Abstract:Converting common biomass materials to high-performance biomedical products could not only reduce the environmental pressure associated with the large-scale use of synthetic materials, but also increase the economic value. Chitosan as a very promising candidate has drawn considerable attention owing to its abundant sources and remarkable bioactivities. However, pure chitosan materials usually exhibit insufficient mechanical properties and excessive swelling ratio, which seriously affected their in vivo stability and integrity when applied as tissue engineering scaffolds. Thus, simultaneously improving the mechanical strength and biological compatibility of pure chitosan (CS) scaffolds becomes very important. Here, inspired by the fiber-reinforced construction of natural extracellular matrix and the porous structure of cancellous bone, we built silk microfibers/chitosan composite scaffolds via ice-templating technique. This biomimetic strategy achieved 500% of mechanical improvement to pure chitosan, and meanwhile still maintaining high porosity (> 87%). In addition, the increased roughness of chitosan pore walls by embedded silk microfibers significantly promoted cell adhesion and proliferation. More importantly, after subcutaneous implantation in mice for four weeks, the composite scaffold showed greater structural integrity, as well as better collagenation, angiogenesis, and osteogenesis abilities, suggesting its great potential in biomedicine.

Preparation and characterization of nano-hydroxyapatite/silk fibroin porous scaffolds.

Abstract:Novel tissue engineering scaffold materials of nano-hydroxyapatite (nHA)/silk fibroin (SF) biocomposite were prepared by freeze-drying. The needle-like nHA crystals of about 10 nm in diameter by 50-80 nm in length, which were uniformly distributed in the porous nHA/SF scaffolds, were prepared by a co-precipitation method with a size. The as-prepared nHA/SF scaffolds showed good homogeneity, interconnected pores and high porosity. XRD and FT-IR analysis suggested that the silk fibroin was in beta-sheet structure, which usually provides outstanding mechanical properties for silk materials. In this work, composite scaffolds containing as high as 70% (w/w) nHA were prepared, which had excellent compressive modulus and strength, higher than the scaffolds at low nHA content level and other porous biodegradable polymeric scaffolds often considered in bone-related tissue engineering reported previously. The cell compatibility of composite scaffolds was evaluated through cell viability by MTT assay. All these results indicated that these nHA/SF scaffold materials may be a promising biomaterial for bone tissue engineering.

Abstract:With the rise of tissue engineering,silk fibroin tissue engineered scaffold has become a hotspot in the field of silk fibroin biomaterials.Silk fibroin has been prepared as various forms of tissue engineering scaffolds to satisfy construction of various cells and tissues.The preparation and the research progress of the silk fibroin tissue engineered scaffolds in bone,cartilage,ligament,tendon and other tissue engineering were summarized in this paper.In addition,the prospect on silk fibroin scaffolds was discussed,providing reference information for further researches on silk fibroin tissue engineered scaffolds.

Abstract:Silk fibroin/chitosan scaffolds were prepared through lyophilization, then hydroxyapatite was deposited by premineralization to obtain porous hydroxyapatite (HA)-silk fibroin/chitosan composite scaffolds.Composite scaffolds were characterized by Fourier transform infrared analysis (FT-IR), X-ray diffraction (XRD), scanning electronic microscopy (SEM), thermogravimetry analysis (TGA), compressive modulus and porosity, as well as the proliferation of MG-63 cells on composite scaffolds was detected by WST-1 method. The results reveal that the structure of deposited HA is a weak crystallinity, which deposits on the wall of pore. With the increase of premineralization cycles, the amount of HA and compressive modulus of premineralized composite scaffolds are significantly increased, the porosity keeps about 90%. WST-1 assay shows that premineralized composite scaffolds deposited with HA dramatically enhance the proliferation of cells, and possess a great cytocompatibility.

Abstract:Silk fibroin (SF) has been one of promising resources of biotechnology and biomedical materials due to its unique properties. Here, different sizes of porous tubular scaffolds were fabricated from a SF aqueous solution with the addition of poly(ethylene glycol diglycidyl ether) (PGDE). The scaffolds were generally flexible and transparent at the wet state with a pore size of 81-128 mu m and porosity of 90-96%, depending on the concentrations of SF and PGDE. The mechanical properties measurement showed that the tubular SF scaffolds had satisfying tensile and compression properties, especially the excellent deformation-recovery ability. FT-IR spectra indicated that the SF in the tubular scaffolds was in a beta-sheet structure, and no PGDE characteristic band was observed, suggesting that the PGDE could be removed from the scaffolds by soaking in deionized water. The cell compatibility of scaffolds was evaluated, and no obvious cytotoxicity to mouse L-929 fibroblasts was detected. (C) Koninklijke Brill NV, Leiden, 2009

Abstract:Silk fibroin (SF) is a structural protein derived from natural silkworm silks. Materials fabricated based on SF usually inherit extraordinary physical and biological properties, including high mechanical strength, toughness, optical transparency, tailorable biodegradability, and biocompatibility. Therefore, SF has attracted interest in the development of sustainable biodevices, especially for emergent bio-electronic technologies. To expand the function of current silk devices, the SF characteristic sequence has been used to synthesize recombinant silk proteins that benefit from SF and other functional peptides, such as stimuli-responsive elastin peptides. In addition to genetic engineering methods, innovated chemistry modification approaches and improved material processing techniques have also been developed for fabricating advanced silk materials with tailored chemical features and nanostructures. Herein, this review summarizes various methods to synthesize functional silk-based materials from different perspectives. This review also highlights the recent advances in the applications of natural and recombinant silks in tissue regeneration, soft robotics, and biosensors, using B. mori SF and silk-elastin-like proteins (SELPs) as examples.

Abstract:To mimic the natural fibrous structure of the tissue extracellular matrix, a nano-fibrous silk fibroin (SF)/sodium alginate (SA) composite scaffold was fabricated by a thermally-induced phase-separation method. The effects of SF/SA ratio on the structure and the porosity of the composite scaffolds were examined. Scanning electron microscopy and porosity results showed that the 5SF/1SA and 3SF/1SA scaffolds possessed an excellent nano-fibrous structure and a porosity of more than 90%. Fourier transform infrared, X-ray diffraction, and differential scanning calorimetry results indicated the physical interaction between SF and SA molecules and their good compatibility in the 5SF/1SA and 3SF/1SA scaffolds, whereas they showed less compatibility in the 1SF/1SA scaffold. Cell culture results showed that MG-63 cells can attach and grow well on the surface of the SF/SA scaffolds. The nano-fibrous SF/SA scaffold can be potentially used in tissue engineering.

Abstract:Regeneration of large bone defects is challenging in the next few years because the age of the population is growing. Bone grafting as a gold standard of bone repairing is limited as a result of many disadvantages. In recent years, many novel materials and processing methods have been introduced to construct the native bone extracellular matrix (ECM) and restore the functions of degenerated bone. Silk fibroin (SF) as a natural polymer, has established a good reputation for bone tissue engineering applications due to its many unique properties, including exceptional biocompatibility, biodegradability, mechanical behavior, and ease of processability. In this review, recent advances in the design and application of SF-based scaffolds for bone regeneration are discussed.

Abstract:Silk fibroin, extracted from the silk of the Bombyx mori silkworm, stands out as a biomaterial due to its nontoxic nature, excellent biocompatibility, and adjustable biodegradability. Porous scaffolds, a type of biomaterial, are crucial for creating an optimal microenvironment that supports cell adhesion and proliferation, thereby playing an essential role in tissue remodeling and repair. Therefore, this review focuses on 3D porous silk fibroin-based scaffolds, first summarizing their preparation methods and then detailing their regenerative effects on bone, cartilage, tendon, vascular, neural, skin, hepatic, and tracheal epithelial tissue engineering in recent years.

Abstract:Preparation of 3D scaffolds for bone tissue engineering (BTE) is a challenging task as it required appropriate pore size and porosities, excellent mechanical properties, controlled rate of bioerosion. The focus of this work is the fabrication of silk fibroin (SF) 3D microparticle scaffolds with the incorporation of hydroxyapatite (HA) and calcium sulfate (CaS) as bioceramics. Physicochemical characterization shows ∽30% filler loading, ∽ 40% optimum porosity with >100 µm pore size for these filled/unfilled scaffolds. Nanoindentation studies show improved Young's modulus at microparticle level with the incorporation of bioceramics. There is a three‐fold increase in Young's Modulus in the case of SF‐HA while two‐fold increase for SF‐CaS. In‐vitro bioerosion study results in early bioerosion with SF‐CaS scaffold while prolonged bioerosion with SF‐HA scaffold. In‐vitro osteo‐regenerative potential is analyzed by estimating ALP, BMP‐2 and OCN. SF‐CaS supports the early stage differentiation as compared to SF‐50%HA scaffolds, which predominantly support at late stage. The expression of TNF‐α, suggesting a reduced risk of immune rejection. This work, therefore, concludes that although SF support bone tissue regeneration, the choice of bioceramic enhances the applicability in various clinical scenarios by providing a controlled rate of bioerosion, tunable speed of osteo‐regeneration and improved load bearing capacities.This article is protected by copyright. Abstract:Silk sericin, a natural protein extracted from silkworm cocoons, has been extensively studied and utilized in the biomedical field because of its superior biological activities and controllable chemical-physical properties. Sericin is biocompatible and naturally cell adhesive, enabling cell attachment, proliferation, and differentiation in sericin-based materials. Moreover, its abundant functional groups from variable amino acids composition allow sericin to be chemically modified and cross-linked to form versatile constructs serving as alternative matrixes for biomedical applications. Recently, sericin has been constructed into various types of biomaterials for tissue engineering and regenerative medicine, including various bulk constructions (films, hydrogels, scaffolds, conduits, and devices) and micro-nano formulations. In this review, we systemically summarize the properties of silk sericin, introduce its different forms, and demonstrate their newly-developed as well as potential biomedical applications.

Abstract:Sericin, a protein derived from silkworm cocoons, was considered as a waste product derived from silk industry for thousands of years due to a lack of understanding of its properties. However, in recent decades, a range of exciting properties of sericin have been studied and uncovered, including cytocompatibility, low‐immunogenicity, photo‐luminescence, antioxidant properties, as well as cell‐function regulating activities. These properties make sericin‐based biomaterials promising candidates for biomedical applications. This review summarizes the properties and bioactivities of silk sericin and highlights the latest developments on sericin in tissue engineering and regenerative medicine. Furthermore, the extended application of sericin in developing flexible electronic devices and 3D bioprinting is also discussed. It is believed that sericin‐based biomaterials have great potential of being developed into novel tissue engineering products and smart implantable devices for various medical applications towards improving clinical outcomes. Abstract:In our recent study, a novel fibroin/collagen scaffold with improved mechanical properties and controllable porous structure was prepared through freeze–drying method. In this research, the cyto-compatibility was further studied, using fibroblast and vascular smooth muscle cells (VSMC) as the model cells. MTT results indicated that the growth of fibroblast and VSMC both further improved in the fibroin/collagen scaffold than in pure fibroin scaffolds. The confocal and SEM results showed that fibroblast cells and VSMCs had better adhesion and spreading properties in the fibroin/collagen scaffolds. Although further studies, such as the extracellular matrix production and the functional gene expression, are necessary to clarify the biocompatibility of the fibroin/collagen scaffolds, the present results indicate that the fibroin/collagen scaffold is a new scaffold material suitable for tissue engineering. On the other hand, the mild and all-aqueous preparation processes also make it possible to embed different growth factors inside the scaffolds to maximize cell functions and the formation of specific tissues.

Abstract:Silk fibroin (SF) is a protein with unique properties that make it an ideal scaffold for tissue engineering. Research in this area has improved its advantages by incorporating many other materials during synthetic stages and helping in use of (SF) in broader biomedical application. Silk is gaining popularity as an encouraging bio-sourced raw material due to superior mechanical qualities, flexibility, as well as bioactivity. This review focus on the current advances in silk-founded biologically inspired active and efficient scaffold in medical fields as well as the most recent applications and advancements in the use of SF as a biocompatible material. The authors begun with a short overview of silk, including its origins, characteristics, source, in addition procedures. The findings of this analysis are crucial in aiding designers in making proper design structure as well as the percentage of porosity required in scaffold for the cell's growth prior to fabrication of the scaffold. Furthermore, SF can be formed into numerous scaffold kinds such as sponges, mats, hydrogels, and films using both traditional and innovative bio-fabrication methodologies.

Abstract:Journal of Bioactive and Compatible Polymers, Ahead of Print. Biomaterials play an important role in biomedical applications serving as scaffolds in tissue engineering and substrates for cell growth by providing structural supports. Signaling cues for cellular events can be also presented, including cell viability, adhesion, spreading, and function developments. In this study, we fabricated a hierarchical nanofibrous scaffold through electrospinning silk fibroin (SF) and decellularized extracellular matrix of the small intestine submucosa (SIS-dECM). The addition of SIS-dECM improved the hydrophilic features of the resultant composite nanofibrous substrates as compared to SF nanofibrous substrates. The biocompatibility of the nanofibrous scaffolds was thoroughly investigated by culturing meningeal fibroblasts on SF and SF/SIS-dECM nanofibrous substrates. Cell viability, proliferation, focal adhesion, and quantitative measurements of cell adhesion-related gene expression levels revealed that the addition of SIS-dECM favored the adhesion and spreading of fibroblast cells. Therefore, the SF/SIS-dECM nanofibers provided a suitable substrate for cell growth and the present study suggested that the nanofibrous substrates hold high potential for bioapplications.

Abstract:Silk sericin is a kind of natural material originated from a wide variety of cocoons and behaves excellent biocompatibility and a series of unique biological properties. The specific amino acid compositions and structural properties of silk sericin endow it with good water solubility, cellular adhesion and proliferation activity, in situ fluorescence, antioxidant and the inhibitory effect on tyrosinase. With the actions of cross-linking agent, chemical active group or just ultraviolet light, silk sericin can be designed into various structural biomaterials, including micro (nano) -structural materials, 2D (patterned) films, hydrogels or even 3D porous scaffolds, denoting broad prospect in biomedical applications, such as wound healing, tissue regeneration, drug delivery, medicine, material coating. Based on the significant studies of silk sericin in recent years, this review summarizes the structure and physicochemical properties of silk sericin, with a focus on the design of sericin-based biomaterials and their relevant biomedical applications. The prospective regarding the future potential of silk sericin is delivered.

Abstract:Silk fibroin is an important natural fibrous protein with excellent prospects for tissue engineering applications. With profound studies in recent years, its potential in tissue repair has been developed. A growing body of literature has investigated various fabricating methods of silk fibroin and their application in tissue repair. The purpose of this paper is to trace the latest developments of SF-based scaffolds for tissue engineering. In this review, we first presented the primary and secondary structures of silk fibroin. The processing methods of SF scaffolds were then summarized. Lastly, we examined the contribution of new studies applying SF as scaffolds in tissue regeneration applications. Overall, this review showed the latest progress in the fabrication and utilization of silk fibroin-based scaffolds.
