Abstract:In the current work, the effects of steam and boiling water blanching on the drying characteristics, water distribution, microstructure, and contents of bioactive substances of Gastrodia elata (G. elata) were explored. Results showed that the degree of steaming and blanching was related to the core temperature of G. elata. The steaming and blanching pretreatment increased the drying time of the samples by more than 50%. The low-field nuclear magnetic resonance (LF-NMR) of treated samples showed that the relaxation time corresponded to water molecule states (bound, immobilized, and free) and G. elata became shorter, which indicated a reduction in free moisture and increased resistance of water diffusion in the solid structure during drying. Hydrolysis of polysaccharides and gelatinization of starch granules was observed in the microstructure of treated samples, which was consistent with changes in water status and drying rates. Steaming and blanching increased gastrodin and crude polysaccharide contents and decreased p-hydroxybenzyl alcohol content. These findings will contribute to a better understanding of the effect of steaming and blanching on the drying behavior and quality attributes of G. elata.

Abstract:To explore the transformation mechanisms of free gastrodin and combined gastrodin before and after steaming of Gastrodia elata (G. elata), a fresh G. elata sample was processed by the traditional steaming method prescribed by Chinese Pharmacopoeia (2015 version), and HPLC-ESI-TOF/MS method was used to identify the chemical composition in steamed and fresh G. elata. Finally, 25 components were identified in G. elata based on the characteristic fragments of the compounds and the changes of the 25 components of fresh and steamed G. elata were compared by the relative content. Hydrolysis experiments and enzymatic hydrolysis experiments of 10 monomer compounds simulating the G. elata steaming process were carried out for the first time. As a result, hydrolysis experiments proved that free gastrodin or p-hydroxybenzyl alcohol could be obtained by breaking ester bond or ether bond during the steaming process of G. elata. Enzymatic experiments showed that steaming played an important role in the protection of gastrodin, confirming the hypothesis that steaming can promote the conversion of chemical constituents of G. elata—inhibiting enzymatic degradation. This experiment clarified the scientific mechanism of the traditional steaming method of G. elata and provided reference for how to apply G. elata decoction to some extent.

Abstract:Gastrodia elata (GE) has a variety of processed products. However, there are few studies on the comparison of the chemical composition and activity differences of various processed products. In order to further study, the raw materials and different processed products of GE were chemically characterized in detail by UPLC-ESI-Q-TOF-MS/MS, and the content of 7 constituents were determined by HPLC-UV. The activities of different products against α-glucosidase, xanthine oxidase (XO) and acetylcholinesterase (AchE) were evaluated. In total, 170 compounds were identified, primarily including 31 phenolics, 109 parishins, 20 amino acids, 5 nucleotides, and 5 organic acids, and first discovery of 4 potential new compounds. The differences in the chemical composition of GE and its products are mainly in phenolics and parishins. Fresh GE (FGE) extract has the highest content of p-hydroxybenzyl alcohol and p-hydroxybenzaldehyde. The parishins content of dried GE processed products were higher than FGE. And, FGE extract showed the strongest inhibitory activity of α-glucosidase (IC50 was 0.47mg/mL) and AchE (IC50 was 0.56mg/mL), and ginger GE extract showed the strongest XO inhibitory activity (IC50 was2.59mg/mLL), all below corresponding standard inhibitors. And this is the first time that the inhibitory activity of GE against XO has been evaluated.

Abstract:Ginkgo biloba is an attractive and traditional medicinal plant, and has been widely used as a phytomedicine in the prevention and treatment of cardiovascular and cerebrovascular diseases. Flavonoids and terpene lactones are the major bioactive components of Ginkgo, whereas the ginkgolic acids (GAs) with strong allergenic properties are strictly controlled. In this study, we tested the content of flavonoids and GAs under ultraviolet-B (UV-B) treatment and performed comparative proteomic analyses to determine the differential proteins that occur upon UV-B radiation. That might play a crucial role in producing flavonoids and GAs. Our phytochemical analyses demonstrated that UV-B irradiation significantly increased the content of active flavonoids, and decreased the content of toxic GAs. We conducted comparative proteomic analysis of both whole leaf and chloroplasts proteins. In total, 27 differential proteins in the whole leaf and 43 differential proteins in the chloroplast were positively identified and functionally annotated. The proteomic data suggested that enhanced UV-B radiation exposure activated antioxidants and stress-responsive proteins as well as reduced the rate of photosynthesis. We demonstrate that UV-B irradiation pharmaceutically improved the metabolic ingredients of Ginkgo, particularly in terms of reducing GAs. With high UV absorption properties, and antioxidant activities, the flavonoids were likely highly induced as protective molecules following UV-B irradiation.

Abstract:This study investigates the optimal extraction conditions, monosaccharide composition, and antioxidant activity analysis of polysaccharides from the aerial parts of three different varietal varieties of Gastrodia (i.e., G. elata Bl. F. elata, G. elata Bl. F. Viridls MaKino , and G. elata Bl. F. Glauca S Chow ). The influence of extraction temperature (30–70 °C), extraction time (15–55 min), and liquid-to-solid ratio (25–65 mL/g) on the yield of polysaccharides was analyzed through single-factor experiments. The response surface methodology was used to optimize the extraction process, and a mathematical model was established to obtain the optimal extraction conditions. The response surface experiment was presented as follows: The optimal extraction conditions for polysaccharides were a liquid-to-solid ratio of 59 mL/g, 56 °C, and 36 min. The polysaccharide yield in G. elata. Bl. F. Glauca S Chow aerial parts under these conditions was 10.90%, which was close to the theoretical value calculated by the model (10.64%). Under the optimal conditions, the yields of polysaccharides followed: G. elata Bl. F. elata (11.32%) > G. elata Bl. F. Glauca S Chow (10.90%) > G. elata Bl. F. Viridls MalKino (10.50%). The polysaccharides were mainly composed of 10 monosaccharides, including glucose, rhamnose, mannose, and xylose. However, the content of monosaccharides in polysaccharides of different varieties of Gastrodia elata Bl aerial parts varied greatly, with glucose and rhamnose being the highest, both exceeding 20%. The polysaccharides in different varieties of Gastrodia elata Bl aerial parts had certain in vitro antioxidant activity. The total reducing power and scavenging rates of 2,2-diphenylpicrylhydrazyl (DPPH) and 2,2′-azinobis-(3-ethylbenzthiazolin-6-sulfonic acid) (ABTS) free radicals increased with the increase of polysaccharide concentration. When the polysaccharide concentration was 1.0 mg/mL, the scavenging rates of DPPH and ABTS free radicals were both over 80%. This study provides a theoretical basis for the further development and utilization of Gastrodia elata Bl aerial part resources.

Abstract:Gastrodia elata Blume (Tianma in Chinese), a myco-heterotrophic orchid, is widely distributed in China. Tubers derived from this orchid are traditionally used as both medicinal and edible materials. At present, five primary varieties of G. elata are recorded in the "Flora of China." Among them, the three main varieties currently in artificial cultivation are G. elata f. elata (GR, red stem), G. elata f. glauca (GB, black stem), and G. elata f. viridis (GG, green stem). In our study, the metabolic profiles and chemical composition of these three varieties were determined via UPLC-MS/MS and HPLC-UV. In total, 11,132 metabolites were detected, from which multiple phytometabolites were identified as aromatic compounds, heteroatomic compounds, furans, carbohydrates, organic acids, and their derivatives. A number of differentially expressed metabolites (DEMs) were annotated as bioactive ingredients. Overall, parishins, vanilloloside, and gastrodin A/B in the GB group were markedly higher, whereas gastrodin, gastrol, and syringic acid were more enriched in the GG or GR groups. Moreover, HPLC fingerprint analysis also found six metabolites used as markers for the identification of Gastrodiae Rhizoma in the Chinese Pharmacopoeia, which were also typical DEMs in metabolomics. Of these, gastrodin, 4-hydroxybenzyl alcohol, citric acid, and adenosine were quantitatively detected, showing a similar result with the metabolomic data. In summary, our findings provide novel insights into the phytochemical ingredients of different G. elata varieties, highlighting diverse biological activities and healthcare value.

Abstract:Numerous sugar radicals have been observed by EPR and ENDOR spectroscopy in X-irradiated nucleosides and nucleotides. However, no sugar radicals have been observed in irradiated DNA. One possibility exists that sugar radicals may be present in relatively small abundance and therefore have so far escaped detection. Another possibility is that each of the five carbon-centered H-abstraction sugar radicals may exist in a wide range of conformations. Adding together various groups of radicals with different hyperfine couplings and anisotropic g factors may result in very broad EPR lines which would be difficult to detect. Using the crystal structure of a B-DNA dodecamer, the conformations of the five H-abstraction sugar radicals have been computed at all sites. The X-band EPR spectra for each radical were then simulated using typical alpha-proton and beta-proton couplings computed from the various radical conformations. The results suggest considerable broadenings of the EPR lines for the C2', C3' H-abstraction radicals. Composite EPR spectra were simulated by adding 15% of an H-abstraction radical to the DNA spectrum. The results indicate that the outer lines of the C1' radical are visible and should be easy to identify. The broad spectra of the C2' and C3' radicals are barely visible. The simulated spectra of the C4' and C5' radicals are basically doublets and are obscured by the DNA signal.

Abstract:In this study, the drying kinetics and multidimensional quality of Gastrodia elata (GE) obtained through five different drying methods were analyzed. The Page model provided the best fit. Though widely used, hot-air drying (HA-D) showed mediocre performance. Due to strong heat penetration, microwave drying (MV-D) and infrared drying (IR-D) effectively reduced drying time and energy consumption. However, they experienced Maillard and caramelization reactions, resulting in notable cell structure shrinkage, severe browning, pronounced caramel taste, and poor retention of active ingredients. Conversely, freeze-drying (F-D) and vacuum drying (V-D), due to the involvement of low-temperature or vacuum environments, demonstrated optimal performance in preserving microstructure, antioxidant activity, active ingredients, and flavor. However, F-D was time-consuming (1320 min) and had an energy consumption of 2.69 times that of HA-D, limiting its large-scale application. V-D struck the best balance between energy consumption and product quality, making it a highly promising drying method for GE.

Abstract:Sulfur is an important industrial raw material. The sulfur dioxide gas (SO2) generated after burning can significantly improve the appearance quality of food and enhance the storage time. However, excessive sulfur dioxide will not only pollute the environment, but also cause the deterioration of food quality, and even the high residual levels can increase the risk of cancer. Traditional processing technology for Gastrodia elata Blume is designed to steam fresh G. elata, followed by fumigation with sulfur, and then dried. However, the effect of this technology on the edible quality of G. elata and its mechanism have yet to be investigated. Starch is the main component of G. elata, which has a significant impact on its morphology and chewiness. Therefore, the effects of traditional processing technology on the morphology, chewiness, and structure of G. elata starch were investigated via spectral analysis and Texture Profile Analysis (TPA). The results showed that compared with G. elata non-steam and non-sulfur fumigation (NSt-NSf), the pH of G. elata after steam and sulfur fumigation (St-Sf) decreased by 0.399, and the starch whiteness, paste transparency and amylose content increased by 1.15-fold, 1.08-fold and 1.10-fold, respectively (p < 0.05). The solubility of starch was reduced by 70%, the swelling degree, starch gel chewiness and the content of rapidly digested starch were increased by 1.66-, 1.17-, and 1.09-fold, respectively. The final viscosity of starch was increased by 5.22-fold (p < 0.05). After steaming, the starch granules form a smooth surface, but the surface of starch granules after sulfur fumigation can see obvious erosion traces, and the starch after sulfur fumigation is easier to gelatinize into a homogeneous solution and improve the cross-linking between molecules. To sum up, sulfur fumigation after steaming (St-Sf) promote the increase of amylose content and the release of molecular chains from starch granules, which are the main reasons for improving the morphology and chewiness of G. elata starch. This study can provide technical and theoretical support for improving the quality of starch-rich foods, replacing sulfur fumigation and reducing potential environmental hazards.

Abstract:ETHNOPHARMACOLOGICAL RELEVANCE: Gastrodia elata Blume (G. elata) has a long historical application in Asian countries and its tubers, seeds, and stalks are capable of being utilized for medicine, food, or health care products.AIM OF THE REVIEW: This study aimed to offer a systematic and up-to-date analysis of the current review of the G. elata research advances in traditional uses, phytochemistry, pharmacology, applications, and quality control, as well as a scientific reference for the development and utilization of this plant.MATERIALS AND METHODS: Electronic databases including PubMed, Web of Science, Google Scholar, ScienceDirect, SciFinder, and CNKI were used for the collection of publications on G. elata. The following keywords of G. elata were used truncated with other relevant topic terms, such as phenolic compounds, polysaccharides, glycosides, neuroprotection, learning and memory improvement effects, cardioprotection, applications, and quality control.RESULTS AND CONCLUSIONS: Approximately 134 chemical components mainly categorizing as phenolic compounds, polysaccharides, glycosides, organic acids, and sterols were reported from this plant. Moreover, preclinical studies indicated that G. elata performs several functions, including neuroprotection, learning and memory improvement effects, cardioprotection, vaso-modulatory effect, anti-depression, anti-cancer, and other effects. Currently, G. elata has been widely applied to clinics and foods. The available literature shows that the quality of G. elata might be affected by factors such as origin, fungus, and harvest time, which will have an impact on the drug efficacy. According to past research, G. elata is a potential medicinal and edible plant with several active components and pharmacological activity that has a high application value in medicine and the food business. Nevertheless, few studies have concentrated on characterization of polysaccharides structure and study of non-medicinal parts, implying that further comprehensive research on its polysaccharides structure and non-medicinal parts is critical for full utilization of resources of G. elata.

Abstract:This study aimed to investigate the impact of slightly acidic electrolyzed water combined with lithium magnesium silicate hydrosol on the quality of fresh slices of Gastrodia elata under varying storage temperatures, including room temperature fresh slices of Gastrodia elata 25 °C and 37 °C. Fresh slices of Gastrodia elata 25 and 37 samples were stored for 13 days and extensively analyzed for color, weight loss, decay index, bacterial count, vitamin C, and polysaccharide contents during different storage periods. The findings revealed that the slightly acidic electrolyzed water + hydrosol treatment notably decreased weight loss and decay index compared to distilled water and slightly acidic electrolyzed water treatments. Moreover, fresh slices of Gastrodia elata treated with slightly acidic electrolyzed water + hydrosol exhibited untraceable bacterial counts after 3 days, with counts starting to increase after 7 days of storage. The bacterial counts rose from 3.25 to 5.36 and from 4.13 to 5.79 log CFU/g under both storage conditions. The application of slightly acidic electrolyzed water + hydrosol resulted in reduced chromaticity values of L*, a*, and b* on the Gastrodia elata surface, along with a lower percentage loss of polysaccharide contents and vitamin C compared to distilled water and slightly acidic electrolyzed water treatments. These results suggested that Gastrodia elata treated with slightly acidic electrolyzed water + hydrosol maintained its quality characteristics and nutritional attributes, exhibiting greater stability during storage.

Abstract:Gastrodia elata (Orchidaceae) is native to mountainous areas of Asia and is a plant species used in traditional medicine for more than two thousand years. The species was reported to have many biological activities, such as neuroprotective, antioxidant, and anti-inflammatory activity. After many years of extensive exploitation from the wild, the plant was added to lists of endangered species. Since its desired cultivation is considered difficult, innovative cultivation methods that can reduce the costs of using new soil in each cycle and at the same time avoid contamination with pathogens and chemicals are urgently needed on large scale. In this work, five G. elata samples cultivated in a facility utilizing electron beam-treated soil were compared to two samples grown in the field concerning their chemical composition and bioactivity. Using hyphenated high-performance thin-layer chromatography (HPTLC) and multi-imaging (UV/Vis/FLD, also after derivatization), the chemical marker compound gastrodin was quantified in the seven G. elata rhizome/tuber samples, which showed differences in their contents between facility and field samples and between samples collected during different seasons. Parishin E was also found to be present. Combining HPTLC with on-surface (bio)assays, the antioxidant activity and inhibition of acetylcholinesterase as well as the absence of cytotoxicity against human cells were demonstrated and compared between samples.

Abstract:Gastrodia elata (G. elata) tuber is a valuable herbal medicine used to treat many diseases. The procedure of establishing a reasonable and feasible quality assessment method for G. elata tuber is important to ensure its clinical safety and efficacy. In this research, an effective and comprehensive evaluation method for assessing the quality of G. elata has been developed, based on the analysis of high performance liquid chromatography (HPLC) fingerprint, combined with the quantitative analysis of multi-components by single marker (QAMS) method. The contents of the seven components, including gastrodin, p-hydroxybenzyl alcohol, p-hydroxy benzaldehyde, parishin A, parishin B, parishin C, and parishin E were determined, simultaneously, using gastrodin as the reference standard. The results demonstrated that there was no significant difference between the QAMS method and the traditional external standard method (ESM) (p > 0.05, RSD < 4.79%), suggesting that QAMS was a reliable and convenient method for the content determination of multiple components, especially when there is a shortage of reference substances. In conclusion, this strategy could be beneficial for simplifying the processes in the quality control of G. elata tuber and giving references to promote the quality standards of herbal medicines.

Abstract:Gastrodia elata (GE) is traditionally subjected to steaming, and steaming duration plays a crucially important role in determining GE quality. This study examined the variations in bioactive components during the steaming process and proposed the utilization of electronic eye and Fourier Transform near-infrared (FT-NIR) spectroscopy for quality assessment. The findings revealed that the levels of parishin E parishin B, parishin A, and gastrodin initially rose and subsequently declined, while 4-Hydroxybenzyl alcohol exhibited a rapid decrease followed by stabilization. With prolonged steaming, the brightness of GE decreased, while the red and yellow tones became more pronounced and the color saturation increased. FT-NIR divided the steaming process into three stages: 0 min (raw GE), 0-9 min (partially steamed GE), and 9-30 min (fully steamed GE), and the partial least squares regression models effectively predicted the levels of five components. Overall, this study provided valuable insights into quality control in food processing.

A total of 345 metabolites were detected, including 76 different amino acids and their derivatives containing all human essential amino acids (e.g., l-(+)-lysine, l-leucine), 13 vitamins (e.g., nicotinamide, thiamine), and 34 alkaloids (e.g., spermine, choline). GEGm has higher amino acid accumulation than GEE y, GEEm and GEGy, and vitamin contents were also slightly different in all four samples. Implying that GE , especially GEGm , is a kind of excellent complementary food as amino acid nutrition provider. From assembled 21,513 transcripts (genes) based on the transcriptome, we identified many genes that encode enzymes (e.g., pfkA, bglX, tyrAa, lysA, his B, aroA), which are responsible for the biosynthesis of amino acids and enzymes (e.g., nadA, URH1, NAPRT1, punA, rsgA) that related to vitamins metabolism. A total of 16 pairs of the differentially expressed genes (DEG) and differentially accumulated metabolites (DAM) (e.g., gene-tia006709 coding GAPDH and l-(+)-arginine, and gene-tia010180 coding tyrA and l-(+)-arginine) and three DEG-DAM pairs (e.g., gene-tia015379 coding NadA and nicotinate d-ribonucleoside) show significant similar positive or negative correlation based on three, and two comparisons of GEEy vs. GEGy , GEGy vs. GEGm , GEEy vs. GEGy and GEEm vs. GEGm , which involved into amino acid biosynthesis, and nicotinate nicotinamide metabolism, respectively. These results prove that the enzyme coded by these DEG promotes (positive correlation) or inhibits (negative correlation) the biosynthesis of parallel DAM in GE . Overall, the data and corresponding analysis in this study provide new insights into the nutritional properties of GE and the related molecular basis.

Abstract:Ethnopharmacological relevance Gastrodia elata Blume is a traditional Chinese medicine with the effects of improving the deficiency of the body and maintaining health, and polysaccharide (GEP) is one of the effective ingredients to play these activities of G. elata . Traditionally, G. elata is orally administered, so the activities of GEP are associated with digestive and intestinal metabolism. However, the digestive behavior of GEP and its effects on the human gut microbiota are unclear and need to be fully studied. Aim of the study This study aimed to investigate the changes in structural characteristics of GEP during digestion and the related impacts of its digestive product on gut microbiota in human fecal fermentation, and to explain the beneficial mechanism of GEP on human health from the perspective of digestive characteristics and "gut" axis. Materials and methods The changes of reducing sugars, free monosaccharides and physicochemical properties of GEP during digestion were investigated by GPC, HPLC, FT-IR, CD, NMR, SEM, and TGA. Moreover, polysaccharide consumption, pH value changes, SCFAs production, and changes in gut microbiota during fermentation were also discussed. Results During digestion of GEP, glucose was partially released causing a decrease in molecular weight, and a change in monosaccharide composition. In addition, the characteristics of GEP before and after digestion, including configuration, morphology, and stability, were different. The digestive product of GEP was polysaccharide (GEP-I), which actively participated in the fecal fermentation process. As the fermentation time increased, the utilization of GEP-I by the microbiota gradually increased. The abundance of probiotics such as Bifidobacterium , Collinsella , Prevotella , and Faecalibacterium was significantly increased, and the abundance of pathogenic Shigella , Dorea , Desulfovibrio , and Blautia was significantly inhibited, thereby suggesting that GEP has the potential to maintain human health through the "gut" axis. In addition, the beneficial health effects of GEP-I have also been observed in the influence of microbial metabolites. During the fermentation of GEP-I, the pH value gradually decreased, and the contents of beneficial metabolites such as acetic acid, propionic acid, and caproic acid significantly increased. Conclusion The structure of GEP changed significantly during digestion, and its digestive product had the potential to maintain human health by regulating gut microbiota, which may be one of the active mechanisms of GEP.

Abstract:Orchidaceae, well known for its fascinating flowers, is one of the largest and most diverse families of flowering plants. There are many kinds of plants in this family; these are distributed practically globally and have high ornamental and medicinal values. Gastrodia elata Blume, a traditional Chinese medicinal herb, is a rootless and leafless achlorophyllous orchid. Phenolic compounds are considered to be the major bioactive constituents in G. elata, with antioxidant, antiangiogenic, neuroprotective, antidepressant, anxiolytic, and sedative activities. In this study, we determined the contents of six main phenolic components in tubers, stems and flowers from G. elata. Meanwhile, the transcriptomes of the tuber, stem and flower tissues of G. elata were obtained using the BGISEQ-500 platform. A total of 58.29 Gb of data and 113,067 unigenes were obtained, of which 74,820 unigenes were functionally annotated against seven public databases. Differentially expressed genes between tuber, stem and flower tissues were identified. A total of 76 DEGs encoding eight key enzymes were identified as candidate genes involved in the biosynthesis of phenolics in G. elata. For further validation, the expression levels of unigenes were measured using quantitative real-time PCR. Our results greatly enrich the transcriptomic data of G. elata and provide valuable information for the identification of candidate genes involved in the biosynthesis of secondary metabolites.

Abstract:Gastrodia elata Bl. still widely known as a medicinal plant due to its anti-inflammatory, neuroprotection, cardiovascular protection etc. Additionally, these medical applications cannot be separated from its antioxidant, anti-aging, regulating cell apoptosis ability, which make it have potential as a functional food as well as it has been eaten for more than 2,000 years in China. At present, although Gastrodia elata Bl. has appeared in a large number of studies, much of the research is based on drugs rather than foods. The review of Gastrodia elata Bl. from the perspective of food is one of the necessary steps to promote related development, by reviewing the literature on analytical methods of Gastrodia elata Bl. in recent years, critical components change in the extraction, analytical methods and improvement of food applications, all of aspects of it was summarized. Based on the report about physical and chemical changes in Gastrodia elata Bl. to discover the pathway of Gastrodia elata Bl. functional food development from current to the future.

Abstract:Abstract In this study Ginkgo biloba leaves (GBL) decoction and commercial capsules were digested using an in vitro model. Thirty-six active compounds were identified and quantified by HPLC-ESI-MS analysis based on the MS/MS patterns (precursor ions and product ions) and retention times, in comparison with reference standards. Most compounds in GBL showed a significant decrease during intestinal digestion, with an exception of vanillic acid and biflavonoids. Bioaccessibility values of chemical compositions varied between decoction and capsules samples. Also, significant reductions of total flavonoids and total phenolic content was observed after in vitro digestion. Both, 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2′-azino-bis (3-ethylbenzothiazo-line-6-sulfonic acid (ABTS) scavenging capacity decreased after gastric digestion, but increased during intestinal digestion. Nevertheless, different behaviour was observed in reducing antioxidant power (FRAP) assay. Compared to the pH of digestion, the influence of digestive enzymes on the chemical composition and antioxidant activity of GBL was relatively minor. Overall, these results may help provide a valid foundation for further investigations on bioactive compounds and the pharmacodynamics of GBL.

Novel Strategies Using Total Gastrodin and Gastrodigenin, or Total Gastrodigenin for Quality Control of Gastrodia elata

Abstract:Gastrodia elata Blume (G. elata), a traditional Chinese medicine, is widely used for treatment of various neuro dysfunctions. However, its quality control is still limited to the determination of gastrodin. In the present study, two novel strategies based on quantitative evaluation of total gastrodin and gastrodigenin with base hydrolysis and total gastrodigenin with base-enzymatic hydrolysis followed by HPLC-FLD were put forward and successfully applied to evaluate the quality of 47 batches of G. elata from eight localities. Meanwhile, a systematic comparison of the novel strategy with the multiple markers and the Pharmacopeia method was performed. The results showed that the parishins category could be completely hydrolyzed to gastrodin by sodium hydroxide solution, and gastrodin could further utterly hydrolyze to gastrodigenin with β-d-glucosidase buffer solution. The contents of total gastrodin and gastrodigenin ranged from 1.311% to 2.034%, and total gastrodigenin from 0.748% to 1.120% at the eight localities. From the comparison, we can conclude that the two novel strategies can comprehensively reveal the characteristics of overall active ingredients in G. elata for quality control. The present study provides a feasible and credible strategy for the quality control of G. elata, suggesting a revision of the latest Chinese Pharmacopoeia or European Pharmacopoeia methods for the modernization of G. elata use.
