Abstract:A new type of gelatin nanofiber has been fabricated by electrospinning using water as the solvent. By using a 33% aqueous solution of gelatin at 40 ℃, gelatin nanofiber mats having a uniform porous structure with fiber diameters in the range 120-210 nm were formed. After crosslinking with an aqueous ethanol solution of 1-ethyl-3-(3-dimethylaminopropyl) carbodiimide/N-hydroxysuccinimide sodium salt (EDC/NHS), the morphology of the fibers in the gelatin nanofiber mats was retained. Crosslinking, led to a decrease in enthalpy but enhanced thermal resistance properties and mechanical strength. Proliferation experiments with periodontal ligament cells (PDLCs) indicated that the cells effectively adhered to, and proliferated on, the nanofiber mats, indicating that the material has considerable potential for use in guided tissue regeneration (GTR) in stomatology.

Abstract:A nanofabrication method for the production of flexible core-shell structured silk elastin nanofibers is presented, based on an all-aqueous coaxial electrospinning process. In this process, silk fibroin (SF) and silk-elastin-like protein polymer (SELP), both in aqueous solution, with high and low viscosity, respectively, were used as the inner (core) and outer (shell) layers of the nanofibers. The electrospinnable SF core solution served as a spinning aid for the nonelectrospinnable SELP shell solution. Uniform nanofibers with average diameter from 301 ± 108 nm to 408 ± 150 nm were obtained through adjusting the processing parameters. The core-shell structures of the nanofibers were confirmed by fluorescence and electron microscopy. In order to modulate the mechanical properties and provide stability in water, the as-spun SF-SELP nanofiber mats were treated with methanol vapor to induce β-sheet physical crosslinks. FTIR confirmed the conversion of the secondary structure from a random coil to β-sheets after the methanol treatment. Tensile tests of SF-SELP core-shell structured nanofibers showed good flexibility with elongation at break of 5.20% ± 0.57%, compared with SF nanofibers with an elongation at break of 1.38% ± 0.22%. The SF-SELP core-shell structured nanofibers should provide useful options to explore in the field of biomaterials due to the improved flexibility of the fibrous mats and the presence of a dynamic SELP layer on the outer surface.
Research Progress on Gelling Mechanism and Modification of Silk Fibroin Hydrogels

Abstract:Hydrogel materials are of great value in medical field due to their similarity to human soft tissue.With silk fibroin as raw materials,organic solvent induction method can be employed to shorten the gelling time of silk fibroin to several hours.Silk fibroin/collagen composite hydrogel prepared with carbodiimide (EDC) as activation cross-linking agent has a storage modulus of up to 10 kPa.In vivo experiments indicate that silk fibroin hydrogel created by enzymatic crosslinking has good biocompatibility and biodegradability.We first explain the gelling mechanism of silk fibroin hydrogels,and then systematically introduce the research progress on preparation and modification methods of various silk fibroin hydrogels.Besides,potential applications of silk fibroin hydrogels as novel biomaterial in clinical applications are analyzed.Basic theoretical and technological issues to be solved for clinical applications of silk fibroin hydrogels are also discussed.

Abstract:In this work, the gelatin/zein nanofibers by hybrid electrospinning were fabricated. The morphology observations by field emission scanning electron microscopy showed that the diameter of gelatin/zein nanofibers was significantly increased with the increasing gelatin ratio. The results of Fourier transform infrared spectroscopy and differential scanning calorimetry indicated that gelatin and zein strongly interacted via hydrogen bonding. The homogeneous mixing resulted in the crystalline structure for the gelatin/zein nanofiber at a 1:1 wt ratio, which showed a hydrophobic surface (water contact angle of 118 similar to). The mechanical tests showed that the gelatin/zein nanofibers with weight ratios of 1:3 and 1:2 gave rise to much higher elongation at break of 87.9% and 69.0%, indicating good deformability and flexibility. In contrast to the pure gelatin or zein nanofibers, the gelatin/zein fibers preserved the 3D porous structures after immersed in water or ethanol for 24 h. (C) 2017 Elsevier Ltd. All rights reserved.
The fabrication and characterisation of surimi gel modified by konjac glucomannan with the different hydrolysis degree

Abstract:The gel performance of surimi-based products is indispensable technological attribute responsible for their high popularity. Here, surimi gels with tunable gel properties were successfully fabricated based on the double network theory. Konjac glucomannan (KGM) hydrolysed for various periods (0, 5, 15, 30, 60 and 90 min) was introduced into surimi systems. Corresponding structural characterisations including FTIR, XRD, SDS-PAGE and grafting degree assessment, were conducted to confirm the presence of double crosslinking reactions. Furthermore, the gel strength, dynamic rheology, thermal stability and in vitro digestibility of the surimi gels were investigated. Red shifts in the amide A band and an increase in relative intensity indicated that hydrogen bonding existed in the complex systems, according to the FTIR spectra. The XRD patterns suggested that the crystalline structures were weakened by interfering with the protein chain arrangement due to the Maillard reaction. In addition, SDS-PAGE and o-phthalaldehyde (OPA) assays also confirmed that myofibrillar protein and hydrolysed KGM were partially linked by covalent bonding. The SK-2 group showed the greatest mechanical strength and lowest digestibility amongst all groups due to its dense hierarchical structure. This work may provide inspiration for the design and development of surimi gels with improved gel properties via microarchitectural modulation.
Superhydrophobic/Hydrophobic Nanofibrous Network with Tunable Cell Adhesion: Fabrication, Characterization and Cellular Activities

Abstract:Superhydrophobic/hydrophobic nanofibrous networks have attracted much attention because of their potential applications in tissue engineering. Cell growth in the scaffold of tissue engineering can be controlled by the hydrophobicity of the scaffold. The superhydrophobic/hydrophobic surfaces are usually made from synthesized polymers, which generally are not biocompatible and biodegradable and, thus, not suitable for biomedical applications. Zein is an amphiphilic protein from corn, and it is potential for hydrophobic surface formation. This work aims to make zein superhydrophobic/hydrophobic nanofibrous network using electrospinning. The formed zein networks show high hydrophobicity with the water contact angles ranging from 130.5 +/- 1.0 degrees to 153.6 +/- 2.1 degrees. The cell attachment and growth on the zein networks are studied. It is observed that the amount of the cells attached and grown in the zein nanofibrous networks are higher than the ones on the conventional zein casting films. The results indicate that the electrospun zein nanofibrous network has great potential as scaffold in tissue engineering to support cell growth and tissue regeneration. (C) 2015 Elsevier B.V. Abstract:Electrospun zein membranes were prepared using DMF as solvent. By changing the solution concentration, the electrospinning voltage and the distance between the spinneret and collector, nanofibrous meshes without bead defects could be obtained. In order to improve the mechanical strength of the hydrated zein meshes, core-shell-structured nanofibrous membranes with PCL as the core material and zein forming the shell were prepared by coaxial electrospinning. The core-shell structure of the composite fibers was confirmed by SEM characterization of the fibers, either extracted with chloroform to remove the inner PCL, or elongated to expose their cross-section. The composition and average diameter of the composite fibers could be modulated by the feed rate of the inner PCL solution. It was found that the core-shell fibrous membranes have similar wettability to the electrospun zein mesh. The presence of PCL in the fibers could significantly improve the mechanical properties of the zein membrane.

Abstract:In this study, the gelatin, zein, and gelatin/zein films were fabricated by electrospinning and solvent casting, respectively. Scanning electron microscopy, differential scanning calorimetry, Fourier transform infrared spectroscopy and water contact angle measurements were performed to characterize the morphology, molecular interactions, thermal behavior, and surface hydrophilicity of the electrospun and casted films. Results showed that the zein particles prevented the gelatin chains from aggregating intra-molecularly in the casted films due to heterogeneous mixing, while strong hydrogen bonding was formed in the nanofibrous film as a result of homogeneous mixing. Interestingly, the gelatin/zein nanofibrous film had a hydrophobic surface (water contact angle of 118.0°), while the casted gelatin/zein film had a hydrophilic surface (water contact angle of 53.5°). The confocal laser scanning microscopy observations suggested that the zein particles were well dispersed in the gelatin network to maintain the nanofibrous structures after immersion in water or ethanol, leading to the improved solvent resistance.

Abstract:Gelatin (GA) is a natural protein widely used in food packaging, but its fabricated fibrous film has the defects of a high tendency to swell and inferior mechanical properties. In this work, a novel spinning technique, solution blow spinning (SBS), was used for the rapid fabrication of nanofiber materials; meanwhile, nylon 66 (PA66) was used to improve the mechanical properties and the ability to resist dissolution of gelatin films. Morphology observations show that GA/PA66 composite films had nano-diameter from 172.3 to 322.1 nm. Fourier transform infrared spectroscopy and X-ray indicate that GA and PA66 had strong interaction by hydrogen bonding. Mechanical tests show the elongation at break of the composite film increased substantially from 7.98% to 30.36%, and the tensile strength of the composite film increased from 0.03 MPa up to 1.42 MPa, which indicate that the composite films had the highest mechanical strength. Water vapor permeability analysis shows lower water vapor permeability of 9.93 g mm/m2 h kPa, indicates that GA/PA66 film's water vapor barrier performance was improved. Solvent resistance analysis indicates that PA66 could effectively improve the ability of GA to resist dissolution. This work indicates that SBS has great promise for rapid preparation of nanofibrous film for food packaging, and PA66 can be applied to the modification of gelatin film.

Abstract:A new method for preparing crosslinked fibrous gelatin membrane was developed in order to circumvent the disadvantages of traditional methods for crosslinking electrospun gelatin fibers. Through a series of experiments, the stable window of one-step electrospinning was found as 3 mL of 10% gelatin solution in TFE with 40 mu L glutaraldehyde added. SEM micrographs showed the diameter of the electrospun fibers ranged from 1 mu m to 1.5 mu m and was thicker than that of the noncrosslinked gelatin fibers. Furthermore, a phenomenon of false core-shell fibers was observed, which might be attributed to the rapid evaporation of glutaraldehyde at the vicinity of the fiber surface. Rheological behavior of the electrospinning solution was studied, indicating that viscosity had a drastic rise upon adding glutaraldehyde and maintained slow increase after 10 min. It was also found that a fibrous structure of the electrospun gelatin membrane could still be preserved after soaked in distilled water at 37 degrees C although the membrane suffered from about 10% weight loss. The one-step electrospinning method should be useful in the fields of biomedical engineering, such as tissue engineering and wound healing.
Study on Wettability, Mechanical Property and Biocompatibility of Electrospun Gelatin/zein Nanofibers Cross-Linked by Glucose

Abstract:In this study, the gelatin/zein nanofibers were electrospun and then cross-linked by glucose via Maillard reaction. The morphology observations showed that the average diameter of the cross-linked gelatin/zein nanofibers decreased with the increasing weight ratio of zein. FTIR results indicated that gelatin and zein molecules were homogenously dispersed within the fibers, and the Maillard reaction occurred between the proteins and glucose. The addition of zein affected the mobility of the gelatin chains, leading to the decreased denaturation temperature. The cross-linked fibers with a 1:1 gelatin/zein ratio showed the most hydrophobic surface (water contact angle of 134 degrees), and comparable mechanical properties to the cross-linked gelatin nanofibers, while a higher ratio of zein (more than 50%) resulted in the decreased elastic modulus. The L929 cell cytotoxicity test suggested that the cross-linked gelatin/zein nanofibers exhibited good biocompatibility and non-cytotoxicity.

Abstract:Electrospinning of natural polymer nanofibers useful for biomedical applications often requires the use of cytotoxic organic solvents. In this study, gelatin nanofibers are electrospun from phosphate buffer saline/ethanol binary mixtures as a benign solvent at ambient temperature. The influences of ionic strength, ethanol concentration, and gelatin concentration on the electrospinnability of gelatin solutions and the fiber microarchitectures are analyzed. The electrospun scaffolds retain their morphologies during vapor-phase crosslinking with glutaraldehyde in ethanol and the subsequent removal of salts contained in the nanofibers via water rinsing. When fully hydrated, the mechanically preconditioned scaffolds display a Young's modulus of 25.5 +/- 5.3 kPa, tensile strength of 55.5 +/- 13.9 kPa, deformability of 160 +/- 15%, and resilience of 89.9 +/- 1.8%. When cultured on the gelatin scaffolds, 3T3 fibroblasts displayed spindle-like morphology, similar to the cell's normal morphology in a 3D extracellular matrix. (c) 2012 Wiley Periodicals, Inc. Abstract:To elucidate the effect of fiber structure on the properties of the electrospun gelatin/PCL hybrid membranes, three types of fibers with different structures, i.e., core-shell, blend, and mixed fibers were fabricated. The crystallinity, wettability, swelling degree, and mechanical properties of the hybrid membranes were compared. It was found that the crystalline characteristics of PCL in the core-shell fibers were different from the fibers fabricated by the other two methods. That is, the orientation degree of the PCL chains in the core-shell fibers was higher than that in both blend and mixed fibers. The wettability of the hybrid membrane was dependent on both the composition and structure of the electrospun fibers. Blended fibers exhibited the highest hydrophobicity because of the enrichment of PCL at the fiber surface. Contrarily, the mixed fibers possessed the highest mechanical strength of 35.18 MPa. Characterization of glycosylated gelatin/pullulan nanofibers fabricated by multi-fluid mixing solution blow spinning

Abstract:In this work, multi-fluid mixing solution blow spinning was applied to develop gelatin/pullulan composite nanofibers, and then the nanofibers were glycated to enhance the physical properties. The results show that the grafting degree of the nanofibers increased significantly from 17.5 % to 36.0 % as the glycation time increased, and the morphology results indicated that 72 h of glycation did not destroy the structure of the nanofibers. FTIR results show that the glycation consumed the the–NH2 groups, cleaved sugar units of polysaccharide, and affected the secondary structure of the protein. The glycation enhanced the thermal stability and improved the rigidity of the nanofibers. Besides, after 120 h of glycation, the water contact angle of nanofibers increased from 0° to 79.1°, and the water vapor transmission rates decreased from 12.49 to 8.97 g mm/m2 h kPa, indicating the enhanced hydrophobicity and barrier properties. In addition, the glycation improved the water stability of the nanofibers, which increased the applicability of the gelatin/pullulan nanofibers in food packaging. The present work provides a green and efficient method for improving the physical properties of gelatin/pullulan nanofibers.

Abstract:In this paper, the Optimization Design and properties of polyvinyl alcohol (PVA)/gelatin (GEL) nanofibers were studied. Different mass fractions of PVA (4, 5, 6 and 7 wt%), GEL (1, 2, 3 and 4 wt%), different spinning distances (8, 15, 22 and 29 cm), different voltages (10, 17, 24 and 31 kV), and different spinning speeds (0.1, 0.2, 0.3 and 0.4 mL) were designed into five factors and four levels by orthogonal experiments. The conductivity and viscosity of the mixed solution was tested. The PVA/GEL nanofibers were prepared by electrospinning and the morphology of composite nanofibers was observed under electron microscope. Finally, the PVA/GEL nanofiber film was tested by infrared spectroscopy and X-ray diffraction. The results showed that the infrared spectral groups and X-ray diffraction angles of nanofibers with different PVA/GEL ratios have little change. PVA/GEL nanofiber materials have great application potential in the field of biomedicine.

Abstract:Despite considerable research effort, the natural hydrogels presently available for tissue engineering suffer from several major drawbacks, one of the significant issue is their poor mechanical strength which are unable to satisfy some mechanical requirements for successful outcomes. Herein, to mimic the composition and structure of the natural extracellular matrix, the micron-sized silk fibers obtained by alkaline hydrolysis were used as a reinforcement phase in a GelMA hydrogel, resulting in a material with significantly greater stiffness than pure GelMA hydrogel alone. In addition, the hydrogel demonstrated tunable compressive strength, swelling capacity, and degradation properties based on the silk fiber length. Experiments with cells indicated that MC3T3-E1 pre-osteoblasts quickly adhered to and proliferated on the surface of the composite hydrogels, as revealed by FDA/PI staining and CCK-8 assays. In addition, various cellular responses, including cell adhesion, changes in cellular morphology and cell proliferation behavior, occurred on the composite hydrogel and varied with fiber length. Overall, this study introduces a series of fiber-reinforced, tunable composite hydrogels that could be useful for various tissue engineering applications.

During forming the gel, the gelator molecules assembled into nano-sized fibrillary network driving by hydrogen bonds of urea groups. In cyclohexane, toluene and their mixed solvents, polymer nanofiber networks were successfully fabricated via Reversible Addition-Fragmentation Chain Transfer (RAFT) polymerization of the xerogels as both templates and monomer source. The solvent polarity was found to affect both the polymerization rate and the microstructures of polymer fibers. As more toluene was used in the mixed solvents, larger-sized fibers with improved surface roughness were obtained, together with the increased product yields. The formation mechanism of the microstructures was discussed based on MALDI-TOF-MS and TEM analytical results. Furthermore, the adsorption to acid indigo blue of the resulting samples was investigated, which indicated that the maximum adsorption capacities were correlated well with the specific surface areas of the samples.
