Abstract:Silk fibroin/chitosan scaffolds were prepared through lyophilization, then hydroxyapatite was deposited by premineralization to obtain porous hydroxyapatite (HA)-silk fibroin/chitosan composite scaffolds.Composite scaffolds were characterized by Fourier transform infrared analysis (FT-IR), X-ray diffraction (XRD), scanning electronic microscopy (SEM), thermogravimetry analysis (TGA), compressive modulus and porosity, as well as the proliferation of MG-63 cells on composite scaffolds was detected by WST-1 method. The results reveal that the structure of deposited HA is a weak crystallinity, which deposits on the wall of pore. With the increase of premineralization cycles, the amount of HA and compressive modulus of premineralized composite scaffolds are significantly increased, the porosity keeps about 90%. WST-1 assay shows that premineralized composite scaffolds deposited with HA dramatically enhance the proliferation of cells, and possess a great cytocompatibility.

Abstract:Novel tissue engineering scaffold materials of nano-hydroxyapatite (nHA)/silk fibroin (SF) biocomposite were prepared by freeze-drying. The needle-like nHA crystals of about 10 nm in diameter by 50-80 nm in length, which were uniformly distributed in the porous nHA/SF scaffolds, were prepared by a co-precipitation method with a size. The as-prepared nHA/SF scaffolds showed good homogeneity, interconnected pores and high porosity. XRD and FT-IR analysis suggested that the silk fibroin was in beta-sheet structure, which usually provides outstanding mechanical properties for silk materials. In this work, composite scaffolds containing as high as 70% (w/w) nHA were prepared, which had excellent compressive modulus and strength, higher than the scaffolds at low nHA content level and other porous biodegradable polymeric scaffolds often considered in bone-related tissue engineering reported previously. The cell compatibility of composite scaffolds was evaluated through cell viability by MTT assay. All these results indicated that these nHA/SF scaffold materials may be a promising biomaterial for bone tissue engineering.

Abstract:An applicable matrix used in tissue engineering should not only have suitable mechanical properties, porous structures and biocompatibility that facilitate the adhesion, growth and proliferation of tissue cells, but also have the ability to release bioactive factors to provide a more conducive and inductive environment for tissue growth. Because of the harsh preparation conditions and deficiency of mechanical properties, it is still difficult for fibroin and collagen matrices to possess these multifunctional properties. In this research, we successfully prepared fibroin/collagen hybrid scaffolds containing heparin that possess multifunctional properties under mild conditions. These scaffolds maintain outstanding mechanical properties and porous structures of fibroin-based scaffolds. Furthermore, the scaffolds keep the bioactivity of collagen, becoming delivering systems that release heparin slowly to make the scaffolds blood compatible. Compared with fibroin/collagen scaffolds, the scaffolds containing heparin further facilitate the growth of HepG2 cells since a more complex, dynamic environment was formed to promote the cell growth. Considering the mild aqueous preparation environment without crosslinking reaction, besides promoting the progress in blood contacting tissue engineering, our research has also opened a door to prepare various multifunctional fibroin/collagen hybrid matrices that combine the advantages of fibroin and collagen.
Three-dimensional fibroin/collagen scaffolds derived from aqueous solution and the use for HepG2 culture

Abstract:Although three-dimensional fibroin scaffolds have been prepared with freeze–drying method, these scaffolds still cannot meet the requirements of tissue engineering. In this article, a new process is described to form fibroin-based porous scaffolds with controllable structure and morphological features. When collagen was added to fibroin solution, the viscosity of the blend solution increased because of the interaction between fibroin and collagen, and then it restrained the unwanted fibroin leaf formation in freezing process that generally appeared in the previous fibroin scaffold preparation. With methanol treatment, the fibroin/collagen scaffolds became water-stable, following the transition from random and α-helix to β-sheet conformation. The aqueous-fibroin porous scaffolds had highly homogeneous and interconnected pores with pore sizes ranging from 127 to 833μm, depending on the fibroin concentration. The porosity of scaffolds was >90%, and the yield strength and modulus were up to 354±25kPa and 30±0.1MPa, respectively, when the blend solution, containing 20% collagen, maintained 4% fibroin concentration. Adhesion, spreading and proliferation of HepG2 cells on fibroin and fibroin/collagen blend scaffolds were also observed to investigate the biocompatibility. Scanning electron microscopy (SEM) and MTT analyses demonstrated that the adding of collagen evidently facilitated HepG2 attachment and proliferation in vitro. These new fibroin based three-dimensional scaffolds provided much more excellent properties due to the greatly improved control of pore size, the uniform pore distribution, the hydrophility, the mechanical properties and the biocompatibility compared with those of reported three-dimensional fibroin scaffolds.

Abstract:One challenge in soft tissue engineering is to find an applicable scaffold, not only having suitable mechanical properties, porous structures, and biodegradable properties, but also being abundant in active groups and having good biocompatibility. In this study, a three-dimensional silk fibroin/chitosan (SFCS) scaffold was successfully prepared with interconnected porous structure, excellent hydrophilicity, and proper mechanical properties. Compared with polylactic glycolic acid (PLGA) scaffold, the SFCS scaffold further facilitated the growth of HepG2 cells (human hepatoma cell line). Keeping the good cytocompatibility and combining the advantages of both fibroin and chitosan, the SFCS scaffold should be a prominent candidate for soft tissue engineering, for example, liver.

Abstract:In bone tissue engineering, a qualified scaffold should provide a three-dimensional porous structure mimicking the extracellular matrix of bone except good biological properties. In this study, biomorphic silk fibroin (SF) scaffolds were fabricated from cane by an innovative biotemplating-negative mold process. The physicochemical properties, in vitro enzymatic degradation behavior and biocompatibility of the biomorphic SF scaffolds were investigated. The results showed that the scaffolds well inherited the original porous morphology of cane, and possessed good mechanical stability. The scaffolds had a compressive modulus of 1.56 +/- 0.08 MPa and a porosity of 82.73%, and exhibited a bimodal pore size distribution (15 and 172 mu m). The degradation ratio of the SF scaffolds increased with prolonging degradation time and reached 29% within 21 days when exposed to 1.0 U/mL collagenase IA. The in vitro cytocompatibility evaluation indicated that the scaffolds could support cell attachment, proliferation, and osteogenic differentiation of osteoblast- like MC3T3-E1 cells, as assessed by SEM, fluorescent staining, MTT, and ALP activity assays. Growth of fibroblast and vascular smooth muscle cells in fibroin/collagen scaffold

Abstract:In our recent study, a novel fibroin/collagen scaffold with improved mechanical properties and controllable porous structure was prepared through freeze–drying method. In this research, the cyto-compatibility was further studied, using fibroblast and vascular smooth muscle cells (VSMC) as the model cells. MTT results indicated that the growth of fibroblast and VSMC both further improved in the fibroin/collagen scaffold than in pure fibroin scaffolds. The confocal and SEM results showed that fibroblast cells and VSMCs had better adhesion and spreading properties in the fibroin/collagen scaffolds. Although further studies, such as the extracellular matrix production and the functional gene expression, are necessary to clarify the biocompatibility of the fibroin/collagen scaffolds, the present results indicate that the fibroin/collagen scaffold is a new scaffold material suitable for tissue engineering. On the other hand, the mild and all-aqueous preparation processes also make it possible to embed different growth factors inside the scaffolds to maximize cell functions and the formation of specific tissues.

Abstract:BACKGROUND Bone tissue engineering, a powerful tool to treat bone defects, is highly dependent on use of scaffolds. Both silk fibroin (SF) and chitosan (Cs) are biocompatible and actively studied for reconstruction of tissue engineering. Gelatin (Gel) is also widely applied in the biomedical field due to its low antigenicity and physicochemical stability. MATERIAL AND METHODS In this study, 4 different types of scaffolds were constructed - SF, SF/Cs, SF/Gel, and SF/Cs/Gel - and we compared their physical and chemical properties as well as biological characterization of these scaffolds to determine the most suitable scaffold for use in bone regeneration. First, these scaffolds were produced via chemical cross-linking method and freeze-drying technique. Next, the characterization of internal structure was studied using scanning electron microscopy and the porosity was evaluated by liquid displacement method. Then, we compared physicochemical properties such as water absorption rate and degradation property. Finally, MC3T3-E1 cells were inoculated on the scaffolds to study the biocompatibility and osteogenesis of the three-dimensional (3D) scaffolds in vitro. RESULTS The composite scaffold formed by all 3 components was the best for use in bone regeneration. CONCLUSIONS We conclude that the best scaffold among the 4 studied for MC3T3-E1 cells is our SF/Cs/Gel scaffold, suggesting a new choice for bone regeneration that can be used to treat bone defects or fractures in clinical practice.
Characterization of Silk Fibroin/Chitosan 3D Porous Scaffold and In Vitro Cytology

Abstract:Bone tissue engineering is a powerful tool to treat bone defects caused by trauma, infection, tumors and other factors. Both silk fibroin (SF) and chitosan (CS) are non-toxic and have good biocompatibility, but are poor biological scaffolds when used alone. In this study, the microscopic structure and related properties of SF/CS composite scaffolds with different component ratios were examined. The scaffold material most suitable for osteoblast growth was determined, and these results offer an experimental basis for the future reconstruction of bone defects. First, via freeze-drying and chemical crosslinking methods, SF/CS composites with different component ratios were prepared and their structure was characterized. Changes in the internal structure of the SF and CS mixture were observed, confirming that the mutual modification between the two components was complete and stable. The internal structure of the composite material was porous and three-dimensional with a porosity above 90%. We next studied the pore size, swelling ratio, water absorption ratio, degradation and in vitro cell proliferation. For the 40% SF-60% CS group, the pore size of the scaffold was suitable for the growth of osteoblasts, and the rate of degradation was steady. This favors the early adhesion, growth and proliferation of MG-63 cells. In addition to good biocompatibility and satisfactory cell affinity, this material promotes the secretion of extracellular matrix materials by osteoblasts. Thus, 40% SF-60% CS is a good material for bone tissue engineering.

Abstract:Three dimensional (3-D) fibroin/collagen scaffolds are the novel fibroin based scaffolds derived from aqueous solution. In this article, we investigated the effect of pH on the formation of fibroin/collagen scaffolds. In the range of pH from 4 to 8.5, the fibroin/collagen scaffolds with good porous structures can be prepared using freeze-drying method, which would facilitate the adding of other biopolymers. The structures of different fibroin-based scaffolds were investigated with FTIR and DSC, which indicated that the interaction of fibroin and collagen affected the methanol-induced transformation of fibroin from random-coil to β-sheet conformation. The mechanical properties were also studied. The results mean that all the fibroin-based scaffolds prepared in various pH values had better mechanical properties than other reported fibroin scaffolds. Since the fibroin/collagen scaffolds can be prepared in the range of pH from 4 to 8.5, it is very easy to prepare different multifunctional scaffolds such as fibroin/collagen/chitosan scaffolds and fibroin/collagen/heparin scaffolds in acidic or neutral conditions. These new fibroin-based blend materials extend the range of biomaterial properties that can promote the use in biomedical applications such as drug release and tissue engineering.
[Primary Study on Histocompatibility of Three Kinds of Collagen-Chitosan Porous Scaffolds].

Abstract:OBJECTIVE:To construct three kinds of collagen-chitosan porous scaffolds with enhanced bio-stability and to investigate the histocompatibility of the scaffolds in vivo.METHODS:Collagen-chitosan porous scaffolds were fabricated by freeze-drying method, cross-linked using dehydrothermal treatment and glutaraldehyde, respectively. The morphology of the uncross-linked scaffold (scaffold 1), dehydrothermal cross-linked scaffold (scaffold 2) and glutaraldehyde cross-linked scaffold (scaffold 3) was studied by scanning electron microscopy. Three kinds of scaffolds were embedded subcutaneously on dorsal surface of 12 rabbit ears. The general and local responses were recorded daily. The bio-stability and histocompatibility of the scaffolds were observed by using HE staining after 3, 7, 14 and 28 days of operation.RESULTS:The scaffolds had three-dimensional porous structures with a porosity of more than 90%, and possessed pore sizes of 120 +/- 10 microm, 80 +/- 15 microm and 170 +/- 20 microm, respectively. All experimental rabbits survived with good general condition during the study. All skin incisions healed well without obvious reactive red or swelling. Histological study showed that scaffold 1 was degraded rapidly with obvious inflammation. The degradation of scaffold 2 was slower than that of scaffold 1 and the inflammation of scaffold 2 was also milder than that of scaffold 1. Scaffold 3 possessed slow degradation property with slight inflammatory reaction, and rapid tissue regeneration.CONCLUSION:The collagen-chitosan porous scaffolds have three-dimensional porous structures that are suitable for tissue regeneration. The bio-stability and histocompatibility of the scaffolds are enhanced after cross-linked. Glutaraldehyde cross-linked is better than dehydrothermal cross-linked, which can facilitate dermal tissue reconstruction.

Abstract:Recombinant human-like collagen (RHLC) was added to fibroin solution to prepare a novel hybrid scaffold material for skin tissue engineering. The morphology of the scaffold had highly homogeneous and interconnected pores with pore sizes 136 +/- 32 mum measured by scanning electron microscopy (SEM). FTIR analysis indicated intermolecular crosslinkages between fibroin and RHLC formed. The viscosity of the blend solution increased because of the interaction between fibroin and RHLC, and then it restrained the unwanted fibroin aggregation in freezing process that generally appeared in fibroin scaffold preparation with freeze drying method. After methanol treatment the fibroin/RHLC scaffold became water-stable. The porosity of scaffolds was >>90%, the compressive strength and modulus were up to 662 +/- 32 KPa and 7.8 +/- 0.64 MPa, respectively. Fibroblasts cultured within fibroin/RHLC scaffolds were investigated by SEM, laser scanning confocal microscopy (LSCM), and MTT assay, which showed that the adding of RHLC significantly enhanced the cells adhesion, proliferation, and viability compare with fibroin scaffolds. These results suggest that the hybrid scaffolds have favorable characteristics for skin tissue engineering.

Abstract:On this basis, A novel recombinant human-like collagen (RHLC)/silk fibroin scaffold material with high porosity and controllable aperture was prepared. The compatibility of osteoblasts (OB) with the blends was tested in vitro. The morphology, adhesion and growth of scaffold cells were observed by scanning electron microscope and laser confocal microscope. Extensive measurements, including 3-[4, 5-dimethylthiazole-2-acyl]-2, 5-diphenyl tetrabrominate assays, intracellular total protein content, and alkaline phosphatase activity assays were performed after 7 days of culture. Survival and protein content increased in RHLC/fibroin stents. LSCM and SEM results confirmed that the cells grew better in the mixed scaffolds than in the pure silk scaffolds, and showed that the cells were easy to adhere and diffuse in the RHLC/ silk scaffolds. Abstract:Silk fibroin is an attractive natural fibrous protein for biomedical application due to its good biocompatibility and high tensile strength. Silk fibroin is apt to form a sheet-like structure during the freeze-drying process, which is not suitable for the scaffold of tissue engineering. In our former study, the adding of chitosan promoted the self-assembly of silk fibroin/chitosan (SFCS) into a three-dimensional (3D) homogeneous porous structure. In this study, a model of the self-assembly is proposed; furthermore, hepatocytes attachment and inflammatory response for the SFCS scaffold were examined. The rigid chain of chitosan may be used as a template for beta-sheet formation of silk fibroin, and this may break the sheet structure of the silk fibroin scaffold and promote the formation of a 3D porous structure of the SFCS scaffold. Compared with the polylactic glycolic acid scaffold, the SFCS scaffold further facilitates the attachment of hepatocytes. To investigate the inflammatory response, SFCS scaffolds were implanted into the greater omentum of rats. From the results of implantation, we could demonstrate in vivo that the implantation of SFCS scaffolds resulted in only slight inflammation. Keeping the good histocompatibility and combining the advantages of both fibroin and chitosan, the SFCS scaffold could be a prominent candidate for soft tissue engineering, for example, in the liver.

Abstract:Through adding insoluble fibroin microparticles into polylactic acid (PLA) solution, stirring intensively and then freeze-drying the blend, we prepared a composite scaffold composed of polylactic acid (PLA) and fibroin. Observation of the PLA/fibroin composite scaffold by scanning electron microscopy (SEM) showed that fibroin microparticles uniformly distributed in PLA matrix and interconnected pore structures were formed in the scaffolds. The in vitro cell culture result showed that the HepG2 (human hepatocellular liver carcinoma cell line) cells attached and spread well on the surfaces of the composite scaffold. Compared with that on PLA scaffolds, the attachment and growth of HepG2 on the composite scaffold were greatly improved. HepG2 cells attached on the hybrid sponges and established cell–cell contacts to form aggregates with a size up to 100μm. Considering that the PLA skeleton facilitated the desired shape formation, and fibroin microparticles improved the cell growth, the PLA/fibroin composite scaffold is a promising matrix for hepatic tissue engineering.

Abstract:In this study, a novel porous 3D composite scaffold based on the biodegradable Poly(ε-caprolactone) (PCL), Polylactide Acid (PLA) and Calcium Citrate (CC) was developed via polymer blends and thermal-induced phase separation. The chemical structure, crystalline structure and micromorphology as well as mechanical strength of the scaffolds were characterized by Fourier Transform Infrared Spectroscopy (FTIR), X-ray Diffraction (XRD), Scanning Electron Microscope (SEM) and tensile tests. The results show that the obtained composite scaffold present a suitable bone-like porous structure and sufficient mechanical strength. Furthermore, the release of calcium ions in Simulated Body Fluid (SBF) indicates that the composite material can provide a stable calcium-ion environment and maintain a constant pH value during the soaking process. The cell proliferation results from CCK-8 and light microscopy show that MG63 cells exhibit excellent adhesion and proliferation on the stent. At the same time, animal implantation histology confirms that the composite scaffolds have good biocompatibility in vivo. The scaffold material has greatly potential application value in the field of bone tissue engineering.

Abstract:To develop an artificial dermal substitute capable of receiving rapid angiogenesis. Collagen/chitosan porous scaffold was fabricated by freeze-drying method, and heparinized using N-(3-dimethylaminopropyl)-N′- ethylcarbodiimide (EDC) and N-hydroxysuccinimide (NHS). The morphology, hydrophilicity, in vitro biodegradation and tissue compatibility of the scaffold were investigated. After combined with angiogenin, the in vivo angiogenesis of the scaffold was studied as well. The results showed that with a suitable three-dimensional porous structure, the heparinized collagen/chitosan porous scaffold possessed good hydrophilicity and stability for collagenase degradation. In vivo study also demonstrated that the heparinized scaffold had good tissue compatibility, and its angiogenesis could be enhanced by angiogenin.

Abstract:Because tissue responses to implants determine the success or failure of tissue engineering products, fibroin/sericin-based scaffolds including bionic silk scaffolds, native silk fibers, fibroin fibers, and regenerated fibroin have been fabricated, and their biocompatibility was investigated. Fibroin/sericin-based scaffolds were characterized by scanning electron microscopy (SEM) and X-ray diffraction (XRD). Bionic silk scaffolds were beneficial to silk fiber formation through self-assembly. Histological and immunofluorescent staining analysis demonstrated that bionic silk scaffolds did not show significant inflammatory responses. Immunization analysis showed that soluble fibroin and sericin did not show obvious immunogenicity. This work supplied an effective approach to design fibroin/sericin-based scaffolds for tissue engineering and drug delivery.
Fabrication of a novel Three-Dimensional porous PCL/PLA tissue engineering scaffold with high connectivity for endothelial cell migration

Abstract:As an artificial extracellular matrix, tissue engineering scaffolds promote tissue regeneration and repair. Therefore, the scaffold’s internal structure is crucial for cell adhesion, growth, and migration. In this paper, based on the supercritical CO2 foaming technology, a novel three-dimensional (3D) porous polycaprolactone/polylactic acid tissue engineering scaffold induced by heterogeneous nanofibres was prepared by introducing the second phase of polylactic acid electrospinning nanofibres. The scanning electron microscopy results showed that the prepared scaffolds exhibited obvious nanofibre structures and 3D reticular pore structures. The influence of foaming temperature on pore morphology was also systematically explored. Tensile tests and water contact angle tests showed that the novel scaffolds exhibited good mechanical properties and hydrophilicity. The results of cross-sectional human umbilical endothelial cell (HUVEC) culture showed that the scaffolds were biocompatible and significantly promote the adhesion, migration and permeability of HUVECs. Single-cell migration assays showed that the 3D porous scaffolds effectively promote HUVEC migration. Therefore, 3D porous scaffolds have enormous application potential in the field of vascular patches.

Abstract:With the increasing occurrence of vascular diseases and poor long-term patency rates of current small diameter vascular grafts, it becomes urgent to pursuit biomaterial as scaffold to mimic blood vessel morphologically and mechanically. In this study, novel human-like collagen (HLC, produced by recombinant E. coli)/chitosan tubular scaffolds were fabricated by cross-linking and freeze-drying process. The scaffolds were characterized by scanning electron microscope (SEM), X-ray photoelectron spectroscopy (XPS), and tensile test, respectively. Human venous fibroblasts were expanded and seeded onto the scaffolds in the density of 1 x 10(5) cells/cm(2). After a 15-day culture under static conditions, the cell-polymer constructs were observed using SEM, confocal laser scanning microscopy (CLSM), histological examination, and biochemical assays for cell proliferation and extracellular matrix production (collagen and glycosaminoglycans). Furthermore, the scaffolds were implanted into rabbits' livers to evaluate their biocompatibility. The results indicated that HLC/chitosan tubular scaffolds (1) exhibited interconnected porous structure; (2) achieved the desirable levels of pliability (elastic up to 30% strain) and stress of 300 +/- 16 kPa; (3) were capable of enhancing cell adhesion and proliferation and ECM secretion; (4) showed superior biocompatibility. This study suggested the feasibility of HLC/chitosan composite as a promising candidate scaffold for blood vessel tissue engineering.
