Cytocompatibility of the nanophase beta-tricalcium phosphate /collagen fibers composite

Abstract:Objective: To investigate the cytocompatibility of the nano-sized beta-tricalcium phosphate /collagen fibers (β-TCP/CF) composite and test a hypothesis that nano-sized composite are suitable for clinical utilization in bone tissue engineering with good cytocompatibility. Methods: the nanophase beta-tricalcium phosphate /collagen fibers (β-TCP/CF) composite was prepared by a wet-chemical method and characterized through X-ray diffractometer and scanning electron microscopy (SEM). Cytocompatibility of the composite was investigated through cell attachment, proliferation, alkaline phosphotatse (ALP) activity and scanning electron microscopy analysis. Results: These assays showed that the β-TCP/CF composite elicited cell adhesion and proliferation better than the controls. Further experiments showed the nanophase structure composite enhanced osteoblast-like cells growth. SEM micrographs also supported the above results. At 1, 3 and 5 days, ALP activity was not significant different for the composite and only cells group, while at 7 day after culture, significantly increased ALP activity was observed for β-TCP/CF than for the controls. Conclusion: The present study demonstrated that the nano-sized β-TCP/CF composite exhibited a remarkable improvement of osteoblast-like cell adhesion and proliferation compared with conventional β-TCP, indicating its excellent cytocompatibility. The composite is expected to be a promising candidate for bone repair substitute or tissue engineering scaffold.
