Abstract:Underwater environment observation with underwater acoustic tomography has been considerably developed in recent years. Moving sound transmission can obtain the observation of entire spatial area with sound station moving. Various internal structures, unique surface and submarine boundaries and changing environment constitutes a complex acoustic propagation channel. This paper focus on the inversion method and signal resampling for sound moving transmission. Also, the current field in three-dimensional (3D) scale is also studied. A five-station sound transmission experiment with four moored station and one moving station that conducted in range of 500m×500m at Huangcai reservoir, Changsha, China is presented. Signal resampling is performed to get correlation of received acoustic data. The vertical layer-averaged flow current results between moving station and moored station are inversed with 2D grid method. 3D flow current field result is composed by grid-averaged inversion current of vertical profile via moving station at different moment. The received results of reciprocal signal transmission between two moored stations and one moving station were used for layer-averaged current variations at vertical scale and grid-averaged current. The feasibility of the method in underwater moving acoustic tomography research is proved and its applicability is discussed. The proposed underwater acoustic tomography technology develops an innovative idea for the further development of temporal- spatial grided tomography observation.

Abstract:Optical process tomography is the developing tomography technology,which is usually used in multiphase flow parameter measurement.A novel infrared laser process tomography prototype system is developed for two-phase flow parameter measurement.The construction,algorithm,performance and characteristics of the system are investigated.The simulation experiment results indicate that optical process tomography technology can get high image reconstruction speed and resolution,and has the development and application potential for multiphase flow parameter measurement.
Transverse flow imaging based on photoacoustic Doppler bandwidth broadening

Abstract:We propose a new method to measure transverse flow velocity based on photoacoustic Doppler bandwidth broadening, which is determined by the geometry of the probe-beam and the velocity of the transverse flow. By exploiting pulsed laser excitation and raster motor scanning, three-dimensional structure and flow velocity can be imaged simultaneously. In addition, the flow direction can be determined with bidirectional scanning. In a flowing suspension of red-dyed microspheres (diameter: 6 microm), transverse flow speeds ranging from 0 to 2.5 mms as well as flow direction were measured. A cross-sectional flow image was also obtained with the tube laid in a zigzag pattern.

Abstract:We present an optically encoded photoacoustic (PA) flow imaging method based on optical-resolution PA microscopy. An intensity-modulated continuous-wave laser photothermally encodes the flowing medium, and a pulsed laser generates PA waves to image the encoded heat pattern. Flow speeds can be calculated by cross correlation. The method was validated in phantoms at flow speeds ranging from 0.23 to 11 mm/s. Venous blood flow speed in a mouse ear was also measured.

Abstract:We present a technique called photoacoustic vector-flow (PAVF) to quantify the speed and direction of flowing optical absorbers at each pixel in an acoustic-resolution photoacoustic image. By varying the receiving angle at each pixel in post-processing, we obtain multiple estimates of the phase difference between consecutive frames. These are used to solve the overdetermined photoacoustic Doppler equation with a least-squares approach to estimate a velocity vector at each pixel. This technique is tested in bench-top experiments and compared to simultaneous pulse-echo ultrasound vector-flow (USVF) on whole rat blood at speeds on the order of 1 mm/s. Unlike USVF, PAVF can detect flow without stationary clutter filtering in this experiment, although the velocity estimates are highly underestimated. When applying spatio-temporal singular value decomposition clutter filtering, the flow speed can be accurately estimated with an error of 17% for USVF and 9% for PAVF for an average flow speed of 2.5 mm/s.

In vivo photoacoustic flowmetry at depths of the diffusive regime based on saline injection

Abstract:We propose a saline injection-based method to quantify blood flow velocity in vivo with acoustic-resolution photoacoustic tomography. By monitoring the saline–blood interface propagating in the blood vessel, the flow velocity can be resolved. We first demonstrated our method in phantom experiments, where a root mean square error of prediction of 0.29 mm/s was achieved. By injecting saline into a mouse tail vein covered with 1 mm chicken tissue, we showed that the flow velocity in the tail vein could be measured at depths, which is especially pertinent to monitoring blood flow velocity in patients undergoing intravenous infusion.

Abstract:We recently proposed photoacoustic correlation spectroscopy (PACS) and demonstrated a proof-of-concept experiment. Here we use the technique for in vivo flow speed measurement in capillaries in a chick embryo model. The photoacoustic microscopy system is used to render high spatial resolution and high sensitivity, enabling sufficient signals from single red blood cells. The probe beam size is calibrated by a blood-mimicking phantom. The results indicate the feasibility of using PACS to study flow speeds in capillaries.

Abstract:Non-invasive imaging of deep blood vessels for mapping hemodynamics remains an open quest in biomedical optical imaging. Although pure optical imaging techniques offer rich optical contrast of blood and have been reported to measure blood flow, they are generally limited to surface imaging within the optical diffusion limit of about one millimeter. Herein, we present photoacoustic vector tomography (PAVT), breaking through the optical diffusion limit to image deep blood flow with speed and direction quantification. PAVT synergizes the spatial heterogeneity of blood and the photoacoustic contrast; it compiles successive single-shot, wide-field photoacoustic images to directly visualize the frame-to-frame propagation of the blood with pixel-wise flow velocity estimation. We demonstrated in vivo that PAVT allows hemodynamic quantification of deep blood vessels at five times the optical diffusion limit (more than five millimeters), leading to vector mapping of blood flow in humans. By offering the capability for deep hemodynamic imaging with optical contrast, PAVT may become a powerful tool for monitoring and diagnosing vascular diseases and mapping circulatory system function.

Abstract:From the flow of a suspension of micrometer-scale carbon particles, the photoacoustic Doppler shift is observed. As predicted theoretically, the observed Doppler shift equals half of that in Doppler ultrasound and does not depend on the direction of laser illumination. This new physical phenomenon provides a basis for developing photoacoustic Doppler flowmetry, which can potentially be used for detecting fluid flow in optically scattering media and especially low-speed blood flow of relatively deep microcirculation in biological tissue.

Abstract:We present a pressure sensor based on a Michelson interferometer, for use in photoacoustic tomography. Quadrature phase detection is employed allowing measurement at any point on the mirror surface without having to retune the interferometer, as is typically required by Fabry-Perot type detectors. This opens the door to rapid full surface detection, which is necessary for clinical applications. Theory relating acoustic pressure to detected acoustic particle displacements is used to calculate the detector sensitivity, which is validated with measurement. Proof-of-concept tomographic images of blood vessel phantoms have been taken with sub-millimeter resolution at depths of several millimeters.

Abstract:We report on experimental demonstration of photoacoustic tomography for simultaneous recovery of the optical and acoustic properties of heterogeneous media. Photoacoustic images are reconstructed from a series of tissue-like phantom experiments using a finite element-based reconstruction algorithm coupled with a scanning photoacoustic imaging system. The results show that the images obtained are quantitative in terms of the shape, size, location and optical and acoustic properties of the heterogeneities examined.

Abstract:Synthetic aperture imaging and virtual point detection have been exploited to extend the depth of view of photoacoustic microscopy. The approach is commonly based on a constant assumed sound speed, which reduces image quality. We propose a new, to the best of our knowledge, self-adaptive technique to estimate the speed of sound when integrated with this hybrid strategy. It is accomplished through linear regression between the square of time of flight detected at individual virtual detectors and the square of their horizontal distances on the focal plane. The imaging results show our proposed method can significantly improve the lateral resolution, imaging intensity, and spatial precision for inhomogeneous tissue.

Abstract:Imaging deep haemodynamics non-invasively remains a quest. Although optical imaging techniques can be used to measure blood flow, they are generally limited to imaging within ∼1 mm below the skin's surface. Here we show that such optical diffusion limit can be broken through by leveraging the spatial heterogeneity of blood and its photoacoustic contrast. Specifically, successive single-shot wide-field photoacoustic images of blood vessels can be used to visualize the frame-to-frame propagation of blood and to estimate blood flow speed and direction pixel-wise. The method, which we named photoacoustic vector tomography (PAVT), allows for the quantification of haemodynamics in veins more than 5 mm deep, as we show for regions in the hands and arms of healthy volunteers. PAVT may offer advantages for the diagnosis and monitoring of vascular diseases and for the mapping of the function of the circulatory system. Successive single-shot wide-field photoacoustic images can be used to estimate the speed and direction of blood flow at more than 5 mm deep from the skin's surface.

Abstract:Speed-of-sound and optical absorption reflect the structure and function of tissues from different aspects. A dual-mode microscopy system based on a concentric annular ultrasound array is proposed to simultaneously acquire the long depth-of-field images of speed-of-sound and optical absorption of inhomogeneous samples. First, speed-of-sound is decoded from the signal delay between each element of the annular array. The measured speed-of-sound could not only be used as an image contrast, but also improve the resolution and accuracy of spatial location of photoacoustic image in inhomogeneous acoustic media. Secondly, benefitting from dynamic focusing of annular array and the measured speed-of-sound, it is achieved an advanced acoustic-resolution photoacoustic microscopy with a precise position and a long depth-of-field. The performance of the dual-mode imaging system has been experimentally examined by using a custom-made annular array. The proposed dual-mode microscopy might have the significances in monitoring the biological physiological and pathological processes. (C) 2017 Optical Society of America

Abstract:Photoacoustic (optoacoustic) tomography, combining optical absorption contrast and highly scalable spatial resolution (from micrometer optical resolution to millimeter acoustic resolution), has broken through the fundamental penetration limit of optical ballistic imaging modalities-including confocal microscopy, two-photon microscopy, and optical coherence tomography-and has achieved high spatial resolution at depths down to the diffusive regime. Optical absorption contrast is highly desirable for microvascular imaging and characterization because of the presence of endogenous strongly light-absorbing hemoglobin. We focus on the current state of microvascular imaging and characterization based on photoacoustics. We first review the three major embodiments of photoacoustic tomography: microscopy, computed tomography, and endoscopy. We then discuss the methods used to characterize important functional parameters, such as total hemoglobin concentration, hemoglobin oxygen saturation, and blood flow. Next, we highlight a few representative applications in microvascular-related physiological and pathophysiological research, including hemodynamic monitoring, chronic imaging, tumor-vascular interaction, and neurovascular coupling. Finally, several potential technical advances toward clinical applications are suggested, and a few technical challenges in contrast enhancement and fluence compensation are summarized.

Abstract:The field of photoacoustic tomography has experienced considerable growth in the past few years. Although several commercially available pure optical imaging modalities, including confocal microscopy, two-photon microscopy, and optical coherence tomography, have been highly successful, none of these technologies can provide penetration beyond $\sim$1 mm into scattering biological tissues, because they are based on ballistic and quasi-ballistic photons. Heretofore, there has been a void in high-resolution optical imaging beyond this penetration limit. Photoacoustic tomography, which combines high ultrasonic resolution and strong optical contrast in a single modality, has broken through this limitation and filled this void. In this paper, the fundamentals of photoacoustics are first introduced. Then, scanning photoacoustic microscopy and reconstruction-based photoacoustic tomography (or photoacoustic computed tomography) are covered.

Abstract:We analyze photo-acoustic signals from capillaries and theoretically demonstrate the quantitative relationship between vascular diameter and spectral slope in a low-frequency band. Phantom experiments validate the theoretical analysis. Based on this finding, spectral slope is proposed as the imaging parameter of a photo-acoustic microscopy. This system effectively quantifies the microvasculature with diameters of 60 and 150 mu m, which are smaller than the wavelength 342 mu m at the central frequency 4.39 MHz of ultrasound transducer. The low frequency also guarantees the imaging depth in the order of centimeters. The proposed scheme could be potential for noninvasive diagnosis of diseases related to abnormal vasoconstriction or angiectasis. (C) 2015 Optical Society of America

Abstract:Photoacoustic tomography is a promising and rapidly developed methodology of biomedical imaging. It confronts an increasing urgent problem to reconstruct the image from weak and noisy photoacoustic signals, owing to its high benefit in extending the imaging depth and decreasing the dose of laser exposure. Based on the time-domain characteristics of photoacoustic signals, a pulse decomposition algorithm is proposed to reconstruct a photoacoustic image from signals with low signal-to-noise ratio. In this method, a photoacoustic signal is decomposed as the weighted summation of a set of pulses in the time-domain. Images are reconstructed from the weight factors, which are directly related to the optical absorption coefficient. Both simulation and experiment are conducted to test the performance of the method. Numerical simulations show that when the signal-to-noise ratio is -4 dB, the proposed method decreases the reconstruction error to about 17%, in comparison with the conventional back-projection method. Moreover, it can produce acceptable images even when the signal-to-noise ratio is decreased to -10 dB. Experiments show that, when the laser influence level is low, the proposed method achieves a relatively clean image of a hair phantom with some well preserved pattern details. The proposed method demonstrates imaging potential of photoacoustic tomography in expanding applications.
Simultaneous Scattering-Absorption Dual-Modal Cell Imaging in a Single Shot by a Transmission-Mode Photoacoustic Microscope.

Abstract:A microscopy scheme is proposed to simultaneously achieve optical scattering-absorption dual-contrast imaging of a transparent or semi-transparent specimen. This scheme is based on a transmission-mode photoacoustic microscope. We find that two peaks exist in the detected photoacoustic signal. One peak is caused by the optical absorption of the specimen, and the other is related to both the optical scattering and absorption of the specimen. Therefore, both the absorption and scattering information can be simultaneously extracted by analyzing the same photoacoustic signal excited by a single-shot laser pulse. After the microscope is validated by imaging a binary mixture consisting of particles with different optical properties, it successfully acquires dual images of red blood cells with different contrasts. Quantitative analysis reveals that the optical absorption and scattering properties of the specimen can be derived from the two images. The proposed dual-modal imaging method would be useful in revealing the structural and functional properties of tissues at the cell level or the clinical assessment of pathological sections.
