Abstract:Lead ion (Pb2+) accumulation in nature can affect the environment and human health severely. Thus, rapid and sensitive detection is of great importance. One-step detection of Pb2+ at attomole levels was realized by using dynamic light scattering (DLS) technique coupled with unmodified gold nanoparticles (AuNPs). Pb2+-dependent DNAzyme was double-stranded and could not adsorb on the surface of AuNPs, while the substrate strand could be cleaved into ssDNA fragments on addition of Pb2+. The ssDNA fragments could adsorb on the surface of AuNPs and prevent them from aggregating in the presence of NaCl. Therefore, the disperse state of AuNPs changed on addition of Pb2+ in the presence of DNAzyme and NaCl, which was estimated with an average hydrodynamic diameter by using DLS. Under optimum conditions, the average diameter of the solution decreased linearly with the concentration of Pb2+ over the range from 10 to 300pM, with a detection limit of 6.2pM. Moreover, satisfactory results were obtained when the proposed method was applied in the detection of Pb2+ in water samples.

Abstract:In the presence of Pb2+, a cleaved enzyme-substrate complex releases ssDNA that adsorbs onto and stabilizes gold nanoparticles (AuNPs) against salt-induced aggregation. In the absence of Pb2+, the uncleaved complex can not stabilize the AuNPs, resulting in purple-blue AuNP aggregates (see figure). The sensor has a low detection limit of 3 nM, a high selectivity, and a tunable dynamic range.

Abstract:A signal-off electrochemiluminescence (ECL) DNA sensor based on gold nanodendrites (Au NDs) modified indium tin oxide (ITO) electrode for the detection of lead ion (Pb(2+)) was developed. Well-defined Au NDs were prepared on ITO electrode using low-potential synthesis, assisted by ethylenediamine. Based on Pb(2+)-specific deoxyribozyme, the silver/zinc oxide (Ag/ZnO) with coupled structure, prepared by one-pot method, was close to the surface of the electrode to catalyze the reduction of part of H2O2, the coreactant for cathodic ECL emission, leading to a decrease of ECL intensity. In addition, taking advantage of the larger surface area to capture a large amount of capture probe as well as excellent conductivity of Au NDs, the sensor could detect Pb(2+) quantitatively in a wider range, and performed excellent selectivity. Furthermore, such simple and sensitive DNA sensor was successfully applied for the detection of Pb(2+) in lake water and human serum samples, respectively.

Abstract:In this paper, we report a novel colorimetric strategy for the detection of small molecules by using Pb(2+) ion as an example. In this strategy, DNAzyme duplex modified gold nanoparticles (GNPs) are designed to be unable to interact with graphene oxide (GO). However, in the presence of Pb(2+), the substrate strand of the DNAzyme is cleaved at its cleavage site, resulting in the disassembly of the DNAzyme duplex modified GNPs into three parts, i.e., the 3'- and 5'-fragments of substrate strand and the DNAzyme strand modified GNPs. By taking advantage of the efficient cross-linking effect of ssDNA-GNPs to GO, colorimetric sensor for the detection of the metal ion can be fabricated with a detection limit of 100 pM, which is much lower than the previous reports. This colorimetric method has also been used for the determination of Pb(2+) in the tap water of the local city and the water from a reservoir with satisfactory results, so it may have potential applications in the future.

Abstract:An electronic DNAzyme sensor for highly sensitive detection of Pb2+ is demonstrated by coupling the significant signal enhancement of the layer-by-layer (LBL) assembled quantum dots (QDs) with Pb2+ specific DNAzymes. The presence of Pb2+ cleaves the DNAzymes and releases the biotin-modified fragments, which further hybridize with the complementary strands immobilized on the gold substrate. The streptavidin-coated, QD LBL assembled nanocomposites were captured on the gold substrate through biotin–streptavidin interactions. Subsequent electrochemical signals of the captured QDs upon acid dissolution provide quantitative information on the concentrations of Pb2+ with a dynamic range from 1 to 1000nM. Due to the dramatic signal amplification by the numerous QDs, subnanomolar level (0.6nM) of Pb2+ can be detected. The proposed sensor also shows good selectivity against other divalent metal ions and thus holds great potential for the construction of general DNAzyme-based sensing platform for the monitoring of other heavy metal ions.

Abstract:Based on target cycling amplification and 3,4,9,10-perylenetetracarboxylic dianhydride derivative functionalized singal probe, an ultrasensitive electrochemiluminescence (ECL) sensor was designed for the detection of lead ions. The hairpin substrate DNA was immobilized on the electrode through molecular self-assembly. In the presence of Pb2+ and DNAzyme, the substrate was cleaved with single strand DNA fragments left on the electrode surface. Meanwhile, the target and DNAzyme was released for another cleaving circularly. As a result, the single strand DNA fragments hybridized with the assist hairpin probe H1, which leaded to the fabrication of H2 labeled with the 3,4,9,10-perylenetetracarboxylic dianhydride derivative functionalized hollow gold nanoparticles. With the increasing concentration of Pb2+, much more signal probe was been captured and the ECL signal of the biosensor in peroxydisulfate (S2O82-) solution would increase. An ECL assay demonstrates that the sensor has a good linear response to Pb2+ concentration in the range of 1x10-(12) mol/L(-1)x10(-6) mol/L, with a detection limit of 1x10(-12) mol/L. The fabricated sensor shows good selectivity toward Pb2+ against other common metal ions.

Abstract:Novel functional oligonucleotides, especially DNAzymes with RNA-cleavage activity, have been intensively studied due to their potential applications in therapeutics and sensors. Taking advantage of the high specificity of 17E DNAzyme for Pb(2+), highly sensitive and selective fluorescent, electrochemical and colorimetric sensors have been developed for Pb2+. In this work, we report a simple, sensitive and label-free 17E DNAzyme-based sensor for Pb(2+) detection using unmodified gold nanoparticles (GNPs) based on the fact that unfolded single-stranded DNA could be adsorbed on the citrate protected GNPs while double-stranded DNA could not. By our method the substrate cleavage by the 17E DNAzyme in the presence of Pb(2+) could be monitored by color change of GNPs, thereby Pb(2+) detection was realized. The detection of Pb(2+) could be realized within 20 min, with a detection limit of 500 nM. The selectivity of our sensor has been investigated by challenging the sensing system with other divalent metal ions. Since common steps such as modification and separation could be successfully avoided, the sensor developed here could provide a simple, cost-effective yet rapid and sensitive measurement tool for Pb(2+) detection and may prove useful in the development of sensors for clinical toxicology and environmental monitoring in the future.

Abstract:Detection of lead(II) (Pb2+) ions in water is important for the protection of human health and environment. The growing demand for onsite detection still faces challenges for sensitive and easy-to-use methods. In this work, a novel surface plasmon resonance (SPR) biosensor based on GR-5 DNAzyme and gold nanoparticles (AuNPs) was developed. Thiolated DNAzyme was immobilized on the gold surface of the sensor chip followed by anchoring the substrate-functionalized AuNPs through the DNAzyme-substrate hybridization. The coupling between the localized surface plasmon (LSP) of AuNPs and the surface plasmon polaritons (SPP) on the gold sensor surface was used to improve the sensitivity. The substrate cleavage in the presence of Pb2+ ions was catalyzed by DNAzyme, leading to the removal of AuNPs and the diminished LSP-SPP coupling. The optimal detection limit was 80 pM for the sensor fabricated with 1 μM DNAzyme, corresponding to two or three orders of magnitude lower than the toxicity levels of Pb2+ in drinking water defined by WHO and USEPA. By tuning the surface coverage of DNAzyme, the sensitivity and dynamic range could be controlled. This sensor also featured high selectivity to Pb2+ ions and simple detection procedure. Successful detection of Pb2+ ions in groundwater indicates that this method has the prospect in the onsite detection of Pb2+ ions in water. Given the variety of AuNPs and metal-specific DNAzymes, this detection strategy would lead to the development of more sensitive and versatile heavy metal sensors.

Abstract:An electrochemical DNAzyme sensor for sensitive and selective detection of lead ion (Pb2+) has been developed, taking advantage of catalytic reactions of a DNAzyme upon its binding to Ph2+ and the use of DNA-Au bio-bar codes to achieve signal enhancement. A specific DNAzyme for Ph2+ is immobilized onto an Au electrode surface via a thiol-Au interaction. The DNAzyme hybridizes to a specially designed complementary substrate strand that has an overhang, which in turn hybridizes to the DNA-Au bio-bar code (short oligonucleotides attached to 13 nm gold nanoparticles). A redox mediator, Ru(NH3)(6)(3+), which can bind to the anionic phosphate of DNA through electrostatic interactions, serves as the electrochemical signal transducer. Upon binding of Pb2+ to the DNAzyme, the DNAzyme catalyzes the hydrolytic cleavage of the substrate, resulting in the removal of the substrate strand along with the DNA bio-bar code and the bound Ru(NH3)(6)(3+) from the Au electrode surface. The release of Ru(NH3)(6)(3+) results in lower electrochemical signal of Ru(NH3)(6)(3+) confined on the electrode surface. Differential pulse voltammetry (DPV) signals of Ru(NH3)(6)(3+) provides quantitative measures of the concentrations of Pb2+, with a linear calibration ranging from 5 nM to 0.1 mu M. Because each nanoparticle carries a large number of DNA strands that bind to the signal transducer molecule Ru(NH3)(6)(3+), the use of DNA-Au bio-bar codes enhances the detection sensitivity by five times, enabling the detection of Pb2+ at a very low level (1 nM). The DPV signal response of the DNAzyme sensor is negligible for other divalent metal ions, indicating that the sensor is highly selective for Pb2+ Although this DNAzyme sensor is demonstrated for the detection of Pb2+, it has the potential to serve as a general platform for design sensors for other small molecules and heavy metal ions.
Visual and Quantitative Detection of Lead Ion Based on G-quadruplex-hemin DNAzyme-mediated Etching of Gold Nanorods

Abstract:A label-free visualized lead ion biosensor was constructed based on G-quadruplex-hemin DNAzyme-mediated etching of gold nanorods (AuNRs). Hemin could induce the transformation of the aptamer PS2. M into a G-quadruplex structure to generate a G-quadruplex-hemin DNAzyme with peroxidase-like activity. 3,3 ',5,5 '-Tetramethylbenzidine (TMB)was oxidized to form a blue diimine derivative TMB+,and the enzymatic reaction was terminated after adding H2SO4,and TMB+ became TMB2+(yellow). Under the action of hexadecyl trimethyl ammonium bromide (CTAB),TMB2+ could effectively oxidize Au(0)to Au?, thereby realizing the etching of AuNRs,and the solution turned yellow. Due to the highly specific interaction between PS2. M and Pb2+,and the ability of Pb2+ to stabilize G-quadruplexes was significantly stronger than that of hemin,G-quadruplexes-hemin DNAzyme with enzyme-like activity could not be formed in the presence of Pb2+,thus unable to etch AuNRs. Therefore,different concentrations of Pb2+ made the solutions produce distinct color changes that could be easily identified with naked eyes. The ratiometric quantitative detection of Pb2+ was achieved by measuring the absorbance of the longitudinal surface plasmon absorption peak of AuNRs and the TMB+ absorption peak at 450 nm,with a linear range of 5-5000 nmol/ L and a detection limit as low as 2. 0 nmol/ L. The method was successfully applied to detection of Pb2+ in Songhua River water and tap water samples,with spiked recoveries were 98. 7%-102. 5% and 94. 8%-108. 2%,respectively,and the results were consistent with the detection results of inductively coupled plasma-mass spectrometry (ICP-MS). The method ingeniously designed the nucleic acid sequence,and realized the rapid and high-sensitivity visual detection of Pb2+.

Abstract:This paper describes a label-free 17E DNAzyme-based time-gated fluorescence sensor for Pb2+ detection by unmodified gold nanoparticles (GNPs) and a terbium ternary complex. The fluorophore that used in this paper is a terbium ternary complex. Its signal can be measured in a time-gated manner which could eliminate most of the unspecific fluorescent background. It is well known that unfolded single-stranded DNA (ssDNA) could be adsorbed on GNPs while double-stranded DNA could not. The cleavage of the substrate by the 17E DNAzyme in the presence of Pb2+ causes the release of ssDNA from the 17E-17S duplex to be absorbed onto GNPs, preventing the aggregation of GNPs and then leading to a fluorescence decrease of terbium ternary complex. By means of this method, the authors have successfully detected Pb2+ over a range of 10 nM to 2500 nM with a detection limit of 1.7 nM. The sensor also exhibited good selectivity. The sensor provided a simple, cost-effective, rapid and sensitive measurement tool for Pb2+ detection.

Abstract:A DNAzyme based method for the sensitive and selective quantification of lead(II) ions has been developed. A DNAzyme that requires Pb2+ for activation was selected. An RNA containing DNA substrate was cleaved by the DNAzyme in the presence of Pb2+. The 2′,3′-cyclic phosphate of the cleaved 5′-part of the substrate was efficiently removed by Exonuclease III. The remaining part of the single stranded DNA (9 or 13 base long) was subsequently used as the primer for the strand displacement amplification reaction (SDAR). The method is highly sensitive, 200pM lead(II) could be easily detected. A number of interference ions were tested, and the sensor showed good selectivity. Underground water samples were also tested, which demonstrated the feasibility of the current approach for real sample applications. It is feasible that our method could be used for DNAzyme or aptazyme based new sensing method developments for the quantification of other target analytes with high sensitivity and selectivity.

Abstract:A novel label-free impedimetric sensing system based on DNAzyme and ordered mesoporous carbon–gold nanoparticle (OMC–GNPs) for the determination of Pb2+ concentration was developed in the present study. Firstly, gold nanoparticles deposited on the modified electrode surface were employed as a platform for the immobilization of thiolated probe DNA, and then hybridized with DNAzyme catalytic beacons. Subsequently, in the presence of Pb2+, the DNAzyme could be activated to cleave the substrate strand into two DNA fragments, which causes differences in the electrical properties of the film. Randles equivalent circuit was employed to evaluate the electrochemical impedance spectroscopy (EIS) result. The charge transfer resistance (RCT) value for the [Fe(CN)6]3−/4− redox indicator was remarkably decline after hybridization with Pb2+. The difference in RCT values before and after hybridization with Pb2+ showed a linear relation with the concentration of the Pb2+ in a range of 5×10−10–5×10−5M, with a detection limit of 2×10−10M (S/N=3). Furthermore, with the application of Pb2+ dependent 8-17DNAzyme, the proposed sensing system exhibited high selectivity without using any labeled probes. This biosensor demonstrated a promising potential for Pb2+ detection in real sample.
Scanometric nanomolar lead (II) detection using DNA-functionalized gold nanoparticles and silver stain enhancement

Abstract:Based on gold label silver stain method, a facile, sensitive and selective scanometric method for the detection of lead (II) was developed. The main components of the method include “8–17” DNAzyme functionalized gold nanoparticles (AuNP1), capture DNA functionalized gold nanoparticles (AuNP2), and silver staining. In presence of lead (II), AuNP1 was cleaved into different pieces that were captured by AuNP2; and became aggregated and formed network structures. These aggregate structures hindered the exposure of gold particles during the silver staining, resulting in a reduced gray values compared with that in the absence of lead (II). Using the gray value of silver stain signals, the developed scanometric assay realized a semi-quantitative detection of nanomolar levels of lead (II), ranging from 2 to 1000nmol/L (about 0.4–200ppb), without significant interference from other metal ions. Results suggested that this scanometric method is a simple, sensitive, selective, and portable tool for the onsite detection of nanomolar levels of lead (II).

Abstract:A DNA electrochemical sensor was fabricated using the modification of a glassy carbon electrode (GCE) with ordered mesoporous carbon nitride (MCN), electrodeposited gold nanoparticles (EAu) and methylene blue (MB)/nanoAu/MCN as the signal amplifier, in which MB was inserted into the nanoAu/MCN composites for Pb2+ detection. The secondary structure of the trans-cleaving 8–17 DNAzyme, composed of the substrate strand (S1) and the enzyme strand (S2), was utilized for the sensor fabrication. S1 immobilized on the signal amplifier could hybridize with S2 and form a DNA double helix structure on account of the principle of complementary base pairing. With the activation of Pb2+, S2 was able to cleave the single RNA linkage with S1 by hydrolysis reaction to break S1 into two fragments. One of the fragments NS1′ that was immobilized on the MB/nanoAu/MCN signal amplifier (S1′/MB/nanoAu/MCN) could hybridize with the DNA probe S3 immobilized on the modified electrode. With the help of the MCN’s large specific surface area and the gold nanoparticles’ good charge-transport capacity, the sensor exhibited excellent sensitivity and selectivity. The proposed sensing strategy represented a wide linear response in the range from 1.0 × 10−3 to 1.0 × 10−14 M. The sensor was also tested with real samples and represented a promising method for detecting Pb2+.

This method is based on gold nanoparticles multi-functionalized by 17E-17DS duplex and barcode DNA, AuNPs@(17E-17DS duplex & barcode DNA). The complex of 17E and 17DS (duplex) was constructed by the hybridization between 17E DNAzyme and its corresponding 17DS cleavage substrate. Because 17E DNAzyme has high specificity for lead(II), the presence of lead(II) induces 17E DNAzyme to specifically cleave 17DS. A barcode DNA complementary to the capture DNA was employed to reduce the amount of 17E-17DS duplex used and increase the capture probability of cleaved AuNPs@(17E-17DS duplex & barcode DNA) conjugates, and thus effectively improve the detection sensitivity. The test results for lead(II) could be visually obtained within 10 min without any instrument and sample pretreatment, with a detection limit 20 nM. In addition, the selectivity of this sensing system was investigated by challenging the sensor with other divalent metal ions, and no significant interference was found. Therefore, the developed test strip could be a potential on-site screening tool for the rapid detection of lead(II).
A Single Fluorophore Ratiometric Nanosensor Based on Dual-Emission DNA-templated Silver Nanoclusters for Ultrasensitive and Selective Pb2+ Detection

Abstract:Lead ion (Pb2+) is known as a major non-degradable environmental pollutant and is quite harmful to human health. In our investigation, a novel ratiometric fluorescent nanosensor with DNA-AgNCs as single fluorophore has been successfully constructed for ultrasensitive and specific detection of Pb2+. The single-strand DNA templated silver nanoclusters present green emission, which can be converted into red emission after approaching a specific DNA segment by forming duplex with its complementary DNA strands (ds-DNA-AgNCs). The ds-DNA-AgNCs contains rA cleavage site of Pb2+-dependent DNAzyme configuration. In the presence of Pb2+, the specific DNA segment would be released from the ds-DNA-AgNCs, resulting in discoloration of DNA-AgNCs from red to green. Since Pb2+ can successively cleave the rA site, the signal change from red to green emission is amplified. Thus, benefiting from the Pb2+-dependent DNAzyme and the charming properties of DNA-AgNCs, the proposed platform exhibits a good linear relationship from 0.001 nM to 10 nM with a detection limit of 1.0 pM for Pb2+ determination, which is lower than most of the reported Pb2+ biosensors. Moreover, this sensitive ratiometric fluorescent probe exhibits excellent selectivity toward Pb2+ detection in real samples, such as lake water, tap water, and human serum samples, illustrating the huge potential applications for complicated samples in the future.
