuNPs) using single-layer carbon dots (s-CDs) as the capping agents has been proposed. The obtained a-AuNPs are mainly composed of several spherical AuNPs of 20-25 nm sized, which aggregate to form nanogaps of ∼1 nm. Furthermore, the obtained a-AuNPs produce a strong localized surface plasmon resonance (LSPR) absorption band centered at around 640 nm, which is quite close to the wavelength of the commonly used 633 nm laser in surface enhanced Raman scattering (SERS). Thus, under the irradiation of 633 nm laser, a lot of electromagnetic field "hot spots" are formed at around the nanogaps, and strong SERS activity is achieved. The obtained a-AuNPs are dropped on tin-foil wafers to fabricate SERS substrates, which show the advantages of high sensitivity, fast response, good repeatability and satisfactory stability. On the basis, a sensitive SERS sensor is developed to detect malachite green in aquaculture water, with a low detection limit of 1 × 10-9 mol/L.

Abstract:Nanoaggregates formed by metal spheres of different radii and interparticle distances represent finite, deterministic, self-similar systems that efficiently concentrate optical fields and act as "nanolenses". Here we verify experimentally the theoretical concept of nanolenses and explore their potential as enhancing nanostructures in surface enhanced Raman scattering (SERS). Self-similar structures formed by gold nanospheres of different sizes are generated by laser ablation from solid gold into water. These nanolenses exhibit SERS enhancement factors on the order of 10(9). The "chemically clean" preparation process provides several advantages over chemically prepared nanoaggregates and makes the stable and biocompatible gold nanolenses potent enhancing structures for various analytical and sensing applications.
