Abstract:The effects of extraction time, temperature, pressure and different concentration of ethanol and their interactions on the yields of amentoflavone, quercetin and ginkgetin extracted from Taxus chinensis by supercritical CO2 were investigated by using a central composite design (CCD). An CCD experimental design with four factors and five levels was used to optimize the extraction parameters. Ultra performance liquid chromatography (UPLC) was used to analyze the content of the tree components in the extracts. Experimental results show that the main effects of factors and their interactions are significant on the yields (p < 0.05). The optimal extraction conditions were established for the three compounds: yield of 4.47 mg/g for amentoflavone at 48 °C, 25 MPa, 2.02 h and 78.5% ethanol, 3.73 mg/g for quercetin at 46 °C, 24 MPa, 2.3 h, 82% ethanol and 3.47 mg/g for ginkgetin at 48 °C, 20 MPa, 2.38 h, 82% ethanol, respectively.

Abstract:The GC-MS analysis of supercritical CO2 extracting products from Tibetan medicine Rhodiola was discussed in this paper.The GC-MS was used to analyze the product which was extracted under these conditions:the concentration of entrainer is 85%(V/V),the extracting temperature is 55 ℃,the extracting pressure is 30 MPa,the extracting time is 2 h.32 kinds of compounds were identified in the product,which are alcohols,esters,alkanes and alkenes,these account for 87.548% of the total components.

Abstract:In this paper, the volatile compounds of Schisandra chinensis obtained by different extraction techniques including supercritical fluid extraction (SFE), steam distillation (SD), Soxhlet extraction (SE) and ultrasound-assisted extraction (UAE) were investigated for the first time. The sample preparation procedure for GC-MS analysis of the volatile compounds was optimized and then 37, 45, 27 and 37 compounds were identified in the samples obtained by SFE, SD, SE and UAE methods, respectively. As the therapeutic effect of the traditional Chinese medicine is usually based on multifarious essential components or the combination of them instead of only one component, the volatile compounds were compared in groups with the extracts by SE, SD and UAE. This would be more reasonable to evaluate the effects of an alternative technique to extracting multifarious essential components. Among the identified components in the SFE extract, 32 compounds were the same as that by three conventional methods, accounting for 90.5% of the volatile compounds identified. However, as the volatile compounds were classed into groups, it was easy to see that the Schisandra chinensis oil extracted by SFE was made up largely of aromatics and sesquiterpenoids (52.1 and 27.6%, respectively), with less amounts of monoterpenoids and other compounds, distinguishing SFE from the conventional extractions.

Abstract:This paper simply studied the extract craft of Fructus Schisandrae Sphenantherae by supercritical CO2 extract, discussed effects of different factors such as extract pressure, extract temperature, extract time, CO2 current capacity condition on the extract efficiency, and employed GC-MS to carry on the qualitative analysis of the volatile constituent extracted by supercritical CO2 extract. The result indicated that, The optimum conditions is extract pressure 30 MPa, the extract temperature 50 ℃, extract time 40 min, CO2 current capacity 30～35 L/H. The volatile constituent by the hydrocarbon compound primarily, accounts for above 60%, in which comparative content highest is Ylangene, accounts for 10.21%, moreover, it also contains the alcohol type, the aldehyde class, the acids and the ester etc.

Abstract:OBJECTIVE: To analyze the constituents of volatile oils extracted from Yuan Zhi (Radix Polygalae), Shi Chang Pu (Acorus Tatarinowii), and a mixture of the two herbs.METHODS: The volatile oils were extracted using supercritical fluid extraction (SFE) with CO2, and the constituents of the volatile oil extracts were analyzed by gas chromatography-mass spectrometry (GC-MS). The relative content of each component was calculated using peak area normalization.RESULTS: The optimized SEE conditions were 45 MPa at 35 degrees C for 2 h. Twenty-four compounds were identified in the extract from the Yuan Zhi (Radix Polygalae) and Shi Chang Pu (Acorus Tatarinowii) mixture, and six of these had relative contents >1. These compounds were 1,2-dimethoxy-4-(2-propenyl)-benzene; 1,2,3-trimethoxy-5-(2-propenyl)-benzene; beta-asarone; (Z,Z) 9,12-octadecadienoic acid; (Z) 6-octadecenoic acid; and ethyl oleate. Combination of the herbs increased the number of pharmacologically active substances in the extract and decreased the number of compounds with one benzene ring compared with the extracts from the individual herbs.CONCLUSION: These results indicate there is a synergistic relationship among the compounds in these herbs. Abstract:Volatile components from Exocarpium Citri Grandis (ECG) were, respectively, extracted by three methods, that is, steam distillation (SD), headspace solid-phase microextraction (HS-SPME), and solvent extraction (SE). A total of 81 compounds were identified by gas chromatography-mass spectrometry including 77 (SD), 56 (HS-SPME), and 48 (SE) compounds, respectively. Despite of the extraction method, terpenes (39.98 similar to 57.81%) were the main volatile components of ECG, mainly germacrene-D, limonene, 2,6,8,10,14-hexadecapentaene, 2,6,11,15-tetramethyl-, (E, E, E)-, and trans-caryophyllene. Comparison was made among the three methods in terms of extraction profile and property. SD relatively gave an entire profile of volatile in ECG by long-time extraction; SE enabled the analysis of low volatility and high molecular weight compounds but lost some volatiles components; HS-SPME generated satisfactory extraction efficiency and gave similar results to those of SD at analytical level when consuming less sample amount, shorter extraction time, and simpler procedure. Although SD and SE were treated as traditionally preparative extractive techniques for volatiles in both small batches and large scale, HS-SPME coupled with GC/MS could be useful and appropriative for the rapid extraction and qualitative analysis of volatile components from medicinal plants at analytical level.

Abstract:Gastrodin, a pharmacologically active constituent, was ultrasonically extracted from gastrodia elata Bl. in the aqueous solution. The effects of six parameters including ethanol-water compositions, extraction time, extraction temperature, particle size, solvent volume, and ultrasonic power on the extraction yield of gastrodin were investigated. According to the orthogonal design, the optimal extraction conditions was explored as extraction temperature 60 degrees C, extraction time 50 minutes, ultrasonic power 126W, solvent volume 8mL center dot g-1, ethanol-water compositions 70%, and particle size 10-20 mesh. Though the yield of gastrodin via ultrasonic extraction was about 0.01% lower than that from the reflux extraction, the extraction time of the ultrasonic extraction was greatly shortened. Therefore, ultrasonic extraction has high efficiency and is proved to be very valuable in the extraction of gastrodin from gastrodia elata Bl.

Abstract:Gastrodia elata (G. elata), as a natural plant with nourishing and edible value, plays a vital role in food and pharmaceutical production. In the present study, a novel approach for the simultaneous determination of six components based on high-performance liquid chromatography (HPLC) coupled with photodiode array detection (PDA) was developed for the quality evaluation of G. elata from different regions. Meanwhile, antioxidant and α-glucosidase inhibitory activities were estimated and compared. The results indicated that the total contents of the six compounds in G. elata from Guizhou and Zhejiang provinces obtained by boiling were higher than those obtained by steaming in Anhui and Yunnan provinces. In addition, samples from Guizhou, Yunnan and Shanxi provinces contained more p-hydroxybenzyl alcohol, and always possessed higher antioxidant activity, while samples collected from Anhui province showed the highest α-glucosidase inhibitory activity with the highest gastrodin content. The results revealed that the quality of G. elata was affected by different regions and different initial processing technologies, which provided a reference for the selection and application of G. elata.

Abstract:The feasibility of active components of Dracocephalum tanguticum Max-im (DtM) extracted by supercritical CO2 was discussed in this paper. The components of the essential oil from the DtM were analyzed by GC-MS. The optimal conditions of supercritical CO2 extraction: pressure 20 MPa, extraction temperature 50 degrees C, extraction time 75 min, CO2 flow 1.0 L(.)min(-1) were obtained. Seventy-six components from DtM were separated and identified by supercritical CO2 extraction. There is an abundance of unsaturated fatty acid, saturated fatty acid, acid, alkane, and ester in the essential oil extracted from the DtM. The study established a good method for further studying medical applications and develoment of the DtM.

Abstract:Objective To analyze the chemical components of essential oil from Congbai. Methods The volatile oil was extracted with supercritical carbon dioxide,the components in which and contents were determined by GC-MS and normalization method. Results 16 components from Congbai by SFE were separated and identified.The main compounds of volatile oil were diterpenoids,fatty acids,alkanes,organosulfur compounds. Conclusion We have investigated the low polarity gradients from the volatile part of Congbai,the way of which benefits for further studying of Congbai.

Abstract:Volatile components was extracted from Tylophora ovata (Lindl.) Hook.ex Steud.by supercritical CO2 fluid extraction. Extraction conditions were: extracting 3 h at 50 ℃, 35 MPa (pressure) and using 20 ml 95 % alcohol as modifier. The volatile components extracted were determined by gas chromatography with area normalization method and identified by gas chromatog- raphy mass spectrometry(GC-MS).The optimal chromatographic conditions were: HP-5 MSsilica capillary column (0.25 mm× 30 m×0.25μm); column temperature: 60℃( hold for 1 min) to 280 ℃ at a rate of 4 ℃/min, and hold for 10 min;split injection, 20:1; injector temperature 250 ℃. Thirty nine components were identified for the constituents extracted by SFE CO2.

Abstract:Technology of supercritical fluid extraction for Tibetan rhodiola was investigated,components of the extract were analyzed by GC-MS.The result shows that the optimum extraction conditions are as follows: the volume percentage of ethanol is 85%,the extraction temperature is 55 ℃,the extraction pressure is 30 MPa,and the extraction time is 2 h.Under these conditions,the extract rate of rhodiola is 5.65%.32 components were identified,the main components of this extract were alcohol,ester,alkane,olefin,etc.
Optimization of Supercritical Carbon Dioxide Extraction of Gardenia Fruit Oil and the Analysis of Functional Components

Abstract:Supercritical carbon dioxide (SC-CO2) extraction of whole fruit oil from Gardenia jasminoides Ellis was performed. The effect of extraction pressure, temperature and CO2 flow rate on the oil yield was investigated by response surface methodology (RSM). The results showed that experimental data had a good fit to the proposed model (R 2 = 0.938). Extraction pressure, CO2 flow rate, the quadratics of pressure, and the interaction between pressure and flow rate showed significant effects on the oil yield (p < 0.05). The optimum parameters that maximized the yield of gardenia fruit oil (GFO) were: extraction pressure of 36.8 MPa, temperature of 65 °C, and CO2 flow rate of 15 kg/h. The main fatty acid of GFO was linoleic acid (about 44%), followed by palmitic acid (about 26.4%) and oleic acid (about 24.6%). α-Tocopherol was dominant in the total tocopherols of GFO, and showed the main antioxidant activity. The fatty acid composition and tocopherols content of GFO were not remarkably affected by the extraction by SC-CO2 and n-hexane.

Abstract:Objective To analyze the chemical componets of burdock root oil.Methods The volatile oil of burdock root was extracted by supercritical fluid CO2 extraction technology,and the chemical components were analyzed using the high pressure GC-MS association with the computer technology.Results 21 chromatographic peaks were obtained.Through the NIST standard spectrum storage retrieval,20 compounds were identified.Conclusion The best extraction condition requires the extraction pressure of 20.0 mPa,extraction temperature of 50℃ and extraction time of 3 h.The yield of volatile oil is 2.7%.

Abstract:An experimental setup for separating ginger essential oil by supercritical fluid extraction is established. The effects of the extraction pressure, temperature, CO2 flow rate and particle size of raw material on the extraction rate are investigated, and the optimum process conditions of supercritical CO2 extraction are determined. A mathematical simulation model is established based on the mass conservation in differential units of extraction bed. The total mass transfer driving force and the equilibrium absorption constant are evaluated by the linear driving force theory. The results from numerical simulation agree well with the experimental data.

Abstract:The volatile constituents of seed oil from Glycyrrhiza uralensis Fisch. were extracted by supercritical CO2 extraction ( SFE-CO2) and analyzed by gas chromatography-mass spectrometry ( GC-MS). For the first time 52 peaks were separated and 16 peaks were identified, the compounds were quantitatively determined by normalization method, and the identified compounds represent 80. 85% of total GC peak areas. The main compounds were dibutyl phthalate and β-sitosterol acetate, which have many physiological activity and provide important foundation for further new medicine exploitation of seed oil from Glycyrrhiza uralensis Fisch.

Abstract:Gastrodia elata Blume (G. elata) is a traditional medicinal and edible plant whose quality is significantly influenced by post-harvest processing. To obtain an optimal post-harvest processing method for G. elata, this study employed sensory evaluation, scanning electron microscopy (SEM), gas chromatography-ion mobility spectrometry (GC-IMS), and non-targeted metabolomics, in conjunction with an in vitro digestion model, to assess the impact of different processing and drying methods on the quality of G. elata. The findings showed that the steam treatment followed by heat pump drying resulted in the highest levels of total phenols, total flavonoids, and polysaccharides in G. elata, and caused more pronounced damage to its microstructure. This treatment also maintained the highest antioxidant activities and optimal acetylcholinesterase (AChE) inhibition capacity throughout in vitro digestion, meanwhile, effectively eliminating the unpleasant odor and achieving the highest sensory scores. Furthermore, non-targeted metabolomic analysis revealed noteworthy alterations in the metabolite profile of G. elata, mainly related to purine metabolism and the biosynthesis of amino acids pathways. This study provides valuable insights into the post-harvest processing of G. elata.
