Abstract:The objective of this study was to evaluate the performance of the UV disinfection combined with chlorination in the inactivation of microorganisms and the formation of trihalomethanes (THMs). Experimental results showed that UV irradiation had obvious advantages in reducing the number of microbial species and community complexity. Under the same initial chlorine concentration, the amount of THMs increased with the UV radiation dose increasing. THMs will be reduced in the UV-chlorine disinfection process compared with the pure chlorine disinfection. In terms of number of surviving microorganisms in water, compared with a single chlorine disinfection, UV irradiation showed a very good disinfection effect. With an average 52.5% decline, the data reflected the sample after UV irradiation in microbial diversity and abundance significantly decreased. the physical and chemical properties of organic matter could be qualitatively and quantitatively described by 3DEEM.

Abstract:Reclamation and re-use of wastewater is one of the most effective ways to alleviate the shortage of water resources, while the safety of reclaimed water becomes one of the critical problems for protecting human health and the ecosystem. While a toxicity test can vividly reflect biological effects of chemicals as a whole, in this study, the Microtox test was used to screen toxicity changes of wastewater during conventional reclamation processes. The results showed that toxicities of water samples decreased continuously along reclamation processes except chlorination/dechlorination in which the toxicity increased significantly. Furthermore, as for different forms of residual chlorine, toxicity of wastewater was quite different with increasing chlorine dosage. NH3-N had a trend to decrease toxicity of disinfected wastewater, while UV254 had a trend to increase toxicity. It was found that there was a good linear relationship between toxicity formation and UV254/NH3-N ratio for wastewater after disinfection with combined chlorine form before peak-point.

Abstract:Chlorination is widely used in wastewater reclamation, however harmful disinfection byproducts (DBPs) may be formed during disinfection. These DBPs are considered as a potential and important source of endocrine-disruption. In this study, the effects of chlorination on estrogenic and antiestrogenic activities in biologically treated wastewater were evaluated by yeast two-hybrid assay. For the first time, chlorination was found to increase the antiestrogenic activity of wastewater notably and decrease the estrogenic activity. By fractionating dissolved organic matter (DOM) in wastewater into different fractions, it was found that the polar compounds (PC) fraction of DOM was the key fraction involved in increasing antiestrogenic activity during chlorination of wastewater. Furthermore, fluorescence spectroscopy analysis on different fractions of soluble organic compounds in wastewater suggested that the PC fraction contained most of the aromatic amino acids and humic/fulvic acid, which were then demonstrated as the precursors of antiestrogenic DBPs through chlorination experiments of tryptophan, humic acid, and tannic acid.
Influence of chlorine disinfection on the concentration of assimilable organic carbon in reclaimed water

Abstract:Chlorine consumption characteristics of reclaimed water and the changes in water quality characteristics during chlorine disinfection process were investigated.The consumption of chlorine was quick in the first five minutes after chlorination,meanwhile the UV absorbance and three-dimensional fluorescence spectra also changed significantly.It was found that the concentration of assimilable organic carbon (AOC) also increased significantly after the disinfection of the reclaimed water,which indicates that the biological stability of the reclaimed water was deteriorated.AOC of recycled water samples in the disinfection process decreaded after the initial increase.The effluents of the advanced treatment processes show lower AOC levels,but higher AOC growth rate than the secondary treatment effluent water.Furthermore,it was found that the increase in AOC was correlated to the decrease in the threedimensional fluorescence integral value after chlorination.

Abstract:Bio-toxicity of disinfected wastewater with chlorination and dechlorination was studied using photobacteria,and the effect of residual chlorine forms,ammonia nitrogen concentration and organic matters content on bio-toxicity in disinfected wastewater was also investigated.The result shows that bio-toxicity in disinfected wastewater increases significantly.When residual chlorine is in combined form,bio-toxicity in wastewater increases along with the increased chlorine dosage,and photobacteria inhibition rate is linear with the logarithm of chlorine dosage;when residual chlorine is in the free form,the peak toxicity occurs with the variation in chlorine dosage,it is followed by the slight reduction of toxicity after increase of chlorine dosage.Ammonia nitrogen shows a tendency to reduce the toxicity in disinfected wastewater,while organic matter shows a tendency to increase the toxicity.

Abstract:By selecting sodium sulfite as dechlorinating agent, photobacteria method was used to investigate the effect of dechlorination on reduction of biotoxication incurred by residual chlorine. It was found that the biotoxication of wastewater after chlorine disinfection is increased remarkably, which mainly results from the disinfection byproducts. Addition of dechlorinating agent can reduce or eliminate the acute toxicity caused by disinfection byproducts. It may be because that the reaction of dechlorinating agent with part of byproduct results in the formation of materials with less toxicity or without toxicity.

Abstract:Reclamation and reuse of wastewater is one of the most effective ways to alleviate water shortage. Disinfection plays a key role in killing the harmful pathogens in reclaimed water, while an unwanted side effect is the formation of disinfection by-products (DBPs). Recently, a number of researches have been conducted on the formation regularities of certain DBPs. However, with current physiochemical techniques, it is impossible to detect all the DBPs. In this study, photobacterium bioassay was used to measure the formation of DBPs and their toxic effect as a whole. The effects of water quality characteristics and operational conditions on the toxicity formation during wastewater chlorination disinfection process were evaluated. A statistical model, depending on chlorine disinfectant dosage, concentration of ammonia nitrogen, and concentration of dissolved organic carbon, was developed to quantitatively estimate the toxicity formation during the disinfection process. It was found that the toxicity of the wastewater samples was positively correlated with chlorine disinfectant dosage, concentration of dissolved organic carbon and UV absorbance at 254nm, while negatively correlated with concentration of ammonia nitrogen.

Abstract:In this work, reclaimed water treated with sodium hypochlorite (NaClO) or chlorine dioxide (ClO2) at 1, 2, and 4 mg/L was operated successively for 30 days respectively, in annular reactors with new cast iron coupons, corresponding to stages I (days 0-30), II (days 31-60), and III (days 61-90). The Illumina HiSeq 2500 sequencing platform was used to analyze the bacterial community composition, scanning electron microscopy and X-ray diffraction analyses were conducted to characterize corrosion scales, and the weight loss method was served to determine the general corrosion rate. Results reveal the precise disinfection effect on biofilm bacteria to be dose dependent and species specific. In stage 1, disinfection caused a reduction in the number of operational taxonomic units, but, had little effect on biofilm composition. In stage II, NaClO and ClO2 induced a reduction of Proteobacteria proportion, but increased the dominance of Firmicutes; the diminished Proteobacteria in NaClO test mainly included Gammaproteobacteria, while, that in ClO2 test mainly included the Gammaproteobacteria and Betaproteobacteria. In stage III, Firmicutes presented a certain resistance to NaClO and ClO2 as the accumulation of corrosion scales. Results also indicated that disinfection enhanced the corrosion process, and the promoting effect of ClO2 was more pronounced than that of NaClO. Moreover, this promoting effect was more obvious in stage I than that in the latter two stages. The strong oxidization effect associated with disinfection in stage I was the dominant factor promoting corrosion, whereas, the bacterial community also played a crucial role in stages II and III. Abstract:Disinfection of reclaimed water prior to reuse is important to prevent the transmission of pathogens. Chlorine is a widely utilized disinfectant and as such is a leading contender for disinfection of reclaimed water. To understand the risks of chlorination resulting from the potential selection of pathogenic bacteria, the inactivation, reactivation and regrowth rates of indigenous bacteria were investigated in reclaimed water after chlorine disinfection. Inactivation of total coliforms, Enterococcus and Salmonella showed linear correlations, with constants of 0.1384, 0.1624 and 0.057 L/(mg·min) and R2 of 0.7617, 0.8316 and 0.845, respectively. However, inactivation of total viable cells by measurement of metabolic activity typically showed a linear correlation at lower chlorine dose (0–22 (mg·min)/L), and a trailing region with chlorine dose increasing from 22 to 69 (mg·min)/L. Reactivation and regrowth of bacteria were most likely to occur after exposure to lower chlorine doses, and extents of reactivation decreased gradually with increasing chlorine dose. In contrast to total coliforms and Enterococcus, Salmonella had a high level of regrowth and reactivation, and still had 2% regrowth even after chlorination of 69 (mg·min)/L and 24 hr storage. The bacterial compositions were also significantly altered by chlorination and storage of reclaimed water, and the ratio of Salmonella was significantly increased from 0.001% to 0.045% after chlorination of 69 (mg·min)/L and 24 hr storage. These trends indicated that chlorination contributes to the selection of chlorine-resistant pathogenic bacteria, and regrowth of pathogenic bacteria after chlorination in reclaimed water with a long retention time could threaten public health security during wastewater reuse.
Effect of Wastewater Characteristics on Acute Toxicity Incurred by Disinfection

Abstract:Photo-bacteria bioassay was used to study the effect of wastewater characteristics on acute toxicity of disinfected wastewater during chlorination/dechlorination. It is found that consideration shall be given respectively to different chlorine addition method to make accurate and objective evaluation. As the characteristic index for expressing the precursors of toxic by-products, UV_(254) is superior to DOC, and DOC is superior to COD; UV_(254) is in good correlation with the quantity of precursors of toxic by-products. Under the standard disinfection conditions, i. e, temperature of 20 ℃, disinfection time of 30 min, and pH of 7.0, when the dosage of available chlorine is 10 mg/L and 50 mg/L, the acute toxicity increment (expressed as Zn~(2+) concentration) of disinfected wastewater can be predicted.

Abstract:The effects of chlorine and chlorine dioxide disinfections on quality and toxicity of wastewater were compared.The experiment results showed that chlorine disinfection had no obvious effect on wastewater color,while chlorine dioxide disinfection decreased wastewater color observably.The DOC of wastewater did not change much after chlorine and chlorine dioxide disinfections.Chlorine disinfection significantly increased UV_(230) of wastewater and chlorine dioxide disinfection slightly decreased UV_(230) of wastewater.When the disinfectants dosage was 30mg/L,UV_(230) increased about 0.7cm~(-1) after chlorine disinfection and decreased about 0.05cm~(-1) after chlorine dioxide disinfection.The acute toxicity of wastewater increased with increasing disinfectants dosage for both chlorine and chlorine dioxide disinfections and the acute toxicity after chlorine disinfection is much stronger than that after chlorine dioxide disinfection.The genotoxicity of wastewater increased slightly after chlorine disinfection and decreased slightly after chlorine dioxide disinfection.
Synergistic effects of UV and chlorine in bacterial inactivation for sustainable water reclamation and reuse

Abstract:Disinfection is a necessity in water and wastewater treatment and reclamation. This study examined the inactivation of a disinfectant resistant but widely existed opportunistic pathogen in reclaimed water, Staphylococcus aureus (S. aureus), by sequential UV and chlorine disinfection or simultaneous UV and chlorine disinfection (UV/Cl). It was identified that UV/Cl greatly promoted the inactivation efficacy and inhibited photoreactivation of S. aureus by the generation of free radicals (i.e. OH and Cl), which reached a 7-log10 reduction at UV and chlorine doses of 18 mJ/cm2 and 2 mg-Cl/L, respectively. The changes of bacterial viability and morphology and the increase of extracellular ATP concentration confirmed the enhancement of cell membranes damages (>21.4 %) due to free radicals generated in UV/Cl process, which caused a dramatic reduction in metabolic activity and suppressed the photoreactivation. Furthermore, this study demonstrated that UV/Cl effectively removed heterotrophic plate count bacteria and aromatic organic fluorophores in reclaimed water samples. This study is of significant theoretical and applicable importance in guaranteeing safe microbial levels for water reclamation and reuse.

Abstract:Disinfection processes were applied in reclaimed water plant to eliminate pathogens and control the related health risk during water reuse. However, extra problems might emerge such as the released free endotoxins from the ruptured cell wall of gram-negative bacteria. Endotoxins can induce lung inflammatory responses after inhalation, which has been neglected in the water quality regulation, and the removal of endotoxin was not under consideration in the process of reclamation. In the present study, two well-known disinfection processes, chlorination and combined UV/chlorine (UV/Cl2), were performed to test the removal efficiency of endotoxin activity, as well as the inflammation inducing ability. In the pure LPS solution, UV/Cl2 treatment significantly reduced both endotoxin activity and lung inflammation responses with better oxidizability of the generated hydroxyl radical. However, its performance on bacteria liquid and real secondary effluent was more complicated. The cell wall-bound LPS have lower endotoxin activities and inflammation inducing ability. Immediately after the cell wall was destroyed, the bound LPS were released to the solution to be free LPS, which dramatically increased both the endotoxin activity and inflammation inducing ability of the water. When these free endotoxins were continuously oxidized, the endotoxin activity and inflammatory response decreased again but not to the background level. Therefore, the inflammation inducing ability of reclaimed water could not be removed efficiently. These results suggest that in spite of its high oxidability, UV/Cl2 treatment is not capable of removing the endotoxin-based toxicity, and other technologies are necessary to control endotoxin levels in reclaimed water.
Effects of operating conditions on THMs and HAAs formation during wastewater chlorination

Abstract:Disinfection is the last barrier of wastewater reclamation process to protect ecosystem safety and human health. However, the chlorination process results in the formation of mutagenic/carcinogenic disinfection by-products (DBPs) deriving from the reaction of the chlorine with organic compounds in wastewater. The effects of operating conditions (chlorine dose, contact time, reaction temperature and pH value) of chlorination on the formation of trihalomethanes (THMs) and haloacetic acids (HAAs) in biologically treated wastewater samples were investigated in this study. The results indicated that the total THMs (TTHM) and total HAAs (THAA) increased exponentially with increasing chlorine dose, but there are discrepancies between the formation rates of TTHM and THAA. The THAA reached a peak at contact time of 2h and thereafter decreased with extended time. The formation time of THMs depends on the wastewater content of quick or slow formers. The yields of bromated HAAs (as MBAA, BCAA, and BDCAA) would decrease markedly after the contact time over 2h during wastewater chlorination, and were favored in low pH values of 4 and high pH values of 9 under certain contact time. In addition, the formation of MBAA, BCAA, BDCAA decreased gradually as reaction temperature increased from 4 to 30 degrees C in the chlorination of wastewater containing a certain concentration of bromide. The effects of operating conditions on THMs and HAAs formation during wastewater chlorination were completely different from those of surface water disinfection.

Abstract:The effects of chlorine dioxide dosage and reaction time on the acute toxicity and genotoxicity of wastewater after chlorine dioxide disinfection were studied using photobacterium bioassay and umu test. The experiment results showed that with the increase in chlorine dioxide dosage, the acute toxicity increased, but the genotoxicity decreased and then became stable. With the increase in reaction time, the acute toxicity increased to a peak and then decreased, but the genotoxicity decreased and then tended to be stable. Because the effects of disinfection conditions on wastewater acute toxicity and genotoxicity are quite different, it was considered that the substance contained in the wastewater with acute toxicity and the substance with genotoxicity were different, and there was a range of disinfection conditions to make both the acute toxicity and the genotoxicity low.

Abstract:Urban wastewater contains a wide range of pathogens and antibiotic resistance genes (ARGs), which are a serious concern if reusing treated wastewater. However, few studies have explored the microbial communities in reclaimed water using ultraviolet (UV)/chlorine treatment and assessed the changes of the resistome. This study investigated the occurrence of typical pathogens, ARGs, and bacterial communities in UV/chlorine-treated reclaimed water samples. The numbers of culturable and viable but non-culturable pathogens were effectively reduced to 0 CFU/mL within 1–10 and 10–30 min after UV/chlorine treatment, respectively. Meanwhile, the physicochemical indices of water quality were not affected. UV/chlorine treatment could significantly change the bacterial community structure of reclaimed water, showing a decrease in bacterial abundance and diversity. Chlorine-resistant Acinetobacter and Mycobacterium were the dominant bacterial genera (> 50%) after UV/chlorine treatment. Moreover, the number of ARGs and mobile genetic elements (MGEs) decreased with an increase in UV/chlorine exposure. However, eight ARGs and three MGEs were consistently detected in more than three seasons, making these major concerns because of their potential role in the persistence and dissemination of antibiotic resistance. Overall, the results of this study suggest that UV/chlorine treatment can potentially improve the microbiological safety of reclaimed water. And more attention should be paid to the pathogens that are both chlorine-resistant and carry MGEs because of their potential for resistance transmission.

Abstract:Several kinds of reclaimed water were used to study the effect of residual chlorine on water bloom alga Microcystis aeruginosa.Results showed that,residual chlorine had strong inhibition effect on Microcystis aeruginosa with an inhibition rate above 60%—80% on the 2nd day and 99% on the 8th day,and the inhibition effect could remain for 60 days.The concentration of ammonia nitrogen in water lowered the inhibition effect.The inhibition rate of residual chlorine on Microcystis aeruginosa decreased when the ammonia nitrogen increased.The inhibition effect of combined chlorine(NH2Cl) is weaker than that of free chlorine(HOCl).

Abstract:Chlorinated reclaimed water is widely used for landscaping and recreational purposes, resulting in human exposure to toxic disinfection byproducts. Although the quality of chlorinated reclaimed water might be affected by sunlight during storage, the effects of solar light irradiation on the toxicity remain unknown. This study investigated the changes in cytotoxicity and total organic halogen (TOX) of chlorinated reclaimed water exposed to solar light. Irradiation with solar light for 12 h was found to significantly reduce the cytotoxicity of chlorinated reclaimed water by about 75%, with ultraviolet light being responsible for the majority of this reduction. Chlorine residual in reclaimed water tended to increase the cytotoxicity, and the synergy between solar light and free chlorine could not enhance the reduction of cytotoxicity. Adding hydroxyl radical scavengers revealed that the contribution of hydroxyl radical to cytotoxicity reduction was limited. Solar light irradiation concurrently reduced TOX. The low molecular weight (<1 kDa) fraction was the major contributor of cytotoxicity and TOX in chlorinated reclaimed water. Detoxification of the low molecular weight fraction by light irradiation was mainly a result of TOX dehalogenation, while detoxification of the high molecular weight (>1 kDa) fraction was probably caused by photoconversion from high toxic TOX to low toxic TOX.
