Abstract:Thrombosis and in-stent restenosis constitute major clinical failures of endovascular stent implantation. In this research, a polyelectrolyte multilayered film of heparin/collagen was constructed onto the endovascular stent via layer-by-layer deposition. The in vitro human platelet adhesion test and blood clotting time assessments demonstrated that the fabricated multilayer took on excellent hemocompatibility. The in vitro human umbilical vein endothelial cell culture results show that the endothelial cell adhesion and proliferation are greatly promoted after the multilayer modification. Such multilayered endovascular stents with excellent hemocompatibilty and human endothelial cytocompatibility may have great potential in clinical applications.

Abstract:Endothelialization and antithrombogenicity are two key issues in stent implantation. The layer-by-layer (LbL) deposition of anticoagulant heparin and cell compatible collagen was explored to develop a multilayered coating with synergic property of antithrombogenicity and fast endothelialization. The quartz crystal microbalance with dissipation (QCM-D), UV spectrometer, spectroscopic ellipsometry, scanning electron microscopy, and confocal laser scanning microscopy investigations indicate that the LbL technique, which based on molecular assembly, provides an easy way to develop a smooth, homogenous, and stable coating onto stents. In vitro blood clotting time tests and the platelet adhesion tests show that the multilayer-modified stents present good hemocompatibility. In vitro endothelial cell (EC) culture results show that the multilayer-modified surfaces accelerate the adhesion and proliferation of ECs. These results illustrate that a stent surface coating with properties of antithrombogenicity and EC preference was obtained via heparin/collagen multilayer modification. In Situ Endothelialization of Intravascular Stents Coated with an Anti-Cd34 Antibody Functionalized Heparin-Collagen Multilayer

Abstract:The in-stent restenosis (ISR) and the late stent thrombosis (LAST) represent the most common failures of stent implantation and are both mediated at the injured endothelium. The natural endothelium healing mechanism provides an approach to achieve in situ endothelialization of the implant by stimulating the neighboring endothelial cells (ECs) migration or capturing the circulating endothelial cells (CEC) directly from the blood circulation. An anti-CD34 antibody functionalized multilayer of heparin/collagen is developed here via layer-by-layer assemble. The ellipsometry and QCM-D results demonstrate that the multilayer coatings with slight glutaraldehyde cross-linking are stable in static incubation and flushing conditions, respectively. The in vitro hemocompatibility tests and cell culture results indicate that both heparin/collagen multilayers with or without the anti-CD34 antibody functionalization not only preserve good hemocompatibility, but also promote cell attachment and growth notably. While the heparin/collagen multilayer coatings show no selectivity in promotion of ECs and smooth muscle cells (SMCs), the anti-CD34 antibody functionalized heparin/collagen multilayers can specifically promote the attachment and growth of the vascular ECs. The metabolic activity assessment and the NO secretion measurements further indicate that the adherent ECs on the anti-CD34 antibody functionalized heparin/collagen multilayer surface have better viability and possess the specific function of the natural vascular ECs. In vivo experiments indicate that the anti-CD34 antibody can enrich and accelerate the attachment of the vascular cells onto the stent and rapid endothelialization is realized. While no significant difference of neointimal hyperplasia is observed between the bare metal stents and heparin/collagen multilayer modified stents, the neointimal hyperplasia on the anti-CD34 antibody functionalized multilayer modified stents is significantly inhibited. The success of the anti-CD34 antibody functionalized heparin/collagen multilayer coating in rapid endothelialization and anti-restenosis might indicate that the immobilization of ECs specific ligand onto a cytocompatible matrix can be a good approach for in situ endothelialization and a possible solution to ISR.

Abstract:In‐stent restenosis (ISR) is seriously affecting the long‐term prognosis of vascular interventional therapy and leading to enormous medical burdens. Great efforts have been devoted to developing functional vascular stents with desired features and properties for effective ISR prevention. Here, we present a multifunctional bionic vascular stent with designed coatings prepared using the microfluidic electrospinning technology. Such stents are composed of biocompatible, drug‐loaded methylacrylated gelatin‐polyethylene glycol diacrylate (GelMA‐PEGDA) and polycaprolactone (PCL) composite nanofibers on the 316L stainless steel stents by an easy‐to‐operate step‐by‐step spraying method. Benefitting from the addition of polydopamine (PDA) during the fabrications, the drug‐loaded composite nanofibers can adhere to both the stent and the vascular wall well. Besides, as the inner fibrous layer of the stent that contacting the lumen is equipped with heparin‐vascular endothelial growth factor (Hep‐VEGF), it plays an anticoagulation role and promotes the growth of endothelial cells (ECs); while the outer layer contacts the vascular wall and releases rapamycin slowly, which can restrain smooth muscle proliferation. By implanting it into the rabbit carotid artery, we have demonstrated that the multi‐functional bionic vascular stent can achieve good anti‐thrombosis and in‐stent restenosis effects, which indicates its potential values in vascular intervention and other biomedical fields. Abstract:Thrombosis and in-stent restenosis are the main obstacles in the healing process after cardiovascular surgery. A promising way to achieve the healing process after percutaneous transluminal coronary angioplasty followed by stenting may rely on the rapid in situ endothelialization on the materials of implants. Several requirements are raised to achieve in situ endothelialization, of which the specifically endothelial cells (ECs) homing and the non-specific cells repulsion come first. In this work, heparin/chitosan multilayer was constructed with thromboresistant and non-specifically cell-resistant properties. The specific ECs adhesive peptide sequence Arg-Glu-Asp-Val (REDV) was then immobilized onto the pristine multilayer and the cell responses of ECs and smooth muscle cells (SMCs) were verified. It is interesting that ECs selective attachment was obtained on the REDV functionalized multilayer, whereas the multilayer maintains resisting to the SMCs. These results show that the REDV functionalized cell-resistant heparin/chitosan multilayer is a ECs selective surface, which may have great potential in cardiovascular biomaterials for in situ endothelialization.

Abstract:Given the risks of poor patient compliance and bleeding associated with current dual antiplatelet therapies, it is urgent to develop the next generation of cardiovascular stents with anticoagulation and rapid endothelialization capabilities. Inspired by the prominent bioactivity and bioavailability of zeolitic imidazolate framework-90 (ZIF-90) in driving endothelial cell (EC) morphogenesis, this research proposes a "synergistic anticoagulant and endothelial regeneration strategy" depending on mussel-inspired phospholipid copolymer (MIPC) and ZIF-90. Depending on the copolymerization of the catechol with dopamine (Dopa) monomers, Dopa/MIPC coating was immobilized on the surface of CoCr via a one-pot process for resisting the initial thrombosis induced by platelets and fibrinogen. Meanwhile, ZIF-90 was loaded on the coating via coordination effect, aiming to accelerate the proliferation and migration of ECs. Compared with CoCr, the well-designed CoCr-Dopa/MIPC@ZIF-90 not only reduced fibrinogen adhesion by approximately 40 % and platelet adhesion by almost 55 %, but also promoted the proliferation and migration of ECs significantly in vitro. Furthermore, the blood flow velocity of CoCr-Dopa/MIPC@ZIF-90 stent was similar to natural aorta and ECs coverage on it was greatly strengthened after 30 days in a rat aorta vascular stent implantation model. Collectively, CoCr-Dopa/MIPC@ZIF-90 exhibited obvious superiority in reducing the formation of thrombus and promoting endothelial regeneration, which might meet the high requirement for the next generation of vascular stent.
Novel iron heparin anticoagulation coating for intravascular stents

Abstract:The experiment was performed at Nanomedicine and Biosensor Laboratory of Harbin Institute of Technology from December 2006 to January 2008. Fe3+/heparin multilayer films were assembled on the surface of Ni-Ti alloy by the layer-by-layer self-assembly, which made the stable glucose-Fe complex coating form on the surface of Ni-Ti alloy. The ultraviolet-visible absorption spectrophotometry, atomic force microscopy and fourier transform infrared spectroscopy proved that the Fe3+/heparin could form uniform multilayer films by alternative deposition on the surface of Ni-Ti alloy, and the coating had good stability. Studies showed that the increase of pH value and ionic strength in solution could obviously improve the increasing speed of the coating. The coating had good anticoagulation property and blood compatibility, which were proved by detection of thrombin inactivation, activated partial thromboplastin time, hemolysis test and platelet adhesion test. The heparin did not decrease its anticoagulation property after combined with Fe3+. The results suggested that the coating could be use for the surface modification of intravascular stents, and prevent thrombosis forming in early stage of stent implantation.
Passivation protein-adhesion platform promoting stent reendothelialization using two-electron-assisted oxidation of polyphenols

Abstract:Superhydrophilic surfaces play an important role in nature. Inspired by this, scientists have designed various superhydrophilic materials that are widely used in the field of biomaterials, such as PEG molecular brushes and zwitterionic materials. However, superhydrophilic coatings with only anti-fouling properties do not satisfy the requirements for rapid reendothelialization of cardiovascular stent surfaces. Herein, a novel polyphenol superhydrophilic surface with passivated protein-adsorption properties was developed using two-electron oxidation of dopamine and polyphenols. This coating has a multiscale effects: 1) macroscopically: anti-fouling properties of superhydrophilic; 2) microscopically: protein adhesion properties of active groups (quinone-, amino-, hydroxyphenyl groups and aromatic ring). Polyphenols not only enhance the ability of coating to passivate protein-adsorption, but also make the coating have polyphenol-related biological functions. Therefore, the polyphenol and passivated protein-adsorption platform together maintain the stability of the scaffold microenvironment. This, in turn, provides favorable conditions for the growth of endothelial cells on the scaffold surface. In vivo implantation of the coated stents into the abdominal aorta resulted in uniform and dense endothelial cells covering the surface of the neointima. Moreover, new endothelial cells secreted large amounts of functional endothelial nitric oxide synthase like healthy endothelial cells. These results indicate that the polyphenol superhydrophilic coating potentially resists intra-stent restenosis and promotes surface reendothelialization. Hence, polyphenol superhydrophilic coatings with passivated protein-adsorption properties constructed by two-electron-assisted oxidation are a highly effective and versatile surface-modification strategy for implantable cardiovascular devices.

Abstract:Drug-eluting stents, vascular prostheses coated with thin polymer-containing medication, have become one of the most effective methods to treat cardiovascular diseases. However, this therapeutic strategy may lead to thrombosis, stent restenosis, and intimal hyperplasia and prevent re-endothelialization. In this study, we selected 3-aminophenylboronic acid-modified hyaluronic acid and carboxylate chitosan as polyelectrolyte layers and embedded an epigallocatechin-3-gallate–tanshinone IIA sulfonic sodium (EGCG–TSS) complex to develop a sandwich-like layer-by-layer coating. The introduction of a functional molecular EGCG–TSS complex improved not only the biocompatibility of the coating but also its stability by enriching the interaction between the polyelectrolyte coatings through electrostatic interactions, hydrogen bonding, π–π stacking, and covalent bonding. We further elucidated the effectiveness of sandwich-like coatings in regulating the inflammatory response, smooth muscle cell growth behavior, stent thrombosis and restenosis suppression, and vessel re-endothelialization acceleration via in vivo and in vitro . Conclusively, we demonstrated that sandwich-like coating assisted by an EGCG–TSS complex may be an effective surface modification strategy for cardiovascular therapeutic applications.

Abstract:A biomacromolecular layer-by-layer coating process of chitosan/heparin onto a coronary stent is designed for the acceleration of the re-endothelialization and healing process after coronary stent deployment. The results of in vitro culturing of porcine iliac artery endothelial cells as well as the measurements of the APTT, PT and TT supported the rationale that the combination of chitosan and heparin could bring both endothelial cell compatibility and haemocompatibility to the stent surface. A porcine coronary injury model and arteriovenous shunt model were used for the further evaluation of the application of this kind of surface-modified stainless steel stent in vivo. The final results proved that this facile coating approach could significantly promote re-endothelialization and was safer compared with bare metal stents for its much improved anticoagulation property.
Anti-fouling plus: Engineered stent coating with inflammation-regulation capability enables enhanced tissue healing

Abstract:Coagulation, inflammation, and hyperplasia frequently restrict the effectiveness of cardiovascular stents. It is critical to modify stent surfaces to allow for in situ tissue healing. Interrelated coagulation and inflammation can impede endothelialization and are critical to the long-term performance of cardiovascular implants. This study proposes a relatively bio-inert "Anti-fouling plus" coating that is effective in anti-coagulation and anti-inflammation at the early stage of implantation and provides mild inflammatory responses during tissue remodeling process. The phosphorylcholine (PC)-based polymer PM(PCLA) is reacted onto a polydopamine-treated polylactic acid (PLA) surface. PC-based polymers that mimic cell membranes present remarkable antiinflammation and anticoagulation properties. The composite of epigallocatechin gallate (EGCG) and tempol is loaded on the PM(PCLA) coating to achieve "anti-fouling plus" which can mediate inflammatory responses by scavenging reactive oxygen species. In vitro hemocompatibility and cell tests demonstrate the effectiveness of coatings in inhibiting thrombus and inflammation. Interestingly, this study originally reveals a novel strategy that the anti-fouling plus coating can direct vascular tissue healing on stent via promoted endothelialization and mediated contractile smooth muscle cell phenotype by switching inflammatory cells to a pro-repair phenotype during tissue healing process, even if the coating is relatively bio-inert.

Abstract:Co-immobilization of two or more molecules with different and complementary functions to prevent thrombosis, suppress smooth muscle cell (SMC) proliferation, and support endothelial cell (EC) growth is generally considered to be promising for the re-endothelialization on cardiovascular stents. However, integration of molecules with distinct therapeutic effects does not necessarily result in synergistic physiological functions due to the lack of interactions among them, limiting their practical efficacy. Herein, we apply heparin and nitric oxide (NO), two key molecules of the physiological functions of endothelium, to develop an endothelium-mimetic coating. Such coating is achieved by sequential conjugation of heparin and the NO-generating compound selenocystamine (SeCA) on an amine-bearing film of plasma polymerized allylamine. The resulting surface combines the anti-coagulant (anti-FXa) function provided by the heparin and the anti-platelet activity of the catalytically produced NO. It also endows the stents with the ability to simultaneously up-regulate α-smooth muscle actin (α-SMA) expression and to increase cyclic guanylate monophosphate (cGMP) synthesis of SMC, thereby significantly promoting their contractile phenotype and suppressing their proliferation. Importantly, this endothelium-biomimetic coating creates a favorable microenvironment for EC over SMC. These features impressively improve the antithrombogenicity, re-endothelialization and anti-restenosis of vascular stents in vivo.

Stable and Thin-Polymer-Based Modification of Neurovascular Stents with 2-Methacryloyloxyethyl Phosphorylcholine Polymer for Antithrombogenicity

Abstract:The advent of intracranial stents has revolutionized the endovascular treatment of cerebral aneurysms. The utilization of stents has rendered numerous cerebral aneurysm amenable to endovascular treatment, thereby obviating the need for otherwise invasive open surgical options. Stent placement has become a mainstream approach because of its safety and efficacy. However, further improvements are required for clinically approved devices to avoid the frequent occurrence of thrombotic complications. Therefore, controlling the thrombotic complications associated with the use of devices is of significant importance. Our group has developed a unique stent coated with a 2-methacryloyloxyethyl phosphorylcholine (MPC)-based polymer. In this study, the surface characteristics of the polymer coating were verified using X-ray photoelectron spectroscopy and atomic force microscopy. Subsequently, the antithrombotic properties of the coating were evaluated by measuring platelet count and thrombin-antithrombin complex levels of whole human blood after 3 h of incubation in a Chandler loop model. Scanning electron microscopy was utilized to examine thrombus formation on the stent surface. We observed that MPC polymer-coated stents significantly reduced thrombus formation as compared to bare stents and several clinically approved devices. Finally, the coated stents were further analyzed by implanting them in the internal thoracic arteries of pigs. Angiographic imaging and histopathological examinations that were performed one week after implantation revealed that the vascular lumen was well maintained and coated stents were integrated within the vascular endothelium without inducing adverse effects. Thus, we demonstrated the efficacy of MPC polymer coating as a viable strategy for avoiding the thrombotic risks associated with neurovascular stents.
Covalently-Attached, Surface-Eroding Polymer Coatings on Magnesium Alloys for Corrosion Control and Temporally Varying Support of Cell Adhesion

Abstract:For degradable magnesium (Mg) alloy-based stents it would be desirable to delay early corrosion to maintain mechanical strength. Similarly, early after stent placement reduced thrombogenicity is an important feature, while chronically, endothelial cell adhesion and vessel integration are desirable. In this study, surface eroding polymers of amino-grafted poly(1,3-trimethylene carbonate) (PTMC-NH2) and PTMC-NH(2)combined with sulfobetaine bearing polymer PSB (PTMC-NHCO-PSB) are developed, and these polymers are covalently attached onto 6-phosphonohexanoic acid (PHA)-coated AZ31 Mg alloy surfaces in sequence. In vitro degradation testing in ovine plasma shows PTMC, PTMC-NH2, and PTMC-NHCO-PSB cast films experience a gradual thickness and mass loss with maintenance of smooth surfaces, confirming surface erosion behavior. The PTMC-NH(2)polymer is firmly bound to the PHA-modified AZ31 surface and demonstrates a resistance to peeling. PTMC, PTMC-NH2, and PTMC-NHCO-PSB coated AZ31 have a lower corrosion rate versus polylactide-co-glycolide coated and untreated AZ31. PTMC-NHCO-PSB coated AZ31 inhibits platelet deposition and smooth muscle cell adhesion and growth, but after 2-week immersion in plasma, this surface supports endothelial cell adhesion and growth. These results suggest PTMC-NHCO-PSB surface eroding coating offers a means of controlling corrosion while providing a temporally varying bio-functionality for biodegradable vascular stent applications.

Abstract:The achievement of the desired hemocompatibility, anti-inflammatory, and rapid endothelialization properties has significantly contributed to the success of implantable blood-contacting materials, especially vascular stents. In this study, we constructed a facile and versatile superhydrophilic coating via the stepwise deposition of metal/phenolic networks (MPNs) using a layer-by-layer assembly technique. Tannic acid and copper ions were employed for the fabrication of MPNs, and the surface roughness and the number of hydrophilic phenolic groups were gradually increased, ultimately leading to the generation of a superhydrophilic coating, where the copper ions were also effectively incorporated. Owing to the superhydrophilicity of the coating, the binding of serum adhesion proteins, such as albumin and fibrinogen, to the surface was strongly inhibited, thereby conferring a potent antithrombotic ability. In addition, a potent antioxidant and anti-inflammatory effects were achieved through the deposition of a large amount of residual phenolic hydroxyl groups on the surface. The incorporated copper ions could also mimic endothelial function through the continuously catalyzed decomposition of endogenous S-nitrothiols (RSNOs) to release nitric oxide (NO) in situ in a controlled manner. The synergistic contribution effect of NO and superhydrophilicity, further inhibited the adhesion/activation of platelets. The optimized coating significantly inhibited the adhesion and activation of platelets, selectively supported the adhesion of endothelial cells, and suppressed the proliferation of smooth muscle cells. Such superhydrophilic coating with the incorporation of copper ions adequately addressed the basic requirements of vascular stents, in terms of antithrombosis, anti-restenosis, and enhanced endothelialization in vivo, demonstrating a promising approach for the clinical application of vascular implants.

Abstract:Heparin, an important polysaccharide, has been widely used for coatings of cardiovascular devices because of its multiple biological functions including anticoagulation and inhibition of intimal hyperplasia. In this study, surface heparinization of a commonly used 316L stainless steel (SS) was explored for preparation of a multifunctional vascular stent. Dip-coating of the stents in an aqueous solution of dopamine and hexamethylendiamine (HD) (PDAM/HD) was presented as a facile method to form an adhesive coating rich in primary amine groups, which was used for covalent heparin immobilization via active ester chemistry. A heparin grafting density of about 900 ng/cm(2) was achieved with this method. The retained bioactivity of the immobilized heparin was confirmed by a remarkable prolongation of the activated partial thromboplastin time (APTT) for about 15 s, suppression of platelet adhesion, and prevention of the denaturation of adsorbed fibrinogen. The Hep-PDAM/HD also presented a favorable microenvironment for selectively enhancing endothelial cell (EC) adhesion, proliferation, migration and release of nitric oxide (NO), and at the same time inhibiting smooth muscle cell (SMC) adhesion and proliferation. Upon subcutaneous implantation, the Hep-PDAM/HD exhibited mitigated tissue response, with thinner fibrous capsule and less granulation formation compared to the control 316L SS. This number of unique functions qualifies the heparinized coating as an attractive alternative for the design of a new generation of stents.

Abstract:Cardiovascular metal stents have shown potential in the treatment of coronary artery disease using percutaneous coronary intervention. However, thrombosis, endothelialization, and new atherosclerosis after stent implantation remain unsolved problems. Herein, a multifunctional coating material based on phase-transited lysozyme was developed to promote stent endothelialization and simultaneously reduce thrombus events by embedding moieties of heparin and co-immobilized copper ions for in-situ catalyzing nitric oxide (NO) generation. The lysozyme-based biomimetic coating is compatible with blood and enables facile loading and sustainable release of copper ions to produce NO with donors via catalytic reaction. The novel coating strategy displayed several bio-effects of anti-thrombosis; it synergistically promoted endothelial cell growth and inhibited smooth muscle cell growth. Thus, this systemic in vitro study will provide a foundation for developing multifunctional cardiovascular stents in clinical settings.

Abstract:Endowing cardiovascular stents with multiple functionalities, such as anti–proliferative, and anti–inflammatory properties, is important for combating restenosis after stent implantation. The surface properties of stents play a key role in obtaining the desired post–implantation performance. Zwitterionic coatings are resistant to non–specific adsorption, and here, a mussel–inspired zwitterionic poly (carboxybetaine acrylate–co–dopamine methacrylate) copolymer (PCBDA) was synthesized and applied to the surface of a biodegradable poly(l–lactide) stent. To obtain a zwitterionic polymer coating, the surface of a PLA stent was pretreated by co–depositing polydopamine (PDA) and polyethyleneimine (PEI). The mussel–inspired catechol moieties in the PCBDA copolymer cross–linked with the PDA–PEI hybrid to form a robust PCBDA/PDA–PEI coating on the PLA stent. The zwitterionic PCBDA formed a hydration layer once it contacted blood, preventing the non–specific adsorption of serum proteins, which further blocked subsequent coagulation and inflammation. The anti–protein adsorption tests showed that the PCBDA/PDA–PEI coating resisted the non–specific adsorption of fibrinogen, which was a positive signal for enhanced anti–coagulation. Moreover, the platelet adhesion/activation tests confirmed its antithrombotic properties. The in vivo tissue response was also greatly suppressed due to the inhibited recognition of PCBDA/PDA–PEI–coated implants by macrophages. Interestingly, unlike traditional zwitterionic polymers, due to the introduction of adhesive catechol moieties, PCBDA supported the growth of endothelial cells, demonstrating its ability to selectively direct the fate of endothelial cells and smooth muscle cells. In vivo stent implantation results showed inhibited intimal hyperplasia due to the protection of PCBDA/PDA–PEI. Overall, due to the specific characteristics of the mussel–inspired PCBDA/PDA–PEI coating, anticoagulation, anti–inflammatory, and anti–proliferation properties were obtained; thus, it is expected that this coating can be used to inhibit restenosis and realize endothelialization after stent implantation.

Abstract:To develop an anticoagulant coating for polyethylene terephthalate (PET) fabric-covered stents, we prepared a series of ultraviolet (UV)-curable coatings with different film-forming compositions and enoxaparin sodium ratios. We then investigated their mechanical properties and hydrophilicity by adhesion, pencil hardness, abrasion resistance tests, and water contact angle measurements. In addition, their biocompatibility and cytotoxicity were evaluated by their protein adsorption, platelet adhesion, and plasma recalcification times, and by CCK-8 assays. Finally, an anticoagulant coating with excellent mechanical properties, biocompatibility, and low cytotoxicity was obtained, offering a feasible scheme for covered stents in clinical practice.
