Healing mechanism of corneal wound of immature rabbits after LASIK

Abstract:Objective: A good therapy effectiveness of laser in situ keratomileusis (LASIK) for anisometropic amblyopia has been proved. However, reaction of developing corneal tissue to LASIK and corneal healing mechanism after LASIK are unclear. In this paper, we studied the response of corneal stroma and the healing mechanism of corneal wound after LASIK in immature rabbit. Methods: Twenty-four immature rabbits and twenty-four adult rabbits were included in this experiment. LASIK was performed on the right eyes of experimental rabbits. The corneal specimen was prepared at the 10th, 30th ,90th and 180 th day after operation. The pathological changes of cornea were investigated using routine histological method and immunohistochemical staining. The expressions of collagen III, proliferating cell nuclear antigen (PCNA) and transforming growth factor-β1 (TGF-β1) in cornea before and after LASIK were analyzed. Results: The interface between the flap and stromal bed of cornea was transparent, and some extent of proliferation and slight recovery response could be seen in the healthy adult rabbits. Expressions of collagen III, PCNA and TGF-β1 in cornea tissue were significantly increased in adult rabbits compared immature rabbits. In the immature rabbits group, the epithelial hyperplasia showed more active than that of the adult rabbits. The expressions of collagen III, PCNA and TGF-β1 were higher than those of the adult rabbits at the 10 th, 30 th and 90 th day following operation. However, no statistically significant differences were found in the expressions of collagen III, PCNA and TGF-β1 in the 180 th day between two groups. Conclusion: Corneal wound healing response of immature rabbit is fiercer in comparison with adult rabbit. The wound healing procedure between these two groups shows a similar outcome. The TGF-β1 takes part in corneal healing in immature rabbits after LASIK.
