Abstract:A well-defined N-heterocyclic carbene (NHC) palladium-catalyzed intramolecular cyclization reaction of aryl-substituted alkylidenecyclopropanes (ACP) to 1-aryl dihydronaphthalenes is described. The NHC salts were found to suppress beta-hydride elimination from the homoallylpalladium intermediate, which may lead to unwanted alkene formation. A plausible mechanism for the cycloisomerization was proposed.

Abstract:A cobalt-catalyzed intramolecular [4 + 2] cycloaddition of cyclopropyl (CP)-capped dienes with ynes/enes/allene was reported, providing an efficient method toward a spiro[2.5]octene ring system found in natural products, such as illudin. The [4 + 2] cycloadducts can be converted into other compounds via CP chemistry. This reaction can also be catalyzed by Ni. A density functional theory study on the reaction mechanisms is also reported here.

Abstract:A tandem coupling-ketooxygenation reaction of readily accessible enediyne-carboxylic compounds with inner alkynes has been developed that utilizes the PdCl2/CuBr2 catalytic system under an O2 atmosphere and assembles a class of isoindolinones and o-acylbenzoic acids. The two oxygen atoms are regioselectively incorporated into enediyne units at the 1- and 6-positions from atmospheric molecular oxygen and H2O, respectively, during the present process. This study uncovered a formal [4C + 2C] benzannulation-diketonization of enediynes and alkynes via a coupling and decoupling strategy.

Abstract:Efficient strategies to assemble enantioenriched pyridine derivatives are highly important, given their significance in both synthetic and medicinal chemistry. Here, we report an enantioselective nickel-catalyzed intermolecular [2 + 2 + 2] cycloaddition of alkyne-tethered malononitriles with alkynes for the synthesis of densely substituted pyridines. The α-all-carbon quaternary center adjacent to pyridine is introduced by desymmetrizing the two cyano groups of disubstituted malononitriles. Notably, terminal alkynes are also tolerated with good regioselectivity to afford tetrasubstituted pyridines. Zinc halide is essential to enable the occurrence of the transformation by promoting the hetero-cyclometallation step. The reaction uses bio-renewable 2-MeTHF as the solvent and features mild reaction conditions, good functional group compatibilities, and enantioselectivities. This study offers a straightforward approach to the valuable enantioenriched heteroarenes from feedstock chemicals by forming three bonds and one quaternary stereocenter simultaneously in a single reaction step.

Abstract:Cross-electrophile coupling reactions of two Csp3-X bonds remain challenging. Herein we report an intramolecular nickel-catalyzed cross-electrophile coupling reaction of 1,3-diol derivatives. Notably, this transformation is utilized to synthesize a range of mono- and 1,2-disubstituted alkylcyclopropanes, including those derived from terpenes, steroids, and aldol products. Additionally, enantioenriched cyclopropanes are synthesized from the products of proline-catalyzed and Evans aldol reactions. A procedure for direct transformation of 1,3-diols to cyclopropanes is also described. Calculations and experimental data are consistent with a nickel-catalyzed mechanism that begins with stereoablative oxidative addition at the secondary center.

Abstract:Developing new transition metal-catalyzed asymmetric cycloadditions for the synthesis of five-membered carbocycles (FMCs) is a research frontier in reaction development due to the ubiquitous presence of chiral FMCs in various functional molecules. Reported here is our discovery of a highly enantioselective intramolecular [3+2] cycloaddition of yne-alkylidenecyclopropanes (yne-ACPs) to bicyclo[3.3.0]octadiene and bicyclo[4.3.0]nonadiene molecules using a cheap Co catalyst and commercially available chiral ligand (S)-Xyl-BINAP. This reaction avoids the use of precious Pd and Rh catalysts, which are usually the choices for [3+2] reactions with ACPs. The enantiomeric excess in the present reaction can be up to 92 %. Cationic cobalt(I) species was suggested by experiments as the catalytic species. DFT calculations showed that this [3+2] reaction starts with oxidative cyclometallation of alkyne and ACP, followed by ring opening of the cyclopropyl (CP) group and reductive elimination to form the cycloadduct. This mechanism is different from previous [3+2] reactions of ACPs, which usually start from CP cleavage, not from oxidative cyclization.

Abstract:A Cu(I)-catalyzed coupling and cycloisomerization of diazo compounds with terminal yne-alkylidenecyclopropanes (ACPs) has been presented. This reaction starts from the formation of an allenic intermediate in the Cu(I)-catalyzed cross-coupling reaction of a diazo compound with terminal alkyne in yne-tethered ACP and then undergoes a domino cycloisomerization of a 6π-electrocyclization and cyclopropane ring-opening rearrangement to give functionalized cyclopenta[ b]naphthalene derivatives in moderate to excellent yields under mild conditions.

Abstract:Herein, we report a [5+2] cycloaddition between readily accessible 1-indanones and internal alkynes through Rh-catalyzed activation of less strained C-C bonds. The reaction is enabled by a strongly σ-donating NHC ligand and a carefully modified temporary directing group. A wide range of functional groups is tolerated, and the method provides straightforward access to diverse benzocycloheptenones that are hard to access otherwise. DFT studies of the reaction mechanism imply the migration insertion as the turnover-limiting step and suggest beneficial π-π interactions in the transition states.

Abstract:A new method of cobalt-catalyzed synthesis of pyrrolidinones from aliphatic amides and terminal alkynes was discovered through a C-H bond functionalization process on unactivated sp(3) carbons with the silver cocatalyst using a bidentate auxiliary. For the first time, a broad range of easily accessible alkynes are exploited as the reaction partner in C(sp(3))-H bond activation to give the important 5-ethylidene-pyrrolidin-2-ones in a site-selective fashion. The reaction tolerates a wide variety of functional groups including -F, -Cl, -Br, -CF3, ether, cyclopropane, and thiophene. Both pyridine ligand and aromatic solvent play the important role for the promotion of reactivity. This cobalt-catalyzed cyclization reaction can be successfully extended to a variety of aromatic amides to afford a variety of isoindolinones. Attractive features of this catalytic system include its low cost, easy operation, and convenient access to a wide range of pyrrolidinones and isoindolinones.

Abstract:A rhodium-catalyzed tandem arylation/cyclization reaction of 3-(ortho-boronated aryl) conjugated enones with unactivated alkynes is reported. By using a rhodium(I)/chiral-diene complex as the catalyst, the protocol was processed smoothly to provide various 2,3-disubstituted indene compounds in high yields with excellent regioselectivities and enantioselectivities. The approach outlined herein is appealing, as simple diarylalkynes, diakylalkynes, and alkyl(aryl)alkynes are the starting materials.

Abstract:As a type of important carbocyclic compounds, indenes and spiroindenes are not only widely found in natural products but extensively employed as synthetic building blocks in materials, pharmaceuticals, and asymmetric synthesis. An efficient and rapid synthesis of indenes and spiroindenes via the annulation of benzvlic alcohols (or aryl-substituted cvcloalkanols) with alkynes in the presence of TiCl4 or AlCl3 was desclosed. This reaction is normally completed within 30 min at room temperature and compatible with a variety of substituted alkynes. Two new C-C bonds are constructed during the reaction. Cycloalkanols such as cyclobutanol, cyclohexanol, cycloheptanol, cyclooctanol, and cyclododecanol are suitable substrates to afford a set of valuable spiroindenes. This method is featured with simple operation, short reaction time, and mild reaction conditions.

Abstract:Zirconacyclopentadienes, prepared from two alkynes or a diyne, reacted with the alkyl-, trimethylsilyl-, or alkoxy-substituted third alkyne as well as an alkyne with an electron-withdrawing group in the presence of a stoichiometric amount of NiBr2(PPh3)2 to give benzene derivatives in good yields. Heteroatom-containing diynes such as dipropargylbenzylamine and propargyl-homopropargylbenzylamine gave isoindoline and tetrahydroisoquinoline derivatives in good to high yields. This procedure was also used for the selective preparation of benzene derivatives from three different alkynes. The use of trimethylsilyl-substituted alkyne as the first, second or third alkyne afforded desilylated benzene derivatives. The reaction of zirconacyclopentadienes with allenes gave benzene derivatives as a mixture of two isomers.

Abstract:Transition metal-catalyzed cycloadditions of cyclopropanes have been well developed over the past several decades, leading to numerous new types of cycloadditions which are complementary to the traditional cycloadditions for the synthesis of carbocycles. Cycloadditions of vinylcyclopropanes (VCPs) and methylenecyclopropanes (MCPs) constitute two main aspects of this field. VCPs can act either as five-carbon synthons or three-carbon synthons, depending on whether the vinyl substituent is acting as an additional two-carbon synthon or not. As five-carbon synthons, VCPs are involved in [5+1], [5+2], [5+2+1], and [5+1+2+1] cycloadditions. As three-carbon synthons, VCPs are mainly involved in [3+2] and [3+2+1] cycloadditions. MCPs mostly act as three-carbon synthons and can have [3+2] cycloadditions with different π systems. Other types of cycloadditions involving MCPs are also reviewed, such as [3+1], [3+2+2], and [4+3+2] cycloadditions. Cycloadditions of some other unusual cyclopropane derivatives are also introduced briefly. The cycloadditions of VCPs and MCPs have found applications in total synthesis and some representative molecules are tabulated as selected examples.
