Experimental study on the keratocyte apoptosis in the stoma following LASIK

Abstract:Objective The aim of this study was to investigate the character of the keratocyte apoptosis after LASIK and to discuss the correlation between the level of apoptosis and the mechanical injury by micro- keratomy, laser ablation respectively. Methods Twenty New Zealand white rabbits (40eyes) were divided into 4 groups. The group A were produced a flap with microkeratome without subsequent laser ablation.The group B were corrected -4.0 diopters with LASIK. The group C were corrected -8.0 diopters with LASIK and the group D were control without any treatment. At 4 hours after surgery the rabbits were enucleated and the corneas were bisected. Celluar morphologic changes were evaluated by transmission electron microscopy. Keratocyte apoptosis were numerated by terminal-deoxynucleotidyl transferase mediated dUTP nick end labeling (TUNEL) to detect DNA fragmentation. Results There was no keratocyte apoptosis expression in stoma of control group, but keratocyte apoptosis was noted in the other three groups. Keratocytes undergoing apoptosis were distributed within a zone approximately 50μm anterior and posterior to the interface and were more in the peripheral than in the central cornea. There were no statistically significant differences in the TUNEL positive cells among the three operated groups(F=1.008,P>0.05). Conclusion There were keratocytes undergoing apoptosis in sroma after LASIK. The keratocytes apoptosis in stroma after LASIK may be related with the production of the flap but there was no significant correlation between keratocytes apoptosis and the depth of laser ablation.
