Abstract:Disulfide bond formation is a common mechanism for regulating conformational changes in proteins during oxidative folding. Despite extensive studies of the use of multiple disulfide bonds to constrain peptide conformation, few studies have explored their usage in developing self-assembling peptides. Herein, we report that a thiol-rich peptide could fold into an amphiphilic β-hairpin conformation through the formation of two hetero-disulfide bonds upon oxidation, subsequently self-assembling into a mechanically rigid hydrogel. Breaking disulfide bonds under reductive condition, the hydrogel exhibited a transition from hydrogel to solution. Molecular simulation revealed that intermolecular interaction between two tryptophan residues was indispensable for hydrogelation. This work is the first case of the use of multiple disulfide bonds to control conformational change and self-assembly, and provides a cell-compatible hydrogel material for potential biomedical application.

Abstract:Comprehensive Summary Hydrogels crosslinked by dynamic covalent bonds can effectively mimic the viscoelastic properties of native extracellular matrix and have been widely explored for 3D cell culture. Disulfide is one of the most common dynamic bonds in biological systems whose formation and cleavage are catalyzed by a set of dedicated enzymes. However, in vitro formation of disulfide bonds is a slow process and requires harsh catalysts. Therefore, it is difficult to use disulfide crosslinked hydrogels for cell culture. In this work, we show that disulfide bonds can be formed by thiol-diselenide (DiSe) exchange under blue light illumination. This reaction is fast, reversible, and biocompatible. Moreover, residual diselenide in the hydrogel network can also accelerate thiol-disulfide exchange reactions leading to faster cell release from the hydrogels upon the addition of thiol-containing agents. We anticipate that disulfide crosslinked hydrogels catalyzed by diselenide can find broad biomedical applications, such as cell culture, cell delivery, and drug-controlled release.

Abstract:This work is aimed to report a new type of multiple stimuli-responsive supramolecular polymers by combining dynamic covalent bonds with cucurbit[8]uril (CB[8])-based host-guest interaction. Supramonomer ( DSN) containing a disulfide bond can bind with CB[8] to fabricate supramolecular polymers by host-enhanced pi-pi interaction. Additionally, the dynamic nature of disulfide bonds and host-guest chemistry endow these supramolecular polymers with redox and guest competitive responsiveness, respectively.

Abstract:Disulfide bonds are a significant motif in life and drug-delivery systems. In particular, steric hindrance and stereoscopic disulfide linkers are closely associated with the stability of antibody-drug conjugates, which affects the potency, selectivity, and pharmacokinetics of drugs. However, limited availability and diversity of tertiary thiols impede the construction of steric and stereoscopic disulfides for cross-linkage in biochemistry and pharmaceuticals. Through modulating the mask effect of disulfurating reagents, we develop a facile and robust strategy for construction of diverse steric and stereoscopic disulfides via N-dithiophthalimides. The practical cross-linkage of biomolecules including amino acids, saccharides, and nucleosides with different drugs and fluorescent molecules is successfully established through hindered disulfide linkers.

Abstract:It is of vital importance to reversibly mask and selectively activate bioactive agents for advanced therapeutic and diagnostic purposes, aiming to efficiently suppress background interferences and attenuate systemic toxicity. This strategy has been involved in diverse applications spanning from chemical/biological sensors and diagnostics to drug delivery nanocarriers. Among these, redox-responsive disulfide linkages have been extensively utilized by taking advantage of extracellular and intracellular glutathione (GSH) gradients. However, direct conjugation of cleavable triggers to bioactive agents through disulfide bonds suffers from bulky steric hindrance and limited choice of trigger-drug combinations. Fortunately, the emergence of disulfide self-immolative linkers (DSILs) provides a general and robust strategy to not only mask various bioactive agents through the formation of dynamic disulfide linkages but also make it possible to be selectively activated upon disulfide cleavage in the reductive cytoplasmic milieu. In this review, recent developments in DSILs are focused with special attention on emerging chemical design strategies and functional applications in the biomedical field.

Abstract:In this work, we studied the linker effect on the nanostructures of a serious of specifically designed, functionalized polyhedral oligomeric silsesquioxane (POSS) based Dendron-like macro-isomers. The varying linkers between hydrophilic and hydrophobic POSS cages lead to topological isomers with identical composition but different molecular shapes. Their unique phase behaviors highlight remarkable effects of molecular architectures on the formation of nanostructures. Due to topological constraints, these macro-isomers assume either a fan- or a cone-molecular shape, which assembles into either hexagonally packed cylindrical phase (HEX) or Frank-Kasper A15 phase. This work provides guiding rules on designing precise molecular nanostructures with desired properties via linker engineering.

Abstract:Beyond the common supramolecular helical polymers in solutions, controlling single-crystal helical self-assembly with precisely defined chirality and architectures has been challenging. Here, we report that simply merging static homochiral amino acids with dynamic chiral disulfides can produce a class of building blocks featuring supramolecular helical single-crystal self-assembly with unusual stereodivergency. Analysis of 20 single-crystal structures of 1,2-dithiolanes gives an atom-precision understanding of the chirality transfer from the molecular to supramolecular level, featuring homochiral and heterochiral helical supramolecular self-assembly in the solid state. The underlying structure-assembly relationship reveals that the synergistic interplay of intermolecular H-bonds and the 1,2-dithiolane ring with adaptive chirality plays a key role in determining the assembly pathway, also involving the effects of residue groups, substituents, molecular stacking, and solvents. The confinement effect in the solid state can stabilize the dynamic stereochemistry of disulfide bonds and selectively result in specific conformers that can minimize the energy of global supramolecular systems. We envision that these results represent a starting point to use dynamic chiral disulfide as a functional entity in supramolecular chemistry and may inspire a new class of supramolecular helical polymers with dynamic functions.

Abstract:Chalcogen-bonding interactions have been viewed as new non-covalent forces in supramolecular chemistry. However, harnessing chalcogen bonds to drive molecular self-assembly processes is still unexplored. Here we report for the first time a novel class of supra-amphiphiles formed by Te center dot center dot center dot O or Se center dot center dot center dot O chalcogen-bonding interactions, and their self-assembly into supramolecular vesicles and nanofibers. A quasi-calix[4]chalcogenadiazole (C4Ch) as macrocyclic donor and a tailed pyridine N-oxide surfactant as molecular acceptor are designed to construct the donor-acceptor complex via chalcogen-chalcogen connection between the chalcogenadiazole moieties and oxide anion. The affinity of such chalcogen-bonding can dictate the geometry of supraamphiphiles, driving diverse self-assembled nanostructures. Furthermore, the reversible disassembly of these structures can be promoted by introducing competing halide Ions or by decreasing systemic pH.

Abstract:Owning to the excellent applications of halogen bonds in supramolecular chemistry, the recognition of halogen bonds is of significance during molecular self-assembly process. The 2D self-assembly of fluorenone molecules (BEDF, BCDF, and BHDF) characterized by different terminal groups have been studied at the 1-phenyloctane/ HOPG interface. The H-bonding motifs relative to the terminal groups determine the bonding motifs of the conjugated cores and serve as the trigger of halogen bonds. The COOC2H5-terminated groups of BEDF sever as hydrogen bond acceptors to bond with the cores, giving rise to the dimer configuration of cores linked by C-H center dot center dot center dot Br hydrogen bonds. By contrast, the COOH-terminated groups of BCDF and the OH-terminated groups of BHDF act as both the electrophile and nucleophile interacted with the cores. On account of the secondary hydrogen bond distinct in strength and the different intersection of the aggregates, the type-I bonds (Br center dot center dot center dot Br) and halogen bonds (Br center dot center dot O=C and Br center dot center dot center dot Br) are observed in the networks of BCDF and BHDF, respectively. The types of halogen bonds could be further modified by changing the concentrations. The work clearly demonstrates that an appreciation of terminal functions is critical to the recognition of halogen bonds, thus establishing the controllable self-assembly morphology.

Abstract:The functionality of metal‐organic frameworks (MOFs) is often encoded by particular chemical moieties found within these architectures. As such, new techniques to install increasingly more complex functionalities in MOFs are regularly being reported in the literature. One such functional group is the disulfide bond. The redox behavior of this covalent linkage renders MOFs responsive to stimuli, often under reducing conditions. Here, we review examples in which disulfide‐containing MOFs are deployed in applications including drug delivery, therapeutic ferroptosis, exfoliation, energy storage, sensing, and others.

Novel Method of Preparation of Diblock Copolymers with Disulfide Bonds at Junction Points

Abstract:Synthesis of diblock copolymers containing disulfides at the junction points was reported. Reversible addition-fragmentation chain transfer (RAFT) polymerization of styrene was employed to prepare polystyrene marco RAFT agent (PS-RAFT). The terminal RAFT agents of PS were reduced to sulfhydryl groups (PS-SH) in the presence of primary amine, and the atom transfer radical polymerization (ATRP) macroinitiator (PS-S-S-ATRP) was prepared by an exchange reaction between PS-SH and an ATRP initiator containing disulfide bonds. PS-S-S-ATRP was used to initiate the polymerization of 2-hydroxyethyl methacrylate (HEMA) and PS-S-S-PHEMA diblock copolymers with disulfides at the junction points were obtained. The structures and molecular weights of the diblock copolymers were characterized by proton nuclear magnetic resonance (H-1 NMR) and gel permeation chromatography (GPC). Kinetics study of ATRP of HEMA shows the characteristic of living polymerization. In methanol, the diblock copolymer self-assembles into polymeric micelle with PS cores and PHEMA coronae. Transmission electron microscopy and dynamic light scattering were used to study the morphology changes of the micelles after cleavage of the disulfide bonds. The result shows a new methd to prepare block copolymers with disulfide bonds at the junction points. The self-assenbly strecture of the copolymers will have a wide application in drug release and biological imaging techniques.

Abstract:The dynamic disulfide linkage plays a vital role in various biological processes as well as drugs and biomaterials. The conversion of thiols to their corresponding disulfides is a hallmark of sulfur chemistry, but notoriously difficult to control. Achieving optimal reactivity and selectivity continues to pose significant challenges. Here, we describe a click chemistry for disulfide formation from thiols in both batch and flow-mode using SO2F2, which display exceptional selectivity toward disulfide formation through an effective nucleophilic substitution cascade. This reaction's unique characteristics satisfy the stringent click-criteria with its high thermodynamic driving force, straightforward conditions, wide scope, quantitative yields, exceptional chemoselectivity, and non-chromatographic purification process. The modular synthesis of symmetrical, unsymmetrical, cyclic and polydisulfides is demonstrated, along with the formation of disulfide cross-linked hydrogels.

Abstract:AbstractThanks to their excellent bond strength, phenyl‐based molecules with thiol anchoring groups are extensively employed to form stable single‐molecule junctions. However, two critical questions are still not answered which seriously hinder high‐yield establishing reliable molecular functional devices: (1) Whether molecular dimer junctions will be formed, and if this is the case, whether the dimerization is caused by intermolecular disulfide bonds or π–π stacking of phenyl rings; (2) Upon a mechanical‐compression force, is it possible that both anchoring groups of the molecule bond to the same electrode instead of bridging two opposite electrodes, which would drastically reduce the yield of the molecular junctions. Here, combining UV‐Vis/Raman spectroscopy of bulk molecules and conductance/flicker‐noise measurements of single molecules, we give compelling evidence that molecular dimers naturally form under ambient conditions, primarily via disulfide bonds rather than by π–π stacking. We further proposed a technique, named electrode‐compression‐hold‐on (ECHO), and reveal that the two thiol groups of phenyl‐backboned molecules will bond to the same electrode upon a compression force with a prolongated ECHO time. In contrast, the compression‐time‐dependent phenomenon is not observed for alkyl‐backboned molecules. The underlying mechanism for these unprecedented observations is elucidated, shedding light on the yield of molecular junctions.
Unveiling the Properties of Sulfhydryl Groups in a Single-Molecule Junction.

Abstract:The metal-thiol interface is ubiquitous in nanotechnology and surface chemistry. It is not only used to construct nanocomposites but also plays a decisive role in the properties of these materials. When organothiol molecules bind to the gold surface, there is still controversy over whether sulfhydryl groups can form disulfide bonds and whether these disulfide bonds can remain stable on the gold surface. Here, we investigate the intrinsic properties of sulfhydryl groups on the gold surface at the single-molecule level using a scanning tunneling microscope break junction technique. Our findings indicate that sulfhydryl groups can react with each other to form disulfide bonds on the gold surface, and the electric field can promote the sulfhydryl coupling reaction. In addition to these findings, ultraviolet irradiation is used to effectively regulate the coupling between sulfhydryl groups, leading to the formation and cleavage of disulfide bonds. These results unveil the intrinsic properties of sulfhydryl groups on the gold surface, therefore facilitating the accurate construction of broad nanocomposites with the desired functionalities.

Abstract:Peptide heterodimers are prevalent in nature, which are not only functional macromolecules but molecular tools for chemical and synthetic biology. Computational methods have also been developed to design heterodimers of advanced functions. However, these peptide heterodimers are usually formed through noncovalent interactions, which are prone to dissociate and subject to concentration-dependent nonspecific aggregation. Heterodimers crosslinked with interchain disulfide bonds are more stable, but it represents a formidable challenge for both the computational design of heterodimers and the manipulation of disulfide pairing for heterodimer synthesis and applications. Here, we report the design, synthesis and application of interchain disulfide-bridged peptide heterodimers with mutual orthogonality by combining computational de novo designs with a directed disulfide pairing strategy. These heterodimers can be used as not only scaffolds for generating functional molecules but chemical tools or building blocks for protein labeling and construction of crosslinking hybrids. This study thus opens the door for using this unexplored dimeric structure space for many biological applications.
