Determination and Investigation of Total Alkaloids and 1-Deoxynojirimycin in Folium Mori

Abstract:OBJECTIVE To establish a method for the determination of the total alkaloids and 1-deoxynojirimycin(DNJ) in Folium Mori and compare the content of alkaloids and DNJ in Folium Mori with different collection time,species,plant site and medicinal part.METHODS The content of alkaloids was measured by Spectrophotometry based on the reaction of alkaloid with Remecke salt and the contents of DNJ was determined by HPLC-ELSD.The HPLC analysis was performed on Agilent ZORBAX NH2 column(4.6 mm×250 mm,5 μm),the mobile phase was MeCN-H2O(80:20),the flow rate was 1.0 mL·min-1,the temperature of column was 30 ℃ and the injection volumn was 20 μL.The ELSD condition were as follows:the temperature of drift tube was 55 ℃,and the gas pressure was 0.15 MPa,and the value of gain was 7.0.RESULTS The methods developed were accurate and repeatable.The contents of total alkaloids and DNJ varied obviously among Folium Mori from different collection time and medicinal part,while no significant difference was found in Folium Mori from different plant sites.CONCLUSION The contents of total alkaloids and DNJ correlated with collection season,plant site and medicinal part,and collection season,species and medicinal part were the major factors.
