Abstract:A new type furo(2,3-d)pyrimidine nucleoside conjugat- ed with various carbohydrates was synthesized using Sonogashira coupling and 'click chemistry'. In the subsequent deprotection of the 4-toluoyl and acetyl groups with catalytic sodium methoxide in methanol, the furo(2,3-d)pyrimidine rearranged to an opened ke- tone-type structure. Their structures were confirmed on the basis of spectroscopy.

Abstract:A series of bicyclic furo[2,3-d]pyrimidine nucleosides conjugated with N-diisopropylphosphoryl amino acids were synthesized from 2′-deoxy-5-iodouridine. The key step is its cross coupling with the 5-phthalimido-1-pentyne under Sonogashira reaction condition. 5-Endo-dig electrophilic cyclization catalyzed by AgNO3 is used to afford the furo[2,3-d]pyrimidines.

Abstract:A Cu-catalyzed tandem synthesis of azole-fused pyrimido[1,2-c]quinazolines and imidazo[1,2-c]quinazolines has been developed. The reaction is based on a C-N cross-coupling/C-H functionalization reaction of 2-(2-bromophenyl)-1,4,5,6-tetrahydropyrimidines with azoles. Abstract:This paper reports syntheses, crystal structures and electrochemical results for two ferrocene(Fc)-modified pyrimidine nucleosides that could potentially be used for investigating electron transfer in DNA. Fc was directly attached to the 5-position of deoxyuridine and deoxycytidine via the Stille coupling reaction. Fc-modified uridine was incorporated into DNA trinucleotides with standard solid-phase synthesis. The structures of corresponding detritylated compounds were determined by single-crystal X-ray analysis. Electrochemical investigations of all compounds by cyclic voltammetry revealed reversible redox processes.

Abstract:An expeditious synthesis of highly functionalized dibenz[b,f][1,4]oxazepin-11(10H)-ones and dibenz[b,f][1,4]-oxazepine-11(10H)-carboxamides has been established via microwave-assisted one-pot U-4CR and intramolecular O-arylation. A Pd-catalyzed intramolecular arnidation was successfully performed under microwave irradiation to furnish the novel conjugates of dibenz[b,f][1,4]oxazepin-11(10H)-ones with a 2-oxindole linked through a C-N single bond.

Abstract:New C-nucleoside analogues were synthesized via the key intermediate 2-(2′ ,3′ 4′-tri-O-benzoyl-β-D-xylopyranosyl)-furan. The stability of the α- and β-isomer was described. The conversion of the furan ring into pyridazine ring was performed by Clauson-Kaas method to give 3-(β-D-xylopyranosyl)-pyridazine and 3-(β-D-xylopyranosyl-4,5-dihydro-6(1H)-pyridazinone.

Abstract:6-Bromofuro[2,3-d]pyrimidine bicyclic nucleosides are the key intermediates for the preparation of novel furo[2,3-d]pyrimidine bicyclic nucleoside derivatives with remarkable antiviral potency. The literature protocol for their syn-thesis involves a multi-step procedure and palladium catalyst. In this paper, an economical and practical method was devel-oped for their preparation via condensation of the easily obtainable 5-formyl pyrimidine nucleosides with carbon tetrabromide and subsequent cyclization promoted by copper iodide.

Abstract:A series of novel 1'-O-alpha-methyl isonucleosides were efficiently and stereoselectively synthesized with 1,2,3,5-tetra-O-acetyl-D-ribofuranose as the starting material. The key steps include regioselective and stereoselective deprotection of the 2,4-dichlorobenzyl group at C2, triflation, and substitution with appropriate nucleobases using cesium carbonate as the base. Removal of the residual 2,4-dichlorobenzyl groups and subsequent transformation afforded the title compounds in 30-37% overall yield. The products represent a new type of isonucleosides with 1'-O-substitution.

Abstract:The pyrrolo[2,3-b] pyridine core structure, a bioisostere of quinolones, is found in several molecules that possess important biological activity. We describe here a new, concise, three-step synthesis of pyrrolo[2,3-b]pyridines starting from L-alanine. A series of 4,7-dihydro-4-oxo-1H-pyrrolo[2,3-b]pyridine-5-carboxylic acid derivatives, which have not been previously reported, were synthesized using this approach.

Abstract:Dedicated to Professor Hiriyakkanavar Ila in great admiration of her creative contributions to heterocyclic chemistry With the motivation to expand the compound library of specifically substituted pyrimidine derivatives, we prepared several pyrimidine-4-carbaldehydes. In most cases, the chemoselective oxidation of 4-hydroxymethyl-substituted pyrimidine derivatives could be achieved in good yields to provide the desired compounds. Alternatively, the aldehydes were prepared by Riley oxidation with selenium dioxide from the corresponding 4-methylpyrimidines. The formyl group of these compounds was employed as handle to generate alkynyl, cyano, oxazol-5-yl or β-ketoester substituents. Furthermore, two methods were found to prepare furo[3,2- d ]pyrimidines. A serendipitously discovered reaction to a 6-(pyrimidin-4-yl)furo[3,2- d ]pyrimidin-7-ol derivative involves a mechanistically interestingly 'dimerization' process with a benzoin addition as key step. Other compounds in this series contain amino, azido, or 1,2,3-triazol-1-yl groups. All these transformations to highly substituted pyrimidine derivatives demonstrate the synthetic versatility of pyrimidine-4-carbaldehydes and similar compounds. Abstract:This paper uses basic-catalysed method to convert 1H-pyrazole-4-carboxamides into tri-fused heterocyclic systems containing two kinds of pyrazolotriazolopyrimidine framework. The synthesis of compounds 5-6 and reactions of them with some electrophiles have been investigated. Preliminary bioassay study showed that some compounds displayed high activity.
