Effects of three defatting ways on porcine bone

Abstract:Background: Defatting of xenogenic bone is the utmost process of bone transplantation. Supercritical CO 2 fluid, chloroform-methanol, ethylether are the common methods of bone defatting. Objective: To compare the effects of supercritical CO 2 fluid, chloroform-methanol, and ethylether on porcine bone defatting, and to explore the optimal method of delipidation. Design, Time and Setting: A randomized controlled study was performed at Key Laboratory of Transplantation Immunity, Sichuan University from September 2006 to April 2007. Materials: Bone was derived from a 6-month-old pig. The soft tissues and periosteum were removed, and the bone samples were cut into sections of 5 mm × 5 mm × 5 mm, 15 sections in total. Methods: Fifteen sections were randomly divided into three groups: supercritical CO 2 fluid, chloroform-methanol, and ethylether, with 5 sections in each group. Bone sections were weighted before defatting. The bone samples in the supercritical CO 2 fluid group were defatted using supercritical CO 2 fluid, with flowing rate of 16-18 kg/h and pressure of 8 MPa, at 50 °C for 5 hours. Bone samples in the chloroform-methanol group were defatted using chloroform-methanol (1:1) at 37 °C for 24 hours. Bone samples in the ethylether group were defatted with ethylether at 37 °C for 24 hours. Main Outcome Measures: Defatted results with the mass loss percentage using infrared spectrum; determination of protein content using Kjeldahl method. Results: The defatted effect of supercritical CO 2 fluid was similar to chloroform-methanol, but both of them were superior to ethylether. Conclusion: As a non-pollution and anoxic defatting method, supercritical CO 2 fluid has a wide prospect in bone defatting.
