Abstract:A mild and efficient 1,4-diazabicyclo[2.2.2]octane (DABCO)-catalyzed three-component domino reaction was developed for the synthesis of highly functionalized NH-pyrroles from arylglyoxal monohydrates, enamino esters, and cyclic 1,3-dicarbonyl compounds in 1,4-dioxane at room temperature for 0.5 hours. Various substituted NH-pyrroles were obtained in moderate to good yields.

Abstract:A convenient and efficient phosphine-catalyzed sequential annulation domino reaction between dienic sulfones and MBH carbonates has been developed. The allylic carbonate, in this reaction, serves as a new kind of 1,1-dipolar synthon. Abstract:A direct method to construct 3,4-dihydropyridin-2-ones by enzymatic condensation of aldehyde with cyanoacetamide and 1,3-dicarbonyl compounds was developed. One ring and four new bonds (two C–C, one C–N, one CC) were formed in one pot. And reaction conditions involving hydrolases, solvents, substrate molar ratios, and hydrolase loading were optimized. A series of new compounds based on the 3,4-dihydropyridin-2-one core were synthesized by the unprecedented three-component domino reaction.

Abstract:A novel NHC-catalyzed three-component domino strategy to access high functionalized cis-ε-ketoesters with excellent yields (up to 98%) and high stereoselectivities (up to 20:1) is documented. The title domino reactions are atom economical and work on a broad range of substrates. The relative stereochemistry could be explained by a cascade crossed-benzoin/oxy-Cope rearrangement/esterification process. The thus-obtained products are of potential synthetic value in the drug research and combinatorial chemistry.

Abstract:The domino cyclization/coupling strategy is one of the most effective methods to produce cyclized and multi-functionalized compounds from olefins, which has attracted huge attention from chemists and biochemists especially for its considerable potential of enantiocontrol. Nowadays, more and more studies are developed to achieve difunctionalization of substituted olefins through an asymmetric domino intramolecular cyclization/cascade reaction, which is still an elegant choice to accomplish several synthetic ideas such as complex natural products and drugs. This review surveys the recent advances in this field through reaction type classification. It might serve as useful knowledge desktop for the community and accelerate their research.

Abstract:An alternative route has been developed for the construction of multisubstituted pyrrole derivatives from readily available, doubly activated cyclopropanes and anilines using an iron-mediated oxidation domino reaction (i.e., sequential ring-opening, cyclization, and dehydrogenation reactions). This reaction uses readily available reactants and is tolerant of a broad range of substrates, with the desired products being formed in good to excellent yields.

Abstract:A silver-catalyzed domino reaction of ortho-carbonylated alkynyl-substituted arylaldehydes with conjugated dienes is described here. Through this reaction, the synthesis of a variety of indanone-fused cyclohexene derivatives can be achieved efficiently. The formation of these tricyclic products could involve a key Diels-Alder reaction of in situ generated indanenone dienophiles with conjugated dienes. Abstract:Highly enantioselective and chemodivergent domino reactions between γ-substituted allenoates and activated alkenes have been developed. In the presence of NUSIOC-Phos, triketone enone substrates smoothly reacted with γ-substituted allenoates to form bicyclic furofurans in good yields with high stereoselectivities. Alternatively, the reaction between diester-activated enone substrates and γ-substituted allenoates formed chiral conjugated 1,3-dienes in good yields with excellent enantioselectivities. Notably, by employing substrates with subtle structural difference, under virtually identical reaction conditions, we were able to access two types of chiral products, which are of biological relevance and synthetic importance.

Abstract:The domino Knoevenagel/Diels-Alder condensation is a highly efficient strategy for the preparation of heterocyclic compounds. In order to study this reaction with an imine catalyst, a three-component reaction involving aromatic aldehydes, benzylideneacetone and meldrum acid catalyzed by a series of new organo-cyclic amino acid catalysts was developed. This simple experiment and environmentally friendly method is beneficial for us to get numerous pharmacological multi substituted spiro[5,5]undecane-1,5,9-triones in moderate to good yields (up to 81%) with excellent diastereolectivities (> 99:1 dr). Moreover, we expanded the reaction substrates, such as barbituric acid, and the catalytic activity was compared with L-proline that is one of the most commonly secondary amine organocatalysts.
