Changes of neuronal nitric oxide synthase-immunopositive neurons in the caudate putamen of spontaneously hypertensive rats

Abstract:Aim: To observe the changes of neuronal nitric oxide synthase (nNOS) - immunopositive neurons in the caudate putamen of spontaneously hypertensive rats. Methods: The experiment was performed at the Department of Human Anatomy, Jinan University in May 2003. Thirty spontaneously hypertensive rats (experimental group) and thirty Wistar-Koyto rats (control group) were detected in blood pressure and sacrificed at the ages of 3 months, 6 months, and 12 months, respectively. The changes of nNOS-immunopositive neurons were observed with ABC immunocytochemical assay. Polyclonal antibody of rabbit anti-nNOS was applied in the experimental group. 0.01 mol/L phosphate buffer was instead of the primary antibody for negative control. Each rat was counted for 20-30 pieces. The amount of nNOS-immunopositive neurons in the caudate putamen was counted based on the mean counting of each rat, which represented the density of nNOS-immunopositive neurons. Results: There was no death in the trial, and all were included in the result analysis. 1 The blood pressure of rats of 3, 6 and 12 months in the control group were [(97.5±12.0),(107.3±9.8),(113.3±12.8)mm Hg, (1 mm Hg=0.133 kPa)], respectively, which had insignificant difference. The blood pressure of the spontaneously hypertensive rats of 3, 6 and 12 months in the experimental group increased gradually, which were [(116.3±13.5),(151.5±8.3),(177.0±9.0)mm Hg], respectively, and had s ignificant difference (P < 0.05). The blood pressure of rats in the experiment group was higher than that of rats of the same age period in the control group, which had obvious difference (P < 0.05). 2 The -nNOS-immunopositive neurons in the caudate putamen were of moderate sizes with many long prominence. The positive neural fibers were mesh. The alteration of the density of nNOS-immunopositive neurons in the caudate putamen of rats: There was no marked change of rats of 3 months in the control group and the experimental group, and it remarkably reduced in the rats of 6 months and 12 months in the experimental group. 3 No significant changes were found in nNOS-immunopositive neurons in the caudate putamen of Wistar-Koyto rats of 3, 6 and 12 months in the control group [(9.41±0.77), (8.64±1.28), (8.51±0.77)piece per mm 2]. The nNOS-immunopositive neurons of spontaneously hypertensive rats of the 6 and 12 months reduced gradually in the experimental group, while compared with the rats of 3 months there was distinct difference [6 months: (6.59±0.70), 12 months: (7.30±0.96), 3 months: (8.90±1.09) piece per mm 2, P < 0.05]. The number of nNOS-immunopositive neurons of rats of 6 and 12 months in the experimental group was lower than that of rats in the same age period in the control group, which had significant difference (P < 0.05). Conclusion: The spontaneously hypertensive rats are good models for imitating human primary hypertension. The blood pressure of spontaneously hypertensive rats is various based on different ages. The alteration of nNOS-immunopositive neurons in the caudate putamen of spontaneously hypertensive rats may indirectly accelerate the development of hypertension by modulating the sympathetic activity in blood pressure centers of accommodation.
