Abstract:We propose a highly sensitive and selective surface-enhanced Raman scattering (SERS) method for determining lead ions based on a DNAzyme-linked plasmonic nanomachine. A metallic nanoparticle-on-a-film structure was built through a rigid double-stranded bridge linker composed of a DNAzyme and its substrate. This DNAzyme could be activated by lead ions and catalyze a fracture action of the substrate. Thus, the double chain structure of DNA would turn into a flexible single strand, making the metal nanoparticles that connected to the terminal of DNAzyme fall to the surface of the metal film. Hereby, a narrow gap close to 2 nm generated between metal nanoparticles and the metal film, exhibiting a similar effect of a "hot spot" and remarkably enhancing the signal of randomly dispersed Raman-active molecules on the surface of metal film. By measuring the improvement of SERS intensity of the Raman-active molecules, we realized the lowest detection concentration of Pb2+ ions to 1.0 nM. This SERS analytical method is highly selective and can be extended universally to other targets via the accurate programming of corresponding DNA sequences.

Abstract:An electronic DNAzyme sensor for highly sensitive detection of Pb2+ is demonstrated by coupling the significant signal enhancement of the layer-by-layer (LBL) assembled quantum dots (QDs) with Pb2+ specific DNAzymes. The presence of Pb2+ cleaves the DNAzymes and releases the biotin-modified fragments, which further hybridize with the complementary strands immobilized on the gold substrate. The streptavidin-coated, QD LBL assembled nanocomposites were captured on the gold substrate through biotin–streptavidin interactions. Subsequent electrochemical signals of the captured QDs upon acid dissolution provide quantitative information on the concentrations of Pb2+ with a dynamic range from 1 to 1000nM. Due to the dramatic signal amplification by the numerous QDs, subnanomolar level (0.6nM) of Pb2+ can be detected. The proposed sensor also shows good selectivity against other divalent metal ions and thus holds great potential for the construction of general DNAzyme-based sensing platform for the monitoring of other heavy metal ions.
DNAzyme-based cascade signal amplification strategy for highly sensitive detection of lead ions in the environment

Abstract:Lead ions are one of many common environmental pollutants, that can cause posing a serious threat to people's health, thus, their efficient and sensitive detection is important. We propose a cascade signal amplification sensor using a DNAzyme-based strand displacement amplification (SDA) and hybridization chain reaction (HCR) for the high-sensitivity detection of Pb2+. In the demonstrated sensor system, the target metal ion can activate DNAzyme to cause a strand displacement reaction. Under the synergistic action of polymerase and nickase, large numbers of DNA strands are generated that can initiate HCR. The subsequent HCR can restore the fluorescence intensity of the FAM quenched for the fluorescence resonance energy transfer effect, which exhibits a strong fluorescence signal. The DNAzyme-based sensor allowed the detection of Pb2+ down to 16.8 pM and resulted in a good dynamic line relationship ranging from 50 pM to 500 nM, and the biosensor also showed good selectivity. Furthermore, we confirmed that the proposed sensor can still detect lead ions in complex environments such as lake water, milk, and serum. We believe these findings will provide new ideas for the detection of toxic elements ions in the environment and food.
Electrochemical DNAzyme sensor for lead based on amplification of DNA-Au bio-bar codes.

Abstract:An electrochemical DNAzyme sensor for sensitive and selective detection of lead ion (Pb2+) has been developed, taking advantage of catalytic reactions of a DNAzyme upon its binding to Ph2+ and the use of DNA-Au bio-bar codes to achieve signal enhancement. A specific DNAzyme for Ph2+ is immobilized onto an Au electrode surface via a thiol-Au interaction. The DNAzyme hybridizes to a specially designed complementary substrate strand that has an overhang, which in turn hybridizes to the DNA-Au bio-bar code (short oligonucleotides attached to 13 nm gold nanoparticles). A redox mediator, Ru(NH3)(6)(3+), which can bind to the anionic phosphate of DNA through electrostatic interactions, serves as the electrochemical signal transducer. Upon binding of Pb2+ to the DNAzyme, the DNAzyme catalyzes the hydrolytic cleavage of the substrate, resulting in the removal of the substrate strand along with the DNA bio-bar code and the bound Ru(NH3)(6)(3+) from the Au electrode surface. The release of Ru(NH3)(6)(3+) results in lower electrochemical signal of Ru(NH3)(6)(3+) confined on the electrode surface. Differential pulse voltammetry (DPV) signals of Ru(NH3)(6)(3+) provides quantitative measures of the concentrations of Pb2+, with a linear calibration ranging from 5 nM to 0.1 mu M. Because each nanoparticle carries a large number of DNA strands that bind to the signal transducer molecule Ru(NH3)(6)(3+), the use of DNA-Au bio-bar codes enhances the detection sensitivity by five times, enabling the detection of Pb2+ at a very low level (1 nM). The DPV signal response of the DNAzyme sensor is negligible for other divalent metal ions, indicating that the sensor is highly selective for Pb2+ Although this DNAzyme sensor is demonstrated for the detection of Pb2+, it has the potential to serve as a general platform for design sensors for other small molecules and heavy metal ions.

Abstract:Ultra-trace lead ion detection has significant implications in various domains, notably in point-of-care testing, environmental monitoring, and cancer-targeted therapies and diagnostics. The demand for rapid and precise lead-ion detection in clinical settings necessitates innovative approaches. In this study, a lead-ion biosensor was designed using DNAzyme-based tilted fiber Bragg grating surface plasmon resonance (TFBG-SPR). The lead ions specifically cleave toward DNAzyme's ribonucleotide adenosine (rA)-containing substrate strand, and a high sensitivity "on-off" sensing structure on the TFBG-SPR surface was established by using the DNA-conjugated AuNPs as a signal amplification element. This "on-off"-type lead ion biosensor has a competitive limit of detection of 3 fM and a wide dynamic measurement range up to ten orders, meeting stringent requirements for ultra-trace, label-free and online clinical detection. Owing to the unique catalytic effect of DNAzyme binding to lead ions, high selectivity was demonstrated. Moreover, to validate its practicality in clinical diagnosis, we successfully achieved spiked-recovery results with 100.39 % average recovery rate and a swift 17-minute analysis time within the colorectal cancer patient samples. By combining DNA biomolecular, TFBG-SPR probe and "on-off" AuNPs-DNA structure, the fM level detection of lead ion in fiber highlighted its potential in vivo and in vitro complex application scenarios.

Abstract:Lead ion (Pb2+, a heavy metal ion existed every aspect of life) is one of the most important hazards for environment and human health. Herein we design an in-situ amplified electronic monitoring system for sensitive determination of Pb2+ on Pb2+-specific DNAzyme-modified sensing interface. The assay consists of target-triggered cleavage of Pb2+-specific DNAzyme, initiator strand-induced DNA hybridization chain reaction, formation of hemin/G-quadruplex -based DNAzyme and its automatically catalyzed polymerization of aniline monomer along the double-stranded DNA. Experimental results indicated that the catalytic currents of the as-produced polyaniline were linearly dependent on target Pb2+ concentrations from 0.05 to 50nM with a detection limit (LOD) of 32pM. Such a synergistic effect of hybridization chain reaction with DNAzyme-catalyzed polymerization provided a universal platform for sensitive screening of target Pb2+, thereby holding great promise for application in practice.

Abstract:An amplified electrochemical biosensing scheme is described for lead(II) ions. It is making use of DNAzyme-assisted target recycling and catalytic hairpin assembly (CHA). The hairpin strand (substrate probe for the Pb2+-based DNAzyme; referred to as SP) is composed of trigger probe (TP) and a capture probe 1 attached to gold nanoparticles (AuNP). In the presence of the enzyme probe that partially hybridizes with SP, the introduction of Pb2+ triggers target recycling and drives the highly amplified translation of target Pb(II) to TP. The CHA reaction is further initiated by TP. The modified AuNP act as the connecting unit, and this leads to the formation of a 3D DNA-AuNP network on the electrode (which is the third amplification step). It can bind the positively charged redox mediator RuHex via electrostatic interaction for electrochemical detection. This biosensor has a low detection limit (95 pM) and any analytical range that covers the 100 pM to 5 μM Pb(II) concentration range. It is selective over other divalent metal ions. It was applied to the determination of Pb2+ in spiked real-world samples.

Abstract:A signal-off electrochemiluminescence (ECL) DNA sensor based on gold nanodendrites (Au NDs) modified indium tin oxide (ITO) electrode for the detection of lead ion (Pb(2+)) was developed. Well-defined Au NDs were prepared on ITO electrode using low-potential synthesis, assisted by ethylenediamine. Based on Pb(2+)-specific deoxyribozyme, the silver/zinc oxide (Ag/ZnO) with coupled structure, prepared by one-pot method, was close to the surface of the electrode to catalyze the reduction of part of H2O2, the coreactant for cathodic ECL emission, leading to a decrease of ECL intensity. In addition, taking advantage of the larger surface area to capture a large amount of capture probe as well as excellent conductivity of Au NDs, the sensor could detect Pb(2+) quantitatively in a wider range, and performed excellent selectivity. Furthermore, such simple and sensitive DNA sensor was successfully applied for the detection of Pb(2+) in lake water and human serum samples, respectively.

Abstract:Lead is one of the toxic heavy metal ions which seriously threatens the environment and human health, even at ultra-trace level. Here, an innovative fluorescent sensor based on DNAzymes coupling with multi-cycle strand displacement amplification (M-SDA) and nano-graphene oxide (GO) was established for the first time to ultra-sensitively detect lead, which is the first attempt to combine biological nucleic acid amplification technology with GO to detect heavy metal ions. There are several advantages. Firstly, the high selectivity of DNAzymes endow the proposed sensor outstanding specificity. Secondly, the introduction of M-SDA let the proposed sensor own the high potential for ultra-sensitive detection of Pb2+. Compared with traditional SDA, the amplification efficiency of M-SDA is greatly enhanced due its intermediate products are able to induce another SDA. Thirdly, the using of GO in fluorescence ion sensor is simple and fast. Under the optimal experimental conditions, the proposed sensor exhibited a good linear from 0.01 nM to 850 nM (R-2=0.99714) with the detection limit as low as 6.7 pM. Additionally, the proposed sensor could be applied to detect the real samples with good accuracy and reliability.
Toehold-mediated Strand Displacement Reaction Triggered by Nicked DNAzymes Substrate for Amplified Electrochemical Detection of Lead Ion

Abstract:Searching for a sensitive analytical method for the detection of lead ion (Pb2+) without involving any enzymes is of great significance in environmental monitoring. Herein, based on the integration of toehold-mediated strand displacement reaction (TSDR) and the electrocatalysis of magnetic Fe3O4 toward electron mediator methylene blue (MB) for signal amplification, a sensitive electrochemical biosensor for Pb2+ was developed by using Pb2+-specific DNAzymes as recognition probe. The electrode surface was firstly modified with Au nanoparticles (AuNPs)-loaded Fe3O4 nanocomposites (AuNPs@Fe3O4) and a DNA duplex containing the capture probe (NH2-CP) with two specific toehold sequences. Fuelled by the cleaved substrate fragments (rSS) of Pb2+-specific DNAzymes, TSDR was activated to allow for the complete assembly of MB-labelled signal probe (MB-SP) through the hybridization with NH2-CP, resulting in the spatial proximity of MB close to Fe3O4 in the modified electrode surface. In this case, the amplified electrochemical output signal was further enhanced due to the robust electrocatalysis of Fe3O4 toward MB reduction. Thus, the developed sensitive biosensor for Pb2+ exhibited a reliable linear range of 1 pM-500 nM with the limit of detection of 0.32 pM. Importantly, by changing the specific DNAzymes, the proposed assay platform would be easily extended to quantitative detection of other metal ions without involving any enzymes and would be potential and promising for environment monitoring. Abstract:A new enzyme-free and ultrasensitive electrochemical Pb2+ biosensor was developed. By coupling the DNA-assisted cascade of hybridization reaction with the quantum dots (QDs) for signal amplification, a detection limit as low as 6.1 pM can be obtained for Pb2+. In this study, the “8-17” DNAzyme was used for specific recognition of Pb2+. In the presence of Pb2+, the DNAzyme was activated and cleaved the substrate strand. And then, the hybridization between the linker probe and signal probe was initiated, which resulted in formation of a long cascade DNA structure as well as assemble of numerous QDs at last. By the use of magnetic beads, the free signal probe can be easily removed by external magnetic field. After acid lysis, a great amount of redox cations can be released from the QDs and eventually result in significantly amplified electrochemical signals. This method is highly sensitive, selective and simple without the participation of any protein based enzyme (nuclease), thereby holds great potential for real sample analysis.

Abstract:Heavy metal lead accumulation in the environment pollutes the ecology systems and further threatens the human health. It is necessary to develop a sensitive method to detect it. Here, we propose a highly sensitive lead detection method by combining DNAzyme and CRISPR system. Once the lead ion is recognized, the substrate chain of DNAzyme is cleaved to produce single strand DNA. The produced single strand DNA can be detected by Cas protein/guide RNA complex and further trigger the collateral cleavage effect of CRISPR system, which can indiscriminately cut short single strand DNA reporters. By this way, the detection signals can be greatly amplified. This method can detect lead ions as low as 0.48 nM. The sensitivity is higher than the DNAzyme method. Furthermore, the portable 3D printing device is designed to observe the fluorescent signals so the end-point detection results can be visualized by the naked eyes. The entire detection process can avoid using bulky and expensive instruments, which can promote on-site lead ion detection.

Abstract:In this work, a sensitive and specific electrochemical biosensor for lead ion (Pb2+) detection was developed by coupling with synergetic catalysis of porous Au–Pd bimetallic nanoparticles and hemin/G-quadruplex-based DNAzyme. In the presence of target Pb2+, the substrate strand was cleaved and the enzyme strand was released. Subsequently, G-rich DNA-labeled Au–Pd bimetallic nanoparticle was linked with the released enzyme strand through the helper DNA. Upon addition of hemin, a large number of hemin/G-quadruplex-based DNAzyme molecules were formed on the electrode to serve as nicotinamide adenine dinucleotide hydrogen (NADH) oxidase and peroxidase mimics. DNAzyme could catalyzed the reduction of H2O2, generated from NADH in the presence of O2, to produce an electrochemical signal when using thionine as the electron mediator. Introduction of porors Au–Pd bimetallic nanoparticles could enhance the detectable signal and cause the increase in the sensitivity. Experimental results showed that the variations (∆I) in the cathodic peak currents of the biosensor were linearly dependent on target Pb2+ concentrations from 1.0pM to 100nM with a detection limit (LOD) of 0.34pM. The excellent performance of the sensing platform indicated its promising prospect as a valuable tool for simple and cost-effective Pb2+ detection in practical application.

Abstract:Lead ions (Pb2+) are one of the most widespread heavy metal contaminants that pose detrimental impact on environment and human health. We demonstrate a highly sensitive and specific liquid crystal (LC)-based sensing platform for detecting Pb2+ assisted by DNAzyme and rolling circle amplification (RCA). Magnetic beads (MBs) are functionalized with DNA duplexes of the catalytic strands (DNAzymes) and the substrate strands. In the presence of Pb2+, the substrate strands are disassembled due to activation of the DNAzyme, which allows initiation of DNA RCA on MBs. The amplified DNA strands can disrupt arrangement of octadecy trimethyl ammonium bromide monolayers (OTAB), thereby inducing planar orientation of LC molecules at the interface of aqueous and LCs. Thus, LCs exhibit bright appearance. In contrast, RCA cannot be triggered in the absence of Pb2+. Therefore, LC molecules adopt perpendicular orientation at the interface, which induces the dark morphology of LCs. The limit of detection reaches as low as 16.7 pM. It is an improvement of more than two orders of magnitude compared to that of previously reported LC-based sensing approaches. This approach also shows excellent performance in monitoring Pb2+ in tap water and lake water.

Abstract:A DNAzyme based method for the sensitive and selective quantification of lead(II) ions has been developed. A DNAzyme that requires Pb2+ for activation was selected. An RNA containing DNA substrate was cleaved by the DNAzyme in the presence of Pb2+. The 2′,3′-cyclic phosphate of the cleaved 5′-part of the substrate was efficiently removed by Exonuclease III. The remaining part of the single stranded DNA (9 or 13 base long) was subsequently used as the primer for the strand displacement amplification reaction (SDAR). The method is highly sensitive, 200pM lead(II) could be easily detected. A number of interference ions were tested, and the sensor showed good selectivity. Underground water samples were also tested, which demonstrated the feasibility of the current approach for real sample applications. It is feasible that our method could be used for DNAzyme or aptazyme based new sensing method developments for the quantification of other target analytes with high sensitivity and selectivity.

Abstract:A label-free sensor was developed for sensitive detection of lead(II), combining high selectivity of a Pb2+-dependent DNAzyme with enormous signal amplification of quantitative polymerase chain reaction (QPCR). Specifically, a substrate strand was designed to have two primer–hybridization sequences at either terminus. The presence of lead ion (Pb2+) catalyzed cleavage of the substrate strands. This resulted in a concentration decrease of the substrate strand that could be detected by QPCR. Compared with existing DNAzyme-based protocols for Pb2+ assay, this strategy circumvented the use of various optical or electrical labels that might be difficult to be synthesized. Also, the incorporation of QPCR furnished our approach with high sensitivity and superb reproducibility. In addition, QPCR allowed an immediate quantification of the cleavage efficiency that could be useful for evaluation of the DNAzyme activity. The results obtained revealed that our approach exhibited a dynamic response toward Pb2+ within a three-decade concentration range from 10nM to 5μM with a detection limit of 1nM. This approach also demonstrated good selectivity against other metal ions that commonly coexisted with Pb2+.
An DNA Enzyme Electrochemiluminescence Biosensor for Ultra-sensitive Detection of Pb2+ Based on CdS/MoS2 Joint Amplification

Abstract:Lead (Pb) is one of the most serious heavy metals to human health and ecological environment. Excessive lead can lead brain retardation and liver and kidney damage. In this work, a novel electrochemiluminescence (ECL) biosensor based on CdS quantum dots (CdS QDs)/MoS2 enzyme assist multiple amplification and DNA enzyme was designed to achieve high sensitivity detection of Pb2+. AuNPs-PDDA-MoS2 complex was synthesized as sensing substrate. The thiol-labeled T30695 DNA was assembled on the surface of the electrode via Au-S bonds, following which the amino-labeled ends of the DNA were linked to the CdS QDs by the formation of amide bonds. In the presence of Pb2+, the G bases of DNA sequence formed G- tetraplex (G4) structure, DNA enzymes were then further synthesized. The catalytic oxidation of H2O2 by DNA enzyme and AuNPs-MoS2 composite resulted in a significant decrease of electrochemiluminescence of CdS QDs with H2O2. The detection range of the biosensor was 1.0×10-14 - 5.0×10-11mol/L (R = 0.9992) with a detection limit of 1×10-15 mol/L. This biosensor can detect Pb2+ in real water samples with satisfactory results.

Abstract:In this work, we develop a dual recycling amplification aptasensor for sensitive and rapid detection of lead ions (Pb2+) using fluorescence and surface-enhanced Raman scattering (FL-SERS). The aptasensor allows targeted cleavage of substrates through specifically binding with the Pb2+-dependent aptamer (M-PS2.M). Ultrasensitive detection of trace Pb2+ has been achieved using an enzyme-free nonlinear hybridization chain reaction (HCR) and the FL-SERS technique. The lower limit of detection (LOD = 3σ/k) is 0.115 pM in FL mode and 1.261 fM in SERS mode. The aptasensor is characterized by high reliability and specificity, among other things, to distinguish Pb2+ from other metal ions. In addition, the aptasensor can detect Pb2+ in actual water with good recovery. Compared with the single-mode aptasensor, the dual-mode aptasensor is characterized by high reliability, an extensive detection range, and high specificity.

Abstract:Lead (Pb2+) is a toxic heavy metal that can severely pollute the environment and cause harm to public health. Therefore, the prompt and accurate monitoring of lead levels in the environment is vital. In this study, a novel DNAzyme-based cascade signal amplification biosensor that could detect Pb2+ with high sensitivity was designed through the combination of the strand displacement reaction (SDR) and rolling circle amplification (RCA). When Pb2+ is absent, RCA is triggered under the synergistic action of T4 DNA ligase and phi29 DNA polymerase with an artificially fluorophore-labeled S-chains being released to replace the upstream products generated by repeated RCA, thereby restoring the quenched fluorescence and emitting a strong fluorescent signal. After adding Pb2+, 8-17 DNAzyme binds specifically to Pb2+ and catalyzes the cleavage of the rA site on a single-stranded DNA with artificially modified rA site to restrict the RCA. The designed sensor provides a linear detection range for Pb2+ from 25 pM to 1 µM, with a low limit of detection 8.3 pM. Significantly, this sensor still demonstrates satisfactory performance when used for detecting Pb2+ in environment samples (e.g., river water). We consider that our study can provide reference values and ideas for the development of heavy metal ion detection methods.
