Expression of Ezrin Protein and Its Relationship with Lung Metastasis and Survival Time of Osteosarcoma Patients

Abstract:Objective:To investigate the expression of Ezrin,a member of ezrin/radixin/moesin family,in osteosarcoma and the relationship between the positive rate of Ezrin and lung metastasis and overall survival time.Methods:The expression of Ezrin was detected in the tissue microarray which contains 56 spots of osteosarcoma tissues,10 spots of chondrosarcoma tissues,and 10 spots of normal tissues by immunohistochemical method.The survival curve analysis and lung metastasis analysis of patients were evaluated based on the clinical follow-up data.Results:Ezrin was over-expressed in osteosarcoma tissues compared with that of chondrosarcoma and normal tissues.The difference was significant(P=0.023 and 0.001).The positive rate of Ezrin had close relationship with Enneking GTM clinical staging.There was significant difference in the positive rate between stage Ⅱb and stage Ⅲ osteosarcoma(61.4% vs 91.7%,P=0.046).But expression of Ezrin protein had no significant correlation with gender,age,location,pathological subtype,and with or without lung metastasis during the primary therapy.Kaplan-Meier analysis showed that Ezrin-positive patients had poor prognosis compared with Ezrin-negative patients.The time of pulmonary metastasis and overall survival was decreased in Ezrin-positive patients.Conclusion:There is a close relationship between Ezrin protein and lung metastasis of osteosarcoma patients.Ezrin will become a useful marker for evaluating prognosis and survival time of osteosarcoma patients.
